The effect of bile deprivation on serum lipid and gastrin contents was investigat\ulcornered after choledocho-urinary cystostomy in Sprague-Dawley rats. Bile deprivated rats were compared with sham operated control group. Gastrin levels in serum and antral tissue were measured and serum lipid concentrations were also measured. Gastrin levels of serum and tissue after bile deprivation were increased significantly compared with those of the controL At the end of 1st and 2nd week after bile deprivation, serum cholesterol and triglyceride contents were significantly lower han those of the control. By 4th week, there was no significant difference between two groups. Increases in serum and antral gastrin levels temporarily coincided well with decreases in serum lipid contents after bile deprivation. These results suggest that there is increase in biosynthesis and release of gastrin and decrease in fat absorption at early stage of bile deprivation.
The bile secretion was accelerated generally by the administration of all derivatives tested: chemodeoxycholate, deoxycholate, cholate, dehydrocholate, 7-ketochenodeoxycholate, and 3, 7-diketochenodeoxycholate in decreasing order. Bile acids content in bile from animals administered with cholate was increased, however, other derivatives did not alter the contents of bile acids and bilirubin. In view of pharmacological point, all derivatives have hydrocholeretic action, however, only cholate exhibits typical choleretic action.

Cholestatic liver injury results from the accumulation of toxic bile salts within the liver. The aim of the present study was to understand the mechanism of bile salts-induced hepatocellular apoptosis in bile duct-ligated (BDL) rats, using Western blot and immunohistochemical analysis.(omitted)
Purpose: The aims of this study are to measure the serum levels of fat soluble vitamins (vitamin A and D) from bile duct ligated rats, and to evaluate the effect of oral bile acids administration to facilitate absorption of fat soluble vitamins. Methods: We measured serum ALT, total bilirubin, vitamin A, and vitamin D of Sprague-Dawley rats 1 week before and 4 weeks after experimental bile duct ligation. Rats were consisted with 3 groups. Group 2 had been fed bile acids and group 3 ursodeoxycholic acid after operation for 4 weeks. Multi-vitamin was given to all groups. Results: 1) Base line (mean value before duct ligation): ALT 74.2 IU, total bilirubin 0.26 mg/dL; vitamin D 13.01 ng/mL vitamin A $0.87\;{\mu}g/mL$, total bile acids $25.16\;{\mu}mol/L$. 2) Four weeks after ligation: ALT 100.7 IU, total bilirubin 2.58 mg/dL; vitamin D 7.89 ng/mL vitamin A $1.37{\mu}g/mL$, total bile acids $278.22\;{\mu}mol/L$. 3) 4 weeks after ligation, each group (group 1, group 2 and group 3) showed vitamin D (7.62, 8.10 and 7.99) ng/mL, vitamin A (1.68, 1.06 and 1.33) ${\mu}g/mL$, total bile acids (233.17, 345.80 and 268.57) ${\mu}mol/L$, which were statistically not significant. Conclusion: Serum level of vitamin A is increased after bile duct ligation although vitamin D is decreased. Oral administration of bile acids does not affect the serum levels of vitamin A and D in bile duct ligated rats.
Bacterial transformation of bile acids is possibly involved in colorectal carcinogenesis. n several epidemiological studies, the fecal bile acid dietary fiber are related to the indicence of colonic cancer. This study investigated the influence of age, dietary fiber intake on fecal bile acid profiles in healthy subject. The dietary fiber were assessed by mean of 24 hour dietary recall method, the subjects consist of 238 members aged 20 to 64 years old and feces are collected from the subjects. Fecal bile acids and neutral sterols were analyzed by gas chromatography. Mean dily crude fiber intake level was 7.7$\pm$1.4g(dietary fiber : 16.7$\pm$3.5g), dietary fiber intake range being 6.5-36.8g. The dietary fiber intake in elederly subject was significantly lower than in the other younger groups. Dietary fiber intakes was negatively correlated with the total bil acid concentation in feces. Probably, a decrease in dietary fiber intake results in higher fecal bile acid concentrations. The secondary bile acid concentration is related to the colon cancer, deoxycholic acid and lithocholic acid were significantly higher in elderly subjects. Concentration of fecal total bile acid, deoxycholic acid, coprostanol, coprostanone were higher in low dietary fiber intake group. These results suggest that the risk factor for colon cancer might be reduced, when dietary fibers are consummed more.
Inspissated bile syndrome (IBS) is a rare condition in which thick intraluminal bile, including bile plugs, sludge, or stones, blocks the extrahepatic bile ducts in an infant. A 5-week-old female infant was admitted for evaluation of jaundice and acholic stool. Diagnostic tests, including ultrasound sonography, magnetic resonance cholangiopancreatography, and a hepatobiliary scan, were not conclusive. Although the diagnosis was unclear, the clinical and laboratory findings improved gradually on administration of urodeoxycholic acid and lipid emulsion containing omega-3 polyunsaturated fatty acids (PUFAs) for 3 weeks. However, a liver biopsy was suggestive of biliary atresia. This finding forced us to perform intraoperative cholangiography, which revealed a patent common bile duct with impacted thick bile. We performed normal saline irrigation and the symptom was improved, the final diagnosis was IBS. Thus, we herein report that IBS can be treated with omega-3 PUFAs as an alternative to surgical intervention.
The present work describes the identification, purification, and characterization of bile salt hydrolase (BSH) from Bifidobacterium animalis subsp. lactis. The enzyme was purified to electrophoretic homogeneity by hydrophobic chromatography, ion-exchange chromatography and ultrafiltration. SDS-PAGE analysis of putative BSH and gel filtration revealed that the analyzed protein is presumably a tetramer composed of four monomers each of about 35 kDa. The purified enzyme was analyzed by liquid chromatography coupled to LTQ FT ICR mass spectrometry and unambiguously identified as a bile salt hydrolase from B. animalis. The isoelectric point of the studied protein was estimated to be around pH 4.9. The pH optimum of the purified BSH is between 4.7 to 6.5, and the temperature optimum is around 50oC. The BSH of B. animalis could deconjugate all tested bile salts, with clear preference for glycine-conjugated bile salts over taurine-conjugated forms. Genetic analysis of the bsh showed high similarity to the previously sequenced bsh gene from B. animalis and confirmed the usefulness of bile salt hydrolase as a genetic marker for B. animalis identification.
Preset study was carried out to evaluate characteristics of lactic acid producing bacteria(LAB) in hen's cecum as probiotics value. Distribution of LAB in intestinal tracts was investigated using 5∼25 weeks - old hens. So, 12 strains to LAB with different morphology were isolated purely. Acid tolerance of LAB tested at pH 1, 2, 3, and 4, and bile resistant also tested at 0, 0.3% and 0.5% bile salt concentration. Growth pattern of LAB observed to 60h. All strains of cecal LAB couldn't survive at pH 1, and decreased linearly survival colony after incubation at pH 2 although some strains could survive for 2h. Most of LAB maintained constant number at pH 3 and 4. The bacterial action could increase linearly at 0% bile salt concentration in all of tested strains. However, only one strain could multiply at 0.3% bile salt, others were influenced by bile salt. That tendency was similar at 0.5% bile salt. Growth was peaked at 12 to 18 h after innoculation. After peak, the decreasing pattern of colony was different to strains which some strains decreased rapidly or maintained for long time. The LAB of hen's cecum was similar to intolerance acidity, but different to resistant to bile salt and growth pattern by strain. So, we choose three strains which have probiocs value, and identified as Lactobacillus amylovorus LLA7, Lactobacillus crispatus LLA9 and Lactobacillus vaginalis LLA11.
As the laparoscopic cholecystectorny is performed more widely, complication associated with the procedure, although rare, are encountered occasionally. Injury to the bile duct occurs somewhat more frequently after the laparoscopic cholecystectorny than the open method. The bile leakage following a bile duct injury can be detected non-invasively either by ultrasonography or radionuclide hepatobiliary scan, but the former is not very specific. Hepatobiliary scan can show the bile leakge and the localization of the bile accumulation. We report two cases of the common bile duct injury following laparoscopic cholecystectorny, accurately detected by hepatobiliary scan using $^{99m}Tc$-diisoprophylimi-nodiacetic acid (DISIDA).
Objective : This study developed species-specific PCR and PCR-RFLP to detect the adulteration of Fel ursi products with cattle and pig bile juices. Methods : All the primers for PCR and PCR-RFLP in this study were designed based on nucleotide sequences of cytochrome b genes in the mitochondria. Results : The species-specific PCR amplified a DNA fragment of 214, 214, 295, and 167 bp from Fel ursi product, bear fur, cattle bile juice, and pig bile juice, respectively. The survey using the speciesspecific PCR indicated that some of commercial Fel ursi products were adulterated with cattle and pig bile juices. PCR-RFLP using the restriction endonucleases, HaeIII and HinfI enabled differentiation among Fel ursi product, cattle bile juice, and pig bile juice. Bear furs from two animals showed variations in PCR-RFLP patterns with HaeIII. Discussion : The detection methods of the species-specific PCR and PCR-RFLP could be useful in eliminating adulterated Fel ursi products from the market.
