Comparison of antioxidative activities from different organs of Rosa rugosa Thunb (해당화의 채취 부위별 항산화 활성 비교)

  • DPPH(1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity and Vitamin contents in different plant parts and harvesting time of Rosa rugosa Thunb were determined. Among the plants parts analysed, the highest DPPH radical scavenging activity in part was root $(below\;0.5\;{\mu}g/ml)$, leaf contained the highest ascorbic acid content of 2999.6 mg/100g. Fruit contained ascorbic acid (713.6 mg/ 100g) 4.2 times less than leaf. Vitamin contents in leaf parts also differed depanding on harvesting time. Vitamin content in leaf harvested in November was much higher than that in September. Spectrophotometric analysis of total ${\beta}-carotene$ in leaf showed lower contents than that in fruit.

  • Citrus phytoene synthase (CitPsy) and ${\beta}$-carotene hydroxylase (CitChx), which are involved in caroteinoid biosynthesis, are distantly related to the corresponding bacterial enzymes from the point of view of amino acid sequence similarity. We investigated these enzyme activities using Pantoea ananatis carotenoid biosynthetic genes and Escherichia coli as a host cell. The genes were cloned into two vector systems controlled by the T7 promoter. SDS-polyacrylamide gel electrophoresis showed that CitPsy and CitChx proteins are normally expressed in E. coli in both soluble and insoluble forms. In vivo complementation using the Pantoea ananatis enzymes and HPLC analysis showed that ${\beta}$-carotene and zeaxanthin were produced in recombinant E. coli, which indicated that the citrus enzymes were functionally expressed in E. coli and assembled into a functional multi-enzyme complex with Pantoea ananatis enzymes. These observed activities well matched the results of other researchers on tomato phytoene synthase and Arabidopsis and pepper ${\beta}$-carotene hydroxylases. Thus, our results suggest that plant carotenoid biosynthetic enzymes can generally complement the bacterial enzymes and could be a means of carotenoid production by molecular breeding and fermentation in bacterial and plant systems.

Effect of Lipoxygenase on Oxidative Stability of ${\beta}-Carotene$ and ${\alpha}-Tocopherol$ (베타-카로틴과 알파-토코페롤의 산화안정성에 대한 리폭시게나아제의 영향)

  • Starch solid system was used to investigate the effects of lipoxygenase, linoleic acid and water activity on the oxidation of ${\beta}-carotene$ or ${\alpha}-tocopherol$. ${\beta}-carotene$ or ${\alpha}-tocopherol$ was co-oxidized severely with linoleic acid by lipoxygenase, and these were reduced to 19% and 5% of initial concentration, respectively, after 2 days storage at $a_w$ 0.72 in the system. The concentration of ${\beta}-carotene$ and the destruction rates were linearly correlated. However, the ${\beta}-carotene$ was very stable in the system without linoleic acid and lipoxygenase. The oxidation products of ${\alpha}-tocopherol$ were considered as ${\alpha}-tocopheryl$ quinone and ${\alpha}-tocopheryl$ dimer, and the level of ${\alpha}-tocopherol$ quinone increased as the reaction time increased.

Anti-wrinkle Activity of $\beta$-carotene Extracted & Purified from Recombinant Escherichia coli (재조합 대장균으로부터 추출.정제된 베타-카로틴의 주름개선 활성)

  • This paper described the extraction/purification of $\beta$-carotene from recombinant E.coli and evaluation of anti-wrinkle activity of purified $\beta$-carotene. No significant differences in extraction yields were observed when hexane or isobutyl acetate was used. However, extraction from wet-cell cake resulted in 2-fold higher amount of $\beta$-carotene than that from dry cells. Disruption of 5 g-wet cells by ultrasonic homogenizer, acetone dehydration, extraction with isobutyl acetate resulted in 36 mg of $\beta$-carotene corresponding to 61.2% of recovery. The formation and separation of $\beta$-carotene crystal improved the purity. 633 mg of $\beta$-carotene crystal with 93% purity was obtained from 223 g/L of wet-cell cake harvested from 2.5-L fed-batch culture broth. The cultures of normal human primary fibroblast were performed to investigate the effect of $\beta$-carotene on cytotoxicity as MTT assay and anti-wrinkle activity as collagen synthesis assays. $1.7{\mu}M$ of $\beta$-carotene was found to be optimal concentration at which 1.4-fold higher amount of collagen was synthesized than that in absence of $\beta$-carotene. This indicates that highly purified $\beta$-carotene can be obtained from recombinant E.coli by applying simple method with less toxic solvent and can be used in functional cosmetics as anti-wrinkle agent.

Overexpression and Characterization of Lycopene Cyclase (CrtY) from Marine Bacterium Paracoccus haeundaensis

  • Lycopene cyclase converts lycopene to ${\beta}$-carotene by catalyzing the formation of two beta-rings at each end of the linear carotene structure. This reaction takes place as a two-step reaction in which both sides of of the lycopene molecule are cyclized into ${\beta}$-carotene rings via the monocyclic ${\gamma}$-carotene as an intermediate. The crtY gene coding for lycopene cyclase from Paracoccus haeundaensis consists of 1,158 base pairs encoding 386 amino acids residues. An expression plasmid containing the crtY gene (pET44a-CrtY) was constructed and expressed in Escherichia coli, and produced a recombinant protein of approximately 43 kDa, corresponding to the molecular mass of lycopene cyclase. The expressed protein was purified to homogeneity by His-tag affinity chromatography and showed enzymatic activity corresponding to lycopene cyclase. We also determined the lycopene substrate specificity and NADPH cofactor requirements of the purified protein. The $K_m$ values for lycopene and NADPH were 3.5 ${\mu}M$ and 2 mM, respectively. The results obtained from this study will provide a wider base of knowledge on the enzyme characterization of lycopene cyclase at the molecular level.

Characterization of Human ${\beta}-Carotene$ 15,15-dioxygenase Isolated from Recombinant Escherichia coli (유전자 재조합 기술에 의하여 제조된 인간 ${\beta}-carotene$ 15,15'-dioxygenase의 반응특성)

  • Characteristics of human ${\beta}-carotene$ 15,15'-dioxygenase isolated by recombinant DNA technology was elucidated. Optimal pH and temperature were 9.0 and $40^{\circ}C$, respectively. Enzyme activity was temperature-sensitive. Enzyme was stable at pH 6.0-9.0 for 24 hr and under $5^{\circ}C$. Half-life of enzyme at $35^{\circ}C$ was 40 min. Crude preparations of enzyme were inhibited by ferrous ion-chelating agent and sulfhydryl-binding agent. GSH offsets inhibitory effect of PCMB. With increasing substrate concentrations, enzyme activity gave typical Michaelis-Menten curve, Based on Hanes-Woolf plot of data, $K_{m}\;and\;V_{max$ were $3.39{\times}10^{6}\;M\;and\;1.2\;pmol/mg$ protein/min, respectively.

Antioxidant and Antiaging Assays of Hibiscus sabdariffa Extract and Its Compounds

  • Skin aging is a complex biological process due to intrinsic and extrinsic factors. Free radical oxidative is one of extrinsic factors that induce activation of collagenase, elastase and hyaluronidase. Natural product from plants has been used as antioxidant and antiaging. This study aimed to evaluate antioxidant and antiaging properties of Hibiscus sabdariffa extract (HSE) and its compounds including myricetin, ascorbic acid, and ${\beta}$ carotene. The phytochemical of H. sabdariffa was determined using modified Farnsworth method and presence of phenols, flavonoids and tannins were in moderate content, whereas triterpenoids and alkaloids were in low content. Total phenolic content performed using Folin-Ciocalteu method, was $23.85{\mu}gGAE/mg$. Quantitative analysis of myricetin, ${\beta}-carotene$, and ascorbic acid of HSE was performed with Ultra-High Performance Liquid Chromatography (UHPLC) that shows $78.23{\mu}g/mg$ myricetin, $0.034{\mu}g/mg$ ${\beta}-carotene$, whilst ascorbic acid was not detected. HSE has lower activity on DPPH ($IC_{50}=195.73{\mu}g/mL$) compared to ${\beta}-carotene$, the lowest in ABTS assay ($IC_{50}=74.58{\mu}g/mL$) and low activity in FRAP assay ($46.24{\mu}MFe(II)/{\mu}g\;$) compared to myricetin, ${\beta}-carotene$. Antiaging was measured through inhibitory activity of collagenase, elastase, and hyaluronidase. HSE had weakest collagenase inhibitory activity ($IC_{50}=750.33{\mu}g/mL$), elastase inhibitory activity ($103.83{\mu}g/mL$), hyaluronidase inhibitory activity ($IC_{50}=619.43{\mu}g/mL$) compared to myricetin, ${\beta}-carotene$, and ascorbic acid. HSE contain higher myricetin compared to ${\beta}-carotene$. HSE has moderate antioxidants and lowest antiaging activities. Myricetin is the most active both antioxidant and antiaging activities.

$\beta-Carotene,\;\alpha$-Tocopherol Concentrations in Maternal-Umbilical Cord Plasma and Their Relationships to Pregnancy Outcome (임신 말 모체와 제대혈의 Retinol, $\beta-Carotene,\;\alpha$-Tocopherol의 농도 및 임신결과와의 상관성)

  • The purpose of this study was to evaluate the intake of antioxidant vitamins and plasma concentrations of those in 60 maternal-infant pairs (30 in normal term delivery group, NT; 30 in preform delivery group, PT). We also investigated the relationship between vitamin levels of maternal-umbilical cord plasma and pregnancy outcome. Mean energy intakes of NT and PT pregnant women were 93.2% and 85.4%, and their protein intakes were 113.3% and 110.9% of the recommended dietary allowance (RDA), respectively. While vitamin A intakes were only 51.2% and 39.6% of the RDA in NT and PT pregnant women. The vitamin E intake was about 50% of the RDA (NT 6.27 mg, PT 7.78 mg). The levels of retinol in maternal plasma of NT and PT were $1.51\mumol/\ell\;and\;1.43\mumol/\ell$, respectively. The retinol levels in umbilical cord plasma in NT and PT were $0.72\mumol/\ell\;and\;0.61\mumol/\ell$, respectively. The level of $\beta-carotene$in maternal plasma of NT was 0.49 $\mu$mol/$\ell$, significantly (p < 0.01) higher than that of PT ($0.31\mumol/\ell$).The $\beta-carotene$ of umbilical cord plasma of NT and PT were $0.702\mumol/\ell\;and\;0.01\mumol/\ell$, respectively. The plasma $\alpha$-tocopherol of maternal of NT and PT were $0.72\mumol/\ell\;and\;0.01\mumol/\ell\;29.51 /mumol/\ell\; and 27.17\mumol/\ell,\;respectively.\; The $\alpha$-tocopherol of umbilical cord plasma of NT and PT were $4.16\mumol/\ell\;and\;3.80\mumol/\ell$, respectively. The antioxidant vitamin levels (retinol, $\beta-carotene,\;and\;\alpha$-tocopherol) in maternal plasma were significantly higher (p<0.0001) than those in umbilical cord plasma. However, there was no correlation between the vitamin levels in maternal plasma and those in umbilical cord plasma. The maternal plasma $\beta$-carotene level showed a positive correlation to gestational age. Also Apgar score at 1 min produced a positive correlation to maternal plasma $\beta$-carotene level.

Formation of Volatile Compounds by the Thermal Degradation of ${\beta}-Carotene$ (${\beta}-Carotene$의 열분해(熱分解)에 의한 휘발성(揮發性) 화합물(化合物)의 생성(生成))

  • Thermal degradation of ${\beta}-carotene$, major carotenoid present in cured tobacco leaves, were carried out at $400,\;600,\;and\;800^{\circ}C$ which are similar to temperatures existing in the combustion zones of cigarettes, and subsequent volatile degradation products were analyzed by combined gas chromatography-mass spectrometry. The volatile compounds identified from degradation products included 36 aromatic hydrocarbons, 10 ${\beta}-ionone-related$ compounds which have trimethylcyclohexane ring, and 7 others. Of these, 37 compounds including ${\beta}-cyclogeraniol$ had not been previously reported in the literature as thermal degradation products of ${\beta}-carotene$. The major compounds of degradation products at $400\;and\;600^{\circ}C$ were ${\beta}-xylene,\;{\alpha}-terpinene,\;{beta}-cyclocitral,\;ionene\;(1,2,3,4-tetrahydro-1,1,6-trimethyl\;naphthalene),\;{\beta}-ionone$, and dihydroactinidiolide. The major compounds at $800^{\circ}C$ were the above six compounds plus toluene.

Determination of β-Carotene and Retinol in Korean Noodles and Bread Products (가공 및 외식식품 중 면류 및 제빵류의 레티놀 및 베타카로틴 함량 조사)

  • The contents of ${\beta}$-carotene and retinol in processed and restaurant foods, such as Korean noodles, mandus, rice cakes and bread products, were quantified by high-performance liquid chromatography (HPLC) with UV/visible and fluorescence detector, respectively. Samples were collected from different local areas (i.e. Gangwon-do, Gyeonggi-do, Gyeongsang-do, Seoul, Jeolla-do, and Chungcheong-do). After homogenization, samples were hydrolyzed by direct alkali saponification; thereafter, fat-soluble components were extracted by a mixture of n-hexane/ethylacetate (85:15, v/v), containing 0.01% butylated hydroxytoluene (BHT). ${\beta}$-carotene and retinol contents in infant formula used as an in-house material for the analytical quality control. Among 14 Korean noodles, high contents of ${\beta}$-carotene were found in Bibim-Guksu (average 442.43 ${\mu}g/100g$) and Jjolmyeon (average 301.39 ${\mu}g/100g$). In 4 Korean mandus, the highest contents of ${\beta}$-carotene was determined in Kimchi-mandu (average 197.64 ${\mu}g/100g$), resulting in 33.3 RE of the converted vitamin A. Among 12 Korean rice cakes, Maeun-Tteokbokki and Modm-Chaltteok contained relatively high content of ${\beta}$-carotene with 205.11 and 41.33 ${\mu}g/100g$, respectively, while retinol was detected only in Maeun- Tteokbokki (1.65~10.45 ${\mu}g/100g$). In addition, among 8 bread products, 77.3 RE of pastry, 51.2 RE of buttercream- bread, and 41.4 RE of morning roll were found as the contents of the converted vitamin A.
