Biosynthesis of Poly(3HB-3HV) and Poly(3HB-4HB) Copolymers in Recombinant Ralstonia eutropha Enforced zwf

  • NADPH has been known as a regulating factor the biosynthesis of polyhydroxyalkanote(PHA), and the flux of NADPH for PHA biosynthesis could be enforced by the amplification of zwf gene encoding glucose 6-phosphate dehydrogenase. The recombinant plasmid pCZWF harboring PHA synthase, phbC from R. eutropha and zwf from E. coli were constructed, and were transformed to R. eutropha by electroporation. The biosynthesis of P(3HB-3HV) copolymer were carried out in transformant R. eutropha through the two-stage cultivation method using valerate as a precursor. The biosynthesis rate and PHA content of transformant R. eutropha harboring pCZWF were increased compared with transformant R. eutropha harboring only phbC. Especially, the molar fraction of 3HV was increased from 68% to 74% due to amplification of zwf gene. And the biosynthesis P(3HB-3HV) and P(3HB-4HB) carried out using propionate and ${\gamma}-butyrolactone$ as a precursor, respectively. But the rate, content, and molar fraction of biosynthesis copolymers were not influenced appreciably. This may be due to the reduced availability of NADPH.

Polyamine에 의한 옥수수 Ribosome의 활성 촉진 (Stimulation of Ribosome Activity of Zea mays by Polyamine)

  • As a part of the study on the relation between exogenous polyamines and various components necessary for protein biosynthesis in the germinating maize seeds, the effects of the polyamines on protein biosynthesis and irbosome activity were investigated. The protein biosynthesis activity by S-30 containing all components necessary for protein biosynthesis was increased by exogenous polyamines, spermidine, spermine and putrescine. As the concentration of polyamine treated was increased, the optimal Mg2+ concentration of in vitro poly U-dependent protein synthesis system was gradually reduced. However, the optimal Mg2+ concentration of poly U-dependent system containing optimal polyamine was 10 mM regardless of the sort of polyamine. It could be infered that polyamines play an important part in protein biosynthesis in the higher plant, and that the role of polyamines take partially the place of Mg2+ action. The activities of ribosome and S-100 in protein biosynthesis were increased by 46.7% and 17.7% with spermidine, and by 44.1% and 16.2% with spermine, and by 29.1% and 19.3% with putrescine. It could be concluded that the increase of protein biosynthesis by polyamines in mainly owing to the ribosome activation.

  • 나방류의 페로몬 생합성은 식도아래 신경절에서 생산된 페로몬 생합성 촉진 신경펩타이드(pheromone biosynthesis activating neuropeptide, PBAN)의 분비로부터 시작된다. 분비된 PBAN은 페로몬샘의 상피세포막에 있는 PBAN 수용체와 결합하고, 신호전달과정을 거쳐 페로몬 생합성과정이 활성화되어, 페로몬이 외부로 방출된다. 본 종설은 콩명나방(Maruca vitrata)의 성페로몬, PBAN과 수용체, 성페로몬 생합성 경로를 소개한다.

A Cholesterol Biosynthesis Inhibitor from Rhizopus oryzae

  • A bile acid derivative, methyl chalate (1), was isolated from EtOAc extract of the fungus Rhizopus oryzae as a cholesterol biosynthesis inhibitor. It showed moderate inhibitory activity on cholesterol biosynthesis in human Chang liver cells. Compound 1 exhibited inhibitory effect on the later step of cholesterol biosynthesis, indicating that its action mode is different from that of statins that act on the HMG-CoA reductase.

Regulation of Ethylene Biosynthesis by Phytohormones in Etiolated Rice (Oryza sativa L.) Seedlings

  • The gaseous hormone ethylene influences many aspects of plant growth, development, and responses to a variety of stresses. The biosynthesis of ethylene is tightly regulated by various internal and external stimuli, and the primary target of the regulation is the enzyme 1-aminocyclopropane-1-carboxylic acid (ACC) synthase (ACS), which catalyzes the rate-limiting step of ethylene biosynthesis. We have previously demonstrated that the regulation of ethylene biosynthesis is a common feature of most of the phytohormones in etiolated Arabidopsis seedlings via the modulation of the protein stability of ACS. Here, we show that various phytohormones also regulate ethylene biosynthesis from etiolated rice seedlings in a similar manner to those in Arabidopsis. Cytokinin, brassinosteroids, and gibberellic acid increase ethylene biosynthesis without changing the transcript levels of neither OsACS nor ACC oxidases (OsACO), a family of enzymes catalyzing the final step of the ethylene biosynthetic pathway. Likewise, salicylic acid and abscisic acid do not alter the gene expression of OsACS, but both hormones downregulate the transcript levels of a subset of ACO genes, resulting in a decrease in ethylene biosynthesis. In addition, we show that the treatment of the phytohormones results in distinct etiolated seedling phenotypes, some of which resemble ethylene-responsive phenotypes, while others display ethylene-independent morphologies, indicating a complicated hormone crosstalk in rice. Together, our study brings a new insight into crosstalk between ethylene biosynthesis and other phytohormones, and provides evidence that rice ethylene biosynthesis could be regulated by the post-transcriptional regulation of ACS proteins.

Chemical Genetics Approach Reveals Importance of cAMP and MAP Kinase Signaling to Lipid and Carotenoid Biosynthesis in Microalgae

  • In this study, we attempted to understand signaling pathways behind lipid biosynthesis by employing a chemical genetics approach based on small molecule inhibitors. Specific signaling inhibitors of MAP kinase or modulators of cAMP signaling were selected to evaluate the functional roles of each of the key signaling pathways in three different microalgal species: Chlamydomonas reinhardtii, Chlorella vulgaris, and Haematococcus pluvialis. Our results clearly indicate that cAMP signaling pathways are indeed positively associated with microalgal lipid biosynthesis. In contrast, MAP kinase pathways in three microalgal species are all negatively implicated in both lipid and carotenoid biosynthesis.

  • The role of S-adenosylmethionine (SAM) as an intermediate in interrelation between polyamine and ethylene biosynthesis was studied in suspension cultures of Nicotiana tabacum L. Exogenous SAM stimulated the polyamine and ethylene biosynthesis in 4 day-cultured cells, which were in active cell divisions, and 10 day cultured cells, which went on with active cell elongation and senescence. SAM-induced ethylene production was more effective in 10 day-cultured cells than in 4 day-cultured cells, but SAM-induced polyamine biosynthesis was more effective in 4 day-cultured cells than in 10 day-cultured cells. Polyamine contents were increased by the blockage of ethylene biosynthetic pathway in the conversion of SAM to ethylene via 1-aminocyclopropane-1-carboxylinc acid (ACC) with aminooxyacetic acid (AOA). Also, ethylene production was increased by the inhibitors of polyamine biosynthesis such as methylglyoxal bis-(guanylhydrazone) (MGBG), dicyclohexylamine (DCHA), $\alpha$-difluoromethylarginine (DFMA) and $\alpha$-difluoromethylorinithine (DFMO). These results suggest that there may be interrelations between polyamine and ethylene biosynthesis for the competition of SAM and the inherent mechanism of switch on-off in polyamine and ethylene biosynthetic activity with the progress of cell growth and senescence.

Streptomyces tubercidicus에서 Adenine과 Histidine에 의한 Tubercidin 생합성 조절 (Regulation of Tubercidin Biosynthesis in Streptomyces tubercidicus by Adenine and Histidine)

  • The regulatory mechanism of tubercidin biosynthesis in Streptomyces tubercidicus was studied. In a wild type strain, addition of adenine and histidine into the medium decreased the tubercidin production by 60-65% and 40%, respectively. The effects of adenine and histidine were alleviated by the addition of inosine monophosphate and 5-aminoimidazole-4-carboxamide ribotide. The production of tubercidin in S. tubercidicus K115 strain ($ade^{-}$ ) was nearly shut off by histidine. In contrast with K115 strain, adenine inhibited the tubercidin biosynthesis in S. tubercidicus K412 strain ($his^{-}$. In S. tubercidicus F667 strain ($ade^{-}$ , $his^{-}$ ), tubercidin production was increased by adenine and histidine. From the effects of adenine and histidine on tubercidin biosynthesis in S. tubercidicus wild type and mutant strains, it became known that feedback control by adenine and histidine of biosynthetic pathwat for purine ribonucleotide and histidine are involved in the regulation of tubercidin biosynthesis.

Screening of Sterol Biosynthesis Inhibitors from Natural Products Using Recombinant Yeast Carrying Human Lanosterol Synthase

  • For the screening of inhibitors of sterol biosynthesis from natural products, a simple and rapid assay method was developed using recombinant yeast carrying human lanosterol synthase, main target of this assay method. Sterol biosynthesis inhibition activity was monitored only by the inhibition of growth of the recombinant yeast. By changing the substrate, this assay method can figure out which step is inhibited in the sterol biosynthesis by the test material. With this assay method total 102 plant samples were screened for their inhibitory activity of sterol biosynthesis. Among plant water extracts screened, 11 plant samples showed inhibitory activity on sterol biosynthesis in ergosterol (-) medium. For selection of the specific inhibitory materials, 11 plant samples were reassayed in ergosterol (+) medium. After all 5 plant samples, Abutilon avicennae Gaertn. (stem), Alnus japonica Steud. (stem), Amaranthus mangostanus L. (aerial part), Philadelphus schrenckii Pupr. (leaf) and Pimpinelia brachycarpa Nakai (aerial part), showed specific inhibitory activity.

Platelet-Activating Factor Antagonistic Activity and ^13C NMR Assignment of Pregomisin and Chamigrenal from Schisandra chinensis

  • In the course of searching for PAF receptor antagonists, pregomisin (1) and chamigrenal (2) were isolated from the fruits of Schizandra chinensis Baill by the bioactivity-guided isolation. Both compounds showed PAF antagonistic activity and the $IC_{50}$ values were $4.8{\times}10^{-5} M and 1.2{\times}10^{-4}M,$ respectively. In addition, the $^{13}C$ NMR assignments of 1 and 2 using DEPT, HMQC, COLOC and HMBC were reported for the first time.
