The Anti-calcification Effect of Dithiobispropionimidate, Carbodiimide and Ultraviolet Irradiation Cross-linking Compared to Glutaraldehyde in Rabbit Implantation Models

  • Background: Glutaraldehyde (GA) is a widely used cross-linking agent for improving mechanical properties and resistance to enzymatic degradation of collagenous tissue, but it has several drawbacks such as calcification and cytotoxicity. The aim of this study was to find the alternative effective cross-linking methods to GA. Materials and Methods: Bovine pericardium was processed with GA with ethanol+octanol and glycine detoxification, and polyethylene glycol (PG) space filler, dimethyl 3,3'-dithiobispropionimidate (DTBP), 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide (EDC) treatment, and the physical fixation of ultraviolet irradiation were done. The biologic material properties of variously treated pericardial tissues were assessed by biochemical, mechanical and histological tests. Treated pericardial tissues were also implanted subcutaneously or intramuscularly into the rabbit for 10 weeks to assess the xenoreactive antibody response of immunoglobulin G and M, their anti-calcification effect. Results: The biochemical and mechanical properties of EDC fixed pericardial tissues were comparable to the GA fixed tissue. The cytotoxicity was lowest in space filler treated GA fixed group. In rabbit subcutaneous or intramuscular implantation models, decellularization, space filler, EDC treatment group showed significantly lower calcium content than GA only and DTBP treatment group (p<0.05, analysis of variance). The titer of anti $Gal{\alpha}1-3Gal{\beta}1$-4GlcNAc-R antibodies did not change in the postimplantation serial enzyme-linked immunosorbent assay. Hematoxylin and eosin and von Kossa staining showed that decellularization, space filler, EDC, and ultraviolet treatment had less inflammatory cell infiltration and calcium deposits. Conclusion: The decellularization process, PG filler, and EDC treatments are good alternative cross-linking methods compared to GA only fixation and primary amine of DTBP treatment for cardiovascular xenograft preservation in terms of the collagen cross-linking stability and in vivo anti-calcification effects.

The Study on Acute and Subacute Toxicity and Anti-Cancer Effects of cultivated wild ginseng Herbal acupuncture (정맥주입용(靜脈注入用) 산양산삼(山養山蔘) 증류약침(蒸溜藥鍼)의 급성(急性).아급성(亞急性) 독성실험(毒性實驗) 및 Sarcoma-180 항암효과(抗癌效果)에 관(關)한 실험적(實驗的) 연구(硏究))

  • Objective : The purpose of this study was to investigate acute and subacute toxicity and sarcoma-180 anti-cancer effects of herbal acupuncture with cultivated wild ginseng (distilled) in mice and rats. Method : Balb/c mice were injected intravenous with cultivated wild ginseng herbal acupuncture for $LD_{50}$ and acute toxicity test. Sprague-Dawley rats were injected intravenous with cultivated wild ginseng herbal acupuncture for subacute toxicity test. The cultivated wild ginseng herbal-acupuncture was injected at the tail vein of mice. Results : 1. In acute $LD_{50}$ toxicity test, there was no mortality thus unable to attain the value. 2. Examining the toxic response in the acute toxicity test, there was no sign of toxication. 3. In acute toxic test, running biochemical serum test couldn't yield any differences between the control and experiment groups. 4. In subacute toxicity test, there was no sign of toxication in the experimental groups and didn't show any changes in weight compared to the normal group. 5. In subacute toxicity test, biochemical serum test showed significant increase of Total albumin, Albumin, and Glucose in the experimental group I compared with the control group. Significant decrease of GOT, ALP, GPT, and Triglyceride were shown. In experiment group II, only Glucose showed significant increase compared with the control group. 6. Measuring survival rate for anti-cancer effects of Sarcoma-180 cancer cell line, all the experimental groups showed significant increase in survival rate. 7. Measuring NK cell activity rate, no significant difference was shown throughout the groups. 8. Measuring Interleukin-2 productivity rate, all the experimental groups didn't show significant difference. 9. For manifestation of cytokine mRNA, significant decrease of interleukin-10 was witnessed in the experimental group compared to the control group. Conclusion : According to the results, we can conclude cultivated wild ginseng herbal acupuncture caused negligible toxicity, and had anti-tumor effects in mice.

Effects of Long-Term Vitamin E and Butylated Hydroxytoluene Supplemented Diets on Murine Intestinal and Hepatic Antioxidant Enzyme Activities

  • The present study was designed to determine long-term feeding effects of vitamin E and BHT (butylated hydroxytoluene) on serum biochemical profiles, organ weight, and intestinal and hepatic antioxidant enzymes including superoxide dismutase (SOD), glutathione peroxidase (GSH-PX), and glutathione-S-transferase (GST) in ICR mice. Four wk old ICR mice (n=8 per group) were fed the diets supplemented with vitamin E (I ; 0.03% and II ; 0.3%) and BHT (I ; 0.05% and II ; 0.5%) for 12 months. Feeding the diets containing vitamin E and BHT had no effects on growth and serum biochemical profiles. However, feeding the diets supplemented with 0.5% BHT for 12 months significantly increased liver weight of the mice. In the small intestine, there were no effects of vitamin E or BHT on SOD and GSH-PX activities in the mucosa. However, the activity of intestinal GST of the mice that received 0.5% BHT was almost twice as high as that of control mice. In the liver, the activity of SOD was not affected by feeding antioxidants for 12 months, whereas GSH-PX activity was significantly increased in mice that received the diets containing BHT (0.05%, 0.5%) and vitamin E (0.03%, 0.3%). In addition, supplementation of 0.5% BHT markedly enhanced hepatic GST activity compared with other groups. Enhanced activity of GSH-PX in response to feeding vitamin E or BHT might aid hepatic enzymes to eliminate active oxygen in organs from mice. However, we could not exclude the possibility of increased lipid peroxidation by high dosage of BHT supplementation. More detailed study is necessary for assessment of preventive or toxicological effects of high dosage of BHT supplementation.

Effect of Heat Treatment on Biochemical Characteristics of Pacific Oyster Crassostrea gigas (열처리에 의한 굴(Crassostrea gigas)의 영양성분 변화)

  • The aim of this study was to investigate changes in the biochemical characteristics of the Pacific oyster Crassostrea gigas in response to boiling water bath ($85^{\circ}C$, $100^{\circ}C$) and autoclaving ($100^{\circ}C$) heat treatments for 5, 10, 15, and 20 min. The weight loss (7.6-9.5%) of oysters exposed to a $100^{\circ}C$ boiling water bath for 15 min was lower compared to those receiving no treatment. Oysters exposed to the $100^{\circ}C$ boiling water bath for 20 min lost 27.8% of their total weight. The $100^{\circ}C$ autoclaving treatment resulted in weight loss of 13.0-26.1%, with the highest weight loss occurring after 10 min of treatment (26.1%). Heat-treated oysters had significantly higher levels of crude lipid and crude protein relative to untreated oysters (P<0.05). In the heat-treated oysters, total amino acids (including proline and glutamic acid) were increased, but they were decreased by the 20-min $100^{\circ}C$ boiling water bath and autoclaving treatments. The fatty acid composition of oysters was not significantly affected by the water bath heat treatment, but oysters treated with $100^{\circ}C$ autoclaving for 20 min had decreased polyunsaturated fatty acids. Oysters exposed to the water bath heat treatment had significantly increased vitamin A content, but $100^{\circ}C$ autoclaving for 20 min was associated with reduced vitamin A content, as compared to all of the other heat treatments. Therefore, oysters were affected by heat treatment, undergoing metamorphosis and weight loss, but nutrient quality did not change significantly compared to controls, except with the 20-min $100^{\circ}C$ boiling water bath and autoclaving treatments.

Growth Performance, Carcass Characteristics, Nutrient Digestibility and Serum Biochemical Parameters of Broilers Fed Low-protein Diets Supplemented with Various Ratios of Threonine to Lysine

  • This study was conducted to evaluate performance, breast (thigh) muscle yield, nutrient digestibility and serum biochemical parameters of broilers fed low-protein diets supplemented with various threonine to lysine ratios (Thr/Lys). Three hundred and twenty four day-old male Arbor Acres broilers were randomly allotted to six dietary treatments with six replicates per treatment and nine broilers per replicate. Six treatments included one control diet [formulated according to NRC (1994)], and five experimental diets (treatments 1-5). In treatments 1-5, the crude protein level was 2% lower than that of the NRC control diet. The Thr/Lys in treatments 1-5 was 0.65, 0.70, 0.75, 0.80 and 0.85 during the starter and grower phases and 0.70, 0.75, 0.80, 0.85 and 0.90 during the finisher phase with other nutrient levels kept consistent. The results showed that: (1) With increasing Thr/Lys, ADG of treatments 1, 3 and 4 were lower (p<0.05) than those of the NRC control diet during the starter phase; ADG of treatments 1-5 were lower (p<0.05) than those of the NRC control during the grower phase, and ADG of treatment 1 was lower (p<0.05) than that of the NRC control during the finisher phase. Average daily feed intake in treatments 1 and 4 were lower (p<0.05) than that of broilers on the NRC control diet during the grower phase. Feed/gain (F/G) of the NRC control diet was lowest (p<0.05) during the starter phase. F/G in treatments 2 and 5 were higher (p<0.05) than that of NRC during the grower phase. (2) Breast muscle proportion on d 21 increased linearly (p<0.05) in response to graded levels of Thr/Lys. (3) No differences were detected for dry matter, energy and crude protein digestibility among all seven treatments. (4) On d 21, serum triglyceride of broilers on dietary treatments 3 and 5 was higher (p<0.05) than that of broilers in control. The results indicate that the low-protein diets supplemented with an appropriate Thr/Lys could not support the same performance that was achieved by the broilers fed the NRC control diet.

The Effects of the Mixture of Fetal Bovine Serum and Poly-glycolic acid in Rabbit Calvarial Model (Poly-glycolic Acid(PGA)와 우태아 혈청(Fetal bovine serum, FBS)의 혼합물이 가토에서 골형성에 미치는 영향)

  • Purpose: This study was undertaken to investigate the osteogenic induction potential of PGA & FBS mixture on a calvarial defect in the rabbit. Methods: Twenty New zealand white rabbit, weighing from 3.5-4kg were allocated into each of the three groups. Four 8 mm sized bone defects were made on the parietal bone by drilling. In group I, the bony defects were implanted with $50{\mu}m$ thickness film containing mixture of PGA and FBS. In group II, with PGA only film, & in group III, the bony defects were left with no implants. Results were evaluated by using morphologic change, radiographic study, biochemical study and histologic examination at 1 week (group I n=7, group II n=7, group III n=14), 2 weeks (group I n=6, group II n=6, group III n=12) and 3 weeks (group I n=7, group II n=7, group III n=14) following implantation. Results: In the morphologic & radiographic study, the formation and corticalization of callus were observed earlier in group I than in groups II and III (p < 0.05). In histological examination, group I showed more abundant and faster new bone formation than in group II and III. In biochemical analysis, group I displayed more activity than in group II and III. Group I also showed more abundant osteopontin, osteocalcin than groups II and III. Conclusion: In conclusion, the results demonstrate that the mixture of PGA and FBS has an effect on osteoblastic formation in the rabbit model. It is considered that further evaluation of long term results on resorption, immunologic tissue reaction and response of applied mixture in the human model will be needed.

Effects of Drinking Ion Water on Performance, Egg Quality and Blood Biochemical Composition of Laying Hens under High Temperature Conditions (고온환경에서 이온수의 급수가 산란계의 생산성, 계란 품질 및 혈액성상에 미치는 영향)

  • This study was conducted to investigate the effect of drinking ion water on the performance, egg quality, and serum biochemical properties of laying hens in a high-temperature environment. A total of 180 laying hens with equal egg production rates were randomly divided into three treatments with five replicates. Treatments were applied from 24 to 27 weeks of age and included the supply of ion water (IW), tap water (TW), and cold water (CW). The results showed that the egg production rate, egg weight, and feed intake of hens were significantly (P<0.01) improved in the IW group compared with those in the TW group but did not differ from those in the CW group. Egg quality parameters such as albumen height, Haugh unit, and eggshell thickness were found to be higher in the IW than in the TW group (P<0.05); however, no significant differences were observed in comparison with the CW group. Serum Ca and P concentrations tended to increase (P<0.10) in the IW group, and serum high-density lipoprotein cholesterol and albumin levels were greater (P<0.05) in response to IW than those to TW. This study provides evidence that the supply of IW can relieve the high-temperature stress during the summer. In particular, because the improved effects of the IW were confirmed to be similar to those of CW, it shows potential as a method to manage the drinking water of hens in the summer season along with the supply of CW.

Development of a Biosensor Using Electrochemically-Active Bacteria [EAB] for Measurements of BOD [Biochemical Oxygen Demand] (전기화학적 활성 미생물을 이용한 BOD 측정용 바이오센서의 개발)

  • A biosensor using electrochemically-active bacteria (EAB) enriched in three-electrode electrochemical cell, was developed for the determination of biochemical oxygen demand (BOD) in wastewater. In the electrochemical cell, the positively poised working electrode with applying a potential of 0.7 V was used as an electron acceptor for the EAB. The experimental results using artificial and raw wastewater showed that the current pattern generated by the biosensor and its Coulombic yield were proportional to the concentration of organic matter in wastewater. The correlation coefficients of BOD vs Coulombic yield and $BOD_5$ vs Coulombic yield were 0.99 and 0.98, respectively. These results indicate that the biosensor enriched with the EAB capable of transferring electrons directly toward the electrode can be utilized as a water-quality monitoring system due to a quick and accurate response.

A Study on GHS Classification of 3-Methylpentane by Subacute Inhalation Toxicity (아급성흡입독성시험을 이용한 3-Methylpentane의 GHS 분류·표시)

  • Objectives : The purpose of this study was to obtain information regarding Globally Harmonized System(GHS) classification and health hazards that may result from a 4 weeks inhalation exposure of 3-Methylpentane in Sprague-Dawley rats. Methods : The testing method was conducted in accordance with OECD guidelines for the testing of chemicals No. 412(Subacute Inhalation Toxicity). The Rats were divided into 4 groups(5 male and 5 female rats in each group) and exposed to 0 ppm, 284 ppm, 1,135 ppm, 4,540 ppm 3-Methylpentane in each exposure chamber for 6 h/day, 5 days/week, for 4 weeks. After two weeks, the test animals were autopsied and carried out blood test and biochemical tests and histopathological examination. We used PRISTIMA (Toxicology data management system) to confirm the system and to have confidence of the raw data. Results : No death and particular clinical presentation including weight change and change of feed rate was observed. Relationship between dose, gender and response was also not significantly changed in hematologic examination, biochemical examination of blood and blood coagulation time. The histopathologic lesions caused by the test substance did not appear. Conclusions : NOAEL(No Observable Adverse Effect Level) of 3-Methylpentane is more than 4,540 ppm in male group and female group and the Ministry of Employment and Labor Guidance Announcement No. 2013-37(criteria for the classification marks and Safety of Chemicals) Specific target organ toxicity(repeated exposure) was determined with a substance that is not the separator material.

Comparison of Blood Gas, Biochemical Factors, and Hormone Concentrations, and Muscle Stress Response Factors of Chickens in General Farms and Animal Welfare Farms (일반농장과 동물복지농장 닭의 혈액 내 gas, 생화학인자, 호르몬 농도 및 근육 내 스트레스 반응인자 비교)

  • It is generally recognized that high stocking density can increase the risk of immune dysfunction and reduction in productivity cause of elevated stress in broiler farm. The domestic livestock industry is implementing a certification system for animal welfare and livestock farms to reduce the problem of dense breeding. This study compared broiler from the general farm and animal welfare farm to confirm the effectiveness of animal welfare farms against stress. Gas concentrations in a farm of general and animal welfare farms were analyzed, blood (n=10) of the 28-day-old broiler was collected, the partial pressure of the gas in the blood was analyzed using whole blood, and biochemical factors and hormones were analyzed using the serum. Glycogen and L-lactate are analyzed using ELISA kit on finely pulverized muscles (n=10). CO2 and NH3 levels in the farm were significantly higher in general farm. Partial carbon dioxide tension (PCO2) in blood was significantly higher and partial oxygen tension (PO2) was significantly lower in the general farm. Creatinine (CREA), glucose (GLU), aspartateaminotransferase (AST), and creatinekinase (CK) in the blood were significantly higher in the general farm, and total protein (TP) and globulin (GLOB) were significantly higher in animal welfare farm. Blood corticosterone and norepinephrine were significantly higher in general farm and epinephrine was significantly higher in animal welfare farm. These results imply that animal welfare farm provides a stress-reduced breeding environment.
