Antioxidant and α-Glucosidase Inhibition Activity of Solvent Fractions from Prunus mume Ethanol Extract (매실 순차분획물의 용매별 항산화 활성 및 α-glucosidase 억제 효과)

  • Prunus mume, known as maesil in Korea, has been widely cultivated in East Asia and used as medication and food. However, because most of the previous studies concerning P. mume had been investigated its under extract state, detailed studies are still required for its extensive utilization. In this study, we evaluated the antioxidant and ${\alpha}-glucosidase$ inhibitory activities of solvent fractions of P. mume ethanol extracts. The ethyl acetate fraction showed higher DPPH radical scavenging activity, ABTS radical scavenging activity, reducing power, and hydrogen peroxide scavenging activity than other fractions. The DPPH radical scavenging activities of ethyl acetate fraction was 67.79%; ABTS radical scavenging activity was 60.03%; reducing power ($OD_{670}$) was 1.26; and hydrogen peroxide scavenging activity was 93.18% at $500{\mu}g/ml$. Also, the ethyl acetate and methanol fraction showed effective levels of ${\alpha}-glucosidase$ inhibition activity (69.25% and 72.29% at $500{\mu}g/ml$). Total polyphenol contents and total flavonoid contents of the ethyl acetate fraction were 88.28 mg/g (gallic acid equivalent) and 70.38 mg/g (quercetin equivalent), respectively. These results suggest that the physiological activities of the ethyl acetate fraction are associated with its polyphenol and flavonoid contents. Therefore, this study can be used as basic data for developing natural antioxidants and potential functional material using P. mume.

An Ethanol Extract of the Brown Seaweed Hizikia fusiformis and Its Active Constituent, Fucosterol, Extend the Lifespan of the Nematode Caenorhabditis elegans (갈조류 톳(Hizikia fusiformis)의 에탄올추출물 및 이의 활성성분 fucosterol에 의한 예쁜꼬마 선충의 수명 연장)

  • The short-lived nematode Caenorhabditis elegans has been used as a model organism for many studies, including lifespan extension. To screen common seaweeds for natural anti-aging agents, the lifespan of C. elegans (N2 wild-type strain) was measured by its hatch rate, growth rate, survival rate, chemotaxis, brood size, and egg-laying time after exposure to nematode growth medium (NGM) containing seaweed extracts. Approximately 30 animals synchronized at the first larval stage were incubated until they reached their adult stages before laying their eggs and were transferred to fresh NGM every 3 days. We also identified the major active compound from the seaweed by gas chromatography-mass spectrometry and tested its optimal dose for longevity. Of 13 common seaweed species, an ethanol extract of the brown seaweed Hizikia fusiformis showed the greatest effect on hatching, growth, and survival rates. The lifespan of C. elegans was significantly expanded 1.54-fold and 1.23-fold in the presence of the ethanol extract (0.05 mg/ml) and the main active component, fucosterol (0.05 mg/ml), respectively. Exposure to the ethanol extract also increased chemotaxis 1.13-fold, decreased brood size 0.74-fold, and shortened egg-laying time 0.96-fold. These results suggest that the aquaculturable H. fusiformis may be a promising source of a diet supplement to support health care.

A Study of the Diversity and Profile for Extracellular Enzyme Production of Aerobically Cultured Bacteria in the Gut of Muraenesox cinereus (갯장어(Muraenesox cinereus) 장으로부터 호기적 조건에서 분리된 미생물의 다양성 및 세포외 효소 생산능 분석에 관한 연구)

  • This research confirmed the diversity and characterization of gut microorganisms isolated from the intestinal organs of Muraenesox cinereus, collected on the Samcheonpo Coast and Seocheon Coast in South Korea. To isolate strains, Marine agar medium was basically used and cultivated at $37^{\circ}C$ and pH7 for several days aerobically. After single colony isolation, totally 49 pure single-colonies were isolated and phylogenetic analysis was carried out based on the result of 16S rRNA gene DNA sequencing, indicating that isolated strains were divided into 3 phyla, 13 families, 15 genera, 34 species and 49 strains. Proteobacteria phylum, the main phyletic group, comprised 83.7% with 8 families, 8 genera and 26 species of Aeromonadaceae, Pseudoalteromonadaceae, Shewanellaceae, Enterobacteriaceae, Morganellaceae, Moraxellaceae, Pseudomonadaceae, and Vibrionaceae. To confirm whether isolated strain can produce industrially useful enzyme or not, amylase, lipase, and protease enzyme assays were performed individually, showing that 39 strains possessed at least one enzyme activity. Especially the Aeromonas sp. strains showed all enzyme activity tested. This result indicated that isolated strains have shown the possibility of the industrial application. Therefore, this study has contributed for securing domestic genetic resources and the expansion of scientific knowledge of the gut microbial community in Muraenesox cinereus of South Korea.

  • Bio-conversion manufacturing technology has been developed to produce ginsenoside Rd which is increasingly in demand as a cosmetic material due to various possibilities related to improving skin function. In order to convert ginsenoside Rb1 which is contained in red ginseng saponin (RGS) into Rd, several commercial enzymes were tested. Viscoflow MG was found to be the most efficient. In order to optimize the conversion of RGS to ginsenoside Rd by enzymatic transition was carried out using response surface methodology (RSM) based on Box-Behnken design (BBD). The main independent variables were RGS concentration, enzyme concentration, and reaction time. Conversion of ginsenoside Rd was performed under 17 conditions selected according to BBD model and optimization conditions were analyzed. The concentration of the converted ginsenoside Rd ranged from 0.3113 g/L to 0.5277 g/L, and the highest production volume was obtained under condition of reacting 2% RGS and 1.25% enzyme for 13.5 hours. Consequently, RGS concentration, enzyme concentration which is 0.05 less than p-value and among the interactions between the independent variables, the interaction between enzyme concentration and reaction time was confirmed to be the most influential.

Analysis of nutrients and antioxidants of sterilized and non-heat-pressed perilla oil (살균 및 비가열압착한 들깨오일의 영양성분 및 항산화 분석)

  • In this study, the nutritional properties of sterilized and non-heat-pressed raw perilla oil (SRPO) were studied and its potential as a functional food was evaluated. The copper, cobalt, and calcium levels were high in sterilized and SRPO. The total polyphenol content and ABTS radical scavenging activity were the highest in SRPO, whereas nitrite scavenging activity was the highest in 45℃ cold pressed perilla oil (CPPO). The above results confirmed that sterilized and non-heat-pressed perilla oil had high mineral and total polyphenol contents, as well as ABTS radical scavenging activity and nitrite scavenging ability. The peroxide value of SRPO decreased as the storage period increased, and the acid value of low-temperature pressed perilla oil over 65℃ (LPPO) significantly increased. This work also provided an opportunity to develop a new method for manufacturing perilla oil, and it is hoped that these experiments will form a basis for the commercialization of perilla oil.

Effect of Different Packaging Atmosphere on Microbiological Shelf Life, Physicochemical Attributes, and Sensory Characteristics of Chilled Poultry Fillets

  • This trial was conducted to evaluate the effect of overwrap, vacuum, and modified atmosphere packaging (MAP) on poultry breast fillets' microbiological, biochemical shelf life and sensory attributes. The fillets were divided into 4 groups, and each of the treatments was replicated 3 times with 60 breast fillets. The first group was a control group with overwrap packaging; the second group was vacuum packed (VP); the third and fourth groups were MAP-1: 0% O2, 40% CO2, 60% N2, and MAP-2: 20% O2, 40% CO2, 40% N2. The microbiological and biochemical analyses were performed for the total viable count, coliform count, Pseudomonas count, Salmonella count, total volatile basic nitrogen (TVB-N), pH, cooking loss, color, lipid oxidation, tenderness, and sensory analysis. The data were analysed through two-way ANOVA by Minitab (Minitab 17.3.1). Meat treated with understudy MAP compositions and vacuum packaging reduced total viable count, Pseudomonas count, and total coliform count than control (p<0.05). TVB-N remained below the recommended limit throughout storage except aerobic packaging (p<0.05). Cooking loss (%) was lowered and showed non-significant results (p>0.05) between vacuum packaging and both MAP concentrations. The meat stored in MAP-2 was characterised by higher (p<0.05) visual scores. Whilst MAP-1 showed higher (p<0.05) L* values and overall acceptability. Sample packaged under aerobic packaging showed significant (p<0.05) results for b* and thiobarbituric acid reactive substances (TBARS). Meat stored in aerobic packaging showed higher (p<0.05) shear force values. The outcome of this trial may help to promote the application of understudy MAP compositions and rapid detection of microbes by biochemical analysis under local conditions.

3-Acetyl-11-Keto-Beta-Boswellic Acid from Boswellia serrata Attenuates Monosodium Iodoacetate-induced Osteoarthritis by Chondroprotective and Anti-inflammatory Effects (Monosodium iodoacetate로 유발된 골관절염 쥐에 유향(乳香) 성분 3-Acetyl-11-Keto-Beta-Boswellic Acid의 연골보호 및 항염증 효과)

  • Objectives : 3-Acetyl-11-keto-𝛽-boswellic acid (AKBA) is a major active compound in Boswellia serrata. We investigated the arthritic changes following AKBA administration in monosodium iodoacetate (MIA)-induced osteoarthritis rats. Methods : All rats were randomly divided into five groups: Normal, Control, INDO (indomethacin 2 mg/kg treated), AKBA30 (AKBA 30 mg/kg treated), and AKBA60 (AKBA 60 mg/kg treated); drugs were given 2 weeks before MIA injection. For all groups except the normal group, 50 µL of sterile saline with MIA (80 mg/mL) was injected into the right knee joint 2 weeks after drug administration. The drug administration was continued for 4 weeks from 1 week after osteoarthritis induction. The histomorphological changes of knee joint cartilage were observed by H&E staining. Also, the levels of glycosaminoglycan (GAG), cartilage oligomeric matrix protein (COMP), 5-lipoxygenase (5-LOX), 5-LOX-activating protein (FLAP), and leukotriene B4 (LTB4) in the knee joint were determined by the ELISA kits. The expressions of mitogen-activated protein kinases (MAPKs), inflammatory cytokines, and matrix metalloproteinases (MMPs) in knee joint were detected by Western blot. Results : Data show that levels of 5-LOX, FLAP, LTB4, and COMP were downregulated significantly in the AKBA treated groups when compared to those in the Control group. On the other hand, GAG levels were significantly elevated. As a result of Western blot, the AKBA-treated groups significantly inhibited phosphorylation of MAPKs. In addition, significant downregulation of the expression of inflammatory cytokines and MMPs was found in the AKBA-treated groups. Conclusion : Our findings suggest that administration of AKBA could exert better chondroprotective and anti-inflammatory effects for MIA-induced osteoarthritis rats.

Toxicological Assessment to Environmental Stressors Using Exoskeleton Surface Roughness in Macrophthalmus japonicus: New Approach for an Integrated End-point Development (칠게 외골격 표면 거칠기를 이용한 노출 독성 평가: 새로운 융합적 연구)

  • Intertidal mud crab (Macrophthalmus japonicus) is an organism with a hard chitinous exoskeleton and has function for an osmotic control in response to the salinity gradient of seawater. Crustacean exoskeletons change in their natural state in response to environmental factors, such as changes in the pH and water temperature, and the presence of pollutant substances and pathogen infection. In this study, the ecotoxicological effects of irgarol exposure and heavy metal distribution were presented by analyzing the surface roughness of the crab exoskeleton. The exoskeleton surface roughness and variation reduced in M. japonicus exposed to irgarol. In addition, it was confirmed that the surface roughness and variation were changed in the field M. japonicus crab according to the distribution of toxic heavy metals(Cd, Pb, Hg) in marine sediments. This change in the surface roughness of the exoskeleton represents a new end-point of the biological response of the crab according to external environmental stressors. This suggests that it may affect the functional aspects of exoskeleton protection, support, and transport. This approach can be utilized as a useful method for monitoring the aquatic environment as an integrated technology of mechanical engineering and biology.

Evaluation of Antithrombosis and Antioxidant Activities of the Ethanol Extract of Different Parts of Hibiscus cannabinus L. cv. 'Jangdae' (케나프 장대 품종의 부위별 에탄올 추출물의 항혈전 및 항산화 활성)

  • Kenaf (Hibiscus cannabinus L.), one of the four major fiber crops, is attracting attention for its efficient CO2-absorbing ability and versatility for producing daily supplies, including textiles. In Korea, a new cultivar 'Jangdae' was established in 2013. The ease of cultivation and seed gathering of 'Jangdae' has led to its nationwide cultivation. However, evaluation of the bioactivities of the different parts of kenaf, and especially the 'Jangdae' cultivar, remains rudimentary. In this study, the antithrombosis and antioxidant activities of extracts prepared from different parts of the 'Jangdae' cultivar were evaluated by determining their effects on blood clot formation. Extracts prepared from seeds (HC-SD), seedpods (HC-SP), leaves (HC-L), stems (HC-S), and roots (HC-R) of the 'Jangdae' cultivar strongly inhibited blood clot formation. In particular, the HC-SD, HC-SP, and HC-S extracts showed strong inhibition against the coagulation factors prothrombin, and thrombin. The HC-SP extract showed strong antioxidant activities, such as scavenging ability against DPPH anion, ABTS cation, nitrite, and reducing power. Since blood clot formation is closely related to oxidative stress, the HC-SP extract could be developed as a novel anticoagulation and antioxidant resource. This is the first report of the antithrombosis activities of different parts of H. cannabinus L. cv. 'Jangdae'.

Hot-melt extruded copper sulfate affects the growth performance, meat quality, and copper bioavailability of broiler chickens

  • Objective: This study was conducted to evaluate the effects of the supplementation of diets of broiler chickens with hot-melt extruded CuSO4 (HME-Cu) on their growth performance, nutrient digestibility, gut microbiota, small intestinal morphology, meat quality, and copper (Cu) bioavailability. Methods: A total of 225 broilers (Ross 308), one-day old and initial weight 39.14 g, were weighed and distributed between 15 cages (15 birds per cage) in a completely randomized experimental design with 3 treatments (diets) and 5 replicates per treatment. Cages were allotted to three treatments including control (without supplemental Cu), IN-Cu (16 mg/kg of CuSO4), and HME-Cu (16 mg/kg of HME processed CuSO4). Results: The HME-Cu treatment tended to increase the overall body weight gain (p<0.10). The apparent digestibility of Cu was increased by supplementation of HME-Cu at phase 2 (p<0.05). The Escherichia coli count in cecum tended to decrease with the supplementation with Cu (p<0.10). In addition, the HME-Cu treatment had a higher pH of breast meat than the control and IN-Cu treatments (p<0.05). Significant increases in the cooking loss, water-holding capacity, and lightness in the breast were observed in the HME-Cu treatment compared to the control (p<0.05). The Cu content of excreta increased with the Cu supplementation (p<0.05). The concentration of excreta Cu in broilers was decreased in the HME-Cu compared to the IN-Cu in phase 2 (p<0.05). The Cu concentration in the liver was increased with the HME-Cu supplementation, compared with the control diets (p<0.05). Conclusion: This study showed that HME-Cu supplementation at the requirement level (16 mg/kg diets) in broiler diets did not affect the growth performance and the physiological function of Cu in broilers. However, supplementation of Cu in HME form improved the meat quality and the bioavailability of Cu.
