Growth Performance of Chinese Cabbage using Soilless Cultivation Method

  • Growing plant in potting media without soil is known as Soilless cultivation. This method is used mostly in greenhouse cultivation to increase horticultural commodities production. Peat moss is commonly utilized as potting media substrate because of its characteristic. However, peat moss price is high because of the quantity of peat moss in nature has been decreased. Recently, most of the research is conducted to find the alternative growing medium to cultivate horticulture plant in potting media. Perlite and rice husk ash were mentioned that had a potent as alternative growing media for seasonal plants to increase agriculture production due to the lack of production area. This research aims to determine the growth of in rice husk ash, perlite and peat moss as growing substrates. The method used was the soilless cultivation. The chinese cabbage was planted in the pot with perlite media, rice husk ash media, and peat moss media. The chinese cabbage was measured after 35 days after planting. The result showed that peatmoss was more potentials in chinese cabbage growth performance than rice husk ash and perlite. Peat moss had the significant result of every research parameters such as plant height, plant weight, number of leaves, plant diameter, root length, and root weight. The best alternative for cultivation chinese cabbage without substrate based on this research was peat moss then rice husk ash and perlite.

Molecular and Phenotypic Investigation on Antibacterial Activities of Limonene Isomers and Its Oxidation Derivative against Xanthomonas oryzae pv. oryzae

  • Xanthomonas oryzae pv. oryzae (Xoo) causes a devastating bacterial leaf blight in rice. Here, the antimicrobial effects of ᴰ-limonene, ᴸ-limonene, and its oxidative derivative carveol against Xoo were investigated. We revealed that carveol treatment at ≥ 0.1 mM in liquid culture resulted in significant decrease in Xoo growth rate (> 40%) in a concentration-dependent manner, and over 1 mM, no growth was observed. The treatment with ᴰ-limonene and ᴸ-limonene also inhibited the Xoo growth but to a lesser extent compared to carveol. These results were further elaborated with the assays of motility, biofilm formation and xanthomonadin production. The carveol treatment over 1 mM caused no motilities, basal level of biofilm formation (< 10%), and significantly reduced xanthomonadin production. The biofilm formation after the treatment with two limonene isomers was decreased in a concentration-dependent manner, but the degree of the effect was not comparable to carveol. In addition, there was negligible effect on the xanthomonadin production mediated by the treatment of two limonene isomers. Field emission-scanning electron microscope (FE-SEM) unveiled that all three compounds used in this study cause severe ultrastructural morphological changes in Xoo cells, showing shrinking, shriveling, and holes on their surface. Moreover, quantitative real-time PCR revealed that carveol and ᴰ-limonene treatment significantly down-regulated the expression levels of genes involved in virulence and biofilm formation of Xoo, but not with ᴸ-limonene. Together, we suggest that limonenes and carveol will be the candidates of interest in the development of biological pesticides.

Optimization of Indole-3-Acetic production by phosphate solubilization bacteria isolated from waste mushroom bed of Agaricus bisporus

  • A total of 35 phosphate solubilizing bacterial strains were isolated from waste mushroom bed of Agaricus bisporus in Buyeo-Gun, Chungnam and screened for the production of indole acetic acid (IAA). The best IAA producing strain was identified as Pantoea rodasii using 16S rRNA analysis. In addition to the IAA production, this strain could act as an efficient phosphate solubilizer (1100 ${\mu}g$ $ml^{-1}$ after 5 days of incubation) also. The selected strain was cultured under different conditions in order to assess the optimum conditions for maximum IAA production. The nutrient broth (NB) medium was recorded as the best medium, where the maximum IAA production (229 ${\mu}g$ $ml^{-1}$) was recorded at the start of stationary phase (12 hours after inoculation) of the bacteria growth. The performance of the strain was found to be maximum at the temperature of $30^{\circ}C$ followed by $25^{\circ}C$. IAA production was found to be increased with increasing tryptophan concentration (from 0.1 to 0.6%), however beyond this limit, a slight reduction in IAA production was observed. The strains' ability to produce IAA was further confirmed by extraction of crude IAA and subsequent TLC analysis. A specific spot from the extracted IAA preparation was found corresponding with the standard spot of IAA with same $R_f$ value. The results of HPLC analysis conducted in identifying and quantifying the IAA production more precisely, are in agreement with the results of the assessment done with colorimetric method. As revealed by the results of the pot experiment, the isolated strain could significantly enhance the growth (as measured by shoot and root growth) of mung bean plants compared to that of non-inoculated plants. Therefore it can be concluded that the present strain, Pantoea rodasii has great potential to be used as bio-inoculants.

EFFECTS OF INTERCROPPING, SEEDLING RATE AND FERTILIZER ON FODDER PRODUCTION IN THE LOW LYING AREA OF BANGLADESH

  • The study was conducted at low lying areas in Pabna Sirajgong districts of Bangladesh. To observe the potentiality of bio-mass production two trials were conducted. In first trial maize intercropped with Khesari taking 15 experimental plots of each size $5m{\times}5m$ were arranged in 5 blocks having homogenous soil characteristics. The study showed that the bio-mass yield of sole maize and sole Khesari were 35.25 t/ha. and 56.80 t/ha. respectively and there was a significant difference (p < 0.01) among them. The results also showed that bio-mass yield of maize and Kherasi was higher ($70.04{\pm}6.25t/ha$, $98.88{\pm}10.77t/ha$ and $80.56{\pm}9.5t/ha$) compared to sole maize and sole Khesari and land equivalent ratio was also lower. For second trial, one hectare of land was divided into 16 experimental plots with 4 replications in each plot. Four levels of urea (0 kg/ha, 30 kg/ha, 45 kg/ha, and 60 kg/ha.) were applied to experimental plot. The seed rates were 98.8 kg/ha (farmer's practice), 86.45 kg/ha, 74.1 kg/ha and 61.75 kg/ha. average bio-mass yield of matikalai at different seed rates along with urea fertilizer ranged from 38.49 t/ha, to 65.35 t/ha. the highest seed rate along with highest fertilizer also correspond to the peak production (65.35 t/ha) and the lowest seed rate (61.75 kg/ha) along with the lowest fertilizer rate (30 kg/ha.) showed lowest production (38.49 t/ha.). Here, it was found that the bio-mass yield of matikalai increased with the incremental amount of seed, indicating significant effect (p < 0.05) of seed rates on the bio-mass yield of matikalai. On the other hand, fertilizer doses in different treatment combinations had significant effect (p < 0.05) on bio-mass yield. Two levels of seed rates at zero level of fertilizer were recommended : 86.45 kg/ha for the resource rich farmers and 61.75 kg/ha for the resource poor farmers.

  • 경제성분석 기능을 가진 생물공정 flowsheeting 소프트웨어인 BioPro Designer를 이용하여 외부단 백질 생산 시스템의 최적 공정설계가 수행되었다. BioPro Designer를 이용하여 Salmonella phage P P22의 tailspike protein의 생산시스템에 적용되었 는데 최적공정을 선택하는 과정에서 투입되는 시간 과 노력에 획기척인 개선이 있었다. 숙주의 내부에 서 생산되는 외부 단백질의 folding 특성이 가장좋 은 생산시스템을 선택하는데 영향을 미 친다는 것을 보여주기위하여 wild type의 tailspike와 두개의 다른 변종 tailspike들, 즉 tsf G244→R과 Su A334 -V을 비교 검토되었다. 회수된 tailspike 단백질의 단위질량 대비 자본투자비를 비교 척도로 하여 최척 의 tailspike 생산 시스템이 선택되었다.
