Effects of Garlic on the Blood Lipids and Other Serum Components in Rats (흰쥐에 마늘 투여로 혈액의 지질량과 그외 혈액성분 변화 고찰)

  • In present study possible hypolipidemic effects of garlic were investigated in an experimental model which consisted of 4 groups of Wistar rats(three weeks old): CO group received a basal diet(as a standard diet) contained 6.3% of corn oil. LO group was taken the experimental diet in which only 6.3% corn oil in basal diet was substituted by 12.6% of lard oil. So the experimental diet was somewhat high saturated fat diet. LG1 group was treated orally with garlic juice as 1% of raw garlic in the diet together with the same diet as the diet for LO group. LG2 group was taken 2% of garlic under the same dietary condition as that of LG1 group. The rat body weights prior and posterior to the experimental period were measured and the amount of the experimental dietary intake was determined at every 3 day interval. After the experimental dietary period of 30 days the blood obtained from all the sacrificed rats were analyzed for the biochemical parameters. Over the one month period of experiment there was no abnormality or apparent change in appearance and activity or diet consumption in all experimental rat groups. LO group fed a diet rich in lard showed significantly increased body weight gain rate by 30% and also elevated levels of triglyceride, total cholesterol and LDL cholesterol by 29.9%, 27.4% and 62.3% respectively and no significant difference in the levels of HDL cholesterol, GOT, GPT, blood glucose and blood urea nitrogen as compared to those of control group. So it seemed that the high calorie diet in LO group mainly contributed to the significant increase in body weight gain rate and other biochemical parameters. In spite of 1% garlic treatment LG1 group had significantly increased body weight gain rate by 25.2%, levels of triglyceride, total choesterol and LDL cholesterol by 25.2%, 24.6% and 50.5% respectively as compared to control and LO group, and no increase in the levels of HDL cholesterol, blood glucose and blood urea nitrogen. At garlic 1% diet there was no hypolipidemic efficacy in rat serum. Under the treatment with garlic 2% in diet LG2 group showed significantly increased body weight gain rate by 19.7% as compared to control and elevated levels of GOT, GPT and BUN by 85.8~96.4%, 127~148% and 88% respectively as compared to control, but LG2 group had significantly reduced levels of triglyceride, total cholesterol and LDL cholesterol as compared to LOand LG1 group. In present study there were no significant change in HDL cholesterol and blood glucose value. In LG2 group GOT, GPT and blood urea nitrogen values were slightly increased presumably due to the inhibiting effects of garlic on the hepatic or renal function of rats. Nevertheless in this study garlic may have some demonstrable hypolipidemic effects in rat.

  • This study was to investigate single and repeated-dose toxicities of Tensolin-$F^{(R)}$, an anti-wrinkle agent, in Sprague-Dawley (SD) rats or ICR mice. In single-dose oral toxicity study, the test materials were administered once by gavage to male and female SD rats at dose levels of 0 and 2,000 mg/kg. No dead animals and abnormal necropsy findings were found in control and Tensolin-$F^{(R)}$ treated group. Therefore, the approximate lethal dose of Tensolin-$F^{(R)}$ was considered to be higher than 2,000 mg/kg in rats. In the 4-week repeated oral toxicity study, the test material was administered once daily by gavage to male and female ICR mice at dose levels of 0, 25, 50 and 100 mg/kg/day for 4-weeks. In the results, no abnormality was observed in mortality, clinical findings, body weight changes, food and water consumptions, opthalmoscopic findings, necropsy findings, histopathological findings. In hematological analysis, there was a trend of increase in reticulocyte at male 25 mg/kg, although such changes were in normal ranges. On the other hand, there was a trend of decrease in hemoglobin at female 50, 100 mg/kg, such changes were in normal ranges. In addition, serum biochemical parameters including sodium, BUN and chloride increased at 25, 50 and 100 mg/kg. Relative organ weights of right testis, brain, lung and left epididymis were increased in 100 mg/kg groups of male rats in contrast to not change in female groups. However, these changes of relative organ weights, hematological and serum biochemical parameters were not accompanied with related signs such as histopathological changes or clinical findings. In conclusion, 4-week repeated oral dose of Tensolin-$F^{(R)}$ to ICR mice did not cause apparent toxicological change at the dose of 25, 50, 100 mg/kg body weight. Consequently the no-observed-adverse-effect level (NOAEL) for Tensolin-$F^{(R)}$ in ICR mice following gavage for at least 4-week is higher than 100 mg/kg/day.

The Extracts from Natural Product Complex (DW) Suppress Lipid reroxidation and Inhibit Liver Fibrosis (Cirrhosis) in Rats (실험동물에서 복합천연물(DW)의 지질 과산화 억제와 간섬유화 저해 효과 검색)

  • The chronic cholestasis induce to biliary liver fibrosis (cirrhosis) and the increased products of ROS(reactive oxygen species) cause to the liver damage. In this study ; the antioxidant and antifibrotic effect of dried extracts of oriental medicine (DW) was investigated under the liver fibrotic (cirrhotic) condition. The female Sprague-Dawley rats were divided in 5 groups (Normal, Op-2, Op-4, OpDW-2, OpDW-4). Except for normal group, the rats were induced to biliary liver fibrosis (cirrhosis) by the operation of bile duct ligation/scission (BDU/S) and were observed in 2 weeks or 4 weeks. And the prepared DW was treated p.o.2 ml/day/rats in 2 weeks or 4 weeks for OpDW groups. At the time of sacrifice, the liver, kidney, spleen were weighed and the ratio of organ weight/body weight was calculated. The MDA, the hyp and biochemical parameters (GOT GTP, ALP, t-bili) were measured in sera and liver tissue of rats. The biochemical change was observed on liver tissue. In the result, the hepatomegaly and spleenomegaly appeared in all BDL/S operated rats, and significantly lower liver weight was observed in OpDW-4 group compared with in Op-4 group (p<0.05). The level of clinical parameters in sera of all liver fibrosis (cirrhosis) developed rats was higher than in normal group. Especial1y, the value of GOT in OpDW-2 group and ALP in OpDW-4 group showed significantly lower than in Op-2 group and Op-4 group (p<0.01, p<0.005). The content of hyp in all operation groups was significantly higher than in normal group (p<0.05∼<0.005), and showed significantly lower value in the OpDW-4 group than in Op-4 group (p<0.05). The product of lipid peroxidationUDA) increased significantly under the fibrotic(cirrhotic) condition (p<0.05∼ <0.005), and the MDA value in OpDW-4 group decreased significantly in Op-4 group (p<0.005). The histological change (bile duct proliferation, fibrosis, collagen bundle) was similarly observed in Op-2 group and in OpDW-2 group, but the weak fibrosis and bile duct proliferation were observed in OpDW-4 group compared with in Op-4 group. In conclusion, lipid peroxidation and severe liver damage were activated by bile duct obstruction, and the measurement of MDA and hap can be useful monitor for the screening of antioxidant and antifibrotic effect in experimental liver fibrosis (cirrhosis). The 4 weeks treatment with DW extracts suppressed lipid peroxidation and inhibited fibrotic (cirrhotic) process in BDL/S operated rats.

A Study on GHS Classification of 3-Methylpentane by Subacute Inhalation Toxicity (아급성흡입독성시험을 이용한 3-Methylpentane의 GHS 분류·표시)

  • Objectives : The purpose of this study was to obtain information regarding Globally Harmonized System(GHS) classification and health hazards that may result from a 4 weeks inhalation exposure of 3-Methylpentane in Sprague-Dawley rats. Methods : The testing method was conducted in accordance with OECD guidelines for the testing of chemicals No. 412(Subacute Inhalation Toxicity). The Rats were divided into 4 groups(5 male and 5 female rats in each group) and exposed to 0 ppm, 284 ppm, 1,135 ppm, 4,540 ppm 3-Methylpentane in each exposure chamber for 6 h/day, 5 days/week, for 4 weeks. After two weeks, the test animals were autopsied and carried out blood test and biochemical tests and histopathological examination. We used PRISTIMA (Toxicology data management system) to confirm the system and to have confidence of the raw data. Results : No death and particular clinical presentation including weight change and change of feed rate was observed. Relationship between dose, gender and response was also not significantly changed in hematologic examination, biochemical examination of blood and blood coagulation time. The histopathologic lesions caused by the test substance did not appear. Conclusions : NOAEL(No Observable Adverse Effect Level) of 3-Methylpentane is more than 4,540 ppm in male group and female group and the Ministry of Employment and Labor Guidance Announcement No. 2013-37(criteria for the classification marks and Safety of Chemicals) Specific target organ toxicity(repeated exposure) was determined with a substance that is not the separator material.

Biochemical Methane Potential of Agricultural Byproduct in Greenhouse Vegetable Crops (국내 주요 시설채소 부산물의 메탄 생산 퍼텐셜)

  • Number of crop residues generated at large amount in agriculture can be utilized as substrate in methane production by anaerobic digestion. Greenhouse vegetable crop cultivation that adopting intensive agricultural system require the heating energy during winter season, meanwhile produce waste biomass source for the methane production. The purpose of this study was to investigate the methane production potential of greenhouse vegetable crop residues and to estimate material and energy yield in greenhouse system. Cucumber, tomato, and paprika as greenhouse vegetable crop were used in this study. Fallen fruit, leaf, and stem residues were collected at harvesting period from the farmhouses (Anseong, Gyeonggi, Korea) adopting an intensive greenhouse cultivation system. Also the amount of fallen vegetables and plant residues, and planting density of each vegetable crop were investigated. Chemical properties of vegetable waste biomass were determined, and theoretical methane potentials were calculated using Buswell's formula from the element analysis data. Also, BMP (Biochemical methane potential) assay was carried out for each vegetable waste biomass in mesophilic temperature ($38^{\circ}C$). Theoretical methane potential ($B_{th}$) and Ultimate methane potential ($B_u$) off stem, leaf, and fallen fruit in vegetable residues showed the range of $0.352{\sim}0.485Nm^3\;kg^{-1}VS_{added}$ and $0.136{\sim}0.354Nm^3\;kg^{-1}VS_{added}$ respectively. The biomass yields of residues of tomato, cucumber, and paprika were 28.3, 30.5, and $21.5Mg\;ha^{-1}$ respectively. The methane yields of tomato, cucumber, and paprika residues showed 645.0, 782.5, and $686.8Nm^3\;ha^{-1}$. Methane yield ($Nm^3\;ha^{-1}$) of crop residue may be highly influenced by biomass yield which is mainly affected by planting density.

  • Recently, anaerobic methane production of agricultural waste biomass has received increasing attention. Until now domestic BMP (Biochemical methane potential) studies concerned with agricultural waste biomass have concentrated on the several waste biomass such as livestock manure, food waste, and sewage sludge from WWTP (Waste water treatment plant). Especially, the lack of standardization study of BMP assay method has caused the confused comprehension and interpretation in the comparison of BMP results from various researchers. Germany and USA had established the standard methods, VDI 4630 and ASTM E2170-01, for the analysis of BMP and anaerobic organic degradation, respectively. In this review, BMP was defined in the aspect of organic material represented as COD (Chemical oxygen demand) and VS (Volatile solid), and the influence of several parameters on the methane potential of the feedstock was presented. In the investigation of domestic BMP case studies, BMP results of 18 biomass species generating from agriculture and agro-industry were presented. And BMP results of crop species reported from foreign case studies were presented according to the classification system of crops such as food crop, vegetables, oil seed and specialty crop, orchards, and fodder and energy crop. This review emphasizes the urgent need for characterizing the innumerable kind of biomass by their capability on methane production.

The Effects of Changing Duty Cycle With Electrical Stimulation on Blood Lactate and Plasma Enzyme (전기자극 시 활동주기 형태의 변화가 혈중젖산과 혈장효소에 미치는 영향)

  • By measuring changes in blood lactate and plasma enzyme (CPK, GOT, GPT) with electrical stimulation applied at two duty cycles, this study is intended to look into which type of duty cycle may have more effects on blood lactate and plasma enzyme constituents through animal experiment so as to determine any duty cycle appropriate for electrical treatment. In this study, electrical stimulation was applied to total 20 Korean house rabbits (weight: 3~3.5 kg) by means of an electrical therapeutic apparatus called TS6000 (made in Netherlands) at duty cycle of 50% and 20% respectively for 30 minutes. Here, 5 cc of blood was collected from their carotid artery before stimulation and in 30 minutes after stimulation respectively to carry out biochemical experiment and analysis. As determined through the above experiment, blood lactate rate was increased to 333.07% at 50% duty cycle after experiment and 185.71% at 20% duty cycle after experiment respectively. In both cases, blood lactate rate was significantly increased to higher level after electrical stimulation than before. Moreover, the rate of change in the average of blood lactate rate at both duty cycles also showed significant differences. CPK rate was boosted to 301.82% at 50% duty cycle after experiment and 321.35% at 20% duty cycle after experiment respectively. In both cases, CPK rate was remarkably boosted to higher level after stimulation than before (p<.05). However, there was not any significant difference in the rate of change in average CPK at both duty cycles (p<.05). GOT rate was significantly boosted up to 38.97% at 50% duty cycle after experiment (p<.05), while it was slightly increased to 1.68% at 20% duty cycle after experiment without any significant difference. Rather, GPT rate dropped slightly at both duty cycles after experiment, but there was not any significant difference. Although blood lactate and GOT were relatively less generated at 20% duty cycle after electrical stimulation than at 50% duty cycle, the change of duty cycle didn't have any significant influence on CPK rate. In this regard, this study failed to come any consistent conclusion about the association between change of duty cycle and muscle fatigue. Therefore, it is advisable that follow-up studies seek various ways to a little more effectively apply electrical stimulation to laboratory animals by avoiding their muscle fatigue. GOT rate was significantly boosted up to 38.97% at 50% duty cycle after experiment (p<.05), while it was slightly increased to 1.68% at 20% duty cycle after experiment without any significant difference. Rather, GPT rate dropped slightly at both duty cycles after experiment, but there was not any significant difference. Effects of a Combined Diet of Jerusalem Artichoke's Inulin, Lotus Leaf and Herb Extracts in Obesity-induced White Rat with Fat Diet (돼지감자의 이눌린, 연잎, 허브의 병합식이가 고지방식이로 유도된 비만흰쥐에 미치는 영향)

  • A preliminary diet experiment utilizing Jerusalem artichoke's inulin, lotus leaf powder, nettle powder and eucalyptus powder extract indicated that combining all four elements gave the most effective result. Therefore, a study was conducted to evaluate the effectiveness of combined diet for weight loss. In this study, Sprague-Dawley, male white rats about 200 g in weight was fed with high fat diet for 8 weeks in order to induce obesity followed by 4 week administration of combined diet to look into the effect of the diet. After a total of 12 weeks of feeding, factors relevant to weight, blood, and lipid metabolism by liver in the body were researched and histologic change was examined with optical microscope. In terms of weight change, both high fat diet group and regular diet group gained weight from high fat diet for 8 weeks compared to normal group. Then, for another 4 weeks, while normal group and high fat diet group kept gaining weight, combined diet group which was provided with high fat diet for 8 weeks, lost weight to the normal group level after 3 week administration of diet. However, after the 4th week of administration, the group weighted significantly less than the normal group and the efficiency of diet also significantly dropped. In the biochemical analysis of blood, total cholesterol, LDL-cholesterol, triglyceride, glucose, GOT, GPT, ${\gamma}-GTP$ and creatine showed significant increase in high fat diet group and there was no significant difference between diet group and normal group except for GPT, ${\gamma}-GTP$ and creatine. In the biochemical analysis of liver, there was significant increase in LDL-cholesterol, triglyceride of high fat diet group compared to normal group, while there was no significant difference in term of total cholesterol and HDL-cholesterol. Compared to normal group, diet group had higher HDL-cholesterol, while total cholesterol dropped significantly. There was no significant difference in terms of LDL-cholesterol and triglyceride. Besides, in high fat diet group, observation of histologic change in liver and change in ultrastructure showed volume increase of hepatic cell and severe fatty degeneration in hepatic cell around hepatic vein. However in diet group, like normal group, no pathological change was observed in terms of cytoplasm, nucleus and capillary in hepatocyte and the alignment of hepatocyte had regularity thanks to the administration of combined diet. Therefore, combined diet utilizing Jerusalem artichoke's inulin, lotus leaf powder, nettle powder and eucalyptus powder was proven to be an effective measure to prevent and improve obesity as a result of abnormal adipose deposition.

Change of the Serum Composition in Irradiated Mice (방사선이 조사된 생쥐의 혈청성분 변화)

  • This study is aimed to investigate the change of serum compositions of mice caused by the irradiation. The 3Gy radiation with 10MeV Linac was once irradiated to whole body of mice and their serum was collected to conduct 14 biochemical analyses. With the collected data, t-test was performed. As the result, the significant change was confirmed in the following 3 compositions. First, the glucose level of the normal control group was $185.43{\pm}14.93$, but the irradiation group was found to be $220.00{\pm}17.58$, which shows significant difference(p<0.001). Second, the BUN(blood urea nitrogen) measurement showed lower value($15.70{\pm}1.48$) in the irradiation group than the normal control group($19.61{\pm}1.65$), which indicates the significant difference in mean value (p<0.01). Third, the measurement of albumin resulted in lower value of $2.89{\pm}0.25$ in irradiation group than $3.19{\pm}0.34$ of the normal control group, which shows the significant difference in mean value(p<0.05). In consequence, the serum of the mice irradiated with 3Gy radiation caused significant change in 3 compositions; glucose, Blood urea nitrogen(BUN) and albumin.

The Effect of Isoflavone Supplementation on Plasma Biochemical Parameters of Women with Polycystic Ovary Syndrome (다낭성 난소 증후군 환자에서 Isoflavone 섭취에 따른 혈중 생화학적 지표의 변화)

  • Objective: This study was performed to evaluate the effect of isoflavone supplementation on hormone levels, lipid profiles and total antioxidant status in patients with polycystic ovary syndrome. Methods: Total 11 women with polycystic ovary syndrome were supplemented daily with 150mg of isoflavone for 6 months. Blood samples were collected 0, 3, and 6 months after supplement of isoflavone for analysis of LH, FSH, E2, testosterone, free testosterone, SHBG levels, serum lipid profiles, and total antioxidant status (TAS). Results: After 6 months isoflavone supplementation, the hormone levels did not change significantly. Serum lipid profiles did not show any significant change in total cholesterol, LDL-cholesterol, triglyceride, lipoprotein(a), and free fatty acid levels. However, there was significant increase in HDL-cholesterol (p<0.05) for 3 months. Total antioxidant status was increased significantly after isoflavone supplementation for 6 months (p<0.05). Conclusion: Isoflavone supplementation showed positive effects on the HDL-cholesterol and total antioxidant status. it is implicated that isoflavone supplementation will may have a effect on cardiovascular disease in patients with polycystic ovary syndrome.
