The Effects of Uncooked Powdered Food on Nutrient Intake, Serum Lipid Level, Dietary Behavior and Health Index in Healthy Women (생식 제품의 섭취가 건강한 성인여성의 영양섭취상태, 식행동, 혈청지질농도 및 건강지표에 미치는 영향)

  • The purpose of this study was to obtain the nutritional values and objective data of uncooked powdered food (UPF) in korea. We interviewed 27 healthy female subjects aged over 25 years living in the lksan area. We just replaced common breakfast and dinner of the subjects with UPF. Their dietary intake status was evaluated by 24-hour recall method. Their body compositions were measured using body fat analyzer. Also we conducted hematological and clinical analysis of blood. The intake of energy, lipid and protein has decreased as people started to take UPF, but the intake of vitamins and minerals has increased. The quality of meals has improved after taking UPF. Due to the energy loss by taking UPF, weight and body fat gradually lessened. Among the lost weight the percentage of the body fat was high and we judged that the process of losing weight was successful. It is hard to predict whether weight loss will occur to people who are already in shape or not, but if overweight and obesity people regularly take UPF instead of other food products, we assume that UPF will help lessening body fat. The most positive change among biochemical changes by taking UPF was decreasing of serum lipid contents. The concentrations of total cholesterol and LDL-cholesterol gradually decreased and decreased intently after 12weeks. Serum HDL-cholesterol gradually increased and serum triglyceride showed gradual decrement. When healthy adult women replaced two of three meals with UPF for 3months, we were able to see some useful changes like decrement of body fat and serum lipid control and during this 3month period, no significant nutritional problems occurred. Complete judgement on UPF may be difficult with these conclusions but if people take a nomal meal once and replenish iron from taking UPF twice a day, we assume that replacing UPF with regular meal not be a problem in nutritional status. If other experiments on the effects of UPF proceed, we believe that those experiments will be very helpful in judging the nutritional value of UPF. (Korean J Nutrition 36(1) : 49-63, 2003)

  • Toxicological studies have an object of detecting adverse effects of a chemical on an organism based on observed toxicity marker (i.e., serum biochemical markers and chemical-specific gene expression) or phenotypic outcome. To date, most toxicogenomic studies concentrated on hepatic toxicity. cDNA microarray analysis enable discrimination of the responses in animals exposed to different classes of hepatotoxicants. In an effort to further characterize the mechanisms of 2, 3, 7, 8,-Tetrachlorodibenzo-p-dioxin (TCDD or dioxin)-mediated toxicity, comprehensive temporal-responsive microarray analyses were performed on hepatic tissue from Sprague-Dawley rats treated with TCDD. Hepatic gene expression profiles were monitored using custom DNA chip containing 490 cDNA clones related with toxicology. Gene expression analysis identified 26 features which exhibited a significant change. In this study, we observed that the genes related with oxidative stress in rats exposed to Dioxin, such as CYPIIA3 and glutathione S-transferase, were up-regulated at 24hr after exposure. In this study, we carried out to discover novel evidence for previously unknown gene expression patterns related to mechanism of hepatic toxicity in rats exposed to dioxin, and to elucidate the effects of dioxin on the gene expression after exposure to dioxin.

The visible injury and physiological responses of three varieties of hot peppers to ozone

  • A growth chamber fumigation was conducted to evaluate the ozone (O3) on the physiology of three hot pepper, Capsicum annuum L. cultivars, 'dabotab', 'buchon' and 'pochungchun'. Thirty-day old plants were exposed to O3 of 120 nl 1-1 in the chambers for 8 h d-1 for 3 days. Foliar damage due to O3 was different from the varieties, 'dabotab'was most sensitive to O3, 'pochungchun' was medium, and 'buchon' was resistant. Ozone symptom on the leaves was bifacial necorsis. Photosynthesis and stomatal conductance were decreased due to O3 treatment, but they were not much different from the variety. Decreases of net photosynthesis by O3 were 56%, 40% and 35% on 'dabotab', 'buchon' and 'pochungchun', respectively Decreases of stomatal conductance by O3 were 66%, 63%, and 50% on each varieties. Ozone closed the stomata and decrease net photosynthesis on hot peppers regardless of the variety. Light curves on the three varieties were showing similar patterns that O3 damage on net photosynthesis were started at the low levels of light with or without the visible injury, Assimilation-internal CO2 concentration curves of the three cultivars were not different due to the treatment. It means there was not significant biochemical damage Inside the leaves by O3. In conclusion, ozone closed the stomata and damaged light capturing system of the pepper leaves with or without the visible damage. Although visible damage of the leaves could be a good indicator of O3 resistance, the ecophysiological change by O3 were not proportional to the amout of visible injuries

AN EXPERIMENTAL STUDY ON TUMOR SPECIFIC ANTIGENS OF RAT SALIVARY GLAND TUMOR INDUCED BY 7, 12-DIMETHYLBENZ(a) ANTHRACENE(DMBA) (7,12-Dimethylbenz(a) anthracene(DMBA)에 의해 유도되는 백서 타액선종양에서의 종양특이항원에 대한 연구)

  • Ever since the expression of new tumor-specific antigens was reported during malignant transformation, studies on separation, purification and characterization of these proteins have been so activated recently. Following experiment was performed to observe tumor-specific antigens by implanting DMBA pellet into submaxillary gland of rat for inducing salivary gland tumor. After dividing 280 rats into 2 groups, in control group, sham operation was performed on right submaxillary gland and, in experimental group, DMBA pellet (5mg) was implanted into right submaxillary gland. Then proteins from excised submaxillary gland by killing 10 rats every two weeks for 28 weeks were extracted with 3M KCl, and SDS-PAGE and PAS-staining were carried out for biochemical examination. The obtained results were summarized as follows; 1) At 12th week since implantation of DMBA pellet, tumor mass formation was inspected. And dysplasia at 6th week and invasive epidermoid carcinoma at 10th week were observed by microscope. 2) In control group, the weight ratio of both submaxillary glands had no any change, however, in experimental group, the ratio was increased remarkably. And at 28th week after DMBA implantation, there was more than 15 times of differences in weight between control and experimental group. 3) There was no DMBA remnant after 22nd experimental week. 4) In the SDS-PAGE, high molecular protein bands (more than 100 kd) were appeared much, and new prominent protein bands (66, 48, 41.5, 39, 37, 37.5 kd) were appeared after 4th week since DMBA implantation. However, 38, 27, 22kd protein bands were disappeared. 5) In PAS-staining, high molecular proteins were proteins were all glycoproteins and 37.5kd protein was proved as to be glycoprotein. And 38kd glycoprotein was disappeared after 4th week since DMBA implantation.

Application of Simple Biosphere Model (SiB2) to Ecological Research (Simple Biosphere Model 2 (SiB2)의 생태학적 응용)

  • The simple biosphere model 2 (SiB2), which is one of the land surface models, simulates the exchange of momentum, energy and mass such as water vapor and carbon dioxide between atmosphere and biosphere, and includes the biochemical sub-model for representation of stomatal conductance and photosynthetical activities. Throughout the SiB2 simulation, the significant information not only to understand of water and carbon budget but also to make an analysis of interaction such as feed-back and-forward between environment and vegetation is given. Using revised SiB2-Paddy, one sample study which is the evaluation of the runoff in Chaophraya river basin according to land use/cover change is presented in this review. Hence, SiB2 is available in order to ecological studied, if revised SiB2 for realistic simulation about soil respiration, computing leaf area index, vegetation competition and soil moisture is improved.

  • Effect of chitosan with sirloin specimens from chitosan-fed and normal-fed pig. The specimens were observed by a optical microscope and their biophoton was measured by a photomultiplier(PMT)tube. Results of experiments showed that specimens of the chitosan-fed pig had lower biophoton rate than that of the normal-fed pig. As a result of the organization were also shown pictures Chitosan as a pork loin tissue and fat in the flesh less delicate and compactness is found that tastes good. Thus by PMT fleshy tissue biochemical measurements to measure the change of state to be one of the new method is to be expected.

Role of hyperforin in diabetes and its associated hyperlipidemia in rats

  • The aim of the present study was to evaluate the possible roles of hyperforin against hyperglycemia, hyperlipidemia and oxidative stress in streptozotocin-induced diabetic rats. Diabetes was induced by a single intraperitoneal injection of streptozotocin (65 mg/kg). Biochemical parameters were measured following hyperforin treatment (10 mg/kg, i.p.) for 7 days. Hyperforin treatment significantly reversed the elevations in plasma glucose, triglycerides, total cholesterol and LDL-cholesterol. Hyperforin also reversed the declines in plasma HDL-cholesterol and liver glycogen, but did not reverse the change in plasma insulin levels when compared to the diabetic control rats. Hyperforin treatment also reversed the oxidative stress induced by streptozotocin. Moreover, the effect of the hyperforin on peripheral glucose utilization in normal rats was evaluated by an oral glucose tolerance test (OGTT). Hyperforin treatment significantly increased (p < 0.05) the glucose tolerance compared to the vehicle in OGTT. The antihyperglycemic, antihyperlipidemic and antioxidant activities of hyperforin (10 mg/kg, i.p.) were comparable qualitatively to glibenclamide (1 mg/kg, p.o.). In conclusion, we report for the first time through an in vivo study that hyperforin is potentially valuable for the treatment of diabetes and its associated hyperlipidemia and oxidative stress by enhancing the glucose utilization by peripheral tissues such as muscle and adipose tissues.

Study on the Resazurin test as a Rapid Method for the Estimation of Maturity of Kimchi (김치숙성도 판정기준을 위한 신속 검사법 Resazurin-test 에 관한 연구)

  • An attempt was made to establish a simple and rapid method for the evaluation of Kimchi quality. Four kinds of Kimchi included one home made traditional Kimchi were stored at $4^{\circ}C$ and$25^{\circ}C$ and biochemical changes, including pH, total acidity, volatile acidity, volatile basic nitrogen, vitamin C, NaCl, glucose, nitrite and resazurin value were determined and tested for the relationship with sensory evaluation. The change in pH, total acidity and resazurin value were highly correlated with the sensory maturity of Kimchi-Resazurin value was strongly influenced by the sample dilution and fermentation temperatures. It was concluded that resazurin test with sample solution of $2{\sim}4%$ could be used as a rapid method for the estimation of the maturity of Kimchi.

Allium Jesdianum Extract Improve AcetaminophenInduced Hepatic Failure through Inhibition of Oxidative/Nitrosative Stress

  • Objectives: Allium jesdianum (Aj) is a medicinal plant that has highlighted pharmacological features. In this study, the effects of Aj extract were examined on acetaminophen (APAP)-induced hepatic failure in rats. Methods: Methanolic fraction of hydro-alcoholic extract of Aj was obtained by silica gel column chromatography method. Animals were randomly divided into four groups each containing six rats and treated by gavage as follows: the first and second groups received normal saline, the third and fourth groups were received with 50 and 100 mg/kg of Aj extract, respectively. After two consecutive weeks, the groups 2-4 were given a single dose of APAP (2 g/kg). After 48 hours, blood and liver samples were collected for biochemical and histological examinations. Results: The findings of the study demonstrated that APAP caused a significant increase in ALT (P < 0.001), AST (P < 0.001), LDH (P < 0.001), ALP (P < 0.001) serum levels, hepatic lipid peroxidation (LPO; P < 0.001) and nitric oxide (NO; P < 0.001). In this regard, APAP led to the depletion of the total antioxidant capacity (TAC; P < 0.001), glutathione and total thiol groups (TTGs; P < 0.001), and structural change in the liver. In the Aj extract groups, a considerable improvement was found in the hepatic function alongside the histopathologic changes. Conclusion: This investigation indicated that the influential effects of Aj extract in APAP-induced hepatic failure might depend on its effect on improving oxidant/antioxidant balance in hepatic tissue.

Twenty-Eight-Day Repeated Inhalation Toxicity Study of Aluminum Oxide Nanoparticles in Male Sprague-Dawley Rats

  • Aluminum oxide nanoparticles ($Al_2O_3$ NPs) are among the most widely used nanomaterials; however, relatively little information about their risk identification and assessment is available. In the present study, we aimed to investigate the potential toxicity of $Al_2O_3$ NPs following repeated inhalation exposure in male Sprague-Dawley rats. Rats were exposed to $Al_2O_3$ NPs for 28 days (5 days/week) at doses of 0, 0.2, 1, and $5mg/m^3$ using a nose-only inhalation system. During the experimental period, we evaluated the clinical signs, body weight change, hematological and serum biochemical parameters, necropsy findings, organ weight, and histopathology findings. Additionally, we analyzed the bronchoalveolar lavage fluid (BALF), including differential leukocyte counts, and aluminum contents in the major organs and blood. Aluminum contents were the highest in lung tissues and showed a dose-dependent relationship in the exposure group. Histopathology showed alveolar macrophage accumulation in the lungs of rats in the $5mg/m^3$ group during exposure and recovery. These changes tended to increase at the end of the recovery period. In the BALF analysis, total cell and neutrophil counts and lactate dehydrogenase, tumor necrosis factor-${\alpha}$, and interleukin-6 levels significantly increased in the 1 and $5mg/m^3$ groups during exposure. Under the present experimental conditions, we suggested that the no-observed-adverse-effect level of $Al_2O_3$ NPs in male rats was $1mg/m^3$, and the target organ was the lung.
