Biosynthesis of lactate-containing polyhydroxyalkanoates (PHAs) was examined in recombinant Escherichia coli W strain from sucrose. The Pseudomonas sp. MBEL 6-19 phaC1437 gene and the Clostridium propionicum pct540 gene, which encode engineered Pseudomonas sp. MBEL 6-19 PHA synthase 1 ($PhaC1_{Ps6-19}$) and engineered C. propionicum propionyl-CoA transferase ($Pct_{Cp}$), respectively, were expressed in E. coli W to construct key metabolic pathway to produce poly(3-hydroxybutyrate-co-lactate) [P(3HB-co-LA)]. The recombinant E. coli W expressing the phaC1437 gene and the pct540 gene could synthesize P(3HB-co-13mol%LA) up to the polymer content of 31.3 wt% when it was cultured in chemically defined MR medium containing 20 g/L of sucrose and 2 g/L of sodium 3-hydroxybutyrate. When Ralstonia eutropha phaAB genes were additionally expressed to provide 3-hydroxybutyrate-CoA (3HB-CoA) from sucrose, P(3HB-co-16mol%LA) could be synthesized from sucrose as a sole carbon source without supplement of sodium 3-hydroxybutyrate in culture medium, but the PHA content was decreased to 12.2 wt%. The molecular weight of P(3HB-co-16 mol%LA) synthesized in E. coli W using sucrose as carbon source were $1.53{\times}10^4$ ($M_n$) and $2.78{\times}10^4$ ($M_w$), respectively, which are not different from those that have previously been reported by other recombinant E. coli strains. Engineered E. coli strains developed in this study should be useful for the production of lactate-containing PHAs from sucrose, one of the most abundant and least expensive carbon sources.
고구마는 21세기 인류가 당면한 식량, 에너지, 환경, 보건문제 등을 해결하는 21세기 구원투수로 등장하고 있다. 고구마는 식량자원 뿐만 아니라 바이오에탄올, 기능성 사료, 항산화물질 등 고부가가치소재를 생산하는 생체반응기로 평가된다. 미국 공익과학단체(The nonprofit Center for Science in the Public Interest, CSPI)는 고구마가 저분자항산화물질, 식이섬유, 칼륨 등을 고함유하고 있는 고구마를 몸에 좋은 10대 슈퍼식품 가운데 하나로 선정하였다. 미국 농무부는 고구마를 전분작물 가운데 식량수급에 영향을 최소화하는 척박한 토양에 가장 적합한 바이오에너지작물로 평가하였다. UN 식량농업기구는 2050년에 세계인구가 97억 명이 될 것이며 지금 추세로 식량을 사용하면 2050년에는 지금의 1.7배의 식량이 필요하다고 전망했다. 어떻게 미래 식량위기를 극복할 것인가? 이러한 측면에서 척박한 토양에서도 어느 정도 수량을 보장하는 고구마가 지구가 당면한 제반 문제를 해결할 수 있을 것으로 기대된다. 본 논문에서는 세계 고구마 유전자원과 생산현황을 살펴보고, 글로벌 식량자원 및 고부가가치 바이오소재 측면에서 새로운 고구마의 북방로드 개척 및 상업적 대량재배를 위한 선결과제를 기술하고자 한다.
본 연구에서는 미생물 검출용 바이오센서 제작을 위해 재조합 람다 파아지 꼬리 단백질 J을 센싱 요소로 활용가능한지를 조사하였다. 융합 단백질의 입체 장애를 최소화하기 위해 J 단백질 절편의 N-말단을 6X His-tag로 융합하고 HisTALONTM 컬럼으로 정제하였다. 정제 단백질은 약 38 kDa 크기를 SDS-PAGE에서 나타내며 anti-His 단클론 항체와 반응하였다. Anti-His 단클론 항체는 6HN-J를 처리한 E. coli K-12와 특이적으로 결합하나 BSA 처리하거나 6HN-J 처리한 다른 미생물들(Salmonella typhimurium, Bacillus subtilis, Pseudomonas aeruginosa)과는 결합되지 않음을 보여주었다. 또한 정제 단백질과 숙주세포의 결합은 온전한 람다 파아지의 in vivo 숙주 표면 흡착을 약 $1{\mu}g/ml$ 단백질 농도에서 50%, $25{\mu}g/ml$ 단백질 농도에서 거의 완전히 방해하였다. 결론적으로 재조합 6HN-J 단백질은 탁월한 선택성과 선별성으로 인해 바이오센서의 제작에서 센싱 요소로 활용 가능함을 시사한다.
비만은 이제 선진국 뿐만 아니라 개발도상국에서도 가장 문제가 되는 대사성 질환이다. 최근 치료보다는 예방에 중점을 두는 예방의학 시대가 도래함에 따라, 인슐린 저항성 비만과 당뇨병 등의 대사 증후군을 예방하기 위한 천연물들이 큰 관심을 받고 있다. 특히, 베타글루칸은 면역계와 혈중 콜레스테롤 조절에 이로운 효과가 있는 것으로 알려져 있지만 비만에 미치는 영향에 대해서는 완전히 밝혀지지 않았다. 이에 본 연구에서는 β-1,3/1,6-glucan의 함량이 전체 글루칸 함량의 90% 이상 되는 자체 개발 흑효모 균주 SM-2001 추출물(폴리칸)이 3T3-L1 지방전구세포의 지방세포 분화에 미치는 영향을 조사하였다. 폴리칸은 지방전구세포에서 지방축적과 GPDH 효소 활성을 억제하는 것으로 나타났다. 이는 폴리칸이 세포 내에서 지방의 축적을 조절하는 전사인자인 PPARγ와 C/EBPα의 하향조절과 세포 내 에너지 센서 역할을 하는 AMPK 효소의 인산화를 유도함으로써 항비만 효과를 발현하는 것으로 확인되었다. 본 연구를 통해 폴리칸의 항비만 효과를 확인하였으며, 향후 비만과 대사성 질환을 예방할 수 있는 식의약 소재로써 그 활용가치를 더욱 높일 수 있을 것으로 기대한다.
3 차원 패키징을 위해 열가압 공정으로 제조된 Cu-Cu 접합 계면의 접합 특성을 평가하기 위해 4 점 굽힘 실험을 수행하였다. Cu가 코팅된 Si 웨이퍼 2 장을 $350^{\circ}C$에서 1 시간 동안 15kN 의 하중으로 접합시킨 후, 동일한 온도에서 1 시간동안 어닐닝을 수행하였다. 접합된 웨이퍼를 $30\;mm\;{\times}\;3\;mm$ 크기로 잘라 시험편을 준비하였다. 시험편의 중심에 깊이 $400\;{\mu}m$의 노치를 가공하였다. 시험기에 광학계를 부착하여 노치에서의 크랙 발생과 계면에서의 크랙 진전을 관찰하였다. 일정한 테스트 속도로 실험을 수행하여, 이에 상응하는 하중을 측정하였다. Cu-Cu 접합 계면 에너지는 $10.36\;J/m^2$ 으로 측정되었으며, 파괴된 계면을 분석하였다. 표면 분석 결과, $SiO_2$와 Ti의 계면에서 파괴가 일어났음을 확인하였다.

Oilseed crop Camelina (Camelina sativa L.) is a suitable for biodiesel production that has high adaptability under low-nutrient condition like marginal land and requires low-input cost for cultivation. Enhanced abiotic stress tolerance of Camelina is very important for oil production under the wide range of different climate. CsRCI2s (Rare Cold Inducible 2) are related proteins in various abiotic stresses that predicted to localized at plasma membrane (PM) and endoplasmic reticulum (ER). These proteins are consist of eight-family that can be divided into tail (CsRCI2D/E/F/G) and no-tail (CsRCI2A/B/E/H) type of C-terminal. However, it is still less understood the function of C-terminal tail. In this study, CsRCI2D/H genes were cloned through gateway cloning system that used pCB302-3 as destination vector. And we used agrobacterium-mediated transformation system for generation of overexpression (OX) transformants. Overexpression of target gene was confirmed using RT-PCR and segregation ratio on selection media. We analyzed physiological response in media and soil under abiotic stresses using CsRCI2D and CsRCI2H overexpression plant. To compare abiotic stresses tolerance, wild type and CsRCI2D/H OX line seeds were sown on agar plate treated with various NaCl and mannitol concentration for 7 days. In the test of growth rate under abiotic stress on media, CsRCI2H OX line showed similar to NaCl and mannitol stress. In the other hand, CsRCI2D OX line showed to be improved stress tolerance that especially increased in 200mM NaCl but was similar on mannitol media. In greenhouse, WT and CsRCI2D/H OX lines for physiological analysis and productivity under abiotic stresses were treated 100, 150, 200mM NaCl. Then it was measured various parameters such as leaf width and length, plant height, total seed weight, flower number, seed number. CsRCI2H OX line in greenhouse did not show any changes in physiological parameters but CsRCI2D OX line was improved both physiological response and productivity under NaCl stress. Among physiological parameters of CsRCI2D OX line under NaCl stress, leaf length and width were observed shorter than WT but it were slightly longer than WT in 200mM NaCl stress. Furthermore, total seed weight of CsRCI2D OX line under stress displayed to decrease than WT in normal condition, but it was gradually raised with increasing NaCl stress then more than WT relatively. These results suggested CsRCI2D might be contribute to improve abiotic stress tolerance. However, function of CsRCI2H is need to more detail study. In conclusion, overexpression of CsRCI2s family can generate various environmental stress tolerance plant and may improve crop productivity for bio-energy production.
본 연구에서는 바이오가스의 연소엔진을 이용한 에너지 전환시에 발생되는 $NO_x$와 CO의 배출특성을 분석하고 나아가 배출계수를 산정하고자 하였다. 바이오가스의 주성분인 메탄을 70%로 한 합성가스를 이용하여 실험한 결과 연소엔진의 표준상태에서 $NO_x$와 CO가 각각 4 ppm과 100 ppm의 배출농도를 나타내었고, 이는 1.29g/MMBtu와 30.86 g/MMBtu의 배출계수 값을 산정할 수 있었다. 바이오가스의 주성분인 메탄을 60%로 한 약간의 과잉공기의 상태에서는 $NO_x$와 CO가 각각 2 ppm과 200 ppm의 배출농도를 나타내었고, 이는 정격 조건에 비해 투입열량이 적어져 연소온도의 저하로 인하여 열적 반응에 의해 생성되는 $NO_x$는 줄고, CO는 증가함을 알 수 있었다. 미국 EPA의 배출계수를 비교하기 위하여 투입된 연료의 열량을 기준으로 비교해 볼 때 본 연구의 결과가 $NO_x$의 경우 근사한 값을 보여 주었고, 이는 국내 바이오 가스 연소시의 오염배출계수로 잘 적용 가능함을 보여 주었다. 본 연구에서 사용한 가스엔진이 오염배출 측면에서 대기 환경보전법상의 배출 허용기준 내에서 가동됨을 알 수 있었고, 가동조건에 따라 연소온도를 증가시킴으로서 CO의 발생을 저감시킬 수 있으리라 사료된다.
An experiment was conducted to investigate the effects of microbial phytase ($Natuphos^{(R)}$) supplementation in combination with carbohydrases (composed of enzymes targeted to soybean meal (SBM) dietary components such as $\alpha$-galactosides and galactomannans; $Endo-Power^{(R)}$) to corn-soybean meal based diet (CSD) and complex diet (CD) with a partial replacement of SBM with rape seed meal (RSM) and cotton seed meal (CSM) on growth performance and nutrient digestibility of growing pigs. A total of 168 growing pigs averaging $13.18{\pm}1.77kg$ of initial body weight was arranged as a $2{\times}2$ factorial design with main effects of diet types (corn-SBM based diet (CSD) and complex diets (CD; 5% of SBM was replaced with 2.5% of RSM and 2.5% of CSM in diet for phase I (0 to 3 weeks) and 6% of SBM was replaced with 3% of RSM and 3% of CSM in diet for phase II (4 to 7 weeks))) and enzyme supplementation (none and 0.1% of phytase (500 FTU/kg diet) and 0.1% of carbohydrases). The diet with enzyme application were formulated to have a 0.18% unit lower aP than diets without enzyme application. Each treatment had three replicates with 14 pigs per replicate. To determine supplementation effect of phytase and carbohydrases on ileal amino acid digestibility of SBM, RSM and CSM, a total of 18 T-cannulated pigs (initial body weight; $13.52{\pm}1.24kg$) were assigned to six dietary treatments in the present study. Dietary treatments in metabolic trial included 1) SBM diet, 2) SBM diet+with enzymes (phytase (500 FTU/kg) and carbohydrases at 0.1%, respectively), 3) CSM diet, 4) CSM diet+enzymes, 5) RSM diet and 6) RSM diet+enzymes. During whole experimental period (0 to 7 wks), there was no difference in growth performance between diets (CSD and CD). However, dietary phytase and carbohydrases supplementation significantly improved gain/feed ratio (G:F) of growing pigs. During the phase II (4-7 weeks), dietary phytase and carbohydrases supplementation significantly improved all fecal nutrient digestibilities (Dry matter (DM), gross energy (GE), crude protein (CP), crude fat (CF), calcium (Ca) and phosphorus (P)). Dietary phytase and carbohydrases supplementation improved significantly overall ileal amino acid digestibilities of SBM, RSM and CSM based diets (p<0.05). The simultaneous inclusion of phytase and carbohydrases in both of CSD and CD reduced feed cost per kg body weight gain (FCG). Also, results suggest that 2.5 to 3% of RSM and CSM, respectively, might be used as a protein source in growing pig diets without having an adverse effect on the growth performance and nutrient digestibility and simultaneous phytase and carbohydrases addition improves nutritional value of SBM, RSM and CSM by improving ileal amino acid digestibilities.

A total of 140 weaning pigs were used to determine the effects of digestive enzyme supplementation to corn-soybean meal diets on growth performance, physiological changes of small intestine, microorganisms and pH in the gastrointestinal tract. Two kinds of enzyme complex (A, B) were used in this experiment. Pigs were allotted in a completely random design (CRD) to five replicates with four pigs per pen. Diets and water were provided for ad libitum consumption. Treatments included 1) Control: without enzyme supplementation, 2) Enzyme A 0.05%, 3) Enzyme A 0.10%, 4) Enzyme A 0.15%, 5) Enzyme B 0.05%, 6) Enzyme B 0.10%, 7) Enzyme B 0.15% in the diets. A total of 24 crossbred barrows 25.78${\pm}$0.55 kg BW fitted with simple ileal T-cannulas were used to evaluate the effect the enzyme addition on the nutrient digstibility. Pigs were allotted 4 treatments (No enzyme, enzyme A 0.05%, enzyme A 0.1%, enzyme A 0.15%), 6 replicates according to a completely random design (CRD). Another digestibility trial was followed for enzyme complex B. Twenty pigs, average 31.92${\pm}$0.37 kg BW, fitted with simple ileal T-cannulas for digestibility trial. Neither enzyme A nor enzyme B affected on fecal or ileal digestibility of dry matter, gross energy, crude protein, crude fat and crude ash (p>0.05). The apparent fecal digestibilities of all the nutrients were higher in total feces collection method than in indirect method. At the end of feeding trial, 21 pigs were slaughtered for examining the morphological changes of small intestine and the concentration of microorganisms in the ileum and the colon. Growth performance, intestinal morphology and pH of ileum and colon were not affected by the either enzyme complex supplementation (p>0.05). These results suggested that enzyme complex A and enzyme complex B were of no benefit to early-weaned pigs when corn-soybean meal based diet was provided.
본 연구에서는 길이 4.5 m와 7.5 m의 다중관 $CO_2$ 급탕 열교환기의 열전달 및 압력강하 특성을 ${\epsilon}-NTU$ 방법을 사용하여 해석하고 결과를 기존의 실험 데이터와 비교하였다. 급탕 열교환기는 쉘측에 물이 흐르고 8개로 구성된 내부 튜브에 $CO_2$를 흐르게 하였으며 열전달 효율을 최대화하기 위하여 대향류로 설계하였다. 각 노드에 대한 물과 $CO_2$ 냉매의 유동에 대한 에너지 평형 방정식은 단면분할법을 이용하여 해석하였다. 열전달율 계산값은 실험값과 ${\pm}5%$ 범위 내에서 잘 일치하였다. 반면에 물의 출구온도는 물 유량이 증가함에 따라 거의 선형적으로 감소하며 계산값과 실험값은 ${\pm}3%$ 내에서 일치하였다. 결과에서 열전달율은 4.5 m와 7.5 m 급탕 열교환기 모두 물 유량 또는 $CO_2$ 입구온도가 증가함에 따라 거의 선형적으로 증가하였으며, 반면에 물 유량이 증가함에 따라 물의 출구온도는 선형적으로 감소하였다. $CO_2$ 압력강하 계산값과 실험값은 $CO_2$가 고유량일 때 5 % 내에서 잘 일치한 반면에 $CO_2$가 저유량일 때 실험값이 약 20 % 높게 나타났다.

제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
