Rhodamine Based Fluorescent Chemosensors for Hg2+ and its Biological Application

  • Two new chemosensors, rhodamine 6G derivative bearing hydroxyethyl group (1) and rhodamine base derivative bearing 15-crown-5 group (2) were synthesized and their sensing behaviors toward various metal ions were investigated by UV/Vis and fluorescence spectroscopies. Addition of $Hg^{2+}$ ion to a $CH_3CN$ solution of 1 and 2 gave visual color changes as well as fluorescent OFF-ON observations. Selectivity and sensitivity of 1 towards $Hg^{2+}$ are excellent enough to detect micromolar level of $Hg^{2+}$ ion, even in equeous media and biological sample (HeLa cell).

In Situ X-ray Absorption Spectroscopic Study for α-MoO3 Electrode upon Discharge/Charge Reaction in Lithium Secondary Batteries

  • In-situ X-ray absorption spectroscopy (XAS) was used to elucidate the structural variation of $\alpha-MoO_3$ electrode upon discharge/charge reaction in a lithium ion battery. According to the XAS analysis, hexavalent Mo atoms in $\alpha-MoO_3$ framework are reduced as the amount of intercalated lithium ions increases. As lithium de-intercalation proceeds, most of pre-edge peaks are restored again. However, according to the Fourier transforms of the extended X-ray absorption fine structure (EXAFS) spectra, lithium de-intercalation reaction is partially irreversible upon the charge reaction, which is one of the main reasons why the capacity of $\alpha-MoO_3$ electrode decreases upon successive discharge/charge cycles.

  • High shear adhesion on wet and rough surfaces and tactile feedback of gripping forces are highly important for realizing robotic gripper systems. Here, we propose a bioinspired robotic gripper with highly shear adhesion and sensitive pressure sensor for tactile feedback systems. To achieve them, we fabricated multi-walled carbon nanotube sensing layer on a thin polymeric adhesive layer of polydimethylsiloxane. With densely hexagonal-packed microstructures, the pressure sensor achieved 9 times the sensing property of a sensor without microstructures. We then assembled hexagonal microstructures inspired by the toe pads of a tree frog, giving strong shear adhesion under both dry and wet surfaces such as silicon (42 kPa for dry and ~30 kPa for underwater conditions) without chemical-residues after detachment. Our robotic gripper can prevent damage to weak or smooth surfaces that can be damaged at low pressure through pressure signal feedback suggesting a variety of robotic applications.

  • In this study, as a part of the study to alleviate the durability degradation due to low tensile performance and cracking of concrete, two kinds of catechol functional chitosan (Cat-Chit) were replaced at a ratio of 5, 10, 15, 20% as a mixed water substitute to evaluate the fluidility and strength characteristics of mortar.

  • Abalone shell is a high strength and light weighted ceramic composite material, which is composed of $CaCO_3$ platelet and protein. Microstructure of abalone shell has a matrix structure that is similar to the bricks and mortar. The technology inspired from nature which consumes low energy at low temperature is called bioinspired technology. In this study, to make high strength and light weighted ceramic composite materials using bioinspired technology, porous green body was prepared with $Al_2O_3$ platelet. PMMA was infiltrated into the porous green body, then warm pressed to eliminate pores present in the composite. The microstructure of the composite was observed with FESEM, and the mechanical/thermal properties were measured.

Plasminogen Activator Inhibitor-1 Antisense Oligodeoxynucleotides Abrogate Mesangial Fibronectin Accumulation

  • Excessive extracellular matrix (ECM) accumulation is the main feature of chronic renal disease including diabetic nephropathy. Plasminogen activator inhibitor (PAI)-1 is known to play an important role in renal ECM accumulation in part through suppression of plasmin generation and matrix metalloproteinase (MMP) activation. The present study examined the effect of PAI-1 antisense oligodeoxynucleotide (ODN) on fibronectin upregulation and plasmin/MMP suppression in primary mesangial cells cultured under high glucose (HG) or transforming growth factor (TGF)-${\beta}1$, major mediators of diabetic renal ECM accumulation. Growth arrested and synchronized rat primary mesangial cells were transfected with $1\;{\mu}M$ phosphorothioate-modified antisense or control mis-match ODN for 24 hours with cationic liposome and then stimulated with 30 mM D-glucose or 2 ng/ml TGF-${\beta}1$. PAl-1 or fibronectin protein was measured by Western blot analysis. Plasmin activity was determined using a synthetic fluorometric plasmin substrate and MMP-2 activity analyzed using zymography. HG and TGF-${\beta}1$ significantly increased PAI-1 and fibronectin protein expression as well as decreased plasmin and MMP-2 activity. Transient transfection of mesangial cells with PAI-1 antisense ODN, but not mis-match ODN, effectively reversed basal as well as HG- and TGF-${\beta}1$-induced suppression of plasmin and MMP-2 activity. Both basal and upregulated fibronectin secretion were also inhibited by PAI-1 antisense ODN. These data confirm that PAI-1 plays an important role in ECM accumulation in diabetic mesangium through suppression of protease activity and suggest that PAI-1 antisense ODN would be an effective therapeutic strategy for prevention of renal fibrosis including diabetic nephropathy.

Bioinspired CuO Hierarchical Nanostructures for Self-cleaning surfaces and SERS substrates

  • Bioinspired hierarchical nanostructures for self-cleaning s-tnwjurface and SERS substrates are investigated. The multi-level hierarchy is combined with CuO nanowire and additional nanoscale structures. CuO nanowire, which has extremely high aspect ratio, serves as a base structure of multi-level hierarchy and additional flower like structures are placed on the CuO nanowires. Since as-fabricated CuO nanostructures are hydrophilic, the surface is coated with perfluorooctyltrichlorosilane in order to change its wetting property to hydrophobic. While those CuO based nanostructures have a sufficient roughness for superhydrophobic characteristics, hierarchical nanoflowers on nanowire structures lead to a self-cleaning surface. Furthermore, flower like nanostructures provide reentrant curvatures, thus enabling oleophobic property. The surfaces has a repellency even for a tiny droplet (10 nL) of low surface tension liquids (~35 mN/m). On the on hands, nanoflowers provide many number of nanoscale gaps. After a thin layer of silver is deposited on the surface of CuO nanostructures, those nanoscale gaps act as hot-spot for surface enhanced Raman scattering (SERS). To analyze SERS enhancement of the surfaces, Raman shift is measured with varying molar density of 4-Mercaptopyridine from mM to pM. From these results, hierarchical CuO nanostructures are suitable for self-maintenance and cost effective SERS sensing applications.
