Prediction of Parathyroid Hormone Signalling Potency Using SVMs

  • Parathyroid hormone is the most important endocrine regulator of calcium concentration. Its N-terminal fragment (1-34) has sufficient activity for biological function. Recently, site-directed mutagenesis studies demonstrated that substitutions at several positions within shorter analogues (1-14) can enhance the bioactivity to greater than that of PTH (1-34). However, designing the optimal sequence combination is not simple due to complex combinatorial problems. In this study, support vector machines were introduced to predict the biological activity of modified PTH (1-14) analogues using mono-substituted experimental data and to analyze the key physicochemical properties at each position that correlated with bioactivity. This systematic approach can reduce the time and effort needed to obtain desirable molecules by bench experiments and provide useful information in the design of simpler activating molecules.

  • A PC based software, the RTP Research Tool Box (RTB), was developed for IMRT optimization research. The software was consisted of an image module, a beam registration module, a dose calculation module, a dose optimization module and a dose display module. The modules and the Graphical User Interface (GUI) were designed to easily amendable by negotiating the speed of performing tasks. Each module can be easily replaced to new functions for research purpose. IDL 5.5 (RSI, USA) language was used for this software. Five major modules enable one to perform the research on the dose calculation, on the dose optimization and on the objective function. The comparison of three cost functions, such as the uncomplicated tumor control probability (UTCP), the physical objective function and the pseudo-biological objective function, which was designed in this study, were performed with the RTB. The optimizations were compared to the simulated annealing and the gradient search optimization technique for all of the optimization objective functions. No significant differences were found among the objective functions with the dose gradient search technique. But the DVH analysis showed that the pseudo-biological objective function is superior to the physical objective function when with the simulated annealing for the optimization.

Z-DNA-Containing Long Terminal Repeats of Human Endogenous Retrovirus Families Provide Alternative Promoters for Human Functional Genes

  • Transposable elements (TEs) account for approximately 45% of the human genome. TEs have proliferated randomly and integrated into functional genes during hominoid radiation. They appear as right-handed B-DNA double helices and slightly elongated left-handed Z-DNAs. Human endogenous retrovirus (HERV) families are widely distributed in human chromosomes at a ratio of 8%. They contain a 5'-long terminal repeat (LTR)-gag-pol-env-3'-LTR structure. LTRs contain the U3 enhancer and promoter region, transcribed R region, and U5 region. LTRs can influence host gene expression by acting as regulatory elements. In this review, we describe the alternative promoters derived from LTR elements that overlap Z-DNA by comparing Z-hunt and DeepZ data for human functional genes. We also present evidence showing the regulatory activity of LTR elements containing Z-DNA in GSDML. Taken together, the regulatory activity of LTR elements with Z-DNA allows us to understand gene function in relation to various human diseases.

Establishment of Baculovirus Infected Insect Cell Line Expressing Porcine Salivary Lipocalin(SAL1) Protein

  • Salivary lipocalin (SAL1) is a member of the lipocalin protein family that has a property to associate with many lipophilic molecules. The importance of SAL1 during pregnancy in pigs has been suggested by our previous study which has shown that SAL1 is expressed in the uterine endometrium in a cell type- and implantation stage-specific manner and secreted into the uterine lumen. However, function of SAL1 in the uterus during pregnancy in pigs is not known. To understand SAL1 function in the uterus during pregnancy, we generated recombinant porcine SAL1 protein in an insect cell line. Porcine SAL1 cDNA was cloned into a baculovirus expression vector using RT-PCR and total RNA from uterine endometrium on day 12 of pregnancy, and the expression vector was used to generate recombinant Bacmid containing the SAL1 gene. The recombinant Bacmid was then transfected Sf9 cell to produce recombinant baculovirus. By infecting Sf9 cell with recombinant baculovirus, we established a SAL1-expressing insect cell expression system. Immunoblot analysis confirmed SAL1 expression in the infected cells. Recombinant SAL1 produced by the Sf9 cell line will be useful for understanding physiological function of SAL1 during pregnancy in pigs.

  • Phospholipids function as major components of biological membranes as well as precursors of biologically active lipid messengers. It is well known that arachidonic acid attached at the sn-2 position of phosphoglycerides serves as a precursor of prostaglandins and leukotrienes. Recently, it has been recognized that lysophospholipids such as lysophosphatidic acid, sphingosine 1-phosphate, lysophosphatidylserine and monoglyceride also function as lipid messengers with a variety of biological activities.   • The purpose of this study was to analyze the concept of physical activity. The method was based on the steps of concept analysis by Walker and Avant(1988). The result of this study were as follows; Upon the concept analysis, physical activity is defined as a series of bodily movements which is performed voluntarily by individual. The defining attributes of physical activity were a series of bodily movements, energy expenditure, goal-directedness, and self-control of the body. The antecedents of physical activity were neuromusculoskeletal function, cardiopulmonary function and cognitive function. And the consequences of physical activity were improvement of physical function, induction of fatigue, and need(physiopsychosocial) satisfaction. Further studies are needed to develop the concept of physical activity using the Hybrid model. In addition, considering personal characteristics it is important to study the facilitating factors of physical activity.

Analysis of Obesity Intervention Programs in Adolescents: Focused on Endothelium Functions (비만 청소년의 비만 중재 프로그램에 대한 국내외 연구 분석: 혈관내피 기능을 중심으로)

  • Purpose: The purpose of this study was to determine the current status of obesity intervention programs, and its effects on endothelium function in adolescents. Method: This was a descriptive research study, that investigated domestic and international literature 2009-2018. Using databases inside and outside of Korea to search for 'adolescent', 'obese adolescent', 'exercise', 'program', or 'intervention', and 'endothelium function', a total of 14 literature have been selected for 6 domestic research and 8 international research, excluding overlapping, case studies, literature reviews, and unrelated studies. Results: Interventions for enhancing endothelium function were exercise program, diet, vitamin D3 treatment, and Orlistat intake relative to international research, and combined exercise and purple sweet potato intake, in most domestic research. Dependent variables included FMD, RHI, NMD, EPCs, and EMPs, for measuring endothelium function in international research, and PWV, FMD and RHI, in domestic research. Conclusion: Results of this study confirmed that application of obese intervention programs, may improve endothelium function in adolescents. Further studies are required, to develop nursing intervention, that would enhance endothelium function in adolescents.
