오리를 이용한 한방식품 추출액이 흰쥐의 지질함량에 미치는 영향 (Effect of Duck Extract on Lipids in Rats)

  • The aim of the present study was to investigate the effect of duck extract on lipids. Rats in the experimental group were orally administered with duck extract for four weeks. The half of the supplementation was composed of duck meat and the other of the supplementation was composed of oriental herbs. Weight gain, diet intake, and food efficiency ratio were compared between control and experimental groups. Organ weight, lengths of small/large intestines, and the following plasma biochemical parameters were also measured: hemoglobin, hemoglobin, GOT, GPT, blood glucose concentration, lipids of plasma, liver, feces. Spleen index (weight/100 g body weight) of the rats in the experimental group was significantly higher than those in the control group due to increase in fat intake from the supplementation of duck meat. The large intestines of the rats in the experimental group was significantly larger due to the supplementation with dietary fiber derived from herbs in the extract. There were significant higher levels of plasma hematocrit, GOT, total cholesterol, and HDL-cholesterol in the rats of the experimental group. From the results, although duck meat is a source of animal food, the fat profile of duck extract, such as the ratio of polyunsaturated fatty acids to saturated fatty acids appears to be beneficial to human health. These findings suggest a possible anti-hyperlipidemic effect of duck extract. (Korean J Nutrition 36(1): 3∼8, 2003)

Occurrence of crown gall of chrysanthemum caused by Agrobacterium tumefaciens.

  • Incidence of crown gall on lower stem of chrysanthemum, Chrysanthemum morifolium Ramat., was first observed at Hwasung, Gyeonggi, Korea in 2001, Tumors on the stem were 1.5-2 cm in size and semi-round with rough surface texture of dark brown color. Four strains of bacteria isolated from the tumor tissues were characterized. Their colonies were convex, glistening, circular with an entire edge, and white to tannish-cream in color on PDA plus CaCO$_3$. They were gram negative, oxidase positive, and growing on DIM agar. The bacterial isolates inducing gall formation in chrysanthemum were identified as Agrobacterium tumefaciens based on biochemical and physiological characteristics, fatty acid profile using Sherlock Microbial Identification System, and substrate utilization patterns using Biolog Identification System. Young chrysanthemum plants inoculated with the bacteria developed typical galls within two to three weeks. Seedlings of tomato and slices of carrot roots also produced typical galls two to three weeks after inoculation. This is the first report on crown gall of chrysanthemum in Korea.

하천(河川)의 수질예측(水質豫測)을 위한 수치모형(數値模型)에 관한 연구(硏究) (-A Study on a Mathematical Model for Water Quality Prediction for Rivers-)

  • The propriety of the numerical model application was examined on Paldang resevoir and its inflow tributaries located in the center of the Korean peninsula and the long term water quality forecast of the oxygen profile was carried out in this syduy. The input data of the model was the capacity of the reservoir, catchment area, percolation, diffusion rate, vertical mixing rate, dissolution rate from the bottom of the reservoir, outflow of the resevoir, water quality measurement and meteorology data of the drainage basin, and the output result was the annual estimation value of the dissolved oxygen concentration and the biochemical oxygen demand. The modeling method is based on the measured or calculated boundary condition dividing the water area into several blocks from the macorscopic aspect and considering the mass balance in these blocks. As the result of the water quality forecast, it was expected that the water quality in Northern Han River and Paldang reservoir would maintain the recent level, but that the water quality in the Southern Han River and its inflow tributary would worsen below the grade 4 of the life environmental standard from around 2000 owing to the decrease of DO concentration and the increase of BOD concentration.

Endocrine Profiles and Blood Chemistry Patterns of Cloned Miniature Pigs in the Post-Puberty Period

  • Although the majority of surviving pigs cloned by somatic cell nuclear transfer (SCNT) appear to be physiologically normal, there is a general lack of detailed hemato-physiologic studies for the period of early adulthood to substantiate this claim. In the present study, we investigated variation in blood chemistry and endocrinological parameters between mesenchymal stem cells (MSCs) derived from cloned and normal age-matched female and male miniature pigs. Cloned females and males showed normal ranges for complete blood count assessments. Biochemical assessments showed that ${\gamma}$-GGT, ALT and cholesterol levels of male and female clones were significantly (P<0.05 or P<0.01, respectively) higher than that of age-matched control miniature pigs. Variations in insulin and IGF-1 were higher in female clones than in male clones and controls. Thus, although female and male cloned miniature pigs may be physiologically similar to normal animals, or at least within normal ranges, a greater degree of physiological and endocrinological variation was found in cloned pigs. The above variation must be taken into account before considering cloned female or male miniature pigs for various biomedical applications.

pH-Controlled Synthesis of Cephalexin by a Purified Acetobacter turbidans Ampicillin Acylase

  • It has been known that, in enzymatic synthesis of cephalexin, the conversion yield was reduced by high loading of ampicillin acylase. In order to elucidate this phenomena, pH-controlled synthesis of cephalexin was examined using a purified Acetobacter turbidans acylase. When the pH of the reaction mixture was maintained at $6.20{\pm}0.04$, the reduction of the maximal conversion rate was not observed even with high enzyme loading. The kinetic parameters also suggest that pH drop during the enzymatic synthesis of cephalexin was mainly attributed to the rapid hydrolysis of D-${\alpha}$-phenylglycine methyl ester to D-${\alpha}$-phenylglycine, rather than the disappearance of 7-amino-3-deacetoxycephalosporanic acid for cephalexin synthesis. At higher molar ratio of two substrates, [D-${\alpha}$-phenylglycine methyl ester]/[7-amino-3-deacetoxycephalosporanic acid], the conversion rate was also elevated under pH-controlled enzymatic synthesis, which implies that the main reason for the pH drop is due to the production of D-${\alpha}$-phenylglycine methyl easter, the effect of a water-methanol cosolvent system on the ester, the effect of a water-methanol cosolvent system on the conversion profile was also examined. Even the though the conversion rate was increased in 10% methanol solution, a higher than 16% methanol in the reaction mixture caused an inactivation of enzyme.

Diclofenac 투여 후 시간경과에 따른 장손상과 장내세균전위의 변화 (The Changes in Intestinal Damage and Bacterial Translocation with Time after Administration of Diclofenac)

  • Non-steroidal anti-inflammatory drug (NSAID)-induced gut damage and bacterial translocation (BT) have not been studies well, especially from the perspective of time after administration of NSAIDs. We therefore examined these changes in animals. The study was performed on 5 groups of rat; a control group (group A) and diclofenac groups (groups B, C, E, and F). Rats in the diclofenac groups were orally administered diclofenac sodium before intestinal permeability (IP) measurement (group B, 1 h before measurement; group C, 10 h before; group D, 22 h before; and group E, 52 h before). The IP, stool pellet number, serum biochemical profile, enteric bacterial number, and BT in the mesenteric lymph nodes (MLNs), liver, spleen, kidney and heart were measured. The administration of diclofenac resulted in significantly increased IP, caused intestinal protein loss, decreased stool pellet number, caused enteric bacterial overgrowth and increased BT in multiple organs in groups A, B, C, and D. IF, intestinal protein loss, and the BT in the liver and the spleen in group E were decreased than those in group D. There were no differences in the other parameters between group D and E. In the recovery phase of the diclofenac-induced gut damage, enteric bacterial overgrowth and BT in the kidneys and the heart did not change while the BT in the reticuloendothelial systems such as in the MLNs and liver was decreased.

In vitro cytotoxicity and in vivo acute toxicity of selected polysaccharide hydrogels as pharmaceutical excipients

  • Polysaccharide hydrogels constitute a structurally diverse class of biological macromolecules with a wide range of physicochemical properties. They also constitute important members of the family of industrial water-soluble polymers. They find application in Pharmacy as binders, disintegrants, suspending, emulsifying and sustaining agents. According to the International Pharmaceutical Excipients Council (IPEC), an excipient must have an established safety profile. Hence, in the present study, in vitro cytotoxicity on Vero and HEp-2 cell lines, and in vivo acute toxicity in rats were carried out to establish the safety of polysaccharide hydrogels from the seeds of Plantago ovata and Ocimum basilicum. The in vitro cytotoxicity was determined by MTT and SRB assays. In the in vivo acute toxicity, the effects of three different doses of hydrogels (100, 200 and 400 mg/kg body weight) on food and water intake, body weight, biochemical and hematological parameters were studied. The results of in vitro did not show any cytotoxicity on both the cell lines used. In the in vivo acute toxicity, the hydrogels did not show any toxic symptoms in all three dose levels. This establishes the safety of the selected hydrogels. Hence, they can be used as excipients in pharmaceutical dosage forms.

김으로부터 분리한 방사선 저항성 세균 (Identification of Radiation-Resistant Bacterium Isolated from Dried Laver (Porphyra tenera))

  • A radiation-resistant bacterium was isolated from gamma irradiated dried laver (Porphyra tenera) and its microbiological characteristics were examined. As a result of resistance test to gamma irradiation, the isolate was survived $10^{3}$ CFU/mL even at 30 kGy and significant shoulder line zone was shown until 20 kGy. The $D_{10}$ value was 11.27 kGy. The isolate was gram-positive, non-motile coccus and catalase-positive. n culture, the red-pigmented smooth colony was observed. The biochemical test in API (analytical profile index) system showed that the isolate fermented glucose and fructose as the carbon source. Therefore, a radiation-resistant bacterium isolated from laver was potentially identified as Micrococcus roseus sp.

Mutation of the lbp-5 gene alters metabolic output in Caenorhabditis elegans

  • Intracellular lipid-binding proteins (LBPs) impact fatty acid homeostasis in various ways, including fatty acid transport into mitochondria. However, the physiological consequences caused by mutations in genes encoding LBPs remain largely uncharacterized. Here, we explore the metabolic consequences of lbp-5 gene deficiency in terms of energy homeostasis in Caenorhabditis elegans. In addition to increased fat storage, which has previously been reported, deletion of lbp-5 attenuated mitochondrial membrane potential and increased reactive oxygen species levels. Biochemical measurement coupled to proteomic analysis of the lbp-5(tm1618) mutant revealed highly increased rates of glycolysis in this mutant. These differential expression profile data support a novel metabolic adaptation of C. elegans, in which glycolysis is activated to compensate for the energy shortage due to the insufficient mitochondrial ${\beta}$-oxidation of fatty acids in lbp-5 mutant worms. This report marks the first demonstration of a unique metabolic adaptation that is a consequence of LBP-5 deficiency in C. elegans.

Expression profile identifies novel genes involved in neuronal differentiation

  • In the presence of NGF, PC12 cells extend neuronal processes, cease cell division, become electrically excitable, and undergo several biochemical changes that are detectable in developing sympathetic neurons. We investigated the expression pattern of the apoptosis-related genes at each stage of neuronal differentiation using a cDNA microarray containing 320 apoptosis-related rat genes. By comparing the expression patterns through time-series analysis, we identified candidate genes that appear to regulate neuronal differentiation. Among the candidate genes, HO2 was selected by real-time PCR and Western blot analysis. To identify the roles of selected genes in the stages of neuronal differentiation, transfection of HO2 siRNA in PC12 cells was performed. Down-regulation of HO2 expression causes a reduction in neuronal differentiation in PC12 cells. Our results suggest that the HO2 gene could be related to the regulation of neuronal differentiation levels.
