BACKGROUND/OBJECTIVES: Cheonggukjang is a traditional fermented soybean paste with significant health-promoting effects. On the other hand, there have been insufficient studies on the safety and efficacy of Cheonggukjang, which is produced using traditional methods containing toxins and biogenic amines (BAs). This study compared the laxative effect of Cheonggukjang, containing high or low levels of toxins and BAs (HTBC or LTBC) in a loperamide (Lop)-induced constipation mouse model. MATERIALS/METHODS: To induce constipation, Lop (5 mg/kg) was administered orally to ICR mice twice a day for 4 days, and the dose was increased to 8 mg/kg after a 3-day rest period. Cheonggukjang (500 mg/kg, HTBC, or LTBC respectively) was administered for four weeks before the Lop treatment. RESULTS: The number of stools, fecal weight, water contents, gastrointestinal transit, and histological alterations were recovered significantly in the HTBC or LTBC groups. HTBC and LTBC administration did not induce significant changes in body weight, dietary intake, and behavior. The opioid-receptor downstream signaling pathway in colon tissues was also evaluated. The c-Kit, stem cell kinase, and mitogen-activated protein kinases subfamilies, including extracellular signal-regulated kinase 1/2, c-Jun N-terminal kinases, and p38, were all downregulated in the HTBC or LTBC-administered mice colon compared to the Lop group. CONCLUSION: These results show that Cheonggukjang, containing high levels of toxins and BAs, have a similar laxative effect in a mouse model of Lop-induced constipation.
This study was conducted to investigate the characteristics of apple fermentation when different types of salt were used during apple fermentation, and when salt was added or not. The pH decreased in all experimental groups during the fermentation process, and the final pH was 4.72-3.31. The sugar content was 17.8-18.5 °Brix, the lowest in the sea salt (FAS), and the highest in the control group (FA). The acidity was measured as 0.48-0.56%, which was high in the control group (FA), and was 0.83-1.06% on the 7th day of final fermentation. The alcohol content was generated from the 5th day of fermentation, and the control group (FA) showed a distribution of 39.23-29.4%, and the alcohol content was higher in the control group than in the experimental group, As for the total polyphenol content and total DPPH content, it was confirmed that the purified salt (3.11 mg GAE/mL) showed the highest total polyphenol content than the control (2.25 mg GAE/mL). When salt was added, the fermentation progress was excellent, and when the salt concentration was high, it was confirmed that the possibility of apple fermentation was high.
This study was conducted experimentally to investigate the surface temperature of the heat sink, the air temperature in the flow channel and the sample temperature by changing the channel number of channel type heat sink and the air flow rate when heating and cooling the bio sample. The target temperature of the sample was 15℃ or less as the minimum value and 82℃ or more as the maximum value. In this study, the channel number of the heat sink(N = 1, 2, 4, 5, 10) and the air flow rate(Q=25, 42, 54m3/min) were varied. The bio sample was replaced with water, and the volume of water is 4mL. The size of the heat sink is 80x73x150mm and the material is aluminum. When cooling the sample, the surface temperature, the air temperature and the sample temperature were highly dependent on the number of channels and the flow rate. However, when the sample is heated, the surface temperature, air temperature and sample temperature do not depend on the number of channels and the flow rate. It was found that the conditions for satisfying the minimum temperature of 15℃ or less when cooling the sample were the number of channels N≥5 and the flow rate Q≥42m3/min. When heating the sample, the conditions to satisfy the maximum temperature of 82℃ or more are the number of channels N≤5 and the air flow rate Q≤42m3/min.
It is important to determine water movement at the growing substrate used in soil-less cultivation for better management of water supply. Numerical simulation is a fast and versatile approach to evaluate highly accurate water distribution. The objective of this study is to simulate the water movement in rockwool as a soil-less medium using HYDRUS-2D. HYDRUS-2D was used to simulate the spatial and temporal water movement in two types of rockwool slabs (Floriculture (FL), high density; Expert (EP), low density). The simulation was performed at two pulse conditions: 10 min ON and 50 min OFF (case A), 20 min ON and 40 min OFF (case B). The total irrigation amounts were the same at both cases. In case A, during the irrigation ON, the water contents at FL increased 1.93-fold faster than the values at EP. Whereas, during the irrigation OFF, the decreasing rate of water contents at FL was almost the same as one at EP. At case B, these values were not changed much from case A. However, the duration of optimum water content (50% - 80%) was 15.0 min and 23.5 min at case A and case B, respectively. Thus, FL and 20 min ON and 40 min OFF (case B) could supply water to rockwool much faster and longer than EP. Once qualitatively validated, this simulation of water movement in rockwool could be used to design an effective optimum irrigation method for vegetables.

Fractionated lipids of Halocynthia aurantium (Pyuridae) have been demonstrated to possess anti-inflammatory properties. However, their modulatory properties have not been reported yet. Thus, the objective of this study was to determine immune enhancing effects of fractionated lipids from H. aurantium tunic on macrophage cells. The tunic of H. aurantium was used to isolate total lipids, which were then subsequently separated into neutral lipids, glycolipids, and phospholipids. RAW264.7 cells were stimulated with different concentrations (0.5, 1.0, 2.0, and 4.0%) of each fractionated lipid. Cytotoxicity, production of NO, expression levels of immune-associated genes, and signaling pathways were then determined. Neutral lipids and glycolipids significantly stimulated NO and PGE2 production and expression levels of IL-1β, IL-6, TNF-α, and COX-2 in a dose-dependent manner, while phospholipids ineffectively induced NO production and mRNA expression. Furthermore, it was found that both neutral lipids and glycolipids increased NF-κB p-65, p38, ERK1/2, and JNK phosphorylation, suggesting that these lipids might enhance immunity by activating NF-κB and MAPK signaling pathways. In addition, H. aurantium lipids-induced TNF-α expression was decreased by blocking MAPK or NF-κB signaling pathways. Phagocytic activity of RAW 264.7 cells was also significantly enhanced by neutral lipids and glycolipids. These results suggest that neutral lipids and glycolipids from H. aurantium tunic have potential as immune-enhancing materials.
본 연구에서는 작약(Paeonia lactiflora Pall.) 종자 오일과 유박의 생리활성의 비교를 통해 작약 종자 유박 활용에 가능성을 평가하고자 수행되었다. 작약 종자 유박 추출물의 추출 수율은 22-45%로 추출용매에 따른 차이를 보였다. 총 폴리페놀 및 플라보노이드와 radical소거능은 오일보다 유박 추출물에서 유의하게 높았다. 작약 유박 추출물의 항균활성은 양성대조군에 비해 우수하였으며, 이는 48시간까지 지속되었다. 또한 LPS로 유도된 염증 반응은 유박 추출물 처리시 현저하게 감소하여 작약 종자 유박의 항염증 활성이 입증되었다. 본 연구를 통해 작약 종자 오일 뿐만 아니라 유박 추출물의 우수한 항산화, 항균 및 항염증 활성을 나타내어 바이오헬스 소재로서 작약 종자 유박의 활용 가능성이 확인되었다.
본 연구에서는 인체 폐암 세포인 A549를 사용하여 택란 메탄올 추출물의 항암활성과 그 분자적 기전에 관하여 연구하였다. 먼저 택란 추출물이 A549의 세포증식에 미치는 영향을 알아본 결과 처리 농도 및 시간 의존적으로 A549의 성장이 저해되었으며, 세포 주기 변화를 분석한 결과 강력한 G1 arrest가 유도되는 것을 확인하였다. 이러한 택란 추출물에 의한 G1 arrest는 세포주기 조절 단백질인 Cyclin D1, Cyclin E 및 Cyclin-dependent kinase인 CDK2, CDK4, CDK6의 발현 감소와 연관되어 있었다. 또한 택란 추출물에 의한 CDK/Cyclin complex의 발현 저해는 DNA 손상에 의해 활성화되는 CHK2의 활성화 형태인 p-CHK2의 발현 증가에 따른 CDK 활성화 효소인 Cdc25A phosphatase의 발현 억제에 의해 나타나는 결과로 사료된다. 반면 종양억제유전자인 p53 및 CDK 억제제인 p21과 p27의 발현량은 증가되지 않았다. 이러한 결과들로부터 택란 추출물은 DNA damage에 의한 ATM/CHK2/Cdc25A/CDK2 pathway를 통해 A549의 G1 arrest를 유도하여 세포의 증식을 억제할 것으로 판단되며, 이때 택란 추출물에 의해 유도되는 G1 arrest는 p53 비의존적인 경로일 것으로 사료된다. 본 연구결과는 택란이 Cdc25A를 target으로 하는 새로운 항암활성 소재로서 사용될 수 있는 가능성을 시사한다. 또한 본 연구결과는 택란 추출물의 세포주기 조절에 의한 항암기전을 이해하고 향후 지속적 연구를 하는 데 있어서 귀중한 기초자료로 사용될 수 있을 것이다.
참취로부터 phenolic compounds를 추출 후 항산화 효과, 건강 기능성 식품 및 미용식품 활성을 검정하여 기능성 소재로 활용 가능성을 살펴보았다. 참취에 함유되어 있는 페놀성 물질은 물과 60% ethanol을 용매로 하여 추출하였을 때 각각 11.1 mg/g, 4.18 mg/g 함량을 나타내었다. 추출물의 phenolic 농도를 $50{\sim}200{\mu}g/mL$로 조절하여 DPPH를 측정한 결과, 물 추출물과 ethanol 추출물 $50{\mu}g/mL$ phenolics 농도에서 각각 87%, 91%였고, ABTS radical decolorization도 $50{\mu}g/mL$ phenolics 농도에서 각각 99%, 99%였다. Antioxidant protection factor는 $200{\mu}g/mL$ phenolics 농도에서 물 추출물과 ethanol 추출물에서 각각 1.87 PF와 2.22 PF로 측정되었으며, TBARs값은 $50{\mu}g/mL$ phenolics농도에서 물 추출물이 95%, ethanol 추출물에서 97%를 나타내었다. Angiotensin converting enzyme(ACE) 저해능은 물 추출물에서 50.8%, ethanol 추출물에서 69.4%의 저해율을 나타내었으며, xanthine oxidase 저해능은 물 추출물은 $50{\mu}g/mL$ phenolics농도에서 91%, ethanol 추출물은 $200{\mu}g/mL$ phenolics농도에서 80%의 높은 효능을 보였다. ${\alpha}$-Amylase 저해능은 물 추출물과 ethanol 추출물에서 각각 24%, 89%로 ethanol 추출물에서 높은 효능을 보였다. Tyrosinase 저해능을 측정한 결과 물 추출물과 ethanol 추출물에서 각각 47%, 25%의 미백 효과를 나타내었다. Elastase 저해능 측정 결과 물 추출물과 ethanol 추출물에서 각각 46%, 14%로 물 추출물에서 주름 개선 효과가 상대적으로 높게 나타내었다. 이러한 결과로 보아 참취 추출물은 항산화 고혈압억제, 통풍억제, 당분해억제, 미백효과, 주름개선효과 등의 건강 기능성 식품 및 미용식품 활성 소재로서 활용 가능성이 큰 것으로 판단되었다.
기능성 천연물 소재로서의 가능성을 검토하고자 캐모마일 추출물의 미백 효과를 조사하고 melanin 생성 반응에 관여하는 물질 억제에 대한 기전을 규명하고자 하였다. 캐모마일을 water와 60% ethanol을 이용하여 추출하였고, 얻어진 추출물을 phenolic 농도별로 설정하여 tyrosinase 저해 활성을 알아보았을 때, water 추출물의 경우 효과가 미비하였고, 60% ethanol 추출물에서는 농도 의존적으로 저해 활성이 나타내어 melanin 생성 저해 효과가 있을 것으로 판단되었다. 캐모마일 60% ethanol 추출물을 동결건조하여 얻어진 분말을 이용하여 B16F10 melanoma cell에 대한 세포 독성을 측정 결과, $75{\mu}g/mL$의 농도에서부터 독성이 관찰되어 농도 구간을 10, 25, $50{\mu}g/mL$으로 선정하였다. ${\alpha}-MSH$로 자극한 B16F10 melanoma cell에서 melanin 생성량을 측정하여 캐모마일의 melanin 성성 억제 효능과 melanin 생성에 영향을 미치는 단백질인 tyrosinase, MITF, TRP-1, TRP-2의 단백질 발현 억제 효과를 알아본 결과, 캐모마일의 농도가 높아짐에 따라 농도 의존적으로 melanin 생성 함량이 감소하였고, tyrosinase, TRP-1, TRP-2, 등의 단백질 발현량 또한 감소하는 것을 확인할 수 있었다. 따라서 캐모마일 추출물은 B16F10 melanoma cell에서 melanin의 생성을 억제하고, melanin 생성 관련 단백질발현을 억제하는 효과가 있음을 확인하였다. 위의 결과들로 인하여 캐모마일은 미백 기능성 식품 산업화를 위한 유용한 자원으로 활용 될 것으로 예상되며, 추후 산업적 응용을 위한 지속적인 연구가 진행되어야 할 것으로 판단되었다.
본 연구에서는 Euptelea pleiosperma 에탄올 추출물(EPEE)의 항산화능과 항염증 생리활성을 in vitro assay system 및 cell culture model system을 이용하여 분석하였다. 먼저 EPEE의 항산화능을 DPPH radical scavenging activity로 분석한 결과 radical 소거능의 정도가 양성 대조군으로 사용한 ascorbic acid 보다 높은 활성을 보여 매우 강한 항산화능을 보유함을 확인하였다. 또한 RAW 264.7 세포주를 이용한 $H_2O_2$ 및 LPS의 유도에 의해 생성된 ROS에 대한 소거능을 분석한 결과에서도 농도의존적인 강한 소거능을 보였다. 뿐만 아니라 대표적인 항산화효소 중 하나로 천연물에 의한 항산화능 활성에 의해 주로 발현이 유도되는 hemeoxygenase 1 (HO-1) 및 그 전사 인자인 nuclear factor-E2-related factor 2 (Nrf2)의 단백질 발현이 EPEE의 처리에 의해 유의적으로 증가됨을 보였으며 이러한 HO-1 및 Nrf2의 발현 변화는 상위신호전달계인 MAPKs 및 PI3K/Akt 에 의해 조절될 가능성을 보였다. 한편 EPEE가 LPS에 의해 유도된 NO 생성에 미치는 영향을 분석한 결과 농도의존적인 NO 생성 저해능을 보였으며 이는 NO 생성 단백질인 iNOS의 발현 저해에서 기인함을 확인하였다. 이와 같은 EPEE의 NO 생성 억제 효과는 염증 상위신호전달계인 NF-${\kappa}B$ 및 AP-1의 조절을 통해 일어날 가능성을 보였다. 이러한 결과를 통해 EPEE의 높은 항산화능과 항염증 활성을 처음으로 확인하였으며 향후 기능성 소재로서 유용하게 활용될 수 있을 것으로 판단된다.
