Isolation and Characterization of Pectinase-Producing Bacillus sp. BS-214 (Pectinase를 생산하는 Bacillus sp. BS-214의 분리 및 특성)

  • A bacterial strain BS-214 producing extracellular pectinase was isolated from soil. The isolated bacterium was identified as a strain of Bacillus so. based on the morphological, biochemical, and physiological characteristics. Cell growth and pectinase activity of Bacillus sp. BS-214 were reached to a mixium in the culture condition of pH 8.5 at 4$0^{\circ}C$. Production of pectinase by the strain was the highest when polygalacturonic acid was added to culture medium as a carbon source, and its optimal concentration was 1%. Also, yeast extract was used as the best nitrogen source for the production of pectinase by the concentration of 0.25%. Decomposition of a constituent of Edzeworthia papyrifera by the strain was observed by scanning electron microscope.

Overexpression and Purification of Monooxygenases Cloned from Arthrobacter chlorophenolicus A6 for Enzymatic Decomposition of 4-Chlorophenol (4-Chlorophenol 분해박테리아 Arthrobacter chlorophenolicus A6로부터의 monooxygenase의 복제 및 대량발현과 정제 그리고 기질분해활성도 분석)

  • Arthrobacter chlorophenolicus A6 possesses several monooxygenases (CphC-I, CphC-II, and CphB) that can catalyze the transformation of 4-chlorophenol (4-CP) to hydroxylated intermediates in the initial steps of substrate metabolism. The corresponding genes of the monooxygenases were cloned, and the competent cells were transformed with these recombinant plasmids. Although CphC-II and CphB were expressed as insoluble forms, CphC-I was successfully expressed as a soluble form and isolated by purification. The specific activity of the purified CphC-I was analyzed by using 4-CP, 4-chlorocatechol (4-CC), and catechol (CAT) as substrates. The specific activities for 4-CP, 4-CC, and CAT were determined to be 0.312 U/mg, 0.462 U/mg, 0.246 U/mg, respectively. The results of this study indicated that CphC-I is able to catalyze the degradation of 4-CC and CAT in addition to 4-CP, which is a primary substrate. This research is expected to provide the fundamental information for the development of an eco-friendly biochemical degradation of aromatic hydrocarbons.

Transformation of dissolved organic matter in a constructed wetland: A molecular-level composition analysis using pyrolysis-gas chromatography mass spectrometry

  • This study investigated the transformation of dissolved organic matter (DOM) in a free-water surface flow constructed wetland. Pyrolysis gas chromatography-mass spectrometry (Py-GC/MS) coupled with preparative high-performance liquid chromatography (prep-HPLC) was used to analyze the compositions of biopolymers (polysaccharides, amino sugars, proteins, polyhydroxy aromatics, lipids and lignin) in DOM according to the molecular size at three sampling points of the water flow: inflow, midflow, and outflow. The prep-HPLC results verified the decomposition of DOM through the decrease in the number of peaks from three to one in the chromatograms of the sampling points. The Py-GC/MS results for the degradable peaks indicated that biopolymers relating to polysaccharides and proteins gradually biodegraded with the water flow. On the other hand, the recalcitrant organic fraction (the remaining peak) in the outflow showed a relatively high concentration of aromatic compounds. Therefore, the ecological processes in the constructed wetland caused DOM to become more aromatic and homogeneous. This indicated that the constructed wetland can be an effective buffer area for releasing biochemically stable DOM, which has less influence on biological water quality indicators, e.g., biochemical oxygen demand, into an aquatic ecosystem.

NMR-based monitoring of the hangover curing effects of deep sea water minerals

  • The term "hangover" refers to symptoms such as headache, heartburn, nausea, and dizziness caused by acetaldehyde created through alcohol decomposition in the body after alcohol intake. Many scientists have conducted research on diverse drugs, foods, and medicinal herbs aimed at eliminating hangovers. However, research on metabolism to objectively verify or measure their effects on hangover symptoms has been lacking. Accordingly, in this study, deep sea water minerals were administered orally at varying concentrations to rats that consumed alcohol, and changes in the levels of amino acids in their bodies were measured using nuclear magnetic resonance spectroscopy to gauge the minerals' effects on hangover symptoms. Thus far, biochemical research on hangover cures has been confined to basic research measuring changes in the levels of alcohol dehydrogenase and acetaldehyde dehydrogenase as well as in the concentrations of ethanol, acetaldehyde, and acetate using spectroscopes such as enzyme-linked immunosorbent assay kits or gas chromatography-mass spectrometers. In comparison, this study presents pharmacokinetic research that simultaneously tracked biomaterials including amino acids and organic acids, metabolites associated with hangover, to clarify hangover mechanisms more specifically. In addition, this study examined hangover mechanisms without an external supply of tracked materials not overlapping with alcohol metabolism-related materials, such as external amino acids and sugars.

A Study on the Hydrogen Degradation of HDPE by Hydrogen Pressure of 90 MPa (90 MPa의 수소 압력에 의한 HDPE의 수소 열화 연구)

  • The physical and chemical changes exhibited by high density polyethylene (HDPE) after treatment with hydrogen at a pressure of 90 MPa followed by rapid release of the hydrogen were studied. X-ray diffraction, differential scanning calorimetry, thermo gravimetric analysis, and attenuated total reflectance (ATR)-fourier transform infrared (FTIR) were used for this purpose. As a result, it was found that the degree of crystallinity of HDPE decreased after hydrogen pressure treatment, while the average thickness of lamellae that constitute the crystals and the melting temperature of the crystalline region actually increased. The decomposition temperature also increased by about 3℃. In addition, it was found that the hydrogen bonding network between -OH groups in the HDPE sample was strengthened and partial chain scission occurred. These cut chains were found to be terminated by oxidative degradation such as cross-linking between chains, -C=O, -C-O, and -CHO, or by the formation of -CH3 at the chain ends, as confirmed by ATR-FTIR.

Adsorption Characteristic of Ammonia by the Cation-Exchange Membrane (양이온 교환막에 의한 암모니아 흡착 특성)

  • In this research, the cation-exchange membrane (SS membrane) containing sulfonic acid group was prepared by radiation induced grafted polymerization onto a porous hollow fiber membrane to effectively remove ammonia which was produced by urea decomposition for peritoneum dialysis system. And the metal ionic cross-linking cation-exchange membrane (SS-M membrane) was prepared by the adsorption of metallic ions (Cu, Ni, Zn) to the SS membranes. The pure water flux and adsorption capacities of ammonia to SS and SS-M membranes were examined. The pure water flux of SS membrane decreased rapidly with the density of $SO_3H$ group increasing. As the metallic ions were adsorbed to the SS membrane, the pure water flux was increased. The adsorption capacities of ammonia at the SS membrane increased with increasing of density of $SO_3H$ group. The ion-exchange capacity of ammonia of the SS membrane was approximately proportional 1 : 1 to the density of $SO_3H$ group. The SS membrane had higher adsorption capacities than the SS-M membrane. The highest adsorption capacities of SS and SS-M membrane appeared the highest pH 9.

Esterase Production and Culture Characteristics of Bacteria Isolated from Acid Hydrolysed Soybean Protein (산분해 대두 단백질로부터 분리된 Esterase 생성균의 생육 및 효소생성 특성)

  • The characteristics of growth and esterase activity of bacterial strains isolated from acid hydrolysed soybean protein were examined. All the isolated strains having decomposition activity of p-hydroxybenzoic acid butyl ester and esterase producing activity were identified as Bacillus sp. by morphological and biochemical methods. The specific growth rates, esterase activities and p-hydroxybenzoic acid butyl ester decomposition activities of isolated strains were $0.844{\sim}1.213\;h^{-1}$, $21{\sim}222\;mU/ml$ and $5.4{\sim}8.1\;mU/ml$, respectively. In the fermentation of Bacillus sp. KB8 strain which had the highest esterase producing activity, growth, extracellular excretion and intracellular synthesis of esterase were inhibited by adding NaCl in the culture broth. Esterase producing activity gradually increased after late exponential growth phase, until maximum value of 420 mU/ml reached after 64 hours culture period. Esterase of Bacillus sp. KB8 strain was stable up to $50^{\circ}C$ for 30 minutes, but was inactivated by heating for 30 minutes at $70^{\circ}C$. The enzyme activity exponentially decreased during the incubation time at the temperatures of $60^{\circ}C$ and $65^{\circ}C$.

Comparison of BOD, COD, TOC and DOC as the Indicator of Organic Matter Pollution of Agricultural Surface Water in Gyeongnam Province

  • This survey was conducted to obtain basic data about organic matter such as BOD (Biochemical Oxygen Demand), COD (Chemical Oxygen Demand), DOC (Dissolved Organic Carbon) and TOC (Total Organic Carbon) in agricultural water in Gyeongnam province. BOD and COD are currently used for water quality indices, but adoption of TOC is being suggested. The surface water samples were collected at 39 locations in Gyeongnam province in April, July and October. Average concentrations of BOD, COD, DOC and TOC were 1.6, 3.3, 5.3 and $5.7mg\;L^{-1}$, respectively. In July, average concentrations of BOD and COD were 2.4 and $3.9mg\;L^{-1}$, respectively, showing these values were higher than other months. Average concentrations of TOC in April, July and October were 4.8, 7.4 and $4.8mg\;L^{-1}$, respectively. This suggested that the highest value of TOC was in July. Average decomposition efficiencies for BOD-C/TOC and COD-C/TOC were 11.4 and 23.9%, respectively. Correlation coefficient of TOC and DOC was higher by 0.995, whereas that of TOC and COD was lower by 0.763. Especially, TOC and DOC were highly related than others.

Kinetics of Removing Nitrogenous and Phosphorus Compounds from Swine Waste by Growth of Microalga, Spirulina platensis

  • Abstract Spirulina platensis was grown in SWlUe waste to reduce inorganic compowlds and simultaneously produce feed resources. Spirulina platensis prefers nitrogenous compounds in Ibe order: $NH_4^{+}-N>NO_3^{-}-N>simple-N$ such as urea and simple amino acids. It even consumes $NH_4^{+}-N$ first when urea or nitrate are present. Therefore, the content of residual $NH_4^{+}-N$ in Spimlina platensis cultures can be determined by the relative extent of the following processes: (i) algal uptake and assimilation; (ii) ammonia stripping; and (iii) decomposition of urea to NH;-N by urease-positive bacteria. The removal rates of total nitrogen ffild total phosphorus were estimated as an indicator of the treatment effIciency. It was found that Spirulina platensis was able to reduce 70-93% of $P_4^{3-}-P$, 67-93% of inorganic nitrogen, 80-90% of COD, and 37-56% of organic nitrogen in various concentrations of swine waste over 12 days of batch cultivation. The removal of inorganic compounds from swine waste was mainly used for cell growth, however, the organic nitrogen removal was not related to cell growlb. A maximum cell density of 1.52 dry-g/l was maintained with a dilution rate of 0.2l/day in continuous cultivation by adding 30% swine waste. The nitrogen and phosphorus removal rates were correlated to the dilution rates. Based on the amino acid profile, the quality of the proteins in the Spirulina platensis grown in the waste was the same as that in a clean culture.ulture.

A Study on the Properties of sPEEK Electrolytic Membranes using Physical Crosslinking (물리적 가교결합을 이용한 sPEEK 전해질막의 특성에 관한 연구)

  • Composite membranes were prepared by the solution casting method from sulfonated poly(etheretherketone)(sPEEK) and imidazole and phosphotungstic acid(PWA) to enhance the electrolytic properties of the membrane. TGA measurements showed that physical crosslinking due to acid-base interactions improved the thermal resistance to the desulfonation of sulfonic acid groups of the composite membrane and the addition of PWA enhanced the resistance to thermal decomposition of the composite membrane. The acid-base interaction decreased the water uptake, proton conductivity and methanol permeability of the sPEEK/imidazole composite membranes. The addition of PWA increased the proton conductivities while it decreased the water uptake and methanol permeability of sPEEK/imidazole/PWA composite membranes. Therefore, the selectivity of the composite membranes was enhanced by the addition of PWA.
