  • The cell spheroid (multicellular mass) is enhanced cell functions because of the cell-cell interaction compared with the individual cell. The objective of this study is synthesis, characterization and evaluation of novel crosslinkers to form spheroid in a short time. Our approach to bridge cells is based on the crosslinking of the cell membrane via the hydrophobic interaction. The crosslinker was prepared by the reaction between ethylenediamine and poly(ethylene glycol) (PEG) derivative with oleyl group as hydrophobic group at the terminal group. The product was characterized with gel permeation chromatography (GPC) and FT-IR. Furthermore, cell culture experiment was also performed to confirm spheroid formation. The function of prepared spheroids was evaluated.

Precise ultrasonic coating and controlled release of sirolimus with biodegradable polymers for drug-eluting stent

  • In the current study, a drug-eluting stent coated with biodegradable polymers and sirolimus was developed by using an ultrasonic nanocoater and characterized in aspects of surface smoothness and coating thickness. In addition, in vitro release profiles of sirolimus by changing top coating layer with different biodegradable polymers were investigated. Smooth surfaces with variable thickness could be fabricated by optimizing polymer concentration, flow rate, nozzle-tip distance, gas pressure, various solvents and ultrasonic power. Smooth surface could be generated by using volatile solvents (acetone, chloroform, and methylene chloride) or post-treating with solvent vapor. Coating thickness could be controlled by varying injection volume or polymer concentration, and higher concentration could reduce the coating time while obtaining the same thickness. The thickness measurement was the most effectively performed by a conventional cutting method among three different methods that were investigated in this study. Release profiles of sirolimus were effectively controlled by changing polymers for top layer. PLGA made the release rate 3 times faster than PDLLA and PLLA and all top layers prevented burst release at the initial phase of profiles. Our results will provide useful and informative knowledge for developing drug-eluting stents, especially coated with biodegradable polymers.

  • Peroxiredoxin 6 (Prx 6) is a member of the thiol-specific antioxidant protein family, which may play a role in protection against oxidative stress and in regulating phospholipid turnover. The aim of this study was to determine whether a human Prx 6/Luc vector was stably expressed and responded to antioxidants in a lung cell line (NCI-H460). To achieve this, the luciferase signal, hPrx 6 mRNA expression, and superoxide dismutase (SOD) activity were measured in transfectants with a hPrx 6/Luc plasmid after treatment with four antioxidant extracts, including Korea white ginseng (KWG), Korea red ginseng (KRG), Liriope platyphylla (LP), and red Liriope platyphylla (RLP). First, the hPrx 6/Luc plasmid was successfully constructed with DNA fragments of human Prx 6 promoter, amplified by PCR using genomic DNA isolated from NCI-H460 cells, and cloned into the pTransLucent reporter vector. The orientation and sequencing of the hPrx 6/Luc plasmid were identified with restriction enzyme and automatic sequencing. A luciferase assay revealed significant enhancement of luciferase activity in the four treatment groups compared with a vehicle-treated group, although the ratio of the increase was different within each group. The KRG- and LP-treated groups showed higher activity than the KWG- and RLP-treated groups. Furthermore, the luciferase activity against RLP occurred roughly in a dose-dependent manner. However, the level of endogenous hPrx 6 mRNA did not change in any group treated with the four extracts. The SOD activity was in agreement with the luciferase activity. Therefore, these results indicate that the hPrx 6/Luc vector system may successfully express and respond to antioxidant compounds in NCI-H460 cells. The data also suggest that the Prx 6/Luc vector system may be effectively applied in screening the response of hPrx 6 to antioxidant compounds in transgenic mice.

  • 대부분의 가정과 요식업체, 식품가공업계에서 이용하고 있는 식용유지는 저장 및 가공과정 중에 산패가 빈번하게 일어나게 된다. 기존에는 유지 산패를 측정하기 위해 산가, 과산화물가 등을 측정하는 이화학적인 적정방법을 이용하였는데 실험자의 숙련도에 따라 결과의 오차가 발생할 수 있고, 반복실험으로 인한 시간과 비용이 많이 소모되는 등 여러 제약사항을 포함하고 있어 식용유지의 산패를 실시간 비파괴적으로 분석할 수 있는 기술의 개발에 많은 관심이 모아지고 있다. 따라서, 본 연구에서는 식용유지의 저장조건에 따른 산패정도를 비파괴적으로 평가하기 위한 근적외선 분광분석과 인공신경망 분석기술을 개발하여 그 실효성을 평가하였다. 식물성 식용유지인 들기름을 특정 온도에서 일정한 시간동안 저장하면서 이화학적 적정방법을 통해 산가와 과산화물가를 측정하였으며 동일한 시료의 근적외선 투과스펙트럼을 획득하였다. 수집된 정보를 이용하여 유지 산패 예측 모델을 개발하기 위해 다변량 분석기법 (주성분 회귀분석, 최소자승 회귀분석과 인공신경망 분석)을 적용하였다. 분석 결과, 인공신경망 분석모델이 산가 ($R^2_{tra}:0.9037$, $R^2_{val}:0.8175$, $R^2_{test}:0.8555$)와 과산화물가 ($R^2_{tra}:0.9210$, $R^2_{val}:0.9341$, $R^2_{test}:0.8286$)의 예측 성능이 가장 우수한 것으로 확인되었다. 본 연구의 결과들은 농산물과 식품의 성분 측정뿐만 아니라 다른 산업분야에서도 유용하게 활용될 수 있을 것으로 기대되어진다.

Preparation and Properties of Regenerated Composite Fibers made from Styela Clava Tunics/PVA Blending(I) (미더덕 껍질과 PVA를 혼합한 재료로부터 제조한 복합섬유의 제법과 성질(I))

  • Regenerated composite fibers are prepared from solution(styela clava tunics /poly vinyl alchol) using N-methylmorpholine-N-oxide(NMMO)/water(87/13)(wt/wt) as a solvent by dry-wet spinning. The chemical cellulose (94%, ${\alpha}$-cellulose content) used for this study is extracted from styela clava tunics (SCT, Midduck), which are treated in chemical process and mechanical grinding. The structure and physical properties of regenerated composite fibers were investigated through IR-spetra, DSC, TGA and SEM. The optimal blend ratio of SCT/PVA for spinning solution was 70/30 and the total weight was 4% concentrations in NMMO/water solvent system. The fiber density, moisture contents and the degree of swelling were $1.5(g/cm^3)$ 10.2(%) and 365(%), respectively. The crystallinity index of composite fibers are decreased as the PVA contents increased. Thermal decomposition of composite fibers took place in two stages at around $250^{\circ}C$ and $550^{\circ}C$. The best thermal stability was obtained with 30% PVA contents.

Structure and physical properties of regenerated composite fibers were investigated through birefrngence, x-ray diffratograms, tenacity, fibrillation and SEM. Optimal blend ratio of SC/PVA for mechanical properties of composite fibers was 70/30 and total weight was 4wt% concentrations in NMMO/$H_2O$ solvent system. Crystallinity index of composite fibers were decreased as the PVA contents increased. Fibrillation of $10{\sim}20wt%$ PVA blended fibers were occurred less than pure SC fiber. Shape of composite fibers were a circle cross section within 10wt% PVA content. But the cross section of fibers were changed as crushed flat with the PVA contents increased.

Effect of Codonopsis lanceolata with Steamed and Fermented Process on Scopolamine-Induced Memory Impairment in Mice

  • Codonopsis lanceolata (Campanulaceae) traditionally have been used as a tonic and to treat patients with lung abscesses. Recently, it was proposed that the extract and some compounds isolated from C. lanceolata reversed scopolamine-induced memory and learning deficits. The purpose of this study was to evaluate the improvement of cognitive enhancing effect of C. lanceolata by steam and fermentation process in scopolamine-induced memory impairment mice models by passive avoidance test and Morris water maze test. The extract of C. lanceolata or the extract of steamed and fermented C. lanceolata (SFCE) was orally administered to male mice at the doses of 100 and 300 mg/kg body weight. As a result, mice treated with steamed and fermented C. lanceolata extract (SFCE) (300 mg/kg body weight, p.o.) showed shorter escape latencies than those with C. lanceolata extract or the scopolamine-administered group in Morris water maze test. Also, it exerted longer step-through latency time than scopolamine treated group in passive avoidance test. Furthermore, neuroprotective effect of SFCE on glutamate-induced cytotoxicity was assessed in HT22 cells. Only SFCE-treated cells showed significant protection at 500 ${\mu}g/ml$. Interestingly, steamed C. lanceolata with fermentation contained more phenolic acid including gallic acid and vanillic acid than original C. lanceolata. Collectively, these results suggest that steam and fermentation process of C. lanceolata increased cognitive enhancing activity related to the memory processes and neuroprotective effect than original C. lanceolata.

Effects of Micro-Electrical Stimulation on Regulation of Behavior of Electro-Active Stem Cells

  • Purpose: Stem cells provide new opportunities in the regenerative medicine for human or animal tissue regeneration. In this study, we report an efficient method for the modulating behaviors of electro-active stem cells by micro-electric current stimulation (mES) without using chemical agents, such as serum or induction chemicals. Methods: Dental pulp stem cells (DPSCs) were cultured on the tissue culture dish in the mES system. To find a suitable mES condition to promote the DPSC functions, the response surface analysis was used. Results: We found that a working micro-current of 38 ${\mu}A$ showed higher DPSC proliferation compared with other working conditions. The mES altered the expressions of intracellular and extracellular proteins compared to those in unstimulated cells. The mES with 38 ${\mu}A$ significantly increased osteogenesis of DPSCs compared with ones without mES. Conclusions: Our findings indicate that mES may induce DPSC proliferation and differentiation, resulting in applying to DPSCs-based human or animal tissue regeneration.

Comparison of D-[18F]FMAU and L-[18F]FMAU as PET Imaging Agents for HSV1-TK Gene Expression

  • D-[$^{18}F$]FMAU and L-[$^{18}F$]FMAU are F-18 labeled nucleoside analogue which have been efficiently synthesized in order to be a PET imaging probe. D-[$^{18}F$]FMAU and L-[$^{18}F$]FMAU were compared as PET imaging agents using HSV1-TK gene expressing tumor-bearing mice. Their cellular uptake profiles were also compared using MCA and MCA-TK cell lines. D-[$^{18}F$]FMAU demonstrated higher cellular uptake and higher accumulation in MCA-TK tumor regions than L-[$^{18}F$]FMAU. On the other hand, L-[$^{18}F$]FMAU showed higher MCA-TK/MCA ratio of %ID/g than that of D-[$^{18}F$]FMAU. L-[$^{18}F$]FMAU can be utilized as a good candidate for HSV1-TK PET imaging. It can be used for antiviral drug evaluation.
