Computer Model for Fibrinogen Adsorption on Polyurethane Surface

  • Protein adsorption on the polyurethane surface was modelled by a modified random sequential adsorption(RSA) process. In this model, polyurethane surface was modelled as a mixed domain of hydrophobic and hydrophilic parts which was implemented by a 2 dimensional $150{\times}150$ lattice in the computer. Protein adsorption was simulated using a small box which represents a particle of the protein, and polyurethane lattice by considering their hydrophobic interaction. In order to validate the model, we perfonned fibrinogen adsorption on polyurethane surface. Isotherms of the adsorbed protein were calculated and compared to the experimental data. The protein adsorption on the polyurethane surface could be well described using this computer model.

  • Construction of the stable monolayer of endothelial cells onto physicochemically modified polymeric surFace is one of the appropriate method to develop the small caliber vascular graft with the long-term patency. In this study, we constructed the monolayer of endothelial cells on the fibronectin rind the extracellular matrix-coated polyurethane surface derived from human fibroblast cells. To elucidate the adhesion strength of endothelial cells on the extracellular matrix-coated polyurethane, a laminar flow chamber apparatus was developed to exposure the shear stress on the apical membrane of ondothelial cells. Endothelial cells show the strongest adhesion after two days of seeding onto the fibronectin-coated polyurethane surface, whereas endothelial cells on the extracellular matrix derived from the human flbroblast cells show the minimal doubling time of cellular growth.

고분자 표면의 혈관내피세포 부착력에 관한 연구 (Evaluation of Endothelial Cell Attachment on Polymer Surface)

  • To improve antithrombogenicity of polymer that used in vascular graft and artificial organs, seeding of human endothelial cells on the polyurethane was studied. Human endothelial cells were ismlated from human umbilical veins, using type I collagenase, and identified with goat anti vWF antibodies. Human endothilial cell seeding was tried upon the polyurethane which has good mechanical property and resists stresses. The hydrophobic polyurethane surface was changed hydrophilic by corona discharf:e treatment. Surface hydrophilicity was measured with Wilhemly plate method and the goniometer. To evaluate matrix protein adsorption, fibronectin adsorption test was done. To eveluate cell adhesion, human endothelial cell attachment forces were measured rising a perfusion chamber of , ism diamter. Less cells were detached from the hydrophilically treated polyurethane. This showed that corona discharge on the polyurethane could improve matrix adsorption and endothelial cell attachment.

  • Many experiments about endothelial cell seeding on artificial vessels were studied and conducted For this one or a combination of the extramatrix was used for the underlying matrix. But we used the whole ECM(extracellular matrix) that made excreated from flbroblasl. In thls study, we obtained human adult omental microvascular endothelium by collagenase digestion and used polyurthane sheets in order to make a new artificial vessel material. We cultured fibroblast on the polyurethane and gelatin - coated polyurethane. After confluent ingrowth we treated the polyure thane with triton in order to destroy the cytoskeleton and nucleus. We observed the preformed extra cellular matrix on the ployurethane and cultured the isolated microvascular endothelium. We also ok served the growth of microvascular endothelium on the polyurethane and gelatin. We conclude that the use of the whole ECM is promising fair as a new underying substrate for endothelial cell seeding on artificial vessels.

Antibacterial Effect of the Surface-Modified Biomedical Polyurethane against Staphylococcus aureus and Staphylococcus epidermidis

  • Staphylococal infection still remains to be one of the most serious infections, having various complications in the clinical use of indwelling polymeric medical devices. However, there are a few promising systems showing a high antibacterial effect without causing any demage of polymer backbone under biological environments such as blood or body fluid. In order to resolve this problem, we have designed a new antibiotic releasing system via a hydrolysis mechanism. The surface of biomedical polyurethane (PU) was modified by using 1,6-diisocyanatohexane (HMDI) to immobilize the rifampicon. Also, the immobilized rifampicin was designed to be released by a selective cleavage of the unstable carbamate linkage that exists on the rifampicin-immobilized polyurethane (PHR). The immobilization of rifampicin on the surface of polyurethane was confirmed by the disappearance of the characteristics IR absorbance peak of the isocyanate (-NCO) group at $2,267\;cm^{-1}$. The PHR showed a continuous rifampicin release profile under an aqueous environment of 10 mM of PBS (phosphate-buffered saline) for ove 6 days. The rifampicin molecules, which are released from PHR under an optimal bacterial infection environment, had a higher antibacterial activity against both S. aureus and S. epidermidis than rifampicin-incorporated polyurethane (RIP). In addition, the PHR maintained a stable antibacterial effect under a blood-mimic aqueous environment such as bovine calf serum.

  • The object of this study is to investigate the potential of dual-electrospun polymer based structure for vascular tissue engineering, especially for the medium or small sue blood vessels. Polyurethane(PU), which is known to be biocompatible in this area, was electrospun with poly(ethylene oxide) (PEO). Concentration of PU was fixed at 20wt%, while that of PEO was set from 15 to 35wt%. Morphological features were observed by SEM image and measurement of porosity and cellular responses were tested before and after extracting PEO from the hybrid scaffolds by immersing the scaffolds into distilled water. The diameter of PEO fibers were ranged from 200nm to 500nm. The lower concentration of PEO tended to show beads. The porosity of the scaffolds after extracting PEO was highly increased with higher concentration of PEO as expected. Also, higher proliferation rate of smooth muscle cells was observed at higher concentration of PEO than at the lower concentration and without PEO. As conclusions, this dual electrospinning technique combined with PU and PEO is expected to overcome the current barrier of cell penetration by providing more space for cells to proliferation.

Plasma Surface Modification of Patterned Polyurethane Acrylate (PUA) Film for Biomedical Applications

  • Polyurethane acrylate (PUA) has been introduced to utilize as a mold material for sub-100 nm lithography as it provides advantages of stiffness for nanostructure formation, short curing time, flexibility for large area replication and transparency for relevant biomedical applications. Due to the ability to fabricate nanostructures on PUA, there have been many efforts to mimic extracellular matrix (ECM) using PUA especially in a field of tissue engineering. It has been demonstrated that PUA is useful for investigating the nanoscale-topographical effects on cell behavior in vitro such as cell attachment, spreading on a substrate, proliferation, and stem cell fate with various types of nanostructures. In this study, we have conducted surface modification of PUA films with micro/nanostructures on their surfaces using plasma treatment. In general, it is widely known that the plasma treated surface increases cell attachment as well as adsorption of ECM materials such as fibronectin, collagen and gelatin. Effect of plasma treatment on PUA especially with surface of micro/nanostructures needs to be understood further for its biomedical applications. We have evaluated the modified PUA film as a culture platform using adipose derived stem cells. Then, the behavior of stem cells and the level of adsorbed protein have been analyzed.

일축 압축하중 하 다공성 폴리우레탄폼의 재료비선형 거동 및 미세구조 변화 (Material Nonlinear Behavior and Microstructural Transition of Porous Polyurethane Foam under Uniaxial Compressive Loads)

  • Porous materials such as polymeric foam are widely adopted in engineering and biomedical fields. Porous materials often exhibit complex nonlinear behaviors and are sensitive to material and environmental factors including cell size and shape, amount of porosity, and temperature, which are influenced by the type of base materials, reinforcements, method of fabrication, etc. Hence, the material characteristics of porous materials such as compressive stress-strain behavior and void volume fraction according to aforementioned factors should be precisely identified. In this study, unconfined uniaxial compressive test for two types of closed-cell structure polyurethane foam, namely, 0.16 and $0.32g/cm^3$ of densities were carried out. In addition, the void volume fraction of three different domains, namely, center, surface and buckling regions under various compressive strains (10 %, 30 %, 50 % and 70 %) were quantitatively observed using Micro 3D Computed Tomography(micro-CT) scanning system. Based on the experimental results, the relationship between compressive strain and void volume fraction with respect to cell size, density and boundary condition were investigated.

Prevention of Catheter-related Infections (CRIs) using Ciprofloxacin

  • Microbial infection provokes one of the most serious complications to the patients with indwelling catheters. Ciprofloxacin (CFX) was added into the catheter materials (polyurethane or silicone) during the manufacturing process to avoid the microbial infection. Efficacy of the catheters containing CFX was investigated by using the in vitro zone of growth inhibition test method. The catheters made of polyurethane or silicone exhibited a strong antimicrobial activity against the major catheter-related microorganisms (S. aureus, S. epidermidis, P. aeruginosa and E. coli), when CFX was incorporated into the catheters. Fetal bovine serum (FBS) did not affected antimicrobial activities of the polyurethane catheters with CFX loading of 0.5 and 1.0% (W/W) against S. aureus and S. epidermidis. However, the polyurethane catheters with 1.0% (W/W) of CFX loading showed a significantly (P<0.05) reduced antimicrobial activity against E. coli when the catheters were exposed to FBS. Silicone catheters with 1.0 and 1.5% (W/W) of CFX loading demonstrated effective antimicrobial activity against S. epidermidis for at least 2 weeks. These results suggest that the use of catheters containing ciprofloxacin could be effective in preventing catheter-related infections.
