Bacillus subtilis YB-70 as a Biocontrol Agent of Fusarium solani causing Plant Root-Rot

  • A bacterial strain YB-70 which has powerful biocontrol activity against Fusarium solani causing plant root-rot resulting in considerable losses of many economical crops was isolated and selected from over 500 isolates from a ginseng rhizosphere in suppressive soil, and identified as a strain of Bacillus subtilis. In several biochemical and in vitro antibiosis tests on F. solani with culture filterates from B. subtilis YB-70, our data strongly indicated metabolites which mediated inhibition of the fungal growth were presumed to be heat-stable, micromolecular, and ethyl alcohol solutable antifungal substances. Suppression of root-rot by B. subtilis YB-70 was demonstrated in pot trials with eggplant (Solanum melongena L) seedlings. Treatment of the seedling with the bacterial suspension (1.7~1.9$\times$$10^5$ CFU/g) in F. solani-infested soil significantly reduced disease incidences by 68 to 76% after 25 to 30 days. The results supported that B. subtilis YB-70 have excellent potentials as a biocontrol agent.

  • Sleep is an essential biological process of which the underlying regulatory mechanisms involve numerous anatomical structures and biochemical substances that can be compromised by stress and the immune system. Gamma aminobutyric acid (GABA) is the main inhibitory neurotransmitter of the central nervous system (CNS). It is well established that activation of $GABA_A$ receptors promotes sleep. L. brevis BJ20 fermentation of sea tangle and oysters resulted in stress reduction and sleep inducing effects. This is the first study to report that GABA has the ability to induce sleep related hormones in mice; therefore, it has potential use as a natural sleep aid. These results suggested that sea tangle and oysters fermented by L. brevis BJ20 can be used as potential agents for stress reduction and sleep promotion.

HPLC Analysis of Free Malonaldehyde in Nine Ginseng Polyacetylene-Treated Liver Microsome (인삼의 9종 폴리아세틸렌으로 처리한 간소포체 중의 유리 말론알데히드의 HPLC에 의한 분석)

  • Free malonaldehyde was determined in nine kinds of ginseng polyacetylene-treated micro- some by HPLC analysis. Antioxidant activities of some phenolic compounds and ginseng saponin were also drtermined both by a new HVLC method and by THA method. A new HPLC system separaterl malonaldehyde at a retention time 5,6 min and showed a linear relationship between the peak are a and malonaldehyde concentration. Panaxnol showed the strongest activity among nine polyacetylenes and the addition of either chlorine or aletyl group reduced polyacetylene's own activity. Since C14-polyacetylenes such as panaxyne and panaxyne-epoxide had little or no antioxidant activities, S17-structure should be preserved to exert a radical-scvenging or trapping activity. The antioxidant activities of chlorogenic acid, ferulic acid and catechol were much weaker than those of C17-polyacetylenes. Ginseng saponin showed no antioxidant activity. Since TBA reactive substances and malonaldehyde contents were almost the same in peroxiedized microsome. TBA value seems a good indicator for lipid peroxidation in this particular Fe+3 ADP/NADPH system.

Antibacterial Activities of Lactobacillus crispatus ATCC 33820 and Lactobacillus gassed ATCC 33323

  • Lactobacillus crispatus ATCC 33820 and L. gasseri ATCC 33323 were grown in MRS broth (pH 6.5) at 37$^{\circ}C$ for 24 h and the antibacterial activities of cell free culture supernatants were determined by the agar well diffusion method. The culture supernatants were inhibitory to Escherichia coli, Klebsiella pneumoniae, Pseudomonas aeruginosa, Bacillus subtilis, Staphylococcus aureus, Enterococcus faecalis, Pediococcus acidilacticii, and Lactobacillus helveticus. The supernatants did not show any lysozyme activity. Addition of catalase did not affect the antibacterial activities of the supernatants. The antibacterial substances were heat stable (100$^{\circ}C$ for 60 min) and sensitive to proteases.

  • The Malaysian biogas upgrading technologies and policies were examined. In Malaysia, the regulation of palm oil mill effluent (POME) has been enforced to reduce the biochemical oxygen demand to 20 ppm and the biogas capture in the palm oil mills have been recently enforced for renewable energy. A huge amount of organic waste is produced from POME, and 80 million tons from palm oil trees, every year. Due to the renewable energy trends, the Malaysian government is modifying the use of biogases as fuels in favor of their conversion into compressed natural gas (CNG) and other chemicals; various green policies are being promoted because of many advantages of the organic substances. The Korean policies for biogas are a good model for exporting environmental plants after upgrading the digestion and purification technologies. Therefore, this article introduces the current status of POME and biogas production in Malaysia, it could encourage creating a new market for biomethane.

In Vivo Effects of Crataegus pinnatifida Extract for Healthy Longevity

  • Aging is a complex series of multi-organ processes that occur in various organisms. As such, an in vivo study using an animal model of aging is necessary to define its exact mechanisms and identify anti-aging substances. Using Drosophila as an in vivo model system, we identified Crataegus pinnatifida extract (CPE) as a novel anti-aging substance. Regardless of sex, Drosophila treated with CPE showed a significantly increased lifespan compared to those without CPE. In this study, we also evaluated the involvement of CPE in aging-related biochemical pathways, including TOR, stem cell generation, and antioxidative effects, and found that the representative genes of each pathway were induced by CPE administration. CPE administration did not result in significant differences in fecundity, locomotion, feeding amount, or TAG level. These conclusions suggest that CPE is a good candidate as an anti-aging food substance capable of promoting a healthy lifespan.

  • Antibiotics are substances produced by microorganisms that kill or inhibit and are essential for infectious diseases management. This study aimed to provide basic data for overcoming antibiotic resistance in the marine bacterium LJ-18. The API 20NE and API 50CH kits were used to identify this microorganism. Morphological, physiological, and biochemical properties were investigated using MacFaddin's manuals. Subsequently, isolated LJ-18 was found to belong to a genus of Streptomyces that forms mycelia. LJ-18 also grew well at 28-32℃ on modified Bennett's agar. To isolate and purify the antibacterial compound, LJ-18 culture was divided into ethyl acetate and distilled water fractions. Considerable antimicrobial activity against various pathogenic microorganisms, including methicillin-resistant Staphylococcus aureus (MRSA), was confirmed in the C18 ODS open column fractions. Peak 2 compound was obtained using reversed-phase HPLC. As a result, this compound had a significant antimicrobial activity against various pathogenic microorganisms. In particular, it showed strong activity against MRSA, Mycobacterium smegmatis, Bacillus subtilis, Bacillus cereus, and Staphylococcus aureus.

  • Monoterpenes, phenolic substances, growth substances and total nitrogen from normal needles and infested needles attacked by Thecodiplosis japonensis were compared for Pinus densiflora, Pinus thunbergii and Pinus rigida. The results obtained in this study were as follows: 1. Major monoterpene components in P. densiflora needles were ${\beta}$-phellandrene, ${\alpha}$-pinene and those in P. rigida needles were ${\beta}$-pinene and ${\alpha}$-pinene. In P. rigida resistant to this insect, infested needles showed higher ${\alpha}$-pinene, ${\beta}$-pinene and myrcene but lower camphene, limonene and ${\beta}$-phellandrene than normal needles. 2. Orcinol, catechol ferulic acid, salicylic acid and five unknowns were detected in P. rigida needles whereas orcinol, catechol, ferulic acid and four unknowns in P. densiflora needles. 3. Tryptophan, a precursor of IAA, was detected in larvae and also in gall tissues of both P. densiflora and P. thunbergii needles. This fact shows that growth substances may involve in gall formation by Thecodiplosis japonensis. 4. Total nitrogen contents per unit needle weight were neither significantly different between P. densiflora and P. rigida, nor between normal and infested needles.

Isolation of Antifungal Substances by Bacillus amyloliquefaciens IUB158-03 and Antagonistic Activity against Pathogenic Fungi (Bacillus amyloliquefaciens IUB158-03이 생산하는 항진균물질의 분리와 항균활성)

  • For the control of pathogenic microorganisms, Bacillus spp. were isolated from diseased pepper fruits in Korea. Among them, Bacillus sp. IUB158-03 showed high inhibitory effect on mycelial growth and spore germination of C. gloeosporioides and Botrytis cinerea. The strain was identified as B. amyloliquefaciens IUB158-03 based on its physiological, biochemical characteristics and Microlog analysis. The highest level of antifungal substances by B. amyloliquefaciens IUB158-03 were obtained when the bacterium was cultured in medium containing 2% soluble starch, 3% yeast extract, 0.5% tryptone, 0.5% $NH_4H_2PO_4$, and 1% NaCl (pH 6.0) at $25^{\circ}C$ for 72 hrs. The antifungal substances were purified by butanol extraction, silica gel column chromatography, preparative thin layer chromatography, and high performance liquid chromatography. The purified antifungal substance was confirmed $R_f$ 0.27 by TLC. This substance exhibited antifungal activity against Fusarium solani, Rhizoctonia solani, Botrytis cineria, Alternata alternaria of plant pathogenic fungi and Trichophyton mentagrophytes, Epidermophyton floccosum, Cryptococcus neoformans of human pathogenic fungi.
