막걸리 열수 추출물의 아질산염 소거능, 알코올 분해능 및 angiotensin converting enzyme 저해 효과 (Nitrite Scavenging and Alcohol Metabolizing Activities of Hot Water Extract from Makgeoly and Its Angiotensin Converting Enzyme Inhibitory Effect)

  • 전통발효주 막걸리의 기능성을 증명하기 위하여 S사막걸리로부터 구입한 막걸리 침전물의 여러 가지 생리활성에 대하여 조사하였다. 우선 막걸리 침전물 열수 추출물의 항산화 효과를 측정하기 위해 DPPH radical 소거능과 SOD 유사활성을 측정하였다. 그 결과 DPPH법을 통해 측정한 막걸리 침전물 열수 추출물의 radical 소거능은 10 mg/ml에서 48.0%으로 나타났으며, 농도가 증가함에 따라 유의적으로 증가하는 경향을 나타내었다. 또한 SOD 유사활성은 10 mg/ml 농도에서 98.7%로 비교적 높은 SOD 유사활성을 보였다. 항고혈압 활성 측정 실험에서는 현재 시판되고 있는 항고혈압제인 captopril은 0.1 mg/ml에서 93.4%의 ACE 억제효과가 나타났고, 막걸리 침전물 열수 추출물 10 mg/ml에서는 74.0%의 높은 저해 활성을 나타내었다. 따라서, 막걸리 침전물 열수 추출물은 인체에 부작용이 적은 천연 항고혈압소재로서 이용가능성이 높은 것으로 사료된다. 아질산염 소거능 측정 실험에서는 positive control인 Vit. C1 mg/ml의 경우 pH 1.2와 3.0에서는 74~64%, pH 6.0에서는 45%의 소거능을 보인반면, 막걸리 침전물 열수 추출물의 경우 pH 1.2와 3.0에서는 51~42%, pH 6.0에서는 28%의 소거능을 나타내었다. 막걸리 침전물 열수 추출물의 숙취해소 효능은 ADH와 ALDH 활성증진에 막걸리 침전물 열수 추출물이 미치는 영향을 조사함으로써 증명하고자 하였다. 그 결과, 알콜과 acetaldehyde 분해능은 높게 나타났다. 이상의 결과들은 막걸리 침전물의 우수한 기능성으로서의 이용 가능성에 대한 기초자료로 그 가치가 기대된다.

알코올성 위염에 대한 조구등(釣鉤藤)의 항염증 효과 (Anti-inflammatory effect of Uncariae Ramulus et Uncus on alcohol-induced gastritis)

  • Objective : Gastritis refers to an inflammatory disease of the gastric mucosa. Alcohol is one of the main aggression factors, causing bleeding and inflammation in the gastric mucosa and it is known to not only increase lipid peroxide levels, but also deplete key antioxidant factors. The purpose of this study was to determine the effect of Uncariae Ramulus et Uncus water extract (URW) in alcohol-induced gastritis. Methods : The total polyphenol and flavonoid contents of URW were confirmed through an in vitro experiment. Also, 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid (ABTS) radical scavenging activity and ferric reducing antioxidant power (FRAP) activity were confirmed. For in vivo experiments, mice were divided into 4 groups (n=8). Also, 1 hr after oral administration of each drug, 50% ethanol was orally administered to induce gastritis. Results : As a result of in vitro experiments, URW showed excellent antioxidant activity. In alcohol-induced gastritis, URW alleviated the damage to the gastric mucosa caused by alcohol. Also, URW decreased reactive oxygen species (ROS) and malondialdehyde (MDA) levels in serum and gastric tissues, and significantly decreased the expression of NADPH oxidases in gastric tissues. In addition, it significantly modulated the nuclear factor erythroid-derived 2-related factor 2 (Nrf2) and nuclear factor-𝜅B p65 (NF-𝜅B) pathways as well as significantly increased the expression of anti-inflammatory proteins. Conclusions : These results suggest that URW not only reduces oxidative stress through excellent antioxidant activity but also relieves gastric mucosal inflammation as a regulator of Nrf2 and NF-𝜅B pathways.

눈꽃동충하초(Paecilomyces japonica)를 이용한 민속주의 제조 및 생리 기능성 (Manufacture and Physiological Functionality of Korean Traditional Liquors by using Paecilomyces japonica)

  • 눈꽃동충하초(Paecilomyces japonica)를 이용한 새로운 약용민속주를 개발하기 위하여 먼저 알콜발효조건을 검토하였다. 1.0%의 눈꽃동충하초를 함유한 덧밥에 코지를 10%첨가하고 Saccharomyces cerevisiae를 이용하여 제조한 주모를 첨가한 후 $25^{\circ}C$에서 15일간 발효 시켰을 때 에탄올이 가장 많이 생성되었다. 눈꽃동충하초를 0.1%, 0.5%, 1.0%, 2.0%씩 각각 첨가하여 제조한 동충하초 민속주들의 관능검사와 생리기능성을 조사한 결과 눈꽃동충하초를 0.1% 첨가하여 제조한 D-1 눈꽃동충하초 민속주가 기호도가 제일 높았고 혈전용해활성과 아질산염 제거 작용이 각각 11.2U와 19.8%로 대조구인 동충하초를 첨가하지 않고 제조한 민속주보다 우수하였다.

Folic acid supplementation reduces oxidative stress and hepatic toxicity in rats treated chronically with ethanol

  • Folate deficiency and hyperhomocysteinemia are found in most patients with alcoholic liver disease. Oxidative stress is one of the most important mechanisms contributing to homocysteine (Hcy)-induced tissue injury. However it has not been examined whether exogenous administration of folic acid attenuates oxidative stress and hepatic toxicity. The aim of this study was to investigate the in vivo effect of folic acid supplementation on oxidative stress and hepatic toxicity induced by chronic ethanol consumption. Wistar rats (n = 32) were divided into four groups and fed 0%, 12%, 36% ethanol, or 36% ethanol plus folic acid (10 mg folic acid/L) diets. After 5 weeks, chronic consumption of the 36% ethanol diet significantly increased plasma alanine transaminase (ALT) (P < 0.05) and aspartate transaminase (AST) (P < 0.05), triglycerides (TG) (P < 0.05), Hcy (P < 0.001), and low density lipoprotein conjugated dienes (CD) (P < 0.05) but decreased total radical-trapping antioxidant potential (TRAP) (P < 0.001). These changes were prevented partially by folic acid supplementation. The 12% ethanol diet had no apparent effect on most parameters. Plasma Hcy concentration was well correlated with plasma ALT (r = $0.612^{**}$), AST (r = $0.652^*$), CD (r = $0.495^*$), and TRAP (r = $-0.486^*$). The results indicate that moderately elevated Hcy is associated with increased oxidative stress and liver injury in alcohol-fed rats, and suggests that folic acid supplementation appears to attenuate hepatic toxicity induced by chronic ethanol consumption possibly by decreasing oxidative stress.

Diaion HP-20 레진을 이용한 천마 추출물의 불쾌취 제거 (Deodorization of Gastrodia elata Blum Extracts using Diaion HP-20 Resin)

  • Background: Although Gastrodia elata contains the valuable, bio-active compounds gastrodin and 4-hydroxybenzyl alcohol (4-HBA), it also contain ρ-cresol, which is a major source of an objectionable pig slurry-like odor. Accordingly, this study was performed to determine the deodorization effect of Diaion HP-20 resin on a G. elata extract. Methods and Results: Using Diaion HP-20 with an increasing concentration of ethanol, an extract of G. elata was fractionated into 15 fractions, which were anlayzed to determine their concentrations of active compounds and ρ-cresol. The gastrodin, 4-HBA, total phenolics, and ρ-cresol concentrations of the G. elata extract were 403.13, 408.15, 2,476.09, and 11.77 ㎍/㎖, respectively. Among the different fractions, gastrodin and 4-HBA were detected in fractions 3 to- 6, and 4 to- 10, respectively whereas total phenolics were detected in all samples, with fractions 5 to- 7 being characterized by higher concentrations than were the other fractions. ρ-Cresol was detected in fractions 12 to- 15, at concentrations ranging from 0.39 ㎍/㎖ to 6.05 ㎍/㎖ with the highest concentration detected in fraction 14. The fractions containing ρ-cresol could be selectively separated from the G. elata extract without the loss of major bio-active compounds such as gastrodin and 4-HBA. Conclusions: The results of this study indicate that a deodorizing method using Diaion HP-20 resin can be considered for improving the sensory qualities of G. elata extract without a loss of the major bio-active compounds.

캐모마일(Matricaria chamomile)을 이용한 전통 민속주의 제조 및 생리기능성 (Manufacture and Physiological Functionality of Korean Traditional Liquor by using Chamomile (Matricaria chamomile))

  • 캐모마일(Matricaria chamomile)을 이용한 새로운 전통민속주를 개발하기 위하여 먼저 알콜 발효 조건을 검토하였고 캐모마일을 각각 1%, 5%, 10%, 20% 첨가하여 발효주를 제조한 다음 관능검사를 실시하여 기호도가 제일 높은 발효주를 선정하였다. 선정된 발효주의 생리 기능성으로 Angiotensin-converting enzyme(ACE)저해활성과 혈전용해 활성 및 전자공여능 등을 조사하였다. 20%의 캐모마일을 함유한 덧밥에 누룩을 15% 첨가하고 Saccharomyces cerevisiae를 이용하여 제조한 주모를 첨가한 후 $25^{\circ}C$에서 10일간 발효시켰을 때 에탄올이 가장 많이 생성되었다. 캐모마일을 1%, 5%, 10%, 20%씩 각각 첨가하여 제조한 발효주의 관능검사를 실시한 결과 캐모마일을 5% 첨가하여 제조한 C-5 캐모마일 발효주가 기호도가 제일 높았고 색상도 미황색으로 우수하였다. 또한, C-5 캐모마일 발효주는 혈전용해활성과 tyrosinase 저해활성 및 아질산염 소거활성 등이 일반 발효곡주보다 우수하였다.

직접분사식 바이오에탄올-가솔린 혼합연료의 분무 및 희박연소 특성에 관한 실험적 연구 (An Experimental Study on the Spray and Lean Combustion Characteristics of Bio-enthanol-Gasoline Blended Fuel of GDI)

  • As a demand for an automobile increases, air pollution and a problem of the energy resources come to the fore in the world. Consequently, governments of every country established ordinances for green-house gas reduction and improvement of air pollution problem. Especially, as international oil price increases, engine using clean energy are being developed competitively with alternative transportation energy sources development policy as the center. Bio ethanol, one of the renewable energy produced from biomass, gained spotlight for transportation energy sources. Studies are in progress to improve fuel supply methods and combustion methods which are key features, one of the engine technologies. DI(Direct Injection), which can reduce fuel consumption rate by injecting fuel directly into the cylinder, is being studied for Green-house gas reduction and fuel economy enhancement at SI(Spark Ignition). GDI(Galoine Direct Injection) has an advantage to meet the regulations for fuel efficiency and $CO_2$ emissions. However it produces increased number of ultrafine particles, that yet received attention in the existing port-injection system, and NOX. As fuel is injected into the cylinder with high-pressure, a proper injection strategy is required by characteristics of a fuel. Especially, when alcohol type fuel is considered. In this study, we tried to get a base data bio-ethanol mixture in GDI, and combustion for optimization. We set fuel mixture rate and fuel injection pressure as parameters and took a picture with a high speed camera after gasoline-ethanol mixture fuel was injected into a constant volume combustion chamber. We figured out spraying characteristic according to parameters. Also, we determine combustion characteristics by measuring emissions and analyzing combustion.

흰쥐의 배양된 간세포에서 ethanol에 의해 유도된 p42/44 MAPkinase가 IGF system에 미치는 효과 (Effects of ethanol-induced p42/44 MAPkinase activity on IGF system in primary cultured rat hepatocytes)

  • Ethanol abuse is associated with liver injury, neurotoxicity, modulation of immune responses, and increased risk for cancer, whereas moderate ethanol consumption exerts protective effects against liver injury. However, the underlying signal transduction mechanisms of insulin-like growth factors (IGFs) which play an important regulatory role in various metabolism mechanisms are not well understood. We investigated the effects of ethanol-induced p42/44 activity on IGF-I secretion, IGF-I receptor and IGFBP-1 secretion using radioimmunoassay and western blotting in primary cultured rat hepatocytes. The p42/44 activity, IGF-I secretion and IGF-I receptor activity significantly accelerated compared to control at 10 and 30 min after 200 mM ethanol treatment, but then it became suppressed at 180 min. In contrast, IGFBP-1 secretion was inhibited compared to control at 30 min after 200 mM ethanol treatment, but increased at 180 min. The IGF-I secretion, IGF-I receptor and p42/44 activity at 30 min after 200 mM ethanol treatment accelerated with increasing ethanol concentration but IGFBP-1 secretion inhibited (p<0.05). The increased IGF-I secretion, inhibited IGFBP-1 secretion and IGF-IR activity by ethanol-induced temporal p42/44 activity at 30 min after ethanol treatment was blocked by treatment with PD98059. Alcohol dehydrogenase (ADH) inhibitor, 4-methylpyramazole blocked the changes of IGF-I secretion, IGFBP-1 secretion, and IGF-IR activity by ethanol-induced p42/44 activity at 30 and 180 min. Taken together, these results suggest that ethanol is involved in the modulation of IGF-I and IGFBP-1 secretion and IGF-IR activity by p42/44 activity in primary cultured rat hepatocytes. In addition, changing of p42/44 activity by ethanol was caused with ADH.

UV/TiO2 허니컴 반응기에서 페놀의 광산화 반응 (Photocatalytic Degradation of Pheonol in UV/TiO2 Honeycomb Reactor)

  • $UV/TiO_2$ 허니컴 광촉매가 고정된 반응기에서 여러 운전 변수에 따른 페놀의 분해반응을 고찰하였다. 광촉매의 종류에 따른 페놀의 분해능을 비교한 결과, 이시하라사 STS-02로 코팅한 허니컴 광촉매는 데구사 P25에 비해 약간 높은 광산화 활성을 나타내 주었다. 반면에 국내 N사의 광촉매로 코팅한 허니컴의 경우에는 페놀의 광분해 효과가 거의 나타나지 않았다. 허니컴 광촉매는 데구사 P25의 코팅량이 증가함에 따라 페놀의 광산화 반응속도가 증가하는 경향을 나타내주었다. 데구사 P25로 코팅된 허니컴 반응기의 경우에 원수의 유량이 증가함에 따라 페놀의 광산화 속도가 지속적으로 증가하다가 500 mL/min 이상의 유속에서는 감소하는 경향을 나타내었다. 광촉매 표면이 페놀에 의해 미리 흡착된 경우에는 초기 반응시간에 자외선 조사에 의한 페놀의 부분산화 반응이 일어나 오히려 269 nm에서 페놀의 흡광도가 상승하는 결과를 나타내 주었다.

수수 입국과 두류 첨가가 전통주의 품질과 엔지오텐신전환효소 저해활성에 미치는 영향 (Effect of Indian Millet Koji and Legumes on the Quality and Angiotensin I-Converting Enzyme Inhibitory Activity of Korean Traditional Rice Wine)

  • 고혈압 예방 효과를 가진 전통주를 개발하기 위하여 우선 엔지오텐신전환효소(ACE)저해 활성이 있다고 알려진 몇 가지 곡류를 이용하여 입국을 만들어 알콜 발효특성을 조사하였다. 또한 선정된 입국과 각종 두류분말을 덧밥에 첨가하여 전통주를 제조한 후 품질특성과 ACE저해 활성 및 기호도 등을 조사하였다. 수수 입국을 이용하여 전통주를 제조하였을 때 에탄올 생성량은 13.8%로 다른 입국으로 만든 술 보다 약간 낮았으나 ACE저해 활성은 43.3%로 제일 높았다. 또한 녹두가루를 덧밥의 50% 첨가하고 민들레꽃을 1% 첨가한 후 수수 입국과 주모로 $30^{\circ}C$에서 10일간 발효 시켰을때 에탄올 생성량과 항고혈압 활성을 나타내는 ACE저해 활성이 약 69%로 크게 증가하였고 맛과 향도 우수하였다.
