  • The purpose of this study was to estimate the iron availability and to analyze dietary factors which influence hematological indices of 130 female adolescents with iron deficiency anemia. Intakes of iron and other nutrients were estimated using a self-administrated questionnaire combined with the 24-hour recall mehtod and iron availability was calculated by Monsen's method. Mean daily intakes of calorie, protein and vitamin C were 1631.0kcal(77.7% of RDA), 54.7g(84.2% of RDA) and 45.7mg(83.0% of RDA), respectively. In terms of iron, mean daily intake was 8.7mg(48.3% of RDA) and heme iron intake was 3.0mg which correspond to 34% of total iron intake. The amount of total absorbable iron was 1.5mg and the estimated bioavailability of dietary iron was 17.2%. In summary, intake of several nutrients for most of the subjects were under RDA. Dietary factors affecting hematological indices were analyzed by stepwise multiple regression. Intake of vitamin C was a major determinant of Hb level, while both intake of enhancing factor and iron availability were major determinants of serum ferritin level. In conclusion proper nutritional education and guidance for iron deficiency anemic female adoalescent needs to be developed and to improve their iron storage should be increased intakes of enhancing factors, female adoalescents.

  • Phytoremediation which uses plants to enhance the bioremediation through stimulation of microbial activity and root uptake, has been a topic of increasing interest. Mathematical model were developed that can be applied to various bioremediation methods in the unsaturated zone, especially phytoremediation, for simulating the fate and transport of contaminants under field conditions. A 2-year field study was conducted using 72 (1.5m long and 0.1 m diameter) column lysimeters with four treatments: Johnsongrass; wild rye grass; a rotation of Johnsongrass and wild rye grass; and unplanted fallow conditions. The developed model represented the fate and transport of contaminant both in vegetated and unplanted soils satisfactorily for field applications. Parameters related to the contaminant concentration in the water phase were the main parameters determining the contaminant fate in the vadose zone and indicated that the bioavailability can be the most important factor in the success of phytoremediation as well as bioremediation applications.

  • The pharmacokinetics and tissue distribution of DA-3030 (recombinant human granulocyte colony-stimulating factor, rhG-CSF, recently manufactured by Dong-A research laboratory of Dong-A Pharmaceutical Company) were compared with reported data in the literature. After intravenous(i.v.) administration of DA-3030, at dose of 5, 10 and 100 $\mu\textrm{g}$/kg to rats, some pharmacokinetic parameters, such as terminal half-lives(1.05, 1.19 and 1.83 hr, respectively) and clearance (84.0, 54.8 and 45.5 mι/hr/kg, repectively), were dose-dependent. This could be due to the saturable metabolism of DA-3030 in rats. Similar results were also reported. After subcutaneous(s.c.) and intramuscular(i.m.) administrations of DA-3030, 10 $\mu\textrm{g}$/kg to rats, the extent of bioavailability(absolute bioavailability) were incomplete; the values were 23.3 and 18.2% after s.c. and i.m. injections, respectively, due to the degradation of DA-3030 by protease. After 7-consecutive day i.v. administrations of DA-3030, 10 $\mu\textrm{g}$/kg/day, to rats, the plasma concentrations and pharmacokinetic parameters of DA-3030 were not significantly different from those in single administration. In mice and dogs at DA-3030 dose of 10 $\mu\textrm{g}$/kg, the plasma concentrations of DA-3030 were also declined rapidly with terminal half-lives of 1.31 and 1.15 hr, respectively. DA-3030 was highly concentrated in the kidney after i.v. administration of DA-3030, 10 $\mu\textrm{g}$/kg, to rats, and the results were similar to those obtained using radiolabelled rhG-CSF in the literature. Above data indicate that DA-3030 has similar properties to rhG-CSF manufactured by other companies in view of pharmacokinetics.

  • Implantation is a most important biological process during pregnancy whereby conceptus establishes its survival as well as maintenance of pregnancy. During the periimplantation period, both uterine endometriurn and conceptus synthesize and secrete a host of growth factors and cytokines which mediate the actions of estrogen and /or progesterone and also exert their steroid-independent actions. Growth factors expressed by the materno-conceptal unit en masse have important roles in cell migration, stimulation or inhibition of cell proliferation, cellular differentiation, maintenance of pregnancy and materno-conceptal communications in an autorcrine /paracrine manner. The present review focuses on the role of the intrauterine IGF system during periimplantation conceptus development. The IGF system comprises of IGF- I and IGF- II ligands, types I and II IGF receptors and six or more IGF-binding proteins(IGFBPs). IGFs and IGFBPs are expressed and secreted by uterine endometrium with tissue, pregnancy stage and species specificities under the influence of estrogen, progesterone and other growth factor(s). Conceptus also synthesizes components of the IGF system beginning from a period between 2-cell and blastocyst stages. Maternal IGFs are utilized by both maternal and conceptal tissues; conceptus-derived growth factors are believed to be taken up primarily by conceptus. IGFs enhance the development of both maternal and conceptal compartments in a wide range of biological processes. They stimulate proliferation and differentiation of endometrial cells and placental precursor cells including decidual transformation from stromal cells, placental formation and the synthesis of some steroid and protein hormones by differentiated endometrial cells or placenta. It is also well-documented in a number of experimental settings that both IGFs stimulate preimplantation embryo development. In slight contrast to these, prenatal mice carrying a null mutation of IGF and /or IGF receptor gene do not exhibit any apparent growth retardation until after implantation. Reason (s) for this discrepancy between the knock-out result and the in vitro ones, however, is not known. IGFBPs, in general, are believed to inhibit IGF action within the materno-conceptal unit, thereby allowing endometrial stromal cell differentiation as well as dampening ex cessive placental invasion into maternal tissue. There is evidence, however, indicating that IGFBP can enhance IGF action depending on environrnental conditions perhaps by directioning IGF ligand to the target cell. There is also a third possibility that certain IGFBPs and their proteolytic fragments may have their own biological activities independent of the IGF. In addition to IGFBPs, IGFBP proteases including those found within the uterine tissue or lumen are thought to enhance IGF bioavailability by degrading their substrates without affecting their bound ligand. In this regard, preliminary results in early pregnant pigs suggest that a partially characterized IGFBP protease activity in uterine luminal fluid enhances intrauterine IGF bioavailability during conceptus morphological development. In summary, a number of in vitro results indicate that IGFs stimulates the development of the rnaterno-conceptal unit during the periimplantation period. IGFBPs appear to inhibit IGF action by sequestering their ligands, whereas IGFBP proteases are thought to enhance intrauterine bioavailability of IGFs. Much is remaining to be clarified, however, regarding the roles of the individual IGF system components. These include in vivo evidence for the role of IGFs in early conceptus development, identification of IGF-regulated genes and their functions, specific roles for individual IGFBPs, identification and characterization of IGFBP proteases. The intrauterine IGF club house thus will be paying a lot of attention to forthcoming results in above and other areas, with its door wide-open!

  • The Codex Committee of Contaminants in Food (CCCF) has been discussing a new standard for arsenic (As) in rice since 2010 and a code of practice for the prevention and reduction of As contamination in rice since 2013. Therefore, our current studies focus on setting a maximum level of As in rice and paddy soil by considering bioavailability in the remediation of As contaminated soils. This study aimed to select an appropriate single chemical extractant for evaluating the mobility of As in paddy soil and the bioavailability of As to rice. Nine different extractants, such as deionized water, 0.01 M $Ca(NO_3)_2$, 0.1 M HCl, 0.2 M $C_6H_8O_7$, 0.43 M $HNO_3$, 0.43 M $CH_3COOH$, 0.5 M $KH_2PO_4$, 1 M HCl, and 1 M $NH_4NO_3$ were used in this study. Total As content in soil was also determined after aqua regia digestion. The As extractability of the was in the order of: Aqua regia > 1 M HCl > 0.5 M $KH_2PO_4$ > 0.43 M $HNO_3$ > 0.2 M $C_6H_8O_7$ > 0.1 M HCl > 0.43 M $CH_3COOH$ > deionized water > 1 M $NH_4NO_3$ > 0.01 M $Ca(NO_3)_2$. Correlation between soil extractants and As content in rice was in the order of : deionized water > 0.01 M $Ca(NO_3)_2$ > 0.43 M $CH_3COOH$ > 0.1 M HCl > 0.5 M $KH_2PO_4$ > 1 M $NH_4NO_3$ > 0.2 M $C_6H_8O_7$ > 0.43 M $HNO_3$ > 1M HCl > Aqua regia. BCF (bioconcentration factor) according to extractants was in the order of : 0.01M $Ca(NO_3)_2$ > 1 M $NH_4NO_3$ > deionized water > 0.43 M $CH_3COOH$ > 0.1 M HCl > 0.43 M $HNO_3$ > 0.2 M $C_6H_8O_7$ > 0.5 M $KH_2PO_4$ > 1 M HCl > Aqua regia. Therefore, 0.01 M $Ca(NO_3)_2$ ($r=0.78^{**}$) was proven to have the greatest potential for predicting As bioavailability in soil with higher correlation between As in rice and the extractant.

  • BACKGROUND: Soil utilization pattern can be the main factor affecting soil physico-chemical properties, especially in soil phosphorus (P). Understanding the distribution and bioavailability of P is important for developing management to minimize P release from arable soils to environment. This study was conducted to evaluate the potential bioavailability of soil organic P by using phosphatase hydrolysis method. METHODS AND RESULTS: Twenty-four soils from onion-rice double cropping and 30 soils from plastic film house were selected from Changyeong and Daegok in Gyeongnam province, respectively. The P accumulation pattern (total P, inorganic P, organic P, residual P) and water soluble P were characterized. Commercial phosphatase enzymes were used to classify water-extractable molybdate unreactive P from arable soils into compounds that could be hydrolysed by (i) alkaline phosphomonoesterase (comprising labile orthophosphate monoesters), (ii) a combination of alkaline phosphomonoesterase and phosphodiesterase (comprising labile orthophosphate monoesters and diesters), and (iii) phytase (including inositol hexakisphosphate). Available P was highly accumulated with 616 and 1,208 mg/kg in double cropping system and plastic film house, respectively. Dissolved reactive P (DRP) and dissolved unreactive P (DUP) had similar trends with available P, showing 24 and 109 mg/kg in double cropping and 37 and 159 mg/kg in plastic film house, respectively, indicating that important role of dissolved organic P in the environments had been underestimated. From the result of phosphatase hydrolysis, about 39% and 66% of DUP was evaluated as bioavailable in double cropping and plastic film house, respectively. CONCLUSION(S): Orthophosphate monoester and orthophosphate diester accounted for high portion of dissolved organic P in arable soils, indicating that these organic P forms give important impacts on bioavailability of P released from P accumulated soils.

  • While ranitidine is well known to be absorbed rapidly, the underlying cause of variable bioavailability in intra- and inter-subjects has not been clarified yet. Intestinal permeability is a key controlling factor for oral absorption of highly soluble drugs, In the present study, intestinal ferfusions have been conducted to determine the intestinal permeabilities(Peffs) of ranitidine in the rats, dogs and humans and compared to the estimated fractions of dose absorbed (FAs) in humans. A new in vivo methodology, using a regional segmental perfusion technique, has been used in the dogs and humans. In situ single-pass perfusion experiments have been performed in the rats. In the dog and human studies, perfusion experiments have been conducted on two periods to determine the intrasubject variability, There was low significant intrasubject variation as compared to intersubject variation. The Peffs of ranitidine were 33%, 51%, and 45% inthe rats, dogs and humans, respectively. The FAs were approximately the same for all three species models, suggesting rats and dogs are good animal models for estimating the oral absorption of ranitidine in humans. In addition, the estimated extent of absorption of this drug is consistent with the average bioavailability, indicating that ranitidine has permeability-limited absorption characteristics. • Anthropogenic polycyclic aromatic hydrocarbons (PAHs) are formed by the incomplete combustion of fuels and industrial waste. PAHs can be widely exposed to the environment (water, soil and groundwater). PAHs are potentially toxic, mutagenic and/or carcinogenic. Fundamental studies such as biota uptake (e.g., earthworm and plant) of PAHs are highly needed. It is necessary to develop alternative ways to evaluate bioavailability of PAHs instead of using living organisms because it is time-consuming, difficult to apply in the field, and also exaction method is tedious and time-consuming. In this study, sorption behaviors of phenanthrene were evaluated to predict the fate of PAHs in soils. Moreover, bioaccumulation of PAHs in an artificially contaminated soil was evaluated using pea plant (Pisum sativum) as a bioindicator. A novel passive sampler, organic-diffusive gradient in thin-film (o-DGT) for PAHs was newly synthesized, tested as a biomimic surrogate and compared with plant accumulation. Sorption partitioning coefficient (KP) and sorption capacity (KF) were in the order of natural soil > loess corresponding to the increase in organic carbon content (foc). Biota-to-soil accumulation factor (BSAF) and DGT-to-soil accumulation factor (DSAF) were evaluated. o-DGT uptake was linearly correlated with pea plant uptake of phenanthrene in contaminated soil (R2=0.863). The Tenax TA based o-DGT as a biomimic surrogate can be used for the prediction of pea plant uptake of phenanthrene in contaminated soil.

  • In this study, we synthesized a glycosylated derivative of caffeic acid phenethyl ester (CAPE) using the amylosucrase from Deinococcus geothermalis with sucrose as a substrate and examined its solubility, chemical stability, and anti-inflammatory activity. Nuclear magnetic resonance spectroscopy showed that the resulting glycosylated CAPE (G-CAPE) was the new compound caffeic acid phenethyl ester-4-O-${\alpha}-{\small{D}}$-glucopyranoside. G-CAPE was 770 times more soluble than CAPE and highly stable in Dulbecco's modified Eagle's medium and buffered solutions, as estimated by its half-life. The glycosylation of CAPE did not significantly affect its anti-inflammatory activity, which was assessed by examining lipopolysaccharide-induced nitric oxide production and using a nuclear factor erythroid 2-related factor 2 reporter assay. Furthermore, a cellular uptake experiment using high-performance liquid chromatography analysis of the cell-free extracts of RAW 264.7 cells demonstrated that G-CAPE was gradually converted to CAPE within the cells. These results demonstrate that the glycosylation of CAPE increases its bioavailability by helping to protect this vital molecule from chemical or enzymatic oxidation, indicating that G-CAPE is a promising candidate for prodrug therapy.
