Osteoblastic differentiation of adult stem cells by Biphasic Calcium Phosphate (이형상 칼슘 인산염(Biphasic Calcium Phosphate)에 의한 성체 줄기세포의 골아세포 분화)

  • The present study was to determine the influence of micro-macro biphasic calcium phosphate(MBCP) on proliferation and differentiation of human marrow-derived mesenchymal stem cells. Primary stem cells were cultured from bone marrow and 3-4 passaged cells were used. This study tested the proliferative effects by cell counting. Collagen sythensis, alkaline phosphatase activity, expression of osteocalcin and bone sialoprotein by Western blot analysis were evaluated. The cellular proliferation of ASC was not influenced by MBCP. Collagen synthesis of ASC cultured on MBCP significantly increased at 5th and 7th days(p<0.05). The ALP activity in ASC cultured on MBCP significantly increased at 5th and 7th days(p<0.05). The expression of OC and BSP incresaed in ASC cultured on MBCP. These results suggest that MBCP may stimulates the osteoblastic activity of ASC.

Review : Clinical application and efficacy of herbal medicines by modulating cytokines in atopic dermatitis-induced animal model (동물모델에서 cytokine 조율을 통한 한약의 항아토피피부염 효능과 임상적 응용에 대한 고찰)

  • Objectives : There is a pressing need to determine the clinical and scientific validity of herbal therapies for animal model with atopic dermatitis since some differences in systemic cytokine polarization between in animal model and in patients with atopic dermatitis has been reported. New studies for tang, medicinal herb itself or effective ingradients of medicinal herb showing anti-atopic dermatitis effectiveness are reviewed in terms of cytokine regulation. Methods : Those herbal therapies used to treat atopic dermatitis in animal model were introduced and the expression pattern of cytokine and the activity of mast cell were compared in both animal model and patients with atopic dermatitis. Results : In case of atopic dermatitis in human, there is a biphasic pattern of cytokine expression in atopic dermatitis, with acute skin inflammation associated with a predominance of IL-4 and IL-13 expression from Th2 cells, and chronic inflammation associated with increased IL-5 from Th2-cells and IFN-${\gamma}$ from Th1-cells. However, a pattern of cytokine expression in animal model with atopic dermatitis is not matched well to the biphasic pattern of cytokine expression in patients with atopic dermatitis. In addition, a kind of cytokine is different by animal model with atopic dermatitis. These differences would make herbal medicines, showing their effectiveness on atopic dermatitis, difficult to apply to patients with atopic dermatitis. Conclusion : The pattern of local cytokine expression plays an important role in modulating tissue inflammation, and in atopic dermatitis this pattern depends on the acuity or duration of the skin lesion. Thus, in order to develop medicinal herb itself or effective ingradients of medicinal herb showing anti-atopic dermatitis effectiveness, biphasic pattern of cytokine expression should be considered in animal model with atopic dermatitis.

Biphasic immunomodulatory effects of ionized biosilica water on the antigen-presenting capability of mouse dendritic cells (마우스 수지상세포의 항원 제시 능력에 대한 이온화 규소수의 biphasic 면역조절 효과)

  • Biosilica is a silica-based substance derived from the cell walls (frustules) of diatoms. Recently, research into biosilica's biological functions is underway, but little has been reported on the effects of biosilica on immune cells. In this study, we investigated the effect of ionized biosilica water (iBW) on dendritic cells (DCs), which play crucial roles as antigen (Ag)-presenting cells. Treatment with iBW increased the expression of immune response-related markers, closely connected to the maturation of DCs, and the production of tumor necrosis factor-alpha. In addition, iBW-treated DCs (iBW-DCs) had a lower uptake of fluorescein isothiocyanate-dextran than that of control DCs. Mixed leukocyte response analysis used for measuring the Ag-presenting capability of DCs, showed iBW-DCs had a higher capability than that of control DCs. Interestingly, DCs treated with lipopolysaccharide (LPS) and iBW had a lower level of Ag-presenting capability than that of LPS-treated DCs. Taken together, the results indicate that iBW alone can mature DCs, but it decreases the Ag-presenting capability of DCs in the presence of LPS, a representative agent of inflammation. This study may provide valuable information regarding the effect of iBW on immune cells. Further research is needed to investigate how iBW induces the observed biphasic immunomodulatory activity.

Stress-induced biphasic ethylene and ROS biosynthesis are synergistically interacted in cell damage (스트레스에 의한 식물세포 손상에서 Biphasic Reactive Oxygen Species(ROS)와 Ethylene 생합성의 Synergism 효과)

  • Although reactive oxygen species (ROS) are inevitable by-products of many redox reactions in eukaryotic cells, they play a crucial role as signaling molecules in many cellular processes for development and defense response to abiotic stresses. The biphasic ROS production which was peaked twice in a first transient phase and a second massive phase was occurred after treatment of abiotic stress such as oxidative stress, high salinity. This biphasic generation of ROS was followed by the biphasic production of stress hormone, ethylene. The mechanism of interactions between ROS and ethylene biosynthesis is studied in tobacco (Nicotiana tabaccum L.) plants under the abiotic stresses. The stress-induced ethylene production was significantly inhibited in RbohD-AS and RbohF-AS, in which antisense expression of NADPH oxidase genes was performed. The accumulation of ROS, which was determined by DAB and DCFH-DA staining, was significantly decreased after abiotic stresses in transgenic plants. The suppression of signaling with ethylene and ROS induced more tolerance in response to abiotic stress. The transgenic plants were more tolerant in MS medium supplemented with salinity stress in contrast with wild-type. Stress-induced cell damage determined by DNA fragmentation was decreased at phase II in those transgenic plants. Therefore, the first burst of ROS is more responsible for making a role as a signaling molecule during stress-induced response. These results suggested that ethylene and ROS act in a positive feedback cycle that results in mutual enhancement of ethylene and ROS production during stress-induced cell death.

Transducer-like Enhancer of Split 1 as a Novel Immunohistochemical Marker for Diagnosis of Synovial Sarcoma

  • Background: Synovial sarcoma is a mesenchymal neoplasm that accounts for around 10% of all soft tissue sarcomas. The diagnosis of synovial sarcoma can be a challenging task, particularly with small biopsy specimens. Aim: We investigated transducer-like enhancer of split 1 (TLE1), monoclonal antibody, expression by immunohistochemical analysis in a group of 74 synovial sarcoma cases, 20 cases of MPNST, 12 cases of neurofibroma, 15 cases of schwannoma, 5 cases of MFH, 10 cases of lieomyosarcoma and 10 cases of solitary fibrous tumor. Materials and Methods: Whole tissue sections were examined: (39 biphasic and 35 monophasic). Nuclear immunoreactivity was scored as negative (<5% of cells positive), 1+(mild /5-25%), 2+ (moderate/25-50%), and 3+ (strong >50%). Results: Overall, 71 (96%) of 74 synovial sarcomas were positive for TLE1, including 37 biphasic (95%) and 34 monophasic (97%) tumors. Other spindle cell tumors showed very low or absent staining of TLE1. Conclusions: We conclude that TLE1 is a sensitive marker and can be a useful diagnostic marker for synovial sarcoma, particularly the monophasic forms.

Effects of nitric oxide on the proliferation and differentiation of human periodontal ligament cells (산화질소가 인간 치주인대세포의 증식과 분화에 미치는 영향)

  • Objective: This study evaluated the effects of nitric oxide (NO) on the proliferation and differentiation of human periodontal ligament cells involved in orthodontic tooth movement. Methods: A range of concentrations of sodium nitroprusside (SNP), a NO donor, were administered to samples of human periodontal ligament cells, followed by measurement of cell viability, alkaline phosphatase (ALP) activity, and expression of osteonectin and bone sialoprotein. Results: Cell viability, ALP activity and the expression of osteonectin and bone sialoprotein were increased in human periodontal ligament cells treated with SNP concentrations of < 0.2 mM compared with controls, but were decreased with SNP concentrations of > 1.0 mM. Conclusion: NO has a biphasic effect on proliferation and differentiation in human periodontal ligament cells, with a stimulatory effect at low concentrations and an inhibitory effect at high concentrations.

Molecular Effects of Genistein on Proliferation and Apoptosis of MCF-7 Cell Line

  • Genistein is a potent, plant-derived isoflavone that displays estrogenic activity at low concentrations but inhibits proliferation at high amounts. However, the molecular mechanism of genistein is not completely understood. In the present study, the biphasic effects (estrogenic and antiestrogenic activity) of genistein on the growth of MCF-7 cells were identified. Genistein within a low range of concentration, $1-10\;{\mu}M$, stimulated proliferation, while $50-100\;{\mu}M$ caused apoptotic cell death. Additionally, genistein at a low concentration induced estrogen receptor (ER)-mediated gene expression and ER phosphorylation. When pre-treated with PD98059, an MEK inhibitor, ER-mediated gene expression and ER phosphorylation by genistein were noticeably increased. However, the increased gene expression and phosphorylation did not enhance cell proliferation. Moreover, it was observed that ER-mediated signaling performs an important role in the MAPK pathway. The proliferation and apoptosis in genistein-treated MCF-7 cells were partially dependent on the Bcl-2 level. The addition of IC1 182, 780, an estrogen receptor antagonist, inhibited Bcl-2 expression induced by genistein. This study suggests that there is a close relationship between Bcl-2 and the ER signaling pathways in MCF-7 cells.

Effect of Low Frequency Electrical Stimulation on VEGF Expression of Suspension Induced Atrophic Muscle (뒷다리 현수 유도 위축근에 대한 저빈도 전기자극의 VEGF 발현에 미치는 영향)

  • Purpose : The purpose of this experiment was to evaluate the pre-application effect of low frequency electrical stimulation(LFES) on VEGF expression of atrophic muscle and to determine the optimal pre-application period of LFES for prevent muscle atropy Methods : Twenty-five adult sprague-dawley rats were randomly assigned to weight bearing group, hindlimb suspension for 14 days group, hindlimb suspension with pre-application of LFES for 14 days group, hindlimb suspension with pre-application of LFES for 11 dsys group and, hindlimb suspension with pre-application LFES for 7 dsys group. 16Hz of Biphasic pulse current was applied to gastrocnemius for 15min per days. Results : VEGF were decreased expression in HSG groups, whereas VEGF were significantly increased in HS+ES14G, HS+ES11G, HS+ES7G groups Conclusion : LFES during the hindlimb suspension showed a positive effect in VEGF induction and early application is strongly encourage VEGF induction. This indicated that pre-application of LFES could prevent muscle atrophy.

Klotho : Expression and Regulation at the Maternal-Conceptus Interface in Pigs

  • Klotho (KL) is a single transmembrane protein composed of KL1 and KL2 repeats possessing ${\beta}$-glucuronidase activity and maintains calcium homeostasis in physiological state. It has been implicated in pigs that calcium is important for the establishment and maintenance of pregnancy, and our previous study has shown that transient receptor potential vanilloid type 6 (TRPV6), a calcium ion transporter, is predominantly expressed in the uterine endometrium during pregnancy in pigs. However, expression and function of KL in the uterine endometrium has not been determined in pigs. Thus, the present study determined expression and regulation of KL in the uterine endometrium during the estrous cycle and pregnancy in pigs. Real-time RT-PCR analysis showed that levels of KL mRNA decreased between Days 12 to 15 of the estrous cycle, and its expression showed a biphasic manner during pregnancy. KL mRNA was expressed in conceptuses and in chorioallantoic tissues during pregnancy. Explant culture study showed that expression levels of KL were not affected by treatment of steroid hormones or interleukin-1beta during the implantation period. Furthermore, levels of KL mRNA in the uterine endometrium from gilts carrying somatic cell nuclear transfer (SCNT)-derived embryos were significantly lower than those from gilts carrying natural mating-derived embryos on Day 12 of pregnancy. These results exhibited that KL was expressed at the maternal-conceptus interface in a pregnancy status- and stage-specific manner, and its expression was affected by SCNT procedure, suggesting that KL may play an important role in the establishment and maintenance of pregnancy in pigs.

  • To maximize the performance of recombinant cell fermentation process through optimizing environmental conditions, the production of invertase from recombinant Saccharomyces cerebisiae Containing SUC2 gene was studied as a model. The recombinant cells showed biphasic growth on glucose. Since the promoter of the SUC2 is regulated by the concentration of glucose in the medium, expression of invertase by recombinant yeast began when the glucose concentration decreased in a range of 0.25-0.4 g/L during the batch culture. Plasmid segregation occured frequently during glucose fermentation, and infrequently during ethanol oxidation. A rapid appearance of invertase activity with glucose was observed under nonaerated condition, and the maximum specific invertase activity was about 1.5 times as high as under aerobic condition, In fed batch culture, when n low level of glucose was continuously supplied to the tormentor after the time of glucose depletion during growth phase, specific and total invertase activity increased about 1.7 and 2.9 fold, respectively, in a batch culture.
