  • The mechanism of how cholelithiasis increases the risk of acute pancreatitis remains obscure. When gallstones obstruct the lower biliary tract, bile acids can enter the pancreas either by luminal diffusion or by interstitial leakage. Here we provide the first evidence that bile acids can be transported into pancreatic acinar cells through the membrane transporters and induce cell death by impairing intracellular Ca$\^$2+/ signals.(omitted)

Dietary ε-Polylysine Decreased Serum and Liver Lipid Contents by Enhancing Fecal Lipid Excretion Irrespective of Increased Hepatic Fatty Acid Biosynthesis-Related Enzymes Activities in Rats

  • ${\varepsilon}$-Polylysine (EPL) is used as a natural preservative in food. However, few studies have been conducted to assess the beneficial functions of dietary EPL. The purpose of this study was to elucidate the mechanism underlying the inhibition of neutral and acidic sterol absorption and hepatic enzyme activity-related fatty acid biosynthesis following EPL intake. EPL digest prepared using an in vitro digestion model had lower lipase activity and micellar lipid solubility and higher bile acid binding capacity than casein digest. Male Wistar rats were fed an AIN-93G diet containing 1% (wt/wt) EPL or L-lysine. After 4 weeks of feeding these diets, the marked decrease in serum and liver triacylglycerol contents by the EPL diet was partly attributed to increased fecal fatty acid excretion. The activities of hepatic acetyl-coenzyme A carboxylase and glucose-6-phosphate dehydrogenase, which are key enzymes of fatty acid biosynthesis, were enhanced in rats fed EPL diet. The increased fatty acid biosynthesis activity due to dietary EPL may be prevented by the enhancement of fecal fatty acid excretion. The hypocholesterolemic effect of EPL was mediated by increased fecal neutral and acidic sterol excretions due to the EPL digest suppressing micellar lipid solubility and high bile acid binding capacity. These results show that dietary EPL has beneficial effects that could help prevent lifestyle-related diseases such as hyperlipidemia and atherosclerosis.

초식동물 쓸개즙 추출물의 천연항산화 성분; 생물학적인 기능 및 특성규명 (Natural Antioxidant Activity of Ethanol Extracted from Bovine Bile ; Biological Effects and Characterization)

  • 본 연구에서는 Bovine bile에서 항산화 물질분획을 분리하고 이에 따른 항산화 효소들의 여러 활성효과를 측정하였으며 GC/FID에 의한 항산화성 물질의 패턴을 확인하였다. 항산화력은 Butylated hydroxy toluene(BHT)에 비해 Bovine bile의 EtOAc추출물(BBE)이 51.2%로 나타났고 xanthine oxidase를 측정 48.3%의 저해효과를 보였으며 glutatione-S-transferase의 활성은 85.7%의 활성이 나타났다. MTT방법에 의한 세포생존율의 조사에서는 6mM glutathione 투여에서 95%라는 생존율과 비교하여 20mg, 60mg의 BBE를 처리했을 때 56%와 65%, 140mg처리에서는 78%의 세포생존율이 나타났다. Bovine bile EtOAc추출물을 TLC로 분석한 결과 Rf값이 0.72에서 항산화 활성이 나타났고 이 분획을 methylation과 trimethysilyl(TMS) 유도체화를 시켜서 Gas Chromatography(FID)와 mass spectrum을 통한 성분 분석을 하고 bile acid의 구성분과 비교하여 계통분석을 실시한 결과 Coprostan-3ol로 추정되었다.

Effects of Intravenous Administration of Taurocholate on Hepatic Catechol-O-Methyltransferase Activity in Common Bile Duct Ligated Rats

  • Possible mechanism of decreased catechol-O-methyltransferase (COMT) activity in cholestatic rat liver was studied. Hepatic and serum COMT activities were determined from the experimental rats with common bile duct ligation (CBDL). The Michaelis-Menten constants in this hepatic enzyme were also measured. The activities of cytosolic, mitochondrial and mircosomal COMT as well as their Vmax values were found to be decreased significantly in CBDL plus taurocholic acid (TCA) injected group than in the control group, such as CBDL alone groups. However, their Km values in the experimental groups did not vary. Serum COMT activity increased slightly in the CBDL plus TCA injected group than in the control group. The above results suggest that TCA represses biosynthesis of the COMT in the liver. The elevated activity of the serum COMT is believed to be caused by the increment of membrane permeability of hepatocytes upon TCA mediated liver cell necrosis.

요구르트에서 분리한 Lactobacillus들의 담즙산염 분해 능력 (Bile Salt Deconjugation Activity of Lactobacillus Strains Isolated from Yogurt Products)

  • To investigate bile salt hydrolase activities of the bacterial strains isolated from fermented milk products, 21 strains of Lactobacillus were tested for their abilities to produced cholic acid from taurocholic and glycocholic acids. The production of cholic acid was measured by HPLC analysis during the growth in broth media for 24hrs. All strains of Lactobacillus acidophilus and L. plantarum deconjugated both taurocholate and glycocholate, whereas none strains of L. delbrueckii subsp. bulgaricus, L. casei subsp. casei, L. casei subsp. rhamnosus, L, reuteri did. L. acidophilus stains isolated from yogurts had the higher decojugation activities on glycocholate than taurocholate, however, L. acidophilus 1009 isolated from the human intestine showed the similar deconjugation activities on both taurocholate and glycocholate.

비소화성 물질과 칼슘 첨가가 흰쥐의 지질대사에 미치는 영향 (Effects of Nondigestable Substances and Calcium on Lipid Metabolism in Rats)

  • This study was conducted to investigate the effect of nondigestable substances and calcium such as oligosaccharide, agar, saponin, tannin and calcium on the reduction of lipid status in rats fed high fat diet. In order to make the observation, the lipid content in plasma, liver and the feces, and bile acid excretion were measured of r 4 weeks. the results obtained from this research are as follows. Concentration of total lipid in plasma seemed highest in the control group and were significantly lower in groups oil-gosaccharide, agar and calcium-tannin, compared to the control group. Concentration of total cholesterol in plasma was significantly lower in groups oligosaccharide, agar, calcium and calcium-saponin, compared to the control group. The improvement in lipid status seems to be insignificant with oligosaccharide, showed increase in total bile acids excretion in feces and decrease in total cholesterol in plasma, compared to the control group. These evidence seems to indicate improvement of the plasma lipid status by calcium and agar supplementation.

  • This study was undertaken to determine the effect of yellow and black soybeans on plasma and hepatic lipid composition and fecal lipid excretion in Sprague Dawley rats. Animals were fed with diets containing 52% yellow soybean, 50% black soybean, or 20% casein for 7 weeks. Feeding efficiency was significantly increased in the animals fed soybeans(p<0.05). Plasma cholesterol concentration was significantly lowered in the group fed yellow soybean compared with black soybean and casein-fed control (p<0.05). Hepatic triglyceride concentration was significantly lowered in soybean groups compared with casein-fed control(p<0.05), whereas hepatic cholesterol concentration was not affected by diet treatments. Soybean feeding significantly increased fecal weight, triglyceride and bile acid contents compared with casein feeding(p<0.05). It is concluded that soybean feeding in rats affects plasma and hepatic lipid levels by increasing the excretion of triglyceride and bile acids.

담즙산과 cholesterol 및 phosphatidylcholine이 vacuolar apparatus에 미치는 영향에 관한 미세구조적 관찰 (Some Observations on the Fine Structure of Vacuolar Apparatus Affected by Dehydrocholic acid, Cholesterol and Phosphatidylcholine)

  • 본 연구에서는 담즙산과 cholesterol 및 phospholipid가 흰쥐 간세포내 소기관에 미치는 영향을 조사하여 이들 담즙물의 수송기전을 알아 보고져 하였다. 정상군과 각 실험군에서 관찰된 Golgi장치는 거의가 cis side를 담세관으로 향하고 있었다. 각 실험군에서는 ER과 Golgi 장치 및 용해소체 등 소기관의 수적 증가, 소조의 팽대, budding 등이 관찰되었다. Dehydrocholic acid 투여군에서는 cis Golgi cistern의 선상 팽대을 많이 볼 수 있었는데 공포(vacuole)들은 담세관과 Golgi 장치사이에서 관찰되었으며 ER과 담세관 사이에서도 관찰할 수 있었다. Cholesterol 투여군과 Phosphotidylcholine 투여군에는 Golgi 장치의 모든 소조들이 팽대되어 있었으며 ER 유래의 공포와 용헤소체가 증가되어 있었는데 용해소체의 증가는 특히 cholesterol 투여군에서 현져하였다. 이상의 소견으로 미루어 담즙성분의 분비에 관여하는 주된 세포내 소기관은 ER과 Golgi 장치 및 용해소체로 추정된다. 그러나 dehydrocholic acid는 ER과 cis Golgi 소조 유래의 공포에 의해 분비되며 Cholesterol과 phosphatidylcholine은 ER에서 또는 trans Golgi 소조을 거친 후 용해소체를 통해서 배출될 것으로 추정되는데 특히 cholesterol은 용해소체를 통해서 빈번히 배출될 것으로 생각된다.

Chenodeoxycholic Acid에 의한 파골전구세포의 증식 조절 (The Regulatory Role of Chenodeoxycholic Acid on the Proliferation of Osteoclast Precursor Cells)

  • We investigated the effect of Chenodeoxycholic acid (CDCA) on the proliferation of osteoclast precursor cells. CDCA decreased the proliferation of osteoclast precursor cells through the control of cell cycle regulators such as cyclin D1, p21 and p27. When we checked the signaling pathway, CDCA decreased Erk activation in osteoclast precursor cells. Furthermore, two bile acid receptors, FXR and TGR5, were involved in the suppressive effect of CDCA. Taken together, this study suggested that bile acid plays an important role in the proliferation of osteoclast precursor cells.
