Probiotics are live microbial feed supplements which beneficially affect the host animal by improving its intestinal microflora. To select the best Lactobacillus spp. as a chicken probiotic, probiotic characteristics of 10 selected Lactobacillus strains isolated from chicken cecum or obtained from KCTC were investigated. The strains were examined for resistance to pH 2.0 and 0.3% oxgall, and adhesion to cecal mucus and cecal epithelial cells. All strains grew in MRS containing 0.3% oxgall. However, Lb. plantarum AYM-10, Lb. fermentum YL-3, AYM-3, and Lb. paracasei YL-6 showed relatively high resistance to 0.3% oxgall. Lb. fermentum YL-3, YM-5, AYM-3, and Lb. paracasei YL-6 survived 4 hours of incubation at pH 2.0. Lb. fermentum YL-3, KCTC 3112, and Lb. plantarum AYL-5 were strongly adhesive to cecal mucus, while the rest showed moderate or low adhesion. Lb. plantarum AYM-10, AYL-1, and AYL-5 had good adhering properties to cecal epithelial cells (30.7$\pm$10.82, 40.2$\pm$20.90, and 14.5$\pm$4.22, respectively). Lb. fermentum YL-3, AYM-3, and KCTC 3547 showed Intermediate adhesion ability, and Lb. plantarum showed better adhesion ability to cecal epithelial cells than Lb. fermentum. Attached Lb. fermentum YL-3 to cecum after 60 min incubation was confirmed using CLSM. Lb. fermentum YL-3 attached to a matrix which was composed of a mucus layer adjacent to intracrypts and pericryptal region. Some Lb. fermentum YL-3 bound to mucosal epithelial cells. From these results, Lb. fermentum YL-3 was selected as a chicken probiotic. In vivo trials of chicks inoculated with Lb. fermentum YL-3 had decreased Salmonella population in cecal contents and livers (p<0.5).

To study the lipid adsorption characteristic of chitosans with different molecular weights and the degrees of deacetylation, in vitro test and near-infrared (NIR) spectroscopic analysis have been performed for the measurement of lipid adsorption characteristics of chitosan. The degrees of deacetylation in chitosans were $70{\%},\;85{\%}\;and\;92{\%}$ at different deacetylation times (1 hr, 2 hrs, 3 hrs), respectively. The molecular weight of each chitosan was controlled by enzymatic hydrolysis, and then the molecular weight of the chitosan was 4 kDa. The bulk density, water holding capacity and fat binding capacity of each chitosan powder were $96.2-504.0{\%},\;374.4-1217.9{\%},\;and\;307.0-659.3{\%}$, respectively. The higher molecular weight of chitosan was exhibited the lower bulk density and the higher water and fat binding capacities. Bindinf capacities of chitosan powders to bile salts, cholesterol and linoleic acid were $41.2-63.3{\%},\;40.8-67.4{\%},\;42.6-72.6{\%}$, respectively. In NIR spectrum of lipid adsorbed chitosan the occurrence static eletronical binding between chitosan and lipid was identified by NIR spectrum peak induced from combination of carboxylic group in lipid and amino group in chitosan. In conclusion, the higher degree of deacetylation and molecular weight of chitosan showed the higher lipid binding capacity and the lipid adsorption of chitosan were occurred by combination of carboxylic group in lipids and amino group in chitosan.

Objective: Dyslipidemia is recognized as a prominent risk factor for cardiovascular and cerebrovascular diseases but it is manageable through therapeutic and lifestyle intervention. Interpreting the latest guidelines is essential for an application of recommendation from guidelines into clinical practice. Therefore, this study aimed to compare the most recent guidelines on dyslipidemia treatment recommendations in Korea and USA. Methods: This study analyzed and compared 2015 Korean guidelines for the management of dyslipidemia, 2013 American College of Cardiology/American Heart Association (ACC/AHA) guideline and 2016 supportive guidelines from ACC. Results: A comparison was made focused on the following: target patients based on cardiovascular risk assessment, target goal, and treatment strategies including statin and non-statin therapies. Four target patient groups by risk were suggested in 2015 Korean guideline and cardiovascular risk factors were also considered for initiation of lipid lowering therapy. Titrated statin regimen was recommended by Korean guideline to reach LDL cholesterol and non-HDL cholesterol target level. In 2013 ACC/AHA guideline, four statin benefit group was introduced considering ASCVD risk and high intensity statin or intermediate intensity statin use were recommended without dose titration. 2016 update was to support non-statin therapy based on updated evidence and new consideration of ezetimibe, PCSK9-inhibitor and bile acid sequestrant was brought up. Conclusion: Guidelines are continuously updating as new and important clinical data are constantly released along with the advent of newly approved drugs for lipid disorder. This article provides resources that facilitates uptake of these recommendations into clinical practice.
The difference in lipid metabolism between breast-fed(BF) and formula-fed(FF) infants were studied in 31 Korean infants at 1, 2 and 3 months postpartum. The formulas had more total lipids(TL), triglycerides(TG) and phospholipids and less cholesterol(CHOL) and free fatty acids(FFA) than the breast milk. The milk consumption of the FF infants was significantly greater than that of the BF infants with a wide individual variation. As a result, the FF infants appeared to consume more TL, TG and PL and less CHOL and FFA than the BF infants during 3 months. The lipid contents of the breast milk tended to decrease in due course of lactation, therefore the intake of lipids of BF infants was reduced during the first three month of lactation. The plasma TG and CHOL levels of infants at 3 month were not significantly different between the BF and FF infants, but the PL level of the BF infants was higher then that of the FF infants. The plasma levels of TG and CHOL were not significantly correlated with the intakes of TL, TG, CHOL, PL and FFA, respectively. The PL level, however, was positively correlated with the intakes of CHOL and FFA, respectively and negatively correlated with PL intake as well as 18 : 2w6 fatty acid content of the breast milk or the formulas. The fecal weight and fecal loss of TL of the FF infants were greater than those of the BF infants during 3 months, however, apparent lipids digestibility was not significantly different between the BF and FF infants. The fecal excretions of CHOL and bile acids of FF infants were substantially higher than those of BF infants during 3 months. New sterol balance showed a particularly large difference between the BF and FF infants. The value of the net sterol balance of the BF infants was negative, but that of the FF infants was positive. This study shows that the consumption pattern of various lipid components of BF and FF infants were different as a result of different lipid compositions between breast milk and formula. The significantly lower sterol balance of the BF infants than the FF infants may have been derived from the unique dietary characteristic of breast milk.
The objective of this study was to develop a paper strip which could determine E. coli qualitatively and quantitatively in water, wastewater, drinks, or food. This paper strip method was a simple and rapid test method that determine E. coli by visual identification. In this study, nutrient culture media were formulated and characterized for optimum conditions. Paper strips were then prepared by impregnating into the media and dried at $60^\circ$C. The test procedure is quite simple to use. The paper strip was dipped into a sample, and excess sample was removed. The strip was then incubated at $37^\circ$C for 16 to 20 hours and the number of colonies on the strip was counted. The color of the colony spots produced by microorganisms varied depending on the media formulation. Violet-red spots were produced by E. coli. The test method was simple, rapid and no special laboratory equipment was necessary for visual identification. Therefore, this test method is applicable to on-site tests such as field tests or home tests. The paper strip method was compared with the standard agar plate method and Japanese commercial product. The method of the economical preparation of test strips was studied for production on industrial scale.
Strain NK34 was characterized for probiotic use. Strain NK34 was named Lactococcus lactis NK34 based on API 50 CHL kit results and 16S rDNA sequencing. L. lactis NK34 was highly resistant to artificial gastric juice (pH 2.5) and artificial bile acid. Based on results from the API ZYM kit, 4 enzymes were produced. L. lactis NK34 was resistant to all antibiotics tested except for $10\;{\mu}g/mL$ roxithromycin and $10\;{\mu}g/mL$ erythromycin. The cholesterol-lowering effect of L. lactis NK34 was about 46.9%. Concentrations of interleukin $(IL)-1{\alpha}$ in the $20{\times}$ concentrated supernatant of L. lactis NK34 was about 361 pg/mL. L. lactis NK34 was also found to inhibit the growth of colon cancer cells due to MNNG-induced DNA damage. These results demonstrate the potential of L. lactis NK34 as a health-promoting probiotic.
The Bacillus KU801 strain, due to its potential in the field of probiotics for animal use, was isolated from chicken feces. Strain KU801 was identified as Bacillus amyloliquefaciens KU801 based on the results of 16S rRNA sequencing. Vegetative and spore cells of B. amyloliquefaciens KU801 were resistant to artificial gastric juice and artificial bile acid. B. amyloliquefaciens KU801 was found to inhibit the production of nitric oxide (NO) and increase the production of Interleukin-1 alpha (IL-1${\alpha}$). DNA damage induced by N-methyl-Ntion of ninitroso-guanidine (MNNG) was significantly inhibited, in a dose dependent manner, by preincubating MNNG together with B. amyloliquefaciens KU801. These results demonstrate the potential use of B. amyloliquefaciens KU801 as a feed additive.
This study was conducted to obtain a good probiotic strain of L. acidophilus from infant feces which have the acid and bile tolerance. The selection criteria for the strain included antimicrobial activity, serum cholesterol reduction, resistance to the hydrogen peroxide, angiotensin converting enzyme (ACE) inhibition activity and iron solubility. To this end, five probiotic Lactobacillus strains have been isolated from infant feces. Especially, L. acidophilus SD 105 had strong antimicrobial activity against Listeria sp., high deconjugation activity in the medium which contained 0.5% of glycocholate (GCA) and high resistance to the hydrogen peroxide. L. acidophilus SD 102 showed the highest ACE inhibition activity among the tested cultures and L. acidophilus SD 103 showed iron solubility of more than 70%.
Intestinal absorption of dietary taurine is one of the regulatory component maintaining taurine homeostasis along with renal reabsorption, bile acid conjugation and secretion, and de nobo synthesis of taurine in mammals. Recent observations of decreased enterocytic levels of taurine in response to trauma, infection and surgical insults, postulate the possibility that intestinal taurine absorption might be impaired in such stressed conditions. The aim of the present study was to evaluate changes in enterocytic taurine transporter activity using the human intestinal colon carcinoma cell line, HT-29, in various stress-induced conditions. Pretreatment of the HT-29 cells with dexamethasone, a stress hormone(0.1,1,10 or 100$\mu$M) for 3 hrs, or with E coli heat-stable enterotoxin(10, 100, or 200nM) for 30 minutes in order to induce the condition of enterotoxigenic infection did not influence taurine uptake as compared to the value found in control cells. In contrast, pretreatment of the cells with cholera toxin(10, 100, 500, or 1000ng/ml)for 3hr or 24hr significantly decreased taurine uptake by HT-29 cells to 40~50% of the value found in untreated control cells. Kinetic studies of the taurine transporter activity were conducted in control and cholera toxin treated HT-29 cells with varying taurine concentrations(2~60$\mu$M) in the uptake medium. Pretreatment of the cells with cholera toxin(100ng/ml) for 3hr did not influence the Vmax, but resulted in a 55% increase in the Michaelis-Menten constant(Km) of the taurine transporter compared to those in control cells. These results suggest that cholera toxin-induced reduction in taurine transporter activity in HT-29 cells is associated with decreased affinity of the taurine transporter without altering the amount of transporter protein. Intestinal taurine absorption appears to be reduced in the condition of water-borne diseases caused by bacteria such as V. cholerae. This might influence the taurine status of infants and young children more readily, an age group in which the prevalence of intestinal infection is high and the role of intestinal absorption is crucial for maintaining the body taurine pool. This study examined the effect of dietary lipids and guar gum on lipid metabolism in ovariectomized rats. The experimental animals received ovarietomy (OVX). Lipids sources were divided into 3 groups (soybean oil(SB), beef tallow(BT)) and fish oil(FO)) and guar gum was supplemented to each lipid diet (SBG, BTG, FOG). Experimental diets were fed to therats for 16 weeks. Serum triglyceride (TG) levels were higher in the BT group and lower in the FO group as compared to the SB group .Serum total cholesterol (Tc) and HDL-C levels were lower in the FO group as compared to the SB group. Serum LDL-C and phospholipid levels were lower in the FO group as compared to the SB group. Serum lipids levels were lowered by guar gum supplement. Serum SFA(saturated fatty acids) contents were not significantly influenced by dietary lipids and guar gum. Serum MUFA(monounsaturated fatty acids) contents were the lowest in the SB group. Fecal weight was highest in the beef tallow group and lowest in the fish oil group. Fecal weight was increased by guar gum supplement in all lipid groups. Total bile acid content in feces was increased by guar gum supplement in the soybean oil and beef tallow groups. The endothelial cells of the beef tallow group changed from a flat shape to distorted round and enlarged shapes. The subendothelial layer was the thickness the thickest in the beef tallow group ; the interspace between elastic lamina was widened and elastic lamina was straightened and partly disrupted . The fish oil group showed more porminient endothelial cells and subendothelial layer. Internal elastic membrane and elastic lamina exhibited regularly wavy shapes. Guar gum supplement showed positive effects in all lipids groups. Based on the above results , it is suggested that beef tallow increased serum TG levels and injured the wall of the aorta. On the other hand, fish oil , which decreased serum lipid levels, has a positive effect on the walls of the aorta. Guar gum protects the aorta from injury by reducing the serum lipid levels. Therefore, it is suggested that soybean oil and beef tallow consumed with guar gum is beneficial.
