Squid ink was purified to melanin powder by repeated treatments with aqueous sodium hydroxide and acetic acid solutions. The exhaustion dyeing conditions of melanin to wool fabrics were investigated in relation with pH, melanin concentration, dyeing temperature and time. The melanin was dyeable to cotton and wool fabrics but higher dyeability of the wool was observed. A K/S of 7 was obtained on the optimally dyed wool fabrics with 5 % owf melanin under pH 4 at $100^{\circ}C$ for 60 minutes. Color fastness to both washing and rubbing was excellent and color fastness to light was also very good probably due to the polymeric nature of the extracted sepia melanin.
The cotton yarn was subjected to bio-polishing treatment with three commercial enzymes(Cellusoft L, Denimax-991L and Denimax-acid) to remove the fuzz on the cotton yarn. Also, enzyme treated cotton yarns were compared with singeing cotton yarns. Experimental variables of enzyme treated cotton yarn were as follow: concentration of enzyme solution and NaOH, dipping time, and processing temperature. The enzymatic treatments were evaluated by analyzing the effect on yarn count, twist contraction, evenness and tenacity. As the results, enzymatic treatment on cotton yarn induced same effects as the traditional singeing treatment. Also, silket treatment of cotton yarn after bio-polishing enhanced the tensile properties of the cotton yarn.
We here demonstrate the evidence of increased anti-tumor and immunostimulating activities of crude extracts (GAL) from Galium. aparine L. In experimental lung metastasis of colon26-M3.1 carcinoma or B16-BL6 melanoma cells, prophylactically intravenous (i.v) administration of GAL significantly inhibited lung metastasis in a dose-dependant manner. In an in vitro cytotoxicity analysis, GAL at the concentration up to $500\;{\mu}g/ml$ did not affect the growth of B16-BL6 melanoma cells. In contrast, GAL showed the enhancement of splenocyte proliferating activity in a dose-dependent manner. Peritoneal macrophages stimulated with GAL produced various cytokines such as $1L-1{\beta},\;TNF-{\alpha},\;IFN-{\gamma}$ and IL-12. These data suggest that GAL has an antitumor activity to inhibit tumor metastasis, and its antitumor effects is associated with activation of nonspecific immnune related cells.
Some fatty acids(palmitic acid, stearic acid, oleic acid, linoleic acid and linolenic acid) in foxtail millet seeds were evaluated. In this study, 32 varieties of accession collected from different place of South Korea were used in order to evaluate their fatty acid composition and concentration. These samples were analyzed by GC with HP20 column. Significant variability in individual fatty acids was observed. The palmitic acid ranged from 10.9 to 13.5%, stearic acid from 2.5 to 8.3%, oleic acid from 10.0 to 15.8%, linoleic acid from 62.5 to 67.9% and linolenic acid ranged from 3.0 to 5.2%. This variability can be exploited for selection of plant and for studying the genetic variability in millet. These selections can also be utilized for genetic improvement of the crops.
Inulo-oligosaccharides (IOS, $F_n$, n=2-6) were purified from enzymatic hydrolysates of water-soluble extract of Jerusalem artichoke tubers. Quantification of inulo-oligosaccharides was done using high pH anion exchange chromatography with pulsed amperometric detector (HPAEC-PAD) at the concentration range of 10-100 mg/L, which was compared with that of fructo-oligosaccharides (FOS, $GF_n$, n=1-7). Peak areas per mg IOS were higher than FOS at the same degree of polymerization (DP). Specific peak areas of IOS increased proportionally as DP increased up to six, in contrast to FOS showing no linearity.
The number of people suffering from diabetes have been increased around the world. In this study, we investigated the antidiabetic and antioxidant effects of Anthriscus sylvestris(L.) Hoffm and its main compounds. It was divided into root(R) and aerial part(AP) for comparative analysis. Total polyphenol, total flavonoid content was higher in AP extract, but nodakenin content was higher in R(1169.13 ± 6.00 mg/g) extract. Antioxidant activity was also higher in AP extract. To compare antidiabetic efficacy, we analyzed the effects of R and AP extracts on ɑ-glucosidase inhibition(AGI), dipeptidyl peptidase-4(DPP-4) and protein tyrosine phosphatase(PTP)1B activity. R and AP extracts showed similar effects on AGI and DPP-4 activity in a concentration dependent manner, and there was no effect on PTP1B activation. Glucose uptake(139.51 ± 3.19%) in 3T3-L1 cells was more effective in the AP extract-treated group than the positive control, rosiglitazone, group. Both R and AP extracts were effective in protecting against pancreatic beta cell damage caused by hyperglycemia. These results suggest that Anthriscus sylvestris(L.) Hoffm. could be used as a candidate for diabetes treatment.
Purine catabolite screening enables reliable diagnosis of certain diseases. In this regard, the development of a facile detection strategy with high sensitivity and selectivity is demanded for point-of-care applications. In this work, the simultaneous detection of uric acid (UA), xanthine (XA), and hypoxanthine (HX) was carried out as model purine catabolites by surface-enhanced Raman Spectroscopy (SERS). The detection assay was conducted by employing high-aspect ratio Au nanopillar substrates coupled with in-situ Au electrodeposition on the substrates. The additional modification of the Au nanopillar substrates via electrodeposition was found to be an effective method to encapsulate molecules in solution into nanogaps of growing Au films that increase metal-molecule contact and improve substrate's sensitivity and selectivity. In complex solutions, the approach facilitated ternary identification of UA, XA, and HX down to concentration limits of 4.33 𝜇M, 0.71 𝜇M, and 0.22 𝜇M, respectively, which are comparable to their existing levels in normal human physiology. These results demonstrate that the proposed platform is reliable for practical point-of-care analysis of biofluids where solution matrix effects greatly reduce selectivity and sensitivity for rapid on-site disease diagnosis.
Transparent exopolymer particles(TEP)는 식물플랑크톤이 배출하는 다당류 물질이 자가응집을 통해 생성되는 끈적거리는 특성을 가진 물질로 해중설 형성에 중요한 역할을 담당한다고 밝혀져 왔다. 본 연구는 동해 북부해역에서 TEP 농도 분포특성과 탄소순환에서 역할을 이해하기에 이산화탄소계 인자, 엽록소-a 농도들을 관측하였다. 동해 북부해역의 TEP는 $0{\sim}338{\mu}g\;Xeq\;l^{-1}$로 연안역의 농도보다 낮으나 다른 외양역의 대번식기와 유사한 값이 나타났다. TEP의 공간분포는 엽록소-a의 공간 분포특성과 유사하게 나타나고 있는 것으로 보아 식물플랑크톤이 TEP의 주요 기원으로 사료된다. 직접 측정한 pH와 총 알칼리도의 값을 이용하여 총 용존 무기탄소($TCO_2$)를 계산하였으며, 염분 35 psu에 해당하는 값으로 변환된 $NTCO_2$는 수온에 대해 선형적으로 감소하고 있다. 수온으로 계산된 $NTCO_2$와 현장에서 측정한 $NTCO_2$와의 차이는 생물적인 과정에 의한 이산화탄소의 제거($NTCO_{2bio}$)를 나타내는데, 난수역에서 $NTCO_{2bio}$값이 음의 값이 나타났고 TEP의 농도가 높은 것으로 보아 생물적인 과정에 의해서 해수중의 이산화탄소가 감소한 것으로 사료된다. 북위 $44^{\circ}-46^{\circ}$ 냉수역은 표층에서 TEP 농도는 낮지만, 엽록소-a 최대층에서 TEP의 농도 또한 높게 나타났고 TEP에 포함된 탄소(TEP-C)가 $NTCO_{2bio}$의 약 40%를 기여하였다. 본 연구에서 TEP농도가 해수 중의 이산화탄소 농도 및 pH와 뚜렷한 상관관계가 나타나지 않아서 이산화탄소 농도에 따른 TEP의 반응은 살펴볼 수 없었으나 해수 중의 탄소순환에서 생물적인 과정을 이해하는데 TEP가 중요하게 고려되어야 함을 시사한다.
나물자원의 수경재배시 양액농도가 생장 및 무기물함량에 미치는 영향을 조사하고자 벌개미취, 원추리 및 질경이를 펄라이트 배지에 추식하여 70일간 재배 한 후 생장정도와 무기물 함량을 분석하였다. 벌개미취와 원추리의 초장, 줄기직경, 잎의 수, 신선중 및 건물중은 1.5배액 처리구에서 확연하게 우수한 결과를 나타내었다. 질경이의 지상부와 지하부 생체중 및 건물중 등은 양액의 농도가 0.2재액에서 1.5배액으로 증가할수록 증가한 반면, 엽수와 근장은 0.5배액과 표준농도에서 가장 높은 것으로 나타났다. Ca Mg 및 Na 함유량은 벌개미취의 경우 1.5배액에서, 원추리와 질경이는 0.25배액에서 가장 많았다. K 함량은 벌개미취와 원추리는 0.5배액에서, 질경이는 1.5배액에서 가장 많았다. $P_2O_5$는 나물종류 및 양액의 농도에 따른 차이가 거의 없었다.
Quercetin is a flavonoid usually found in fruits and vegetables. Aside from its antioxidative effects, quercetin, like other flavonoids, has a various neuropharmacological actions. Quercetin-3-O-rhamnoside (Rham1), quercetin-3-O-rutinoside (Rutin), and quercetin-3-(2(G)-rhamnosylrutinoside (Rham2) are mono-, di-, and tri-glycosylated forms of quercetin, respectively. In a previous study, we showed that quercetin can enhance ${\alpha}7$ nicotinic acetylcholine receptor (${\alpha}7$ nAChR)-mediated ion currents. However, the role of the carbohydrates attached to quercetin in the regulation of ${\alpha}7$ nAChR channel activity has not been determined. In the present study, we investigated the effects of quercetin glycosides on the acetylcholine induced peak inward current ($I_{ACh}$) in Xenopus oocytes expressing the ${\alpha}7$ nAChR. $I_{ACh}$ was measured with a two-electrode voltage clamp technique. In oocytes injected with ${\alpha}7$ nAChR copy RNA, quercetin enhanced $I_{ACh}$, whereas quercetin glycosides inhibited $I_{ACh}$. Quercetin glycosides mediated an inhibition of $I_{ACh}$, which increased when they were pre-applied and the inhibitory effects were concentration dependent. The order of $I_{ACh}$ inhibition by quercetin glycosides was Rutin${\geq}$Rham1>Rham2. Quercetin glycosides-mediated $I_{ACh}$ enhancement was not affected by ACh concentration and appeared voltage-independent. Furthermore, quercetin-mediated $I_{ACh}$ inhibition can be attenuated when quercetin is co-applied with Rham1 and Rutin, indicating that quercetin glycosides could interfere with quercetin-mediated ${\alpha}7$ nAChR regulation and that the number of carbohydrates in the quercetin glycoside plays a key role in the interruption of quercetin action. These results show that quercetin and quercetin glycosides regulate the ${\alpha}7$ nAChR in a differential manner.
