Changes of Physicochemical Characteristics of Acanthopanax senticosus Extract during Storage (오가피 추출액의 저장조건에 따른 이화학적 특성변화)

  • This study was conducted to investigate the effects of temperature and packaging on the quality characteristics of hot-water extract from Acanthopanax senticosus. The herb samples, ground and sifted($8{\sim}20$ mesh), were extracted by a boiling($100^{\circ}C$) water under the atmospheric pressure for 2 hrs. The extract was packed with out nylon PE(0.08 mm) pouch and kept under the condition(40 and $60^{\circ}C$) for 10 days. The reduction of turbidity, pH, polyphenol amount and DPPH radical scavenging activity of extract were retarded by packaging under the low temperature. Titratable acidity was increased with packaging at high temperature. Soluble solid content was not affected by packaging and temperature condition. The changes of quality characteristics followed the first-order reaction rate. Arrhenius equation was applied to determine the effects of temperature on the reaction rate constant, showing high $R^2$. These results suggested that packaging under lower temperature can be utilized for maintaining the quality of A senticosus extract.

Quality Change of Beverage Containing Muskmelon Vinegar and Concentrated Muskmelon Juice during Storage (참외 식초 및 농축액 함유 음료의 저장 중 품질 변화)

  • Response surface methodology(RSM) was used for monitoring the quality change of muskmelon beverage with storage temperature and time. Muskmelon beverage was prepared with the ratio of liquid fructose($11\%$), muskmelon vinegar($3\%$), concentrated muskmelon juice($1\%$) and refined water($84\%$). To examine physicochemical properties of muskmelon beverage, various factors such as color, brown color intensity and sugar content were determined using physicochemical methods. Physicochemical properties of muskmelon beverage were more affordable with the increased storage temperature. While, organoleptic properties decreased with the increased storage time and temperature. The result of this study indicated that the quality of muskmelon beverage was mostly influenced by the storage temperature. Muskmelon beverage showed the good score under $60^{\circ}C$ as a storage temperature.

Occurrence of Tetracyclines Resistant Bacteria in the Soil Applied with Livestock Manure Compost (가축분 퇴비 시용 농경지 내 Tetracyclines 내성균 분포 특성)

  • BACKGROUND: Large amount of veterinary antibiotics have been used in the livestock industry to prevent diseases and promote growth. These antibiotics are excreted through feces and urine in unchanged form and reach to agricultural fields via application of the livestock manure based composts. The purpose of this study was to evaluate the occurrence of tetracyclines-resistant bacteria in the soil received livestock manure compost for a long term. METHODS AND RESULTS: Tetracyclines (tetracycline TC, chlortetracycline CTC, and oxytetracycline OTC) resistance bacteria in the soil of rice-onion field applied pig manure compost (PM), in the soil of grass-rye field received cow manure compost (CM), and in the soil of rice field applied inorganic fertilizer (NPK) were determined. The soil received livestock manure composts clearly showed higher number of TC, CTC, and OTC resistance bacteria compared with the soil treated with inorganic fertilizer. The antibiotic resistant bacteria recovered appeared at 80 mg/L of tetracyclines was identified 1 specie, 6 genera 7 species, and 6 genera 7species in the soils received CM, PM, and NPK, respectively. The dominant resistant bacteria with the CM and PM application were Ochrobactrum and Rhodococcus. CONCLUSION: The application of livestock manure compost in the agricultural field is likely to contribute the occurrence of antibiotic resistance bacteria in the agricultural environment.

  • In this study, ozone oxidation experiment was carried out for the removal of fluorescent whitening agent which is widely used in textile dyeing and paper industry. The stilbene fluorescent whitening agent has been industrialized since the earliest, and the amount of current production is the highest. Due to the characteristics of the fluorescent whitening agent that can not be removed by conventional wastewater treatment methods, the fluorescent whitening agent in wastewater treatment has difficulty in using as recycled water in the process. Pre-treatment ozone oxidation experiment was conducted prior to the introduction of Membrane Bio Reactor(MBR) treatment process by converting biodegradable materials into biodegradable materials. The removal efficiencies of fluorescent whitening agents, a diaminostilbenedisulfonic acid derivative by ozone oxidation were evaluated by $UV_{254}$ Scan, $COD_{Mn}$, T-N and color using a synthetic wastewater sample ($COD_{Mn}=433.0mg/{\ell}$) and paper and paper mill wastewater ($COD_{Mn}=157.2mg/{\ell}$).

Induction of Apoptosis and Inhibition of NO Production by Piceatannol in Human Lung Cancer A549 Cells (A549 인체 폐암세포에서 piceatannol에 의한 apoptosis 유발과 NO 생성의 억제)

  • Piceatannol (trans-3,4,3',5'-tetrahydroxystilbene), a natural stilbene, is an analogue of resveratrol. Although recent experimental data have revealed the health benefit potency of piceatannol, the molecular mechanisms underlying the anti-cancer activity have not yet been studied in detail. In the present study, the further possible mechanisms by which piceatannol exerts its pro-apoptotic action in cultured human lung cancer A549 cells were investigated. Exposure of A549 cells to piceatannol resulted in growth inhibition and induction of apoptosis. Apoptosis induction of A549 cells by piceatannol showed correlation with proteolytic activation of caspase-3, -8, and -9, and concomitant degradation of activated caspase-3 target proteins such as poly (ADP-ribose) polymerase, phospholipase C-${\gamma}1$, ${\beta}$-catenin, and Inhibitor caspase-activated DNase. The increase in apoptosis by piceatannol treatment was also associated with an increase of pro-apoptotic Bax expression and decrease of anti-apoptotic Bcl-2 and Bcl-xL expression, and caused down-regulation of the inhibitor of apoptosis protein family members and up-regulation of Fas and Fas legend. In addition, piceatannol treatment markedly inhibited the expression of mRNA and proteins of inducible nitric oxide (NO) synthase, and the levels of NO production were progressively down-regulated by piceatannol treatment in a dose-dependent fashion. The results indicate that piceatannol may have therapeutic potential against human gastric cancer cells.

Inhibition of Cell Invasion by Ethyl Alcohol Extracts of Hizikia fusiforme in AGS Human Gastric Adenocarcinoma Cells (AGS 인체 위암세포에서 톳 에탄올 추출물에 의한 침윤성 저해)

  • In this study, we investigated the effects of ethyl alcohol extracts of Hizikia fusiforme (EHF) on the correlation between tightening of tight junctions (TJs) and anti-invasive activity in human gastric adenocarcinoma AGS cells. Inhibitory effects of EHF on cell proliferation, motility, and invasiveness were found to be associated with increased tightness of the TJs, which was demonstrated by an increase in transepithelial electrical resistance. Activities of matrix metalloprotease (MMP)-2 and -9 in AGS cells were dose-dependently inhibited by treatment with EHF, and this was also correlated with a decrease in expression of their mRNA and proteins; however, tissue inhibitor of metalloproteinase (TIMP)-1 and -2 mRNA levels were increased. Additionally, immunoblotting results indicated that EHF repressed the levels of claudin proteins (claudin-1, -3, and -4), major components of TJs that play key roles in control and selectivity of paracellular transport. Furthermore, EHF decreased expression of insulin such as growth factor-1 receptor proteins, while concurrently increasing that of thrombospondin-1 and E-cadherin. In conclusion, these results suggest that EHF treatment may inhibit tumor cell motility and invasion, and therefore act as a dietary source to decrease the risk of cancer metastasis.

Molecular Cloning and Expression of a Laccase from Ganoderma lucidum, and Its Antioxidative Properties

  • Laccases are multicopper-containing oxidases that catalyze the oxidation of many aromatic compounds with concomitant reduction of oxygen to water. Interest in this enzyme has arisen in many fields of industry, including detoxification, wine stabilization, paper processing, and enzymatic conversion of chemical intermediates. In this study, we cloned a laccase gene (GLlac1) from the white-rot fungus Ganoderma lucidum. The cloned gene consists of 4,357 bp, with its coding region interrupted by nine introns, and the upstream region has putative CAAT and TATA boxes as well as several metal responsive elements (MREs). We also cloned a full-length cDNA of GLlac1, which contains an uninterrupted open reading frame (ORF) of 1,560 bp coding for 520 amino acids with a putative 21-residue signal sequence. The DNA and deduced amino acid sequences of GLlac1 were similar but not identical to those of other fungal laccases. GLlac1 was released from the cells when expressed in P. pastoris, and had high laccase activity. In addition, GLlac1 conferred antioxidative protection from protein degradation, and thus may be useful in bio-medical applications.

A Study on Biodiesel Fuel of Engine Performance and Emission Characteristics in Diesel Engine (디젤엔진 성능에 미치는 바이오디젤 연료에 관한 연구)

  • Diesel engines have the superior combustion efficiency and fuel economy that they are widely used for industry, heavyduty vehicles, etc. However, its exhaust emissions have become the major concerns due to their environmental impacts. Moreover, the depletion of fossil fuels is the main issue. Therefore, it is important to look for alternative sources of energy. Bio-diesel is one of the ideal energy which has proved to be ecofriendly for more than fossil fuels. The experimental tests analysed the engine performance and emission characteristics of a diesel engine using diesel and biodiesel blended of BD25, BD45 and BD65, in order to study the use of clean fuel to meet the increasingly stringent emission regulations. The engine performance was examined by using engine dynamometer while an exhaust gas analyzer was used to examine the emission characteristics. The effect of biodiesel on engine performance were lower to diesel through comparing their HP and torque but fuel consumption was slightly increased because of biodiesel has lower heating value and higher density than diesel. However, due to the better lubricity, the brake thermal efficiency of biodiesel was higher than diesel. The emission characteristics were strongly affected by the blending ratio of diesel and biodiesel. The results showed that the smoke opacity, hydrocarbons (HC) and carbon monoxide (CO) emissions decreased while the nitrogen oxides (NOX) slightly increased.

Influence of Prenatal Noise and Music on the Expressions of c-Fos and Nitric Oxide Synthase in the Hippocampus of Rat Pups

  • The expressions of c-Fos and nitric oxide synthase (NOS) represent neuronal activity and play' a crucial role in the shaping of the development of brain. During the late pregnancy, stresses may influence neuronal activity of prenatal rats. In the present study, the effects of prenatal noise and music on the expressions of c-Fos and NOS in the hippocampus of rat pups were investigated. Exposure to the noise during pregnancy decreased c-Fos and NOS expressions in the hippocampus of rat pups, whereas exposure to music during pregnancy increased c-Fos and NOS expressions in the hippocampus of rat pups. The present results show that prenatal music stimulation may increase neuronal activity of rat offspring, whereas exposure to noise during pregnancy may reduce the neuronal activity of offspring. The present study suggests that prenatal stimuli including noise and music could affect the fetal brain development.

Therapeutic Potential of the Rhizomes of Anemarrhena asphodeloides and Timosaponin A-III in an Animal Model of Lipopolysaccharide-Induced Lung Inflammation

  • Investigations into the development of new therapeutic agents for lung inflammatory disorders have led to the discovery of plant-based alternatives. The rhizomes of Anemarrhena asphodeloides have a long history of use against lung inflammatory disorders in traditional herbal medicine. However, the therapeutic potential of this plant material in animal models of lung inflammation has yet to be evaluated. In the present study, we prepared the alcoholic extract and derived the saponin-enriched fraction from the rhizomes of A. asphodeloides and isolated timosaponin A-III, a major constituent. Lung inflammation was induced by intranasal administration of lipopolysaccharide (LPS) to mice, representing an animal model of acute lung injury (ALI). The alcoholic extract (50-200 mg/kg) inhibited the development of ALI. Especially, the oral administration of the saponin-enriched fraction (10-50 mg/kg) potently inhibited the lung inflammatory index. It reduced the total number of inflammatory cells in the bronchoalveolar lavage fluid (BALF). Histological changes in alveolar wall thickness and the number of infiltrated cells of the lung tissue also indicated that the saponin-enriched fraction strongly inhibited lung inflammation. Most importantly, the oral administration of timosaponin A-III at 25-50 mg/kg significantly inhibited the inflammatory markers observed in LPS-induced ALI mice. All these findings, for the first time, provide evidence supporting the effectiveness of A. asphodeloides and its major constituent, timosaponin A-III, in alleviating lung inflammation.
