Association of lnc-LAMC2-1:1 rs2147578 and CASC8 rs10505477 Polymorphisms with Risk of Childhood Acute Lymphoblastic Leukemia

  • Long non-coding RNAs (lncRNAs) are a novel class of non-protein coding RNAs that are involved in a wide variety of biological processes. There are limited data regarding the impact of lnc-LAMC2-1:1 rs2147578 as well as CASC8 rs10505477 T>C polymorphisms on cancer development. Here we examined for the first time whether rs2147578 and rs10505477 polymorphisms are associated with childhood acute lymphoblastic leukemia (ALL) in a total of 110 cases and 120 healthy controls. Genotyping was achieved by the polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method. The rs2147578 variant increased the risk of ALL in codominant (OR=4.33, 95%CI=2.00-9.37, p<0.0001, CG vs CC, and OR=5.81, 95%CI=2.30-14.69, p=0.0002, GG vs CC), dominant (OR=4.63, 95%CI=2.18-9.86, p<0.0001, CG+GG vs CC), overdominant (OR=1.74, 95%CI=1.02-2.97, p=0.0444, CG vs CC+GG) and allele (OR=1.91, 95%CI=1.32-2.77, p=0.0008, G vs C) inheritance models tested. No significant association was found between the CASC8 rs10505477 T>C variant and risk of childhood ALL. In conclusion, the present study revealed that the lnc-LAMC2-1:1 rs2147578 polymorphism may be a risk factor for developing childhood ALL. Further studies with larger sample sizes with different ethnicities are now required to confirm our findings.

A Non-invasive Real-time Respiratory Organ Motion Tracking System for Image Guided Radio-Therapy (IGRT를 위한 비침습적인 호흡에 의한 장기 움직임 실시간 추적시스템)

  • A non-invasive respiratory gated radiotherapy system like those based on external anatomic motion gives better comfortableness to patients than invasive system on treatment. However, higher correlation between the external and internal anatomic motion is required to increase the effectiveness of non-invasive respiratory gated radiotherapy. Both of invasive and non-invasive methods need to track the internal anatomy with the higher precision and rapid response. Especially, the non-invasive method has more difficulty to track the target position successively because of using only image processing. So we developed the system to track the motion for a non-invasive respiratory gated system to accurately find the dynamic position of internal structures such as the diaphragm and tumor. The respiratory organ motion tracking apparatus consists of an image capture board, a fluoroscopy system and a processing computer. After the image board grabs the motion of internal anatomy through the fluoroscopy system, the computer acquires the organ motion tracking data by image processing without any additional physical markers. The patients breathe freely without any forced breath control and coaching, when this experiment was performed. The developed pattern-recognition software could extract the target motion signal in real-time from the acquired fluoroscopic images. The range of mean deviations between the real and acquired target positions was measured for some sample structures in an anatomical model phantom. The mean and max deviation between the real and acquired positions were less than 1mm and 2mm respectively with the standardized movement using a moving stage and an anatomical model phantom. Under the real human body, the mean and maximum distance of the peak to trough was measured 23.5mm and 55.1mm respectively for 13 patients' diaphragm motion. The acquired respiration profile showed that human expiration period was longer than the inspiration period. The above results could be applied to respiratory-gated radiotherapy.

A Study on Cost-effective Treatment of Wastewater and Odor Reduction for Southeast Asian Market Entry

  • Purpose - The purpose of this study is to apply a cost effective ultrasonic odor reduction method that generated micro-bubbles using ejector to the Southeast Asian wastewater market. Research design, data, and methodology - A leather maker located in Ansan-city, Gyunggi-do, South Korea was sampled from the collection tank to select experimental materials. Experimental setup consisted of circulating water tank-air ejector-ultrasonic device, and circulating wastewater. Sample analysis was performed by CODcr, T-N, T-P, and turbidity by the National Environmental Science Institute. Results - Experimental results show that it is most effective in removing odors when the frequency range of ultrasonic wave is 60~80 Khz and the output is 200 W. It showed that the concentration of complex odor dropped from a maximum of 14,422 times to a minimum of 120 times. Also, analysis of ammonia and hydrogen sulfide in specific odor substances has shown that they were reduced from 1.5 ppm to 0.4 ppm and from 0.6 ppm to 0.1 ppm, respectively. Conclusions - It is possible to shorten more than 12 hours in the treatment of micro-organisms. It can be seen that the processing time of odor after ultrasonic treatment in the pre-treatment facility is reduced by 25% when compared to the resultant micro-organisms after the chemical treatment, that is, the time of the bio-treatment of micro-organisms. Based on the results, it was confirmed that the pre-treatment method using the ultrasonic and the air ejector device of the experiment shows the effect of reducing the water pollutants and odor more effectively in a relatively short time than the conventional advanced oxidation method.

Anti-proliferative and Antioxidant Activities of 1-methoxy-3-methyl-8-hydroxy-anthraquinone, a Hydroxyanthraquinoid Extrolite Produced by Amycolatopsis thermoflava strain SFMA-103

  • Actinobacteria are prolific producers of a large number of natural products with diverse biological activities. In the present study, an actinobacterium isolated from sunflower rhizosphere soil sample collected from Medak, Andhra Pradesh, South India was identified as Amycolatopsis thermoflava strain SFMA-103. A pigmented secondary metabolite in culture broth was extracted by using methanol and it was further purified by silica gel column chromatography with methanol-chloroform solvent system. Structural elucidation studies based on UV-visible, 1D and 2D-NMR, FT-IR, and mass spectroscopic analyses confirmed the structure as 1-methoxy-3-methyl-8-hydroxy-anthraquinone. It showed significant in vitro anticancer activity against lung cancer and lymphoblastic leukemia cells with $IC_{50}$ values of 10.3 and $16.98{\mu}M$, respectively. In addition, 1-methoxy-3-methyl-8-hydroxy-anthraquinone showed good free radical scavenging activity by DPPH method with an $EC_{50}$ of $18.2{\mu}g/ml$. It also showed other promising superoxide radical scavenging, nitric oxide radical scavenging and inhibition of lipid peroxidation activities. This is a first report of anti-proliferative and antioxidant activities of 1-methoxy-3-methyl-8-hydroxy-anthraquinone isolated from A. thermoflava strain SFMA-103 which may find potential application in biotechnological and pharmaceutical fields.

Search Space Reduction Techniques in Small Molecular Docking (소분자 도킹에서 탐색공간의 축소 방법)

  • Since it is of great importance to know how a ligand binds to a receptor, there have been a lot of efforts to improve the quality of prediction of docking poses. Earlier efforts were focused on improving search algorithm and scoring function in a docking program resulting in a partial improvement with a lot of variations. Although these are basically very important and essential, more tangible improvements came from the reduction of search space. In a normal docking study, the approximate active site is assumed to be known. After defining active site, scoring functions and search algorithms are used to locate the expected binding pose within this search space. A good search algorithm will sample wisely toward the correct binding pose. By careful study of receptor structure, it was possible to prioritize sub-space in the active site using "receptor-based pharmacophores" or "hot spots". In a sense, these techniques reduce the search space from the beginning. Further improvements were made when the bound ligand structure is available, i.e., the searching could be directed by molecular similarity using ligand information. This could be very helpful to increase the accuracy of binding pose. In addition, if the biological activity data is available, docking program could be improved to the level of being useful in affinity prediction for a series of congeneric ligands. Since the number of co-crystal structures is increasing in protein databank, "Ligand-Guided Docking" to reduce the search space would be more important to improve the accuracy of docking pose prediction and the efficiency of virtual screening. Further improvements in this area would be useful to produce more reliable docking programs.

A Study on the Serum Lipid, Digestive Enzymes and Thyroid Hormone in Sasang Constitutional Medicine (사상체질별 혈중지질, 소화효소 및 갑상선 호르몬의 비교 연구)

  • 1. Objectives: The purpose of this study was to analyze the serum lipid, digestive enzymes, thyroid hormone of each Sasang type groups which were classified by Sasang medicine specialist. 2. Methode: The subjects were 1084 (male:female=399:685) person with 371 of Taeeum type, 324 of Soeum type, 389 of Soyang type after the Sasang type classification. The blood samples of the 1084 subjects were to analyze the serum lipid(triglyceride, total cholesterol, HDL-cholesterol, LDL-cholesterol), digestive enzymes(lipase, amylase), thyroid hormone(T3, TSH). The general linear model (SPSS 12.0) were used for the analysis. 3. Results: Taeeum type group ($133.50\pm89.84$, $189.41\pm33.11$, $107.29\pm28.66$) showed significantly higher triglyceride, total cholesterol, LDL-cholesterol than other two group. In digestive enzymes(lipase, amylase), we found significant differences in the order of Soeum>Soyang>Taeeum. Soeum type group($116.23\pm21.38$) has lower T3 than Soyang type group ($119.90\pm21.60$) with marginal significance (p=0.066). 4. Conclusions: The Results of present study showed that Taeeum type showed significantly highest in triglyceride, total cholesterol, LDL-cholesterol. Soeum type showed significantly highest in digestive enzymes. Blood sample of each Sasang type group will serve as an important evidence for further study showing that the Sasang typology has biological basis.

Molecular biological characterization of transmissible gastroenteritis viruses isolated in Korea (돼지 전염성 위장염 바이러스(국내분리주)의 분자생물학적 특성 규명)

  • Sixteen Korean field transmissible gastroenteritis viruses (TGEVs) were isolated using swine testicular cell (STC) and the genomic diversity of them was analyzed. All TGEV isolates produced a typical cytopathic effect in STC and were confirmed as TGEV by immunofluorescence assay using monoclonal antibody against TGEV and PCR using TGEV specific primers. RNAs from TGEV field isolates and vaccine TGEV were extracted and amplified by RT and PCR. The RT-PCR products were digested with selected restriction enzymes and analyzed RFLP patterns. The N-terminal end region of S gene and ORF 3 and 3-1 genes of TGEV amplified by TGEV specific primer pairs seemed to be conserved. Most specific variations were detected in S gene amplified by TGEV 4/6 primer pairs which includes antigenic sites A and D. When the PCR products were treated with Sau3AI and Ssp I, Bvac(vaccine strain), field isolates 133 and 347 were differentiated from Miller and Purdue types. In the case of D5 field isolates, it was classified into Purdue type by Sau 3AI but classified into independent TGEV by Ssp I. Two different TGEV strains from D2 sample were confirmed by plaque purification and RT-PCR-RFLP analysis. To investigate the change occurring in TGEV genome after serial passage, the TGEV P44 strain was passaged through STC. There were specific changes in S gene and a large deletion was observed in ORF 3 and 3-1 genes. These studies showed that a distinct difference in genome exists among TGEV field isolates.

Changes of Biological Activities of Rhizome and Leaves of Polygonatum sibiricum Redoute according to Steaming Time and Temperature (층층갈고리둥굴레 (Polygonatum sibiricum Redoute) 뿌리줄기와 잎의 증숙 시간과 증숙 온도에 따른 생리활성 변이 연구)

  • Background: The aim of this study was to analyze the total phenol and total flavonoid contents and antioxidant activity of steam-treated leaves and rhizomes of Polygonatum sibiricum Redoute. In addition, we aimed to confirm their potential use as cosmetic materials by investigating their anti-aging and skin-whitening activity. Methods and Results: The leaves and rhizomes of P. sibiricum were treated with steam at different temperatures for different durations, and the antioxidant activity (DPPH and ABTS radical scavenging activity) and total phenol and total flavonoid contents of each sample were tested. The steam temperature and treatment duration siginificantly affected the antioxidant activity and, total phenol and total flavonoid content of the leaves and rhizome of P. sibiricum. Treating the P. sibiricum samples with steam at 120℃ for 12 h, yielded higher total phenol and total flavonoid contents. Comparatively, the samples treated with steam at 120℃ for 12 to 24 h showed significantly higher antioxidant activity. Further, the steamed samples of P. sibiricum demonstrated collagenase and tyrosinase inhibition activity, which indicated their anti-aging and skin-whitening properties. The samples steamed at 120℃ for 12 h, exhibited higher collagenase and tyrosinase inhibition activity. Conclusions: Leaves and rhizomes of P. sibiricum steamed at 120℃ for 12 h, showed highest antioxidant activity and, total phenol and total flavonoid contents than all other samples. Our results indicate the potential of using P. sibiricum as a cosmetic material by confirming its excellent anti-aging and whitening activity.

Temporary Persistance of Pesticide Residue of Diazinon, Iprodione and Chlorfenapyr during the Cultivation Periods in Shallot (Allium ascalonicum L.) (쪽파(Allium ascalonicum L.) 재배기간 중 Diazinon, Iprodione 및 Chlorfenapyr의 잔류량 변화)

  • In order to know the residual pattern of pesticides and predict to the degradation period until below MRL, we experimented diazinon, iprodinon and chlorfenapyr for shallot. They were the most detected pesticide in shallot by NAQS (National Agricultural product Quality management Service) survey. In this experiment, we sprayed those pesticides 15days before harvest and analyzed 0, 1, 2, 3, 5, 7, 10, 14 day sample to establish logical equation and to calculate $DT_{50}$. During the cultivating period, the residue amount of diazinon was changed from 1.02 mg/kg (0 day) to 0.01 mg/kg (14 day), $DT_{50}$ was 2.19 days, and iprodione was changed from 1.45 mg/kg (0 day) to 0.14 mg/kg (14 day), $DT_{50}$ was 4.15 days, and chlorfenapyr was changed from 1.5 mg/kg (0 day) to 0.01 mg/kg (14 day), $DT_{50}$ was 1.97 days. The $DT_{50}$ of double amount in those pesticides, diazinon was 2.24 days, iprodione was 4.82 days, and chlorfenapyr was 2.24 days, respectively.

Extraction of Genistein and Formononetin from Sophoraflavescens Aiton using Ultrasonic wave (초음파를 이용한 고삼에 포함된 Genistein 및 Formononetin의 추출)

  • In this work, we the ettect on extraction amounts and general composition content of phytoestrogen genistein and formononetin extracted from Sophoraflavescens Aiton by various ultrasonic waves(35, 72, and 170 KHz) and extraction time(30, and 60 min) were compared using extraction solvent water 100%. The pretreatment step was composed of ultrasonic waves extraction, filtration, concentration, and membrane filtration. The extracted sample was analyzed by reversed-phase high performance liquid chromatography(RP-HPLC). And the mobile phase applied was linearly changed with A/B of 80/20~65/35 vol% for 60 min(A water/acetic acid, 99.9/0.1 vol%, B acetonitrile/acetic acid, 99.9/0.1 vol%). The experimental results, general composition carbohydrate(0.255 to 0.413%) excepts, other ingredients was confirmed almost similarly. Also, The highest yield of extraction amount 3.17g was obtained by ultrasonic waves with a frequency of 170 KHz and an extraction time of 60 min. This work offers would be useful for chemical and biological studies of natural plants and its products.
