Serum Concentrations of $\alpha$-Tocopherol, Carotenoids and Retinol of Normal Koreans

  • Five hundred and seventy-eight healthy subjects (351 men and 227 women) with a mean age of 44.8 years (45.2 for men, 44.3 for women) participated in this study. The serum concentrations of $\alpha$-tocopherol, \beta$-carotene, $\alpha$-carotene, zeaxanthin + lutein, cryptoxanthin, lycopene and retinol of normal Koreans were measured, and their relation to gender, age, BMI, alcohol consumption, cigarette smoking, and menopausal status were evaluated. The concentrations of $\alpha$-tocopherol, carotenoids and retinol in serum were measured simultaneously by reverse phase HPLC (multi-wavelength, gradient and computerized automatic system). Average serum levels of $\alpha$-tocopherol, \beta$-carotene and retinol were 10.1$\pm$0.41\mu$g/dl, 33.1$\pm$1.24\mu$g/dl, and 82.0$\pm$1.63\mu$g/dl for men and 11.1$\pm$0.74\mu$g/ml, 48.1$\pm$1.60\mu$g/dl and 64.5$\pm$1.96\mu$g/dl for women, respectively, Serum concentrations of $\alpha$-tocopheol \beta$-carotene, $\alpha$-carotene, cryptoxanthin and lycopene appeared to be higher in women than in men. The serum concentrations of zeaxanthin + lutein increased with the increase in age of men and those of $\alpha$-carotene and \beta$-carotene increased with the increase in age of women. For men, current smokers showed significantly lower serum concentrations of \beta$-carotene and $\alpha$-carotene than ex-smokers (p <0.05) . Current drinkers also showed significantly lower serum \beta$-carotene and zeaxanthin + lutein concentrations than ex-drinkers. For women, current smokers showed significantly love. serum concentrations of zeaxanthin+lutein and cryptoxanthin than ex-smokes (p<0.05). Men with BMI$\geq$24 showed significantly lower serum concentrations of \beta$-carotene, $\alpha$-carotene, lycopene and cryptoxanthin than men with BMI < 20. The mean concentrations of $\alpha$-tocopherol, \beta$-carotene, $\alpha$-carotene and lycopene far postmenopausal women were higher than those for premenopausal women (p<0.05). In conclusion, there were obvious differences in serum $\alpha$-tocopherol and carotenoids levels depending on gender, age, cigarette-smoking habits, alcohol consumption, BMI and menopausal status. further studies are required to establish the normal levels of these vitamins for children and the elderly and to elucidate their roles in different disease states.

Effects of Dietary $\beta$-Crotene Substitution for Vitamin A and Chronic Consumption of Ethanol on Folate Metabolism in Rats ($\beta$-Carotene 대체 급여 및 에탄올의 만성적 급여가 흰쥐가 엽산대사에 미치는 영향)

  • The effects of $\beta$-carotene substitutionl for vitamin A and the chronic consumption of ethanol of ethanol on hepatic folate metabolism were studied it rats. The substitution of $\beta$-carotene for vitamin A depressed hepatic 10-formyl-tetreahydrofolate dehydrogenase(10-formyl-tetrahydrofolate : NADP oxidoreductase, E.C. 1.5. 1.6)activity to 65% of controls(p<0.001) and enhanced hepatic 5, 10-methy-lenetetrahydrofolate reductase(E. C. 6.3.3.2)activity by 56% with respect to control levels(p<0.001). Hepatic activity of 10-formyltertrahydrofolate dehydrogenase was depressed to about half that of control levels by ethanol administration to rats(36% ethanol diet, p<0.001). The activity of 5, 10-methyleneterahydrofolate reductase was not changed by ethanol consumption. The increased activity of 5, 10-methyleneterahydrofolate reductase and the decreased activity of 10-formyltetrahydrofolate dehydrogenase appeared to decrease the level of nonmethyl folate conezyme and the rate of one-carbon metabolism. Plasma homocysteine concentrations were significantly higher in rats fed ethanol(p<0.01) o $\beta$-carotene(p<0.001) than in controls, which suggests that increased activity of 5, 10-methylenetetrahydrofolate reductase can depress homocysteine metabolism. We concluded that dietary substitution of $\beta$-carotene for vitamin A or chronic administration of ethanol resulted in changes in the activity of hepatic folate-dependent enzymes, which could affect the distribution of folate derivatives, plasma homocysteine levels and one-carbon metabolism.

Effect of Dietary Genetically Modified ${\beta}$-Carotene Biofortified Rice on Immune in Rats

  • This study aims to examine the effect of Genetically Modified ${\beta}$-Carotene Biofortified Rice rice developed by simultaneous expression technology in NAAS on biological immunity. Accordingly, this study added Genetically Modified ${\beta}$-Carotene Biofortified Rice 25, 50% and general rice 50% as control group into diet and provided rats with the prescribed feeds and then measured the contents of immunoglobulin and cytokine in blood. As a result, male and female IgM, IgE, male IgG1, female IgG2a and TNF-a, IL5 and IL12 showed no significant difference; male IgG2a tended to decrease dependently on the combined concentration of Genetically Modified ${\beta}$-Carotene Biofortified Rice; female IgG1 showed significance with control group, but its association with diet was not found. The higher the dietary mixing ratio, the more the male and female IFN-a and female IL-4 contents, regardless of rice variety, and it was found that female IL6 content decreased significantly, but its association with diet was not found. The risk of beta carotene-enriched rice into environment and human body has not been reported yet. The digestion of Genetically Modified ${\beta}$-Carotene Biofortified Rice can be seen as "safe" as this test result showed no big difference between general rice and Genetically Modified ${\beta}$-Carotene Biofortified Rice, and its usability is full of suggestions.

Analysis of Serum Antioxidant Materials Concentration and Their Relation with Blood Lipids and Anthropometric Indices in Middle-Aged Adults in Korea (중년기 성인의 혈중 항산화 물질 분석 및 혈중 지질과 신체계측치와의 상관성)

  • The serum levels of antioxidant materials (${\alpha}$-carotene, ${\beta}$-carotene, ${\beta}$-cryptoxanthin, lutein, lycopene, ${\alpha}$-tocopherol, ${\gamma}$-tocopherol, retinol) of the healthy Korean middle-aged adults (n = 373) were measured and their relationships with the serum lipids and anthropometric indices were analyzed. The serum levels of ${\beta}$-cryptoxanthin, ${\beta}$-carotene, lutein were higher than those of lycopene and ${\alpha}$-carotene. The levels of all measured carotenoids except lutein were significantly higher in females than in males, but retinol level was vice versa. There was a tendency of increase in serum levels of antioxidant vitamins with increasing age. The serum carotenoid levels had a positive correlation with serum cholesterol and a negative correlation with serum triglyceride. The serum levels of tocopherols or retinol showed a significantly positive relationship with blood cholesterol or triglyceride. In overall, anthropometric indices showed negative relations with serum carotenoids levels, but vice versa with serum tocopherol or retinol levels. Particularly, ${\beta}$-carotene and lutein levels showed a significantly negative relation with blood pressure in male subjects. The serum levels of ${\alpha}$-carotene, ${\beta}$-carotene and ${\beta}$-cryptoxanthin had significantly negative relations with body fat-related indices in female subjects. The results demonstrated that blood antioxidants levels differed by sex and age, and had significant relations with blood lipid levels and anthropometric indices. Therefore, the rationale and significance of the relationships need to be elucidated in the future study related to dietary intakes and life style.

  • Carotenoids are abundant int he Korean food supply. The intake of foods rich in carotenoids appears to be associated with optimal health, and a reduction in the risk of cancer, cardiovascular disease, macular degeneration and cataract formation. Specific dietary carotenoids may be responsible for these specific protective effects. Hydrocarbon carotenoids such as $\alpha$-,$\beta$-carotenes and lycopene may reduce the risk of cancer and heart disease, whereas oxygenated carotenoids, such as lutein and zeaxanthin, may be important in protection of the eye. Dietary carotenoids, such as lutein, cryptoxanthin, $\alpha$-carotene, $\beta$-carotene and lycopene can be readily obtained from the diet, Green leafy vegetables, such as spinach and broccoli, contain both oxygenated and hydrocarbon carotenoids ; yellow or orange vegetables, such as carrots, have high levels of $\alpha$-carotene and $\beta$-carotene ; and tomatoes contain high amounts of lycopene. Besides being important vitamin A sources, provitamin A carotenoids such as $\alpha$-carotene, $\beta$-carotene and cryptoxanthin, participate in the cell defense systems that are associated with radical quenching. Non-provitamin A carotenoids, such as lutein and lycopene, major carotenoids in human plasma, have also been reported to possess strong antioxidant capability. The alteration of dietary sources of carotenoids can modify their levels in the circulation and target tissues, and thus prevent or delay the onset of these chronic diseases.

Comparison of Antioxidant Level of the Elderly Living in Institutions: Vegetarian and Non-vegetarian Diet (채식·비채식 식단에 따른 시설거주노인의 항산화 상태비교)

  • Purpose: The purpose of this study was to compare the antioxidant level, serum cholesterol and skin ${\beta}$-carotene, of female elderly who had been living more then 10 months in institutions which offered either vegetarian or non vegetarian diet. Methods: Total of 110 female elderly, 56 from institution serving vegetarian diet and 54 from nonvegetarian diet, were recruited from institutions located in S and N city in Korea. ${\beta}$-carotene level was measured on their palm using Pharmanex Biophotonic Scanner and annual health evaluation data was utilized for the analysis of serum cholesterol. Results: The subjects on a vegetarian diet had a significantly higher level of phosphorus, carotene, and folic acid. The elderly who were living in an institution offering a vegetarian diet had a lower level of serum cholesterol and higher level of skin ${\beta}$-carotene, compared to those living in an institution offering a non-vegetarian diet. Conclusion: This study reveals that a vegetarian diet is beneficial in increasing skin ${\beta}$-carotene level, decreasing serum total cholesterol level. The skin ${\beta}$-carotene measurement appear valuable as a bio-marker of antioxidant intake. Further study on antioxidant food and effective serving strategy for elderly are recommended.

라드를 이용한 열처리 온도 변화에 따른 ${\beta}-carotene$의 산화 안정성에 대한 연구

  • 0%, 0.0012%, 0.004%의 ${\beta}-carotene$을 lard를 첨가한후 $80^{\circ}C$, $130^{\circ}C$, $180^{\circ}C$의 온도처리로 2분간 열처리를 하고 $50^{\circ}C$에서 보관시키면서 각 기간별로 총 균수검사, 과산화물가(Peroxide Value), TBArs를 측정하여 비교하였다. 1) 미생물 총 균수 실험에서 ${\beta}-carotene$의 첨가량이 0%, 0.0012%, 0.004%로 증가함에 따라 균의 증식이 둔화되었다. 2) Peroxide Value의 측정 실험에 ${\beta}-carotene$의 첨가량이 0.004%일 때가 0%, 0.0012%에서 보다 항산화 능력이 더 높은 것으로 나타났다. 3) TBA가 측정실험에서 control과 Sample과의 비교에서 온도가 증가함에 따라 그 증가폭 커지므로 ${\beta}-carotene$의 항산화 능력이 있다는 것을 알 수 있다.

Variation of β-carotene Concentration in Soybean Seed and Sprout

  • In this study, ${\beta}$-carotene concentrations was determined in soybean cultivar according to seed size, usage, seed coat color and cotyledon color as well as the process of seed germination. The total average concentration of ${\beta}$-carotene was $6.6{\mu}g/g$ in soybean seed, $33.3{\mu}g/g$ in soybean sprout. According to seed size, the total ${\beta}$-carotene concentration of soybean was $6.9{\mu}g/g$ in large soybean seed, $6.7{\mu}g/g$ in medium soybean seed, and $6.31{\mu}g/g$ in small soybean seed. In soybean sprout, the total ${\beta}$-carotene concentration was $21.4{\mu}g/g$ in large soybean sprout, $30.5{\mu}g/g$ in medium soybean sprout, and $43.5{\mu}g/g$ in small soybean sprout. According to the utilization of seed, the total ${\beta}$-carotene concentration of soybean seed was $7.2{\mu}g/g$ in cooked with rice soybean seed, $6.1{\mu}g/g$ in paste and curd soybean seed, and $6.3{\mu}g/g$ in sprout soybean seed. In soybean sprout, the total ${\beta}$-carotene concentration was $25.9{\mu}g/g$ in cooked with rice soybean sprout, $32.4{\mu}g/g$ in paste and curd soybean sprout, and $41.9{\mu}g/g$ in sprout soybean sprout. When comparison with seed coat color, the total ${\beta}$-carotene concentration of soybean with brown seed coat ($8.8{\mu}g/g$) was slightly higher than those of soybean with yellow ($6.1{\mu}g/g$). In soybean sprout, the total ${\beta}$-carotene concentration was $21.8{\mu}g/g$ in black seed coat sprout, $38.7{\mu}g/g$ in brown seed coat sprout, $34.1{\mu}g/g$ in green seed coat sprout, $39.5{\mu}g/g$ in yellow seed coat sprout, and $30.5{\mu}g/g$ in mottle seed coat sprout. The results of this study suggested the functional characteristics of soybean through quantitative analysis of ${\beta}$-carotene.

Identification of Carotenoid Oxidation Products in Pigment Extracts from Star Ruby Grapefruit Pulp at Different Temperatures with Exposure to Light

  • Pigment extracts obtained from Star Ruby grapefruit pulp were stored at different temperatures (4.5$^{\circ}C$, 23$^{\circ}C$) and exposed to light. many carotenoid oxidation products were formed due to light-exposure during storage periods. They were monitored by using HPLC with photodiode array detection and tentatively identified. Including (all-E)-lycopene and trans-$\beta$-carotene as predominant carotenoids in red grapefruit, 5Z-lycopene, 9Z-lycopene, 13Z-lycopene, and 15-cis-$\beta$-carotene were formed at 4.5$^{\circ}C$, 23$^{\circ}C$. Degradation of all-tarns lycopene was more susceptible to light-exposure and temperature a than that of all-trans $\beta$-carotene. The formation of lycopene cis isomers was favored under lighted condition. Respectively, (5Z)-lycopene was formed in greater amounts than other isomers at 23$^{\circ}C$ storage. The concentration of 15-cis-$\beta$-carotene was significantly increased during storage at 23$^{\circ}C$ storage. The concentration of 15-cis-$\beta$-carotene was significantly increased during storage at 23$^{\circ}C$.

Metabolism of Dietary Carotenoids and Effects to Improve the Body Color of Cultured Flounder and Red sea bream (양식 넙치, 참돔의 사료 Carotenoids 대사와 체색개선에 미치는 영향)

  • To investigate the effects on pigmentation and carotenoid metabolism of red sea breams Pagrus major and flounders Paralichithys olivaceus by the supplemented carotenoids, fishes wire fed the diet each containing ${\beta}$-carotene, lutein ester, astaxanthin, astaxanthin monoester, astaxanthin diester and ${\beta}$-apo-8'-carotenal for 8 weeks. Carotenoids in the integuments were analyzed. In cultured red sea breams with supplemented carotenoids, carotenoid deposition and pigmentation were higher in order of astaxanthin diester group, ${\beta}$-apo-8'-carotenal group and astaxanthin monoester group. The main carotenoids of red sea breams were astaxanthin diester, tunaxanthin and ${\beta}$-carotene. Difference in the content of astaxanthin diester and ${\beta}$-carotene was observed from natural and cultured red sea breams. In cultured flounders with supplemented carotenoids, carotenoid deposition and pigmentation were higher in order of ${\beta}$-carotene group and lutein ester group. The main carotenoids of flounders were zeaxanthin and lutein. Difference in lutein and ${\beta}$-carotene contents was observed from the natural and cultured flounders. Based on the contents and composition of carotenoids in each group after feeding experimental diet, carotenoid metabolism in red sea breams were presumed the reductive metabolic pathway, astaxanthin to tunaxanthin, and likewise, in flounders, lutein to tunaxanthin.
