β-Carotene, Cucumisin Content and Fruit Morphology of Melon (Cucumis melo L.) Germplasm Collections

  • Melon fruits are a year-old plant in the family Cucurbitae and are the most cultivated fruit in tropical countries. Melon flesh is an important source of anti-cancer, antioxidant effects, attracting attention as a functional food. We investigated the morphological properties, β-carotene content, and cucumisin activity of seeds of 58 melon genetic resources. Melon resources have shown various morphological properties. Melons grouped by morphological properties were the five groups. β-carotene content varied between 82.34 mg/kg, 86.75 mg/kg, 25.56 mg/kg, 86.25 mg/kg, and 54.65 mg/kg. Between β-carotene, cucumisin activity and other quantitative fruit morphological properties, the color of the pulp and the firmness of the pulp had a significant amount of correlation between the β-carotene content. However, cucumisin activity and β-carotene content had significant negative correlation, and the color of the fruit and shape of the fruit had significant negative correlation with β-carotene content and activity of cucumisin. Our study vindicated that high diversity in melon morphological characters of genetic sources that provide beneficial baseline data for the future and in the activity of β-carotene and cucumisin, and these results could predict indicators of β-carotene content by the length of leaf, the width of fruit, the length of joint, and the length of seed, and collectively the morphological properties of melons could help predict indicators of β-carotene content and help develop functional sarcoma and farmhouse cultivation.

High Serum Level of Retinol and α-Tocopherol Affords Protection Against Oral Cancer in a Multiethnic Population

  • Background: A comparative cross-sectional study involving oral cancer patients and healthy individuals was designed to investigate associations between retinol, ${\alpha}$-tocopherol and ${\beta}$-carotene with the risk of oral cancer. Materials and Methods: This study included a total of 240 matched cases and controls where subjects were selected from the Malaysian Oral Cancer Database and Tissue Bank System (MOCDTBS). Retinol, ${\alpha}$-tocopherol and ${\beta}$-carotene levels and intake were examined by high-performance liquid chromatography (HPLC) and food frequency questionnaire (FFQ) respectively. Results: It was found that results from the two methods applied did not correlate, so that further analysis was done using the HPLC method utilising blood serum. Serum levels of retinol and ${\alpha}$-tocopherol among cases ($0.177{\pm}0.081$, $1.649{\pm}1.670{\mu}g/ml$) were significantly lower than in controls ($0.264{\pm}0.137$, $3.225{\pm}2.054{\mu}g/ml$) (p<0.005). Although serum level of ${\beta}$-carotene among cases ($0.106{\pm}0.159{\mu}g/ml$) were lower compared to controls ($0.134{\pm}0.131{\mu}g/ml$), statistical significance was not observed. Logistic regression analysis showed that high serum level of retinol (OR=0.501, 95% CI=0.254-0.992, p<0.05) and ${\alpha}$-tocopherol (OR=0.184, 95% CI=0.091-0.370, p<0.05) was significantly related to lower risk of oral cancer, whereas no relationship was observed between ${\beta}$-carotene and oral cancer risk. Conclusions: High serum levels of retinol and ${\alpha}$-tocopherol confer protection against oral cancer risk.

Dietary Antioxidant Vitamins Intakes and Plasma Antioxidant Levels in Korean Elderly with Diabetes Living in Ulsan (울산 지역 당뇨 질환 노인의 항산화 영양소 섭취실태 및 혈액 항산화 영양상태)

  • Diabetic mellitus in an older population is associated with increased basal oxidative stress and free radical accentuated by hyperglycemic challenge. Enhanced free radical in diabetic elderly can cause the oxidative damage and such damage can be protected by antioxidant defense system. It is believed that vitamin C, A and E are the most abundant and effective antioxidants in human plasma. The purpose of this study was to determine the antioxidant status in Korean diabetic elderly using the case-control study. The antioxidant status was examined by determining plasma levels of antioxidant vitamins (vitamin C, A, E, ${\beta}$-carotene), total antioxidant status (TAS) and thiobarbituric acid reactive substance (TBARS) and intakes of vitamin C, A, ${\beta}$-carotene and retiol. Fasting glucose and HbA1c levels and serum lipid profiles (triglyceride (TG), total cholesterol, HDL-cholesterol and LDL-cholesterol) were also determined. Diabetic subjects were 122 elderly persons over 60 years old, visiting public health center, and control subjects were 96 healthy elderly persons living in Ulsan, Korea and they were matched by age, gender, smoking and drinking status. The diabetic and control subjects were divided into sub-groups according to the status of using diet therapy and vitamin supplement. The subjects were interviewed to collect data on their general characteristics, disease history, vitamin supplement, diet therapy and health-related habits by questionnaires. Their dietary intakes were obtained by means of semi-quantitative food frequency questionnaires (SQFFQ). Fasting plasma glucose and HbA1c levels were significantly higher in diabetes than in control subjects, and plasma total cholesterol level of diabetes was not significantly different from that of control subjects. However serum HDL cholesterol level of diabetes was significantly lower and serum TG level of diabetes was significantly higher than those of control group. The average vitamin A and ${\beta}$-carotene intakes of diabetes were significantly higher than those of control subjects. There was no significant difference in plasma vitamin C, ${\beta}$-carotene, and TBARS levels between two groups, but plasma vitamin A, E and TAS levels were significantly higher in diabetes than those in control group. Plasma vitamin A and TAS levels of diabetic subjects using diet therapy were higher than those of control using diet therapy, and plasma vitamin E, ${\beta}$-carotene and TAS levels of diabetic subjects using vitamin supplements were significantly higher than those of controls using vitamin supplements. These results suggested that diabetic mellitus could enhance antioxidant defences against reactive oxygen species and interest in healthy eating such as consumption of more antioxidant nutrients.

Breeding on High Lycopene and Beta Carotene with Multi-Disease Resistance in Tomato

  • This study was carried out to breed and develop high quality and functional nutrient tomato with multi disease resistance as well as a stable growing adaptation for fresh market usage under protected plastic houses cultivation. The materials were used 5 inbred lines and their 6 hybrids of large tomato group, which have been bred and developed from 1999 to 2007 in Division of Plant Resource Department of Chungnam National University. Fruit weight showed hybrid vigor effect that $F_1$ hybrids weighed more than their parent lines, fruit shape formed three type of oblate, deep oblate and globe shape, in firmness and pericarp thickness have got a high significant correlation, inbred DN611 line was measured the most firm fruit with 6.04 mm pericarp thickness. In fruit color at maturity, pink color crossed to red color appeared all red fruit color in the $F_1$ hybrids, it means red skin color is a dominant gene compared to pink skin color is a recessive gene in tomato, while between fruit skin color and shoulder part color showed no any co-relationship. The sugar content and titratable acid of $F_1$ hybrids inherited an intermediate data of their parent lines, the flavor of KP543 inbred line and the hybrid (JB535 x KP543) revealed the better taste with high brix and proper titratable acid content$^{*}$. In beta carotene content DN611 line showed 2~3 times higher than other materials so that its 3 hybrids contained an increased level of beta carotene, lycopene content was not so much difference among inbred lines and $F_1$ hybrids, of them MD508 contained higher of 8.72 mg and hybrid (JB535 x JA517) had 8.05 mg lycopene content per 100 g fruit, overall pink skin color and red skin color measured a higher lycopene content than yellow and orange skin color at ripe stage. In disease resistance test by PCR marker for Fusarium race2 (I2), Nematode (Mi1), ToMV ($Tm2^2$), Cladosporium (Cf9), (JB535 x JA517) hybrid have got multi-resistance with homozygote band in Nematode, ToMV, Cladosporium and heterozygote band in Fusarium race2. Through this breeding program we could select high quality and functional nutrient with multi resistant $F_1$ hybrids and inbred lines in tomato which are two best hybrids (JB535 x MD508), (JB535 x JA517), additionally developed high beta carotene inbred line DN611 and increased the level of lycopene inbred line MD508. These results will be very useful to make a high quality tomato variety continuously.

Comparison of Carotenoid Pigments on Manchurian Trout, Brachymystax lenok and Masu Salmon, Oncorhynchus macrostomus in the Family Salmonidae (연어과에 속하는 열목어와 산천어의 Carotenoid 색소성분의 비교)

  • Carotenoids in integument of wild manchurian trout, Brachymystax lenok, and wild and cultured masu salmon Oncohynchus macrostomus, which are all the Korean native cold fresh water fish, were investigated by thin layer chromatography, column chromatography and HPLC. The total carotenoid contents of the wild manchurian trout were $3.72\;mg\%$ which is relatively higher compare to other species of salmonidae. The carotenoids were composed of $36.9\%$ zeaxanthin and $14.7\%$ $\beta-carotene$ as the major compounds, $7.8\%$ triol $7.3\%$ isocryptoxanthin, $5.7\%$ 4-hydroxy echinenone, $4.7\%$ lutein, $4.5\%$ salmoxanthin and $2.2\%$ astaxanthin as minor compounds, and other carotenoids such as canthaxanthin, tunaxanthin A, tunaxanthin B, tunaxanthin C, $\beta-cryptoxanthin$ and $\alpha-cryptoxanthin$ as minute carotenoids. Wild masu salmon contained more total carotenoids than cultured one and the contents were $0.82\;mg\%$ and $0.66\;mg\%$, respectively. The composition of the carotenoids from wild masu salmon were $20.7\%$ xeaxanthin, $17.0\%$ isocryptoxanthin and $15.8\%\;\beta-carotene$ as major compounds, and $6.2\%$ triol, $6.1\%$ 4-hydroxy echinenone, $6.1\%$ salmoxanthin, $5.9\%$ canthaxanthin, $5.8\%$ lutein, $4.9\%$ $\alpha-cryptoxanthin$ and $1.0\%$ astaxanthin as minor compounds. The composition of the carotenoids from cultured masu salmon were $19.7\%$ isocryptoxanthin, $18.0\%$ $\beta-carotene$ and $10.3\%$ zeaxanthin as the major compounds, and $8.9\%\;\beta-cryptoxanthin$, $8.5\%\;\alpha-cryptoxanthin$, $8.0\%$ lutein, $7.6\%$ canthaxanthin, $5.1\%$ triol and $2.0\%$ astaxanthin as minor carotenoids. Based on these data, wild masu salmon contained more zeaxanthin, salmoxanthin and 4-hydroxy echinenone while cultured masu salmon contained more $\alpha-cryptoxanthin$, indicating that carotenoid pigment of masu salmon depends on their living conditions. Unlike wild masu salmon, 4-hydroxy echinenone and salmoxanthin which are the characteristic carotenoids of salmons, were not found in the integument of cultured masu salmon. Unlike manchurian trout, both wild and cultured masu salmon did not contain tunaxanthin A, tunaxanthin B and tunaxanthin C.

Changes of chemical components and antioxidant activities in Hizikia fusiformis(Harvey) OKAMURA with blanching times (데침시간에 따른 톳의 주요성분 및 항산화 활성의 변화)

  • This study was performed to investigate the change of chemical components and antioxidant activities in Hizikia fusiformis(Harvey) OKAMURA with blanching times. For chemical composition, the contents of minerals (K, Ca, Na, Mg, Fe, Cu, Mn and Zn), vitamins (vitamin C, ${\beta}$-carotene and ${\alpha}$-tocopherol) and total polyphenol were analyzed. The antioxidant activity was determined through free radicals (DPPH radical, superoxide anion radical, hydroxyl radical and hydrogen peroxide) scavenging activity and linoleic acid peroxidation inhibitory activity. As the blanching time increased, the contents of all minerals, vitamin C and total polyphenol decreased. However ${\beta}$-carotene and ${\beta}$-tocopherol were not affected by the blanching time. In addition antioxidant activities decreased with increasing blanching time.

Comparison of Antioxidant Activity of $\alpha$-, $\beta$-Carotene, Lutein and Lycopene by High Pressure Liquid Chromatography (고압 액체 크로마토그라피에 의한 알파, 베타, 카로텐, 루테인 및 리코펜의 항산화효과 비교 연구)

  • A new HPLC method for determining malonaldehyde content in lipid peroxidation systems was developed and antioxidant activities of $\alpha$-, $\beta$- carotene, lutein and lycopene were compared by a newly developed HPLC analysis and by TBA value. In addition, malonaldehyde forming ability of rat liver microsome was determined depending on thawing numbers. As results, malonaldehyde was eluted at a retention o f5.60 min and showed a linear relationship between peak area and concentration in standard curve. The MA content of microsome decreased with thawing numbers possible by destruction of cellular membranes. Lycopene, lutein and $\alpha$-carotene showed stronger antioxidant activities than $\beta$-carotene of DL-$\alpha$-tocopherol both in Fe+3-ADP/NADPH and in paraquat/NADPH system. The inhibitory effects of carotenoids and DL-$\alpha$-tocopherol on Fe+3-ADP/NADPH lipid peroxidation system was similar by TBA value and by the HPLC analysis for malonaldehyde.

  • This was aim to study the changes of components by different parts and maturity of tomato. Tomato include the green pigment chlorophyll, the yellow pigment-carotene. the red pigment lycopene and the other component. These colors of tomatoes are reported to have important functional roles to promote health in human. The main objective of this study was to define changes in the content of the four tomato maturation that is ascorbic acid, chlorophyll, carotene, lycopene. A ascorbic acid contents of tomato fruits was the highest at 50th day. Chlorophyll contents of tomatoes fruits was the highest at 10th days after flowering. ${\alpha}$-carotene contained low-quantity while all growth period. ${\beta}$-carotene contained the highest at 50th days after flowering, $1036.97{\mu}g$. Lycopene contained the highest at 50th days after flowering, $5800{\mu}g$.

Influence of Processing on Quality of Carrot Juice (가공공정에 의한 당근쥬스의 품질변화에 관한 연구)

  • This study was conducted to determine the organoleptic and physical properties and carotenoid of commercial canned, frozen and freshly-made carrot juice. Samples were evaluated by sensory panel and measured for viscosity and acidity. For carotene analysis, HPLC of alpha- and beta-carotene, and spectrophotometry of total carotenoid content were used. Sensory evaluation indicated that the canned sample was less acceptable, especially for flavor and texture, than other juices, while forzen juice was considered as acceptable as freshly-made carrot juice. The canned product showed about 10 times higher viscosity and lower acidity than others. Between two kinds of frozen samples, one sample was the same as freshly-made sample for all parameters while the other showed less alpha-carotene content which was 2 times higher than that of canned one. Canned sample contained 70-77% of freshly-made or frozen samples in total carotenoid and beta-carotene content and 24% of freshly-made one in alpha-carotene. These results suggest that freezing process is a good preservation method for carrot juice with respect to sensory evaluation, physical property and carotenoid content.

Effects of Paprika Extract and Its Components on Cell Death and Expression of p53 and GADD45 Genes in Ultraviolet B- Exposed HaCaT Cells (UVB를 조사한 HaCaT 세포의 세포사멸과 p53 및 GADD45 유전자 발현에 대한 파프리카 추출물 및 성분들의 효과)

  • In the present study, the effects of paprika extract and its components including vitamin C, lycopene and beta-carotene on cell death in ultraviolet B (UVB)-exposed HaCaT cells were investigated. The cell viability upon treatment for 24 hr with either paprika extract or vitamin C alone was similar to or greater than that of the untreated control. However, the viability of the cells treated with lycopene or beta-carotene decreased to about 20% of that in the untreated control. When UVB-exposed cells were post-incubated for 24 hr in medium containing paprika extract or vitamin C, cell viability increased in a concentration dependent manner as compared to those post-incubated in a normal growth medium. In contrast, post-incubation of UVB-exposed cells with lycopene or beta-carotene decreased cell viability in a concentration dependent manner as compared to those post-incubated in a normal growth medium. The nuclear fragmentation analysis showed that paprika extract or vitamin C decreases UVB-induced apoptosis. The apoptotic nuclear fragmentation resulting from UVB exposure was also protected by the paprika extract or vitamin C post-incubation. However, the UVB-induced apoptotic nuclear fragmentation of the cells treated with lycopene or beta-carotene increased in a concentration dependent manner. Western blot analysis showed that either paprika extract or vitamin C treatment alone did not significantly change the level of p53 and GADD45 protein. Interestingly, post-incubation of UVB-exposed cells with paprika extract or vitamin C decreased the p53 and GADD45 protein level as compared to those post-incubated in a normal growth medium. In contrast, incubation of UVB-exposed or non-irradiated cells with lycopene or beta-carotene increased the p53 and GADD45 protein levels in a concentration dependent manner as compared to those incubated in a normal growth medium. All these results suggest that paprika extract and vitamin C help the survival of the UVB-exposed cells, while lycopene and beta-carotene potentiate the apoptotic death of UVB-exposed cells, in accordance with the respective changes in p53 and GADD45 protein levels.
