Expression of Heat Shock Protein and Antioxidant Genes in Rice Leaf Under Heat Stress

  • We have previously investigated the proteome changes of rice leaves under heat stress (Lee et al. in Proteomics 2007a, 7:3369-3383), wherein a group of antioxidant proteins and heat shock proteins (HSPs) were found to be regulated differently. The present study focuses on the biochemical changes and gene expression profiles of heat shock protein and antioxidant genes in rice leaves in response to heat stress ($42^{\circ}C$) during a wide range of exposure times. The results show that hydrogen peroxide and proline contents increased significantly, suggesting an oxidative burst and osmotic imbalance under heat stress. The mRNA levels of chaperone 60, HSP70, HSP100, chloroplastic HSP26, and mitochondrial small HSP responded rapidly and showed maximum expression after 0.5 or 2 h under heat stress. Transcript levels of ascorbate peroxidase (APX), dehydroascorbate reductase (DHAR) and Cu-Zn superoxide dismutase (Cu-Zn SOD) showed a rapid and marked accumulation upon heat stress. While prolonged exposure to heat stress resulted in increased transcript levels of monodehydroascorbate reductase, peroxidase, glyoxalase 1, glutathione reductase, thioredoxin peroxidase, 2-Cysteine peroxiredoxin, and nucleoside diphosphate kinase 1, while the transcription of catalase was suppressed. Consistent with their changes in gene expression, the enzyme activities of APX and DHAR also increased significantly following exposure to heat stress. These results suggest that oxidative stress is usually caused by heat stress, and plants apply complex HSP- and antioxidant-mediated defense mechanisms to cope with heat stress.

틸리피아에서 분리(分離)한 Edwardsie tarda의 성상(性狀)과 항원성(抗原性)에 대하여 (Characteristics and Antigenicity of Edwardsiella tarda Isolated from Tilapia (Oreochramis niloticus))

  • 1987년(年) 3(월)月 부산수산대학(釜山水産大學) 어병학(魚病學) 실험실(實驗室)에서 사육중(飼育中)이던 틸라피아 (Oreochromis niloticus)에 세균성(細菌性) 질병(疾病)이 발생(發生)하여 만성적(慢性的)으로 몇마리씩 폐사(斃死)하였다. 본(本) 연구(硏究)는 이러한 질병(疾病)의 원인(原因)을 밝히기 위하여 원인균(原因菌)을 분리(分離)하였으며, 형태적(型態的), 생화학적(生化學的) 성상(性狀)을 관찰(觀察)하고 병원성(病原性), 약제감수성(藥劑感受性) 실험(實驗)을 하였으며 시험항원(試驗抗原)을 제작(制作)하여 그 항원성(抗原性)을 조사하였는바, 그 결과(結果)를 요약(要約)하면 다음과 같다. 1. 병어(病魚)에서 분리(分離)된 원인균(原因菌)은 Edwardsiella tarda로 동정(同定)되었다. 2. 분리균(分離菌)을 틸라피아에 접종하여 재현성실험(再現性實驗)을 한 결과(結果) 전형적인 E. tarda의 감염증상을 나타내고 사망(死亡)했으며, 어류(魚類)로부터 동일균(同一菌)을 얻을 수 있었다. 3. 분리균(分離菌)은 oxytetracycline에서 감수성(感受性)이 높았고, MIC는 6.9로 나타났다. 4. 분리균(分離菌)은 병원성(病原性)이 강한 균(菌)이었으며, 48-LD50은 $10^{6.4}cell$/fish 이었다. 5. 시험항균(試驗抗原)으로, 포르말린사균항원, 초음파처리항원, 균여과액항원을 사용하였으며, 이 세가지를 사용한 항원성시험(抗原性試驗) 결과 포르말린사균항원의 시험구에서 높은 응집가와 방어력을 나타내었다.

  • n-3HUFA가 조피볼락의 성장, 영양소축적효율 및 체성분에 미치는 영향을 구명하기 위해 오징어 간유 및 대두유를 지질원으로 사료에 첨가하여 사료의 n-3HUFA 함량이 $0\~1.5\%$가 되도록 조정하여 사육한 결과는 다음과 같다. 성장, 사료효율 및 영양소 축적효율 모두 n-3HUFA 무첨가 사료에서 가장 낮은 값을 나타내었으며, 사료의 n-3HRFA 함량의 증가와 더불어 그 효과가 현저히 개선되다가 $1.2\%$ 이후에는 더이상 개선효과가 없었다. n-3HUFA함량이 높은 실험구의 간지질(비극성 지질) 함량은 감소한 경향을 보였으나 간의 glycogen 함량은 증가하였으며, HSI 값도 다소 증가하였다. 전어체의 지질함량 및 내장의 지질함량은 모두 사료의 n-3HUFA 함량이 높은 실험구에서 높게 나타났으며, 이러한 증가는 비극성 지질의 변화 경향과 일치하였다. 간 극성 지질 중의 지방산조성은 사료의 지방산조성에 따라 변화하여, 사료의 n-3HUFA가 증가함에 따라 n-3HUFA와 18:1은 증가하였고, 18:2n-6과 18:3n-3은 감소하였다.

수온 상승에 따른 북방전복, Haliotis discus hannai과 둥근전복, H. discus discus 체액의 생리학적 변화 (Influence of Elevated Temperatures on the Physiological Response of Hemolymph from Two Species of the Abalone, Haliotis discus hannai and H. discus discus)

  • 본 연구에서는 수온 증가에 따른 북방전복 (Haliotis discus hannai) 과 둥근전복 (H. discus discus) hemolymph의 생리 및 면역학적 변화를 관찰하기 위하여 위 두 종을 20, 22, 24, 26 및 $28^{\circ}C$ 수온에 각각 4 일간 노출시켰다. 노출 결과, hemolymph의 total-protein (TP), glucose, 칼슘 (Ca) 은 둥근전복이 북방전복보다 높은 값을 보였으나, 마그네슘 (Mg), alkaline phosphatase (ALP) 및 lysozyme은 두 종에서 유사한 값을 보였다. 수온 증가에 따른 hemolymph의 TP, glucose 및 마그네슘은 두 전복 모두에서 유의한 변화가 관찰되지 않았다. 그러나 칼슘과 면역관련 인자인 ALP 및 lysozyme은 둥근전복에서 유의한 차이가 없었으나, 북방전복에서는 $26^{\circ}C$ 또는 $28^{\circ}C$의 고수온에 노출되었을 때, 유의하게 높은 활성을 보였다. 한편 phenoloxidase (PO)는 북방전복에서 높은 값을 보였으며, 수온이 높을수록 증가하는 경향을 보였다. 결론적으로 북방전복은 둥근전복과 달리 여러 지표에서 유의한 변화가 관찰된 것으로 보아 북방전복이 둥근전복보다 고수온에 보다 민감한 것으로 사료된다.

Arabidopsis thaliana의 Ethylene Triple Response Mutant에서 에틸렌 생합성 과정의 생리 생화학적 특성

  • Arabidopsis thaliana의 에틸렌 관련 돌연변이체인 에틸렌 과다 생성 돌연변이체(eto1-1)와 에틸렌에 대한 반응이 둔감한 돌연변이체(etr1-3과 ein2-1)의 생리·생화학적 특성을 분석하였다. 2∼3주된 Arabidopsis 식물로부터 얻은 성숙한 잎을 재료로 이용하였다. 에틸렌 생성량은 eto1-1은 야생형의 2배, etr1-3은 야생형의 4배, 그리고 ein2-1은 야생형의 4.5배 더 많았다. eto1-1에서의 ACC synthase와 ACC oxidase 활성은 야생형과 비슷하였으나 ACC 함량은 야생형보다 4.5배 더 많았다. ACC-N-malo-nyltransferase의 활성은 eto1-1이 야생형보다 3배 더 높았으며 SAM synthetase의 활성은 야생형보다 1.5배 더 높았다. 이들 결과로부터 eto1-1로의 변형은 SAM에서 ACC로 전환하는 과정 이전에 있음을 추측할 수 있다. etr1-3과 ein2-1에서 ACC synthase의 활성은 야생형보다 높았으나 ACC oxidase의 활성은 야생형보다 낮았다. 그러나 SAM synthetase 활성은 etr1-3에서는 야생형과 비슷하였고 ein2-1에서는 야생형보다 1.7배 높았다. 이것은 etr1-3과 ein2-1이 에틸렌에 대한 반응에 결함이 있기 때문에 그것으로 인하여 자가 조절이 되지 않았기 때문으로 추정된다. etr1-3의 ACC 함량은 야생형보다 2.3배 더 많았으며 ACC N-malonyltransferase의 활성은 야생형보다 3.9배 더 높았다. 그리고 ein2-1의 ACC 함량은 야생형보다 1.7배 더 많았으며 ACC N-malonyltransferase의 활성이 촉진된 것으로 추정된다. In vitro kinase assay를 한 결과 eto1-1과 ein2-1에서 36 kDa 단백질의 인산화를 관찰할 수 있었다.

  • 기침, 호흡곤란 및 성한 전신 쇄약감을 주소로 내원하여 기관지 내시경 및 경부 림프절 생검을 통한 조직검사로 소세포 폐암을 확진하고, 병력상 심한 전신무력감 및 체중감소를 호소해며, 검사상 성한 저칼륨혈증 및 대사성 알칼리증의 소견이 있으며, 최근 발생된 고혈당, 고혈압 등의 증상이 있어 시행한 내분비학적 생화학 검사상 혈중 cortisol이 상승하고, cortisol의 일간 변동(diurnal variation)이 소실되었으며, 24시간 뇨 free cortisol의 현저한 상승으로 Cushing 증후군이 paraneoplastic syndrome으로 소세포 폐암에서 동반되었음을 확인한 1예를 경험하였기에 보고하는 바이다.

오공약침(蜈蚣藥鍼)의 안전성(安全性)에 관한 연구(硏究) (The Study on Safety of Scolopendrid Aqua-acupuncture)

  • Objective : Recently scolopendrid aqua-acupuncture has been a good effect on pain control but it has not been known about clinical safety. The purpose of this study was to investigate acute toxicity of scolopendrid aqua-acupuncture. Method : In order to prove the clinical safety of scolopendrid aqua-acupuncture, We have observed a bacteriological examination and clinical pathology test after scolopendrid aqua-acupuncture treatment. Balb/c mice were injected intravenous with Scolopendrid aquaacupuncture treatment for $LD_{50}$ and acute toxicity test. We analyzed physical reaction(side effect)and clinical pathology test before and after Scolopendrid aqua-acupuncture treatment of mice and 20 patients suffering from pain, who admitted department of Acupunture and Moxibustion, College of Oriental Medicine, Won-Kwang University Kwangju hospital. Results : In the Blood agar plate and Nutrient agar plate, a bacteriological examination did not show a bacillus. In acute $LD_{50}$ toxicity test, there was no mortality thus unable to attain the value. Examining the toxic response in the acute toxicity test, there was no sign of toxication. In acute toxic test, running biochemical serum test couldn't yield any differences between the control and experiment groups. In the 20 patients treated with Scolopendrid aqua-acupuncture, hematologic test did not show remarkable change. In the 20 patients treated with Scolopendrid aqua-acupuncture, Liver function test(AST, ALT, ALP) showed a slight decrease on the contrary, and abnormal rate showed a decrease of 5.0% compared with previous study. Reanl function test(BUN, Cr) and abnormal rate showed a decrease of 5.0% compared with previous study. In the 20 patients treated with Scolopendrid aqua-acupuncture, Electrolyte were normal range before and after treatment. In the Urine analysis of 20 patients, Leukocyte, Protein, Glucose, Keton, Bilirubin, U-bilinogen were not detected before and after Scolopendrid aqua-acupuncture treatment, and the rest almost made no difference.

Rice OsACDR1 (Oryza sativa Accelerated Cell Death and Resistance 1) Is a Potential Positive Regulator of Fungal Disease Resistance

  • Rice Oryza sativa accelerated cell death and resistance 1 (OsACDR1) encodes a putative Raf-like mitogen-activated protein kinase kinase kinase (MAPKKK). We had previously reported upregulation of the OsACDR1 transcript by a range of environmental stimuli involved in eliciting defense-related pathways. Here we apply biochemical, gain and loss-of-function approaches to characterize OsACDR1 function in rice. The OsACDR1 protein showed autophosphorylation and possessed kinase activity. Rice plants overexpressing OsACDR1 exhibited spontaneous hypersensitive response (HR)-like lesions on leaves, upregulation of defense-related marker genes and accumulation of phenolic compounds and secondary metabolites (phytoalexins). These transgenic plants also acquired enhanced resistance to a fungal pathogen (Magnaporthe grisea) and showed inhibition of appressorial penetration on the leaf surface. In contrast, loss-of-function and RNA silenced OsACDR1 rice mutant plants showed downregulation of defense-related marker genes expressions and susceptibility to M. grisea. Furthermore, transient expression of an OsACDR1:GFP fusion protein in rice protoplast and onion epidermal cells revealed its localization to the nucleus. These results indicate that OsACDR1 plays an important role in the positive regulation of disease resistance in rice.

Prognostic Value of Biochemical Response Models for Primary Biliary Cholangitis and the Additional Role of the Neutrophil-to-Lymphocyte Ratio

  • Background/Aims: Recently reported prognostic models for primary biliary cholangitis (PBC) have been shown to be effective in Western populations but have not been well-validated in Asian patients. This study aimed to compare the performance of prognostic models in Korean patients and to investigate whether inflammation-based scores can further help in prognosis prediction. Methods: This study included 271 consecutive patients diagnosed with PBC in Korea. The following prognostic models were evaluated: the Barcelona model, the Paris-I/II model, the Rotterdam criteria, the GLOBE score and the UK-PBC score. The neutrophil-to-lymphocyte ratio (NLR) was analyzed with reference to its association with prognosis. Results: For predicting liver transplant or death at the 5-year and 10-year follow-up examinations, the UK-PBC score (areas under the receiver operating characteristic curve [AUCs], 0.88 and 0.82) and GLOBE score (AUCs, 0.85 and 0.83) were significantly more accurate in predicting prognosis than the other scoring systems (all p<0.05). There was no significant difference between the performance of the UK-PBC and GLOBE scores. In addition to the prognostic models, a high NLR (>2.46) at baseline was an independent predictor of reduced transplant-free survival in the multivariate analysis (adjusted hazard ratio, 3.74; p<0.01). When the NLR was applied to the prognostic models, it significantly differentiated the prognosis of patients. Conclusions: The UK-PBC and GLOBE scores showed good prognostic performance in Korean patients with PBC. In addition, a high NLR was associated with a poorer prognosis. Including the NLR in prognostic models may further help to stratify patients with PBC.

Effects of method and duration of restraint on stress hormones and meat quality in broiler chickens with different body weights

  • Objective: The study was designed to investigate the effects of restraint method, restraint duration, and body weight on stress-linked hormones (corticosterone, adrenaline, and noradrenaline), blood biochemical (namely glucose and lactate), and the meat quality in broiler chickens. Methods: A total of 120 male broiler chickens (Cobb 500) were assigned to a $2{\times}3{\times}2$ factorial arrangement in a completely randomized design using two restraint methods (shackle and cone), three durations of restraint (10, 30, and 60 s), and two categories of live body weight ($1.8{\pm}0.1kg$ as lightweight and $2.8{\pm}0.1kg$ as heavyweight). Results: Irrespective of the duration of restraint and body weight, the coned chickens were found to have lower plasma corticosterone (p<0.01), lactate (p<0.001), lower meat drip loss (p<0.01), cooking loss (p<0.05), and higher blood loss (p<0.05) compared with their shackled counterparts. The duration of restraint had significant effects on the meat initial pH (p<0.05), ultimate pH (p<0.05), and yellowness (p<0.01). The lightweight broilers exhibited higher (p<0.001) blood loss and lower (p<0.05) cooking loss compared to the heavyweight broilers, regardless of the restraint method used and the duration of restraint. However, the interaction between the restraint method, duration of restraint, and body weight contributed to differences in pre-slaughter stress and meat quality. Therefore, the interaction between the restraint method and the duration of restraint affected the meat shear force, lightness ($L^*$) and redness ($a^*$). Conclusion: The duration of restraint and body weight undoubtedly affect stress responses and meat quality of broiler chickens. Regardless of the duration of restraint and body weight, the cone restraint resulted in notably lower stress, lower meat water loss, and higher blood loss compared to shackling. Overall, the findings of this study showed that restraint method, duration of restraint, and body weight may affect the stress response and meat quality parameters in broilers and should be considered independently or interactively in future studies.
