Monitoring of Organoleptic and Physical Properties on Preparation of Oriental Melon Jelly (참외젤리 제조에 따른 관능적 및 물리적 특성 모니터링)

  • To prepare oriental melon jelly having highly favorite, oranoleptic and physical properties of jelly with special to the content of glucomannan, oriental melon concentration juice and vinegar was monitored by four-dimensional response surface methodology. Mouth-feel showed maximum sensory score in 0.09% glucomannan, 3.17% oriental melon concentration juice and 1.25% oriental melon vinegar. Organoleptic taste showed maximum sensory score in 0.12% glucomannan, 3.64% oriental melon concentration juice and 0.91% oriental melon vinegar. Overall palatability showed also maximum sensory score in 0.10% glucomannan, 3.07% oriental melon concentration juice and 1.34% oriental melon vinegar. The sensory socore of overall palatability was more than 7.0 in the texture ranges of 0.20~0.31 cm/g (softness) and 8.0~12.5 gㆍcm (jelly strength).

In vivo Comet Assay on Flounder and Clam Exposed to BaP and TBT (BaP 및 TBT에 노출된 넙치와 개조개의 in vivo Comet assay)

  • The comet assay, also called single-cell electrophoresis (SCGE) assay, is a potential sensitive monitoring tool for DNA damage in cells. The primary objective of this study was to use comet assay to ascertain if the blood cells of flounder (Pleuronichthys olivaceus) and muscle cells of clam (Saxidomus purpurata) are suitable for genotoxicity screening. This was achieved by initially exposing blood and muscle cells under in vitro conditions to the reference genotoxin hydrogen peroxide ($H_2O_2$); strong correlation between $H_2O_2$ concentration and comet values were found. Subsequently, the identification of DNA damage in isolated cells from flounder and clam was performed under in vivo exposure to benzo(a)pyrene (BaP) and tributyltin (TBT). Flounder and clam were exposed to different concentrations (1, 10, 50, 100 ${\mu}g/L$) of BaP or TBT for 4 days. Regardless of treated chemicals, blood cells of flounder were more prone to DNA breakage compared to muscle cells of clam. In conclusion, in vivo genotoxicity of BaP and TBT can be biomonitored using the comet assay. This study suggests that flounder and clam do show potential as mediums for monitoring genotoxic damage by comet assay.

Sex Differentiation and Early Gonadogenesis in Sebastes inermis Cuvier

  • Early gonadal development and sexual differentiation of dark-banded rockfish (Sebastes inermis Cuvier) were followed from parturition to 400 days post parturition (dpp). During this period, average total length (TL) increased from 0.57 to 13.18 cm. Primordial germ cells (PGCs) were first detected at 0.68 cm TL (10 dpp). When fish reached 1.52 cm TL (50 dpp), initial stages of ovarian differentiation were identified by the presence of PGCs containing condensed chromatin and their transformation into meiotic oocytes. At 10.23 cm TL (300 dpp), the ovaries gradually developed into oocytes in the primary yolk stages. Ovary growth was rapid after sex differentiation, but testis tissue continued to multiply without growing until fish reached 6.97 cm TL (200 dpp), after which the production of spermatocytes, spermatogonia, and cyst cells was apparent. Histological analysis of gonadal structure suggested a gonochoristic sexual development pathway. Our analysis of the sex ratio at 400 dpp showed a significantly higher proportion of males.

  • Influenced by Constructivism Art, Constructivism theatre projected the future with the help of participating artists and the development of new technologies. Therefore, this thesis studied the stage costumes in the plays of Meyerhold who represented Constructivism Play and showed its characteristics. The stage costumes in Meyerhold's plays followed the rules of Constructivism Play and showed several unique characteristics, such as the Bio-mechanic as a characteristic in his plays, functionality that facilitated the movement based on Taylorism, simple shapes, and easy cutting. Constructive artists saw the stage of constructive plays as a laboratory where they can display their designs, directly portraying future life forms through stage costumes and props. Stage costumes were no more a supporting tool that had described the play. Following the new concept of stage costumes, working costumes and stage costumes came to be regarded as the same. In turn, this led to new models and special costumes such as sportswear or specialized garments. Constructivism artists foresaw the future through the plays, with their fundamental basis for stage costumes shown in the later works of ordinary costumes.

Cytotoxic Compounds from the Roots of Pulsatilla koreana

  • Seven compounds including hederagenin 3-[(O-${\alpha}$-L-rhamnopyranosyl-($1{\rightarrow}2$)-${\alpha}$-L-arabinopyranosyl) (1), $3{\beta}$-[(O-${\alpha}$-L-rhamnopyranosyl-($1{\rightarrow}2$)-${\alpha}$-L-arabinopyranosyl)oxy]olean-12-en-28-oic acid (2), caffeic acid methyl ester (3), ferulic acid (4), orebiusin A (5), latifonicinin C (6) and 5-(hydroxymethyl)-2-furfuraldehyde (7) were isolated from the ethyl acetate fraction of the roots of Pulsatilla koreana. Their chemical structures were established based on physicochemical and spectroscopic data analyses. All isolates were investigated for their cytotoxic activity against cancer cell lines. Among them, compound 1 showed inhibitory activity against A549, COLO 205, and L1210 cancer cell lines with $IC_{50}$ values of 15.8, 36.5, and 22.8 ${\mu}g$/mL, respectively.

Inhibitory Activity against Helicobacter pylori of Isolated Compounds from Pinus koraiensis Siebold et Zucc Leaves

  • A phenol substance was extracted from Pinus koraiensis Siebold et Zucc leaf extracts and its biological efficacy was measured. The highest content of the phenol substance contained in Pinus koraiensis Siebold et Zucc leaves was 13.5 mg/g, which was obtained when it was extracted with 80% ethanol. At a concentration of 200 mg/mL, the phenolic substances extracted with 80% ethanol and water showed antimicrobial activities against Helicobacter pylori, producing clear zones of 10 and 12 mm diameter, respectively. Pinus koraiensis Siebold et Zucc. leaf extracts were separated using a Sephadex LH-20 column and 4 fractions were obtained (fractions A-D). Fractions C and D showed the greatest inhibitory activity against Helicobacter pylori producing 10.1 and 12.3 mm clear zones, respectively. These two fractions were purified using a Sephadex LH-20 and MCI-gel column ($H_2O{\rightarrow}100%$ ethanol). Purified compounds A and B were identified as syringic acid and compound C was identified as p-coumaric acid based on $^1H$-nuclear magnetic resonance (NMR), $^{13}C$-NMR, and fast atom bombardment mass spectrometry spectra. When two or more purified compounds were mixed, a synergistic effect of anti-Helicobacter pylori activity was evident. This result indicates that extracts of Pinus koraiensis Siebold et Zucc leaves could be considered a functional food because of their high antimicrobial properties.

Development of Microarrayer for Manufacturing DNA Chip (DNA 칩 제작을 위한 로봇 시스템의 개발)

  • This study exploits the robot system which is necessary in gene study and bio-technology industry. As well, a DNA chip, which of use has been increased recently, can be manufactured with this system. The robot consists of a device spotting DNA on the silylated slide, a well plate, a bed for fixing well plates, devices of washing and drying the pin in DNA spotting .device, a distillation-water vessel, and a discharge vessel of wash water. We made the period of sticking DNA to the pin on the well plate to be 15 seconds. The spot size of DNA was set to be 0.28 mm on the average by bringing the slide into contact with pin during 1 second. If DNA is spotted in minimum space possible about 0.32mm, this system can stick about 8,100 DNA spots on the well plate with this rate. Analyzing the procedure: Movement starts, Pin washes, dries, and smears DNA on the well plate. Spotting DNA onto 12 chips took 2 minutes and 50 seconds.

  • This study examined the effectiveness of micro-current stimulation (MCS) to improve the facial skin qualities by performing clinical test. The MCS is generally known that provide healing response in the damaged tissue and pain relief through activating the adenosine triphosphate (ATP) and protein synthesis. In here, we can hypothesize that the improvement of facial skin qualities may occur due to MCS, since the induction of micro-current from outside may enhance the cellular activity according to ATP activation. From the clinical test, our results showed that a variety of evaluating categories, which is able to assess the skin qualities, significantly enhanced due to stimulation of micro-current after 7 and 14 days. Therefore, we can estimate that MCS in human facial skin may be effective to improve the skin qualities.

Effect of Soybean Oil Supplementation on the Contents of Plasma Cholesterol and Cis9, trans11-CLA of the Fat Tissues in Sheep

  • A feeding trial was conducted with 10 sheep for 12 weeks to examine the effect of soybean oil (SBO) supplementation on long-chain fatty acids composition, especially cis9,trans11-conjugated linoleic acid (c9,t11-CLA) in fat tissues. Sheep were fed either a SBO supplemented diet (5%, DM basis) or a control diet without SBO. Chopped rye grass hay was fed as roughage. Concomitant increases in contents of total cholesterol (T-C) and HDL-cholesterol (HDL-C) in the plasma of sheep were observed from the SBO supplementation. The supplementation of SBO reduced (p<0.05) the proportions of $C_{16:1}$, $C_{17:0}$ and $C_{17:1}$ but increased (p<0.05) the proportions of $C_{18:0}$ and octadecenoic acid (t11-$C_{18:1}$) in the intramuscular fat. The $C_{18:0}$ proportion only in the subcutaneous fat was increased (p<0.05) by the SBO supplementation. The SBO supplementation slightly increased CLA proportion in the intramuscular fat and subcutaneous fat.

Molecular Characterization of Some Antilisterial Bacteriocin Genes from Enterococcus faecium and Pediococcus pentosaceus

  • Food bio preservation is of major interest in the food industry. Many types of antimicrobial compounds can be produced by lactic acid bacteria (LAB), including bacteriocins. Bacteriocins increase the shelf-life of food by decreasing some food-borne diseases. In this study, a multi-coding sequence of bacteriocin genes was used for primer design to produce bacteriocin genes in Enterococcus faecium AH2 strain and Pediococcus pentosaceus AH1. Multi-coding sequences were aligned to detect conserved sequences in the bacteriocin gene. Eight genes encoding proteins involved in bacteriocin production were isolated and sequenced, including six from E. faecium AH2 (entA, entI, entF, entR, orfA2, orfA3) and two from P. pentoceseus AH1 (papA, pedB), and all gene sequences were deposited in the Gen Bank database under accession numbers LC064146-LC064151, LC101300, and LC101789, respectively. P. pentosaceus AH1 and E. faecium AH2 strains displayed bacteriocin activities of $2610AU\;mL^{-1}$ and $690AU\;mL^{-1}$ and inhibition zones of 26 mm and 19 mm, respectively. Overexpression of entA in E. faecium AH2 increased the bacteriocin and antimicrobial activities.
