Bio-vector Generation Framework for Smart Healthcare

  • In this paper, by managing the biometric data is changed with the passage of time, a systematic and scientifically propose a framework to increase the bio-vector generation efficiency of the smart health care. Increasing the development of human life as a medicine and has emerged smart health care according to this. Organic and efficient health management becomes possible to generate a vector when the biological domain to the wireless communication infrastructure based on the measurement of the health status and to take action in accordance with the change of the physical condition. In this paper, we propose a framework to create a bio-vector that contains information about the current state of health of the person. In the proposed framework, Bio vectors may be generated by collecting the biometric data such as blood pressure, pulse, body weight. Biometric data is the raw data from the bio-vector. The scope of the primary data can be set to active. As the collecting biometric data from multiple items of the bio-recognition vectors may increase. The resulting bio-vector is used as a measure to determine the current health of the person. Bio-vector generating the proposed framework, it can aid in the efficiency and systemic health of healthcare for the individual.

Expression of Bacillus macerans Cyclodextrin Glucanotransferase in Bacillus subtilis

  • A plasmid vector was constructed for the expression and secretion of Bacillus macerans cyclodextrin glucanotransferase (CGTase) in Bacillus subtilis. The vector, pUBACGT, was composed of the ribosome-binding sequence, signal sequence, and cgt gene from B. macerans under the control of amyR2, the promoter of amyE gene coding for $\alpha$-amylase from B. subtilis var. natto. Bacillus subtilis LKS88, a mutant strain lacking genes for an amylase and two proteases, was used as a host for the transformation of the plasmid vector. The transformants were selected on kanamycin-containing Luria-Bertani plates. The starch hydrolyzing activity was observed on the starch-containing plates by the iodine method and cyclodextrin-forming activity was detected in the culture medium. A SDS-PAGE analysis showed that most of the expressed CGTase in the recombinant B. subtilis was secreted into the medium at a high expression level.

Bacillus subtilis as a Tool for Screening Soil Metagenomic Libraries for Antimicrobial Activities

  • Finding new antimicrobial activities by functional metagenomics has been shown to depend on the heterologous host used to express the foreign DNA. Therefore, efforts are devoted to developing new tools for constructing metagenomic libraries in shuttle vectors replicatable in phylogenetically distinct hosts. Here we evaluated the use of the Escherichia coli-Bacillus subtilis shuttle vector pHT01 to construct a forest-soil metagenomic library. This library was screened in both hosts for antimicrobial activities against four opportunistic bacteria: Proteus vulgaris, Bacillus cereus, Staphylococcus epidermidis, and Micrococcus luteus. A new antibacterial activity against B. cereus was found upon screening in B. subtilis. The new antimicrobial agent, sensitive to proteinase K, was not active when the corresponding DNA fragment was expressed in E. coli. Our results validate the use of pHT01 as a shuttle vector and B. subtilis as a host to isolate new activities by functional metagenomics.

Construction of a Hexapeptide Library using Phage Display for Bio-panning

  • Random hexapeptide library on the surface of filamentous bacteriophage was constructed using the SurfZAP vector. The size of the library was approximately 105. The peptide insert was flanked by two cysteines to constrain the peptide structure with a disulfide bond. This library was screened for the topoisomerase II binding peptide. Dramatic enrichment of the fusion phage over the VCS M13 helper phage was demonstrated by bio-panning affinity selection.

  • Xenopus oocyte을 이용하여 식물 aquaporin 단백질의 물 흡수 활성을 측정하기 위한 최적의 조건을 확립하기 위하여 애기장대의 AtPIP2-1유전자를 클로닝하여 cRNA 제작용 vector, buffer osmolarity, hypoosmotic shock 처리시간, 발현 단백질의 localization등을 검토한 결과, Xenopus ${\beta}$-globin 유전자의 5'과 3' UTR(untranlation region)'염기서열을 갖고 있는 pGEMHE vector가 단백질 생산에 더욱 효과적이며, 이 vector를 시용하였을 경우 hypoosomotic stress는 1/2ND buffer에서 6분간 처리시 가장 큰 차이를 볼 수 있었으며, 애기장대 AtPIP2-1단백질과 GFP를 결합시켜 발현시킬 경우 GFP가 plasmamembrane에 위치하는 것을 보아 올바른 subcelluar localization이 이루어지는 것을 확인할 수 있었다.

  • 본 논문은 사용자의 생체 정보(맥박, 이완기 혈압, 수축기 혈압, 혈당)를 무선 센서들을 통하여 획득한 후 스트레스 지수에 따른 감성을 인식하여 대응되는 컬러와 음원을 분류하는 시스템으로서, 맥박 센서, 혈압 센서, 혈당 센서 등의 입력치를 받아 데이터베이스에 저장한 후 다중 SVM(Support Vector Machine) 알고리즘을 이용하여 스트레스 지수에 따른 감성을 분류한다. 2,000개의 데이터 집합을 사용하여 다중 SVM 알고리즘을 학습한 결과 약 87.7%의 정확도를 가졌다.

  • Drug delivery to the brain is facilitated by the synthesis of chimeric peptides, where in neuropharmaceuticals are linked to a vector such as an antibody to the transferrin receptor that mediates transcytosis through the blood-brain barrier (BBB). When disulfide linkers are used in the chimeric peptide, it is crucial that the S-S bridge is stable during transit and that cleavage does not occur prematurely within endothelial cells, as the peptide drug moiety would then be sequestered by the BBB instead of passing through it. The present study addressed that problem. As a model drug a metabolically stable opioid peptide, [$^3$H]DALDA (Tyr-dArg-Phe-Lys-NH$_2$), was used. It was monobiotinylated with NHS-SS-biotin to yield bio-[$^3$H]DALDA. The biotinylated peptide was bound to the vector OX26-SA which is a covalent conjugate of OX26 and streptavidin (molar ratio = 1: 1). In vitro treatment of the chimeric peptide, bio-[$^3$H]DALDA/OX26-SA, with a reducing agent, dithiothreitol, released the labeled peptide from the vector by conversion of bio-[$^3$H]DALDA to the desbiotinylated derivative, desbio-[$^3$H]DALDA.

  • Conventional support vector machines (SVMs) find optimal hyperplanes that have maximal margins by treating all data equivalently. In the real world, however, the data within a data set may differ in degree of uncertainty or importance due to noise, inaccuracies or missing values in the data. Hence, if all data are treated as equivalent, without considering such differences, the optimal hyperplanes identified are likely to be less optimal. In this paper, to more accurately identify the optimal hyperplane in a given uncertain data set, we propose a membership-induced distance from a hyperplane using membership values, and formulate three kinds of membership-induced SVMs.

Biological Synthesis of 7-O-Methyl Apigenin from Naringenin Using Escherichia coli Expressing Two Genes

  • Within the secondary metabolite class of flavonoids, which consist of more than 10,000 known structures, flavones define one of the largest subgroups. The diverse function of flavones in plants as well as their various roles in the interaction with other organisms offers many potential applications including in human nutrition and pharmacology. We used two genes, flavone synthase (PFNS-l) that converts naringenin into apigenin and flavone 7-O-methyltransferase (POMT-7) that converts apigenin into 7-O-methyl apigenin, to synthesize 7-O-methyl apigenenin from naringenin. The PFNS-l gene was subcloned into the E. coli expression vector pGEX and POMT-7 was subcloned into the pRSF vector. Since both constructs contain different replication origins and selection markers, they were cotransformed into E. coli. Using E. coli transformants harboring both PFNS-l and POMT-7, naringenin could be converted into 7-O-methyl apigenin, genkwanin.

  • In this research, the effect of normal load, sliding velocity, and texture density on thefriction coefficient of surfaces micro-textured on AISI 4140 under paraffin oil lubrication were investigated. The predicted tribological behavior by numerical calculation can be serves as guidance for the designer during the machine development stage. Therefore, in this research friction coefficient prediction model based on response surface methodology (RSM), support vector machine (SVM), and artificial neural network (ANN) were developed. The experimental result shows that the variation of load, speed and texture density were influence the friction coefficient. The RSM, ANN and SVM model was successfully developed based on the experimental data. The ANN model can effectively predict the tribological characteristics of micro-textured AISI 4140 in paraffin oil lubrication condition compare to RSM and SVM.
