Identification of Physiologically Active Compounds from Purslane(Portulaca oleracea L.) (쇠비름(Portulaca oleracea L.)에 함유(含有)된 생리활성물질(生理活性物質) 탐색(探索))

  • This experiment was conducted to determine the presence of allelopathic substance in Portulaca oleracea L. Water and methanol extract from P. oleracea markedly inhibited the germination of lettuce, rice, raddish etc., indicating the presence of biological substances. The biochemical substances such as ferulic, p-coumaric, salicylic, vanillic, p-hydroxybenzoic acid etc., belonging to phenolic compounds were detected in a large amount, which may be responsible for exhibiting inhibitory effects. Various phenolic compounds were detected from different samples such as freshly, dried plants, seeds and callus. The highest amount of tannic and gallic acids were detected in dried samples of P. oleracea, the highest grantity of salicylic and vanillic acids in fresh samples, the largest amount of ferulic acid in seed sample, the highest amount of p-hydroxybenzoic and p-chlorobenzoic acids in callus. Linolenic acid was presented in amount of 9.203 mg/g in dried plant of P. oleracea as one of the major fatty acids and oxalic acid presented 27.941 mg/g as one of the major organic acids. These compounds seemed to be related to inhibitory effect of P. oleracea which needs further study.

Prediction of Chemical Organic Composition of Manure by Near Infrared Reflectance Spectroscopy

  • The organic materials included in excreta of livestock are important resources for organic manure and for improving soil quality, although there is still far from effective using. One reason for this is still unclearly standard of quality for evaluation of manure made from excreta of livestock. Therefore, the objective of this study is to develop rapid and accurate analytical method for analyzing organic compositions of manure made from excreta of livestock, and to establish quality evaluation method based on the compositions predicted by near infrared reflectance spectroscopy (NIRS). Sixteen samples of manure, each eight samples prepared from two treatments, were used in this study. The manure samples were prepared by mixing 560 kg feces of swine,60 kg sawdust with moisture content was adjusted to be 65%. The mixture was then keep under two kinds of shelter, black and clear sheets, as a treatment on the effect of sunlight. Samples were taken in every week (form week-0 to 7) during the process of manure making. Samples were analyzed to determine neutral detergent fiber (NDF), acid detergent fiber (ADF) and acid detergent lignin (ADL) by detergent methods, and organic cell wall (OCW) and fibrous content of low digestibility in OCW (Ob) by enzymatic methods. Biological oxygen demand (BOD) was analyzed by coulometric respirometer method. These compositions were carbohydrateds and lignin that were hardly digested. Spectra of samples were scanned by NIR instrument model 6500 (Pacific Scientific) and read over the range of wavelength between 400 and 2500nm. Calibration equations were developed using eight manure samples collected from black sheet shelter, while prediction was conducted to the other eight samples from clear sheet shelter. Accuracy of NTRS prediction was evaluated by correlation coefficients (r), standard error of prediction (SEP) and ration of standard deviation of reference data in prediction sample set to SEP (RPD). The r, SEP and RPD value of forage were 0.99, 0.69 and 7.6 for ADL, 0.96, 1.03 and 4.1 for NDF, 0.98, 0.60 and 4.9 for ADF, 0.92, 1.24 and 2.6 for Ob, and 0.91, 1.02 and 7.3 for BOD, respectively. The results indicated that NIRS could be used to measure the organic composition of forage used in manure samples.

Isolation and Purification of Fucoidans from Laminaria religiosa and Undaria pinnatifida in Korea (한국산 다시마 및 미역으로 부터 Fucoidan의 추출 및 정제)

  • The objective of this research was to characterize fucoidans isolated from Laminaria religiosa and Undaria pinnatifida in Korea to obtain basic data for Production of soluble dietary fiber materials with biological functionality. Fucoidans were successively extracted 3 times at $65\%$ for 1hr with arid solution of pH 2.0, and cetylpyridinium chloride was used for partial purification. The yields of partially purified fucoidans were $2.71\%$ for L. religiosa, $6.65\%$ for sporophylls of U. pinnatifida and $0.40\%$ for blade of U. pinnatifida. The yield from sporophylls of U. pinnatifida was highest among the sample tested, whereas the yield from blade of U. pinnatifida was lowest. It appeared that the fuconidans content in different parts of U. pinnatifida varied. Partially purified fucoidans were separated into 3 fractions by DEAE-Sephadex A-25 ion exchange column and the maior fractions were refractionated with tractional precipitation with ethanol. $60-70\%$ ethanol precipitated fractions of 1. religiosa and sporophylls of U. pinnatifida turned out to be homogeneous by cellulose acetate electrophoresis and gel filteration chromatography. The molar ratios of fucose, galactose, and sulfate in the purified fucoidans(ethanol precipitated fractions) were 1 : 0.31 : 2.43 for L. religiosa and 1 : 0.97 : 1.99 for sporophylls of U. pinnatifida. The averaged molecular weights of the purified fucoidans from L. religiosa and sporophylls of U. pinnatifida were 31,000 and 38,000, respectively.

Evaluation of Workers' Exposures to Airborne Lead chromate in the Producing and Using Industries (국내 무기안료 제조 및 취급 공정에서의 공기 중 크롬산연 노출 평가)

  • Lead chromate is made by sodium dichromate and lead acetate, and has being used widely in the part of pigment, paints, inks, plastics and so on. Even though lead chromate has health hazards which like both lead and chromium, there are a few study about pigment workplaces using lead chromate in Korea. The purpose of this study is to evaluate workers' exposure levels and airborne lead and chromium concentration in the pigment workplaces using lead chromate. There are 20 workers in the total 5 workplaces. 10 workers(50%) have been exposed to lead and 3 workers(15%) have been exposed to chromium, which exceeded the American Conference of Governmental Industrial Hygienists(ACGIH) Threshold Limit Value (Pb: $0.05\;mg/m^3$, Cr: $0.012\;mg/m^3$) and Korean Ministry of Labor's Standard. Geometric mean (GM) of airborne lead was highest in pigment ($0.0421\;mg/m^3$), paint ($0.0020\;mg/m^3$) and PVC coloring ($0.0007\;mg/m^3$), respectively(p<0.05). The result of airborne chromium concentration was paint ($0.0033\;mg/m^3$), paint ($0.0004\;mg/m^3$) and PVC coloring ($0.0003\;mg/m^3$). Also the lead and chromium concentration in the manual process is each 30 times and 10 times higher than the value in automatic process(p<0.01). In the classified process by detail, the concentration of airborne lead was $0.0638\;mg/m^3$ in grinding & packaging, mixture & after-measuring ($0.0436\;mg/m^3$), filtration & drying ($0.0402\;mg/m^3$), lead nitrate & dissolution($0.0129\;mg/m^3$), pigment commitment & mixture ($0.0013\;mg/m^3$) and dispersion & grinding ($0.0010\;mg/m^3$) (p<0.05). Moreover the concentration of a sample in weighting & packaging was $0.0023\;mg/m^3$. The concentration of lead in workers' blood was pigment (15.12 ug/dl), paint (4.74 ug/dl) and PVC coloring (2.50 ug/dl), and some samples have exceeded biological exposure limit. In conclusion, the depending on their work industry and process, workers have been exposed to the high lead chromate.

Adsorption Characteristics of Cadmium ions from Aqueous Solution using by-product of Brewing (주정오니를 활용한 수중의 카드뮴(Cd) 흡착 특성)

  • Biosorption is considered to be an alternative method to replace the present adsorbent systems for the treatment of metal contaminated wastewater. In this study, by-product which was abandoned from brewing factory was used to remove metal component in aqueous solution. The experimental results showed that the range of the removal efficiency is 60~91% and adsorption equilibrium was reached in about 3 hr. FT-IR and stereo microscope has been used to observe the surface conditions and changes in functional groups by calcination. At the end of elution, the amount of nitrogen and phosphorus in water was increased 11 and 7 times compare raw sample to calcinated samples. The Langmuir isotherm adequately described the adsorption of waste materials and the maximum adsorption capacity was 28.17 mg/g for Cd. The overall results suggested that waste material might can be used for biosorption of Cd.

Applicability test of broad leaf samples stored at the NESB for biomonitoring of airborne pollutants (국가환경시료은행 활엽 시료를 활용한 지역별 대기침적 오염물질 모니터링 활용성 검토)

  • The National Environmental Specimen Bank (NESB) has been collecting broad leave samples to monitor environmental pollution from five different designated sampling areas. In order to ensure the reproducibility and comparability of the results, all the procedures from selecting trees and pooling leaves to make the representative sample are defined in the standard operation procedures(the SOP). The representative samples were subjected to the chemical analyses for some heavy minerals and Polycyclic Aromatic Hydrocarbons(PAHs). The uncertainty levels involved in each step of the SOP, that is, the sampling and the chemical analysis, were derived using the Robust ANOVA, which enables the relative comparison among the different levels of pollutants concentrations with confidence. Furthermore, the effect of the varying degrees of precipitation on the pollutants concentration of the leaves was also examined. Overall, the biological difference estimated from the duplicate samples was found to exceed the variation across the site, implying even aerial deposition over site. Samples from Gwanak Mt. showed highest heavy metal concentrations than the other sites. Washing off effect of the pollutants adhering in the form of particles on the leaf surface was found to be affected by the cumulative precipitation.

  • The precise measurement of human emotion is of pivotal importance in the field of emotion science. Based on the perspective of psychoneuroimmunoendocrinology, human emotion is now considered to be measurable by body fluid. The biological molecule cortisol, which is known for the stress hormone, has been widely investigated to help researchers to estimate the stress loaded on human body. Until now, several measurement techniques such as High Performance Liquid Chromatography (HPLC), fluorometric assay, and reverse phase chromatography have been developed. However, since these measurements are expensive, take relatively long time for an operation, and they are not portable, they are not appropriate for POCT (point of care testing). In this paper we demonstrate the performance of a miniaturized-microwave resonant device in the measurement of cortisol. Our method has many advantages in that it requires a small volume of sample, has fast response time, is easy to operate and needs no labeling process. Besides, it will shed a light on the measurement techniques for emotion science.

Evaluation of Enzymatic Method using an Automated Chemistry Analyzers for Homocysteine Measurement

  • In clinical practice, homocysteine has gained popularity because its elevated values are strongly associated with an increased risk of cardiovascular disease. More recently, a new enzymatic colorimetric assay for homocysteine in biological sample, suitable for automated clinical analyzers, has been proposed. To evaluate one of these enzymatic methods and compare the results obtained with this method with those of an immunoenzymatic method, thirty-two samples were analyzed for total homocysteine by HiSens$^{(R)}$ homocysteine reagent on the automated chemistry analyzers TBA 200FR and compared to the widely used immunoenzymatic method ADVIA Centaur. In TBA 200FR, the within-run CVs of two control materials were 3.23% and 0.92%, respectively; the between run CVs were 4.58% and 2.55%, respectively. And in ADVIA 1650, the within-run CVs were 6.81% and 0.99%, respectively; the between run CVs were 9.0% and 3.9%, respectively. The recovery for homocysteine was 100% ($60.8{\mu}mol/L$), 99.1% ($48.64{\mu}mol/L$), 96.3% ($36.48{\mu}mol/L$), 96.1% ($24.32{\mu}mol/L$), and 92.1% ($12.16{\mu}mol/L$). The regression equation of TBA 200FR vs. ADVIA Centaur was y=0.9095x-2.5086 (r=0.9632). And the regression equation for the ADVIA 1650 chemistry vs. Immulite 2000 was y=0.8418x + 0.3207 (r=0.9625). In conclusion, this enzymatic method using automated chemistry analyzer for homocysteine assay shows acceptable analytical performance. I suggest that this assay will be suitable for routine analysis.

Study on the Bio-chemical Safety of Ganjang Gejang Distributed in Korea (국내 유통 간장게장 제품 안전성 조사 연구)

  • To evaluate biochemical safety of gejang distributed in Korea, 33 samples were collected and analyzed for indicator microorganisms, pathogen, parasite, pH, volatile basic nitrogen(VBN), salinity, 3-MCPD, histamine, and synthetic food colors. Total plate counts of gejang were the range of 3 to 7 log cfu/g and coliform were the range of ND to 4 log cfu/g. Vibrio parahaemolyticus was detected from 4 samples, and Escherichia coli(1 log cfu/g) and copepoda were detected from 1 sample among those. pH and VBN value of samples were the range of 6.69 to 8.10 and 21.99 to 94.55 mg%, respectively. The ranges of salinity concentration for ganjang chamgejang, ganjang ggokgejang, and ganjang dolgejang were $11.6{\sim}32.6%,\;2.8{\sim}20.5%$, and $11.6{\sim}13.3%$, respectively. But, the range of salinity concentration of yangnum gejang was $2.6{\sim}9.4%$, which was lower than ganjang gejang. Besides, 3-MCPD, histamine and synthetic food colors were not detected. From the results, most of gejang was appropriate for Korea national standard for food regulation, but still biological hygiene control for raw materials, processing, and distribution should be improved.

Development of Immunochromatography for the Rapid Detection of Listeria monocytogenes (Listeria monocytogenes 신속 검출을 위한 면역크로마토그래피법의 개발)

  • The objective of this study was the development of immunochromatography (ICG) for the rapid and accurate detection of Listeria monocytogenes. Here, monoclonal antibodies (MAb) were conjugated with 40 nm colloidal gold particles, where the conjugate was used as the detection reagent in the ICG. The ICG was composed of three pads (sample, conjugate, and absorbance pads) and one nitrocellulose membrane. The colloidal gold-MAb conjugate was applied to the conjugate pad, and the test line and control line on the membrane were treated with MAb (FKLM-3BI2-37) and anti-mouse IgG, respectively. The detection limit of the ICG was $10^{5}$ cell/mL and it showed no cross-reaction to food borne pathogens. We inoculated meat and lettuce samples with various counts of L. monocytogenes, and analyzed them by ICG. All the inoculated meat samples gave positive results after enrichment for 24 h in LEB. These results indicate that ICG was able to serve as a primary screening tool for L. monocytogenes in various foods and agricultural products within 20 min after enrichment.
