  • In cosmetics industry, we notice that the research should be focused on developing well-being related products such as SPA, esthetique, mass-customization, organics, naturals besides inner beauty and body shaping products. These well-being related products should satisfy customers' expectation about not only concern about health but also safety, reliability, and ethos about products. In that context, the systematic way for well-being related research in cosmetics should be carried out concerning anti-aging with fat-decom-position, vitamin, polyphenol, and organics. At the same time, there are several important factors in this research like augmenting efficacy, controlling skin-absorbency, skin-adaptability, forming, stabilization of activated com-ponents, improving sustainability, and developing new materials based on dermatologic and emotional science. It should be also emphasized that recent technologies such as bio-technology, nano-technology, and natural-material-technology are the cornerstones of developing well-being products.

Characteristics of Elsholtzia splendens Extracts on Simultaneous Steam Distillation Extraction Conditions (수증기증류조건에 따른 꽃향유 추출물의 품질특성)

  • This study was performed to develop extract materials from Elsholtzia splendens by analyzing the functionality and aroma profile by the simultaneous steam distillation extraction. The qualities of extracts such as total yield, total phenolic compound and electron donation ability were affected by extraction temperature than time. The main flavor compounds were analyzed as estragole, thymol and beta-caryophyllene. The response variables had significant with extraction temperature than tim. The established model was suitable to predict calculated value in experimental ranges. The optimum extinction conditions, which were limited of maximum value for dependent variables were $108^{\circ}C$ and 2.1 hr in the simultaneous steam distillation extraction method.

  • To evaluate the human risk of long-term intake of genetically modified (GM) rice, we carried out RT-PCR of housekeeping genes. Housekeeping genes, which show highly uniform expression in living organisms during various stages of development and under different environmental conditions, were normalized by RT-PCR. We assessed the expression of 10 common housekeeping genes (18s rRNA, 25S rRNA, UBC, UBQ5, UBQ10, ACT11, GAPDH, eEF-$1{\alpha}$, ${\beta}$-TUB, GAPDH, ${\beta}$-actin, B2m, G6pd2, Gyk, Gus, Hprt, Cyclophlin A, Tfrc, ${\alpha}$-tubulin and RPL13A) in the liver, stomach, small intestine, large intestine, kidney and spleen of mice fed GM or non-GM rice. We found no significant differences in the expression of housekeeping genes between the two groups of mice.

Effect of High Vanillin Treatment on Storage Quality of Fresh-cut Apples

  • The effect of post-cut vanillin treatment at high concentrations on changes of quality and microorganism in fresh-cut apples was studied. Apples (Malus domestica Borkh. cv. Fuji) were sliced, treated by dipping in different vanillin solution, 0, 40, 80, and 120 mM, packed in polyethylene bag, and then stored for up to 3 weeks at $4^{\circ}C$. Changes in total aerobic bacteria, yeast and molds, browning, soluble solids, and titratable acidity during storage were investigated. Growth of total aerobic bacteria throughout storage was strongly inhibited by vanillin regardless of treatment concentrations. Growth of yeast and molds was inhibited by vanillin of all concentrations until 2 weeks of storage. Levels of browning index, soluble solids, and titratable acidity were not significant difference among the treatment conditions until 2 weeks of storage. However, when stored for 3 weeks, browning index increased more at 80 or 120 mM vanillin, while soluble solids and titratable acidity more be decreased by 120 mM vanillin as compared with other treatment conditions. These results show that the usage of vanillin in processing of fresh-cut apples had a limitation for maintaining quality.

Analysis of Anti-Allergic Activities by Extracts from Persimmon Sap-Stained Rayon and Cotton Fabrics (감물염색 직물의 추출물에 의한 항알러지 활성의 분석)

  • Allergic dermatitis is one of the most prevalent diseases in young/juvenile children worldwide. In this research, extracts with persimmon sap-stained fabrics (rayon and cotton) exhibited an elevation in $CD4^+$ cell numbers. MMP-2 and MMP-9 expressions by Hematoxylin-Eosin staining and immunohistochemistry revealed that the expressions were decreased by addition of the extracts. The present results collectively suggest that the active ingredients of persimmon sap-stained fabrics play an important role in inhibition of DNFB-induced-atopic symptoms in vivo.

Live Cell Detection of Monoclonal Antibody Light and Heavy Chain mRNAs using Molecular Beacons (분자 비컨을 이용한 살아 있는 세포에서 단일클론항체 경쇄와 중쇄 mRNA 검출에 의한 세포주 선별방법)

  • Developing the method for the selection of animal cell line producing therapeutic monoclonal antibody (mAb) is invaluable as its market is rapidly growing. Although the quality of produced mAb is as important as quantity, however there is no method developed for the selective screening of cell lines on the basis of both quantity and quality. From recent reports, the ratio of light and heavy chain mRNAs of mAb in the cell is a key parameter for the indication of product quality. Therefore, it is obvious that developing the novel method that can detect both light and heavy chain mRNAs in single live cell will provide unprecedented opportunities in bio-industry. Here, we have constructed oligonucleotide probes, molecular beacons for the detection of light or heavy chain mRNAs, respectively, in the live cells producing mAbs. Both beacons showed increased fluorescent intensity after transient transfection of plasmid expressing mAbs analyzed by fluorometer. Flow cytometric analysis clearly demonstrated that both molecular beacons can simultaneously detect the expression of light and heavy chain mRNAs of mAb in the same cell. The technique described in the thesis provides the new direction and concept for developing the method for the smart selection of cell lines producing recombinant proteins including therapeutic mAbs.

Culture Condition for Biomass of Pleurotus eryngii (새송이버섯 biomass를 위한 최적배양 조건)

  • For the purpose of application for biomass of Pleurotus eryngii, the optimum culture condition were tested. It was found that the optimum culture condition for spot culture of pleurotus eryngii were 24$^{\circ}C$ for 18 days with PDA medium. And the optimum culture condition of bioreactor for biomass were pH 5.5, 18$^{\circ}C$ and 27 days with PDMP broth. It was possible to artificial cultivation of mycelial from Pleurotus eryngii using bioreactor for biomass under the optimum conditions, and it was also possible for Pleurotus eryngii biomass because the forming of fruiting body when Pleurotus eryngii was cultivated using mass artificial cultivated mycelial in the bioreactor.

Fabrication of a high performance microvalve using a multilayer piezoelectric actuator and its characteristics (적층형 압전 엑츄에이터를 이용한 고성능 마이크로 밸브의 제작과 그 특성)

  • This paper describes the design, fabrication and characteristics of a micromachined piezoelectric valve utilizing a multilayer ceramic actuator (MCA). The micromachined MCA valve, which uses a buckling effect, consists of three separate structures: the MCA, the valve actuator die and the seat die. The valve seat die with 6 trenches was made, and the actuator die, which is driven by the MCA under optimized conditions, was also fabricated. After Si wafer direct bonding between the seat die and the actuator die, the MCA was also anodically bonded to the seat/actuator die structure. A polydimethylsiloxane (PDMS) sealing pad was fabricated to minimize the leak rate. Finally, the PDMS sealing pad was also bonded to the seat die and the stainless steel package. The MCA valve shows a flow rate of 9.13 sccm at an applied DC voltage of 100 V with a 50% duty cycle and a maximum non-linearity of 2.24% FS. Therefore, the fabricated MCA valve is suitable for a variety of flow control equipment, as a medical bio-system and in the automobile industry.

  • Micro-injection molding and microfluidic devices with the help of MEMS technologies including the LIGA process are expected to play important roles in micro-system industries, in particular the bio-application industry, in the near future. Understanding fluid flows in micro-channels is important since micro-channels are typical geometry in various microfluidic devices and mold inserts for micro-injection molding. In the present study, Part 1, an experimental investigation has been carried out to understand the detailed flow phenomena in micro-channel filling process. Three sets of micro-channels of different thickness (40um,30um and 2011m) were fabricated using SU-8 on silicon wafer substrate. And a flow visualization system was developed to observe the filling flow into the micro-channels. Experimental flow observations are extensively made to find the effects of pressure, inertia force, viscous force and surface tension. A dimensional analysis for experimental results was carried out and several relationships A dimensionless parameters are obtained.

Backbone 1H, 15N and 13C Resonance Assignment and Secondary Structure Prediction of HP0062 (O24902_HELPY) from Helicobacter pylori

  • HP0062 is an 86 residue hypothetical protein from Helicobacter pylori strain 26695. HP0062 was identified ESAT-6/WXG100 superfamily protein based on structure and sequence alignment and also contains leucine zipper domain sequence. Here, we report the sequence-specific backbone resonance assignment of HP0062. About 97.7% of all $^1H_N,\;^{15}N,\;^{13}C_{\alpha},\;^{13}C_{\beta}\;and\;^{13}C=O$ resonances were assigned unambiguously. We could predict the secondary structure of HP0062 by analyzing the deviation of the $^{13}C_{alpha}\;and\;^{13}C_{\beta}$ chemical shifts from their respective random coil values. Secondary structure prediction shows that HP0062 consist of two ${\alpha}$-helices. This study is a prerequisite for determining the solution structure of HP0062 and can be used for the study on interaction between HP0062 and DNA and other Helicobacter pylori proteins.
