Complement component 9 (C9) 유전자의 단일염기다형성과 버크셔 돼지 육질 형질과의 연관성 분석 (Association between a non-synonymous single nucleotide polymorphism in the Complement component 9 (C9) gene and meat-quality traits in Berkshire pigs)

  • 본 연구는 Berkshire 간 조직을 이용하여 RNA-sequencing 분석을 통해 돼지 육질 연관 단일염기다형성을 발굴하기 위해 수행되었다. 그 결과, C9 유전자의 cDNA 942번 G 서열이 T 서열로 변환되어 라이신(lysin)이 아스파라진(asparagin)으로 변하는 non-synonymous SNP를 확인하였다. Berkshire 돼지 405두에서 C9 단일염기다형성의 유전자형을 분석한 결과 major allele는 G, minor allele은 T였다. Berkshire 돼지 405두의 육질 형질을 분석하여 C9 단일염기다형성의 유전자형과 육질 형질과의 연관성 분석한 결과 우성 모델의 경우 육색의 명도, 콜라겐, 수분, 도축 후 24시간 뒤 pH ($pH_{24h}$) 육질 형질에서 유의성이 확인되었고, 열성 모델의 콜라겐 함량, 공우성 모델의 육색의 명도(CIE L), 단백질, 콜라겐 함량에서 유의성을 가졌다. 성별에 따른 C9 유전자형과 육질 형질 간의 연관성을 분석한 결과 거세돈에서 도체중, 콜라겐에서 유의성이 있었으며, 암퇘지의 경우 육색의 명도, 단백질, $pH_{24h}$ 육질 형질에서 유의성이 있었다. 육질 형질 중 $pH_{24h}$ 형질은 육질을 결정하는 중요한 형질로 C9 유전자의 유전자형이 다른 유전자형들에 비해 $pH_{24h}$가 증가되고 육즙 손실이 감소되는 것으로 확인되어 C9 유전자의 TG 유전자를 가진 돼지가 더 좋은 육질을 가지는 것으로 판단된다. 본 결과를 바탕으로 C9 유전자의 단일염기다형성을 육질을 판단하는 생물마커(biomarker)로의 활용이 기대된다.

Genetic Parameters of Reproductive and Meat Quality Traits in Korean Berkshire Pigs

  • Genetic parameters of Berkshire pigs for reproduction, carcass and meat quality traits were estimated using the records from a breeding farm in Korea. For reproduction traits, 2,457 records of the total number of piglets born (TNB) and the number of piglets born alive (NBA) from 781 sows and 53 sires were used. For two carcass traits which are carcass weight (CW) and backfat thickness (BF) and for 10 meat quality traits which are pH value after 45 minutes (pH45m), pH value after 24 hours (pH24h), lightness in meat color (LMC), redness in meat color (RMC), yellowness in meat color (YMC), moisture holding capacity (MHC), drip loss (DL), cooking loss (CL), fat content (FC), and shear force value (SH), 1,942 pig records were used to estimate genetic parameters. The genetic parameters for each trait were estimated using VCE program with animal model. Heritability estimates for reproduction traits TNB and NBA were 0.07 and 0.06, respectively, for carcass traits CW and BF were 0.37 and 0.57, respectively and for meat traits pH45m, pH24h, LMC, RMC, YMC, MHC, DL, CL, FC, and SH were 0.48, 0.15, 0.19, 0.36, 0.28, 0.21, 0.33, 0.45, 0.43, and 0.39, respectively. The estimate for genetic correlation coefficient between CW and BF was 0.27. The Genetic correlation between pH24h and meat color traits were in the range of -0.51 to -0.33 and between pH24h and DL and SH were -0.41 and -0.32, respectively. The estimates for genetic correlation coefficients between reproductive and meat quality traits were very low or zero. However, the estimates for genetic correlation coefficients between reproductive traits and drip and cooking loss were in the range of 0.12 to 0.17 and -0.14 to -0.12, respectively. As the estimated heritability of meat quality traits showed medium to high heritability, these traits may be applicable for the genetic improvement by continuous measurement. However, since some of the meat quality traits showed negative genetic correlations with carcass traits, an appropriate breeding scheme is required that carefully considers the complexity of genetic parameters and applicability of data.

Meat quality of pork loins from Hereford×Berkshire female and intact male pigs reared in an alternative production system

  • Objective: The objective of the present study was to investigate pork quality from Hereford${\times}$Berkshire female and intact male pigs reared outdoors in an alternative production system. Methods: Berkshire purebred sows were artificially inseminated, once in the fall and again in the spring of the following year, with semen from Hereford boars and managed free of antibiotics in an outdoor hoop structure until the last month of pregnancy, after which they were moved to a pasture-based unit of 0.8 hectares with individual lots with a farrowing hut, shade, and water ad libitum. Piglets were weaned at 4 weeks of age and housed in a deep-bedded hoop structure, grouped by sex. Animals were harvested at market weight of 125 kg, approximately 200 days of age. Hot carcass weight was collected at the time of the harvest. After 24 hours of refrigeration, carcass characteristics were measured. Longissimus dorsi samples collected from the right side loin. Loins were cut into 2.54-cm thick chops and were used to measure marbling score, color score, drip loss, and ultimate pH. Sensory panel tests were conducted as well at North Carolina State University. For pork characteristics and sensory panel data, trial and sex were included in the statistical model as fixed effects. Hot carcass weight was included in the model as a covariate for backfat thickness. Results: Neither the subjective nor the objective color scores displayed any differences between the boars and the gilts. No difference was found for pH and marbling score between trials or sexes. Gilts had a thicker backfat measurement at the last lumbar and a narrower longissimus muscle area measurement when compared to the boars. The only difference in the sensory characteristics was found between the trials for texture and moisture scores. Conclusion: Consumers were not able to detect boar taint under the condition of this study, which is that the intact males were reared outdoors. Additional trials would be necessary; however, based on the results of the present study, outdoor rearing can be suggested as a solution to the issue of boar taint.

버크셔 돼지 육질 형질과 Enoyl-CoA delta isomerase 2 (ECI2) 유전자 nsSNP의 연관성 분석 (The identification of non-synonymous SNP in the Enoyl-CoA delta isomerase 2 (ECI2) gene and its Association with Meat Quality Traits in Berkshire pigs)

  • 본 연구는 돼지 농가의 생산성 향상 및 수익성 증대를 위한 분자 육종 기술에 적용할 유전자 마커를 개발하기 위해 수행되었다. 분석 결과에 따르면, 돼지의 간 조직을 이용하여 RNA-Sequencing을 수행한 결과 ECI2 유전자의 SNP를 발견하였고 발견된 SNP chr 7:g.2302809는 c.608로 608번째 C가 G로 변환되어 Threonine에서 Serine으로 아미노산이 치환되는 단일염기다형성임을 확인하였다. Berkshire 돼지 430으로 ECI2 유전자형과 육질 형질과의 연관성 분석 결과 도 체중, 적색도, 육즙 손실, 수분 함량 및 $pH_{24hr}$에서 유의성을 확인할 수 있었다. 그중 GG 유전자형은 $pH_{24hr}$에서 다른 유전자형에 비해 수치가 증가시키는 경향을 확인하였다. 성별에 따른 유전자형과 육질 형질과의 연관성은 거세돈에서 GG 유전자형이 육즙 손실의 감소와 $pH_{24hr}$에서 유의성이 확인되었고, 암퇘지의 GG 유전자형도 수분함량이 증가되었다. 따라서 ECI2 유전자의 GG 유전자형을 가진 돼지가 육질이 더 좋은 것으로 판단된다. 이러한 결과를 바탕으로 ECI2의 GG 유전자형을 고정시킨다면 육질이 우수한 돼지고기 생산이 가능할 것이다. 또한 ECI2 유전자를 이용하여 품종개량 및 조기 선발 기술에 바이오마커로 활용한다면 농가의 경쟁력 강화 효과에 도움이 될 것으로 사료된다.

일본버크셔의 도살체중이 혈액성상과 돈육품질에 미치는 영향 (Effect of Slaughter Weight on the Blood Profile and Pork Qualities of Japan Berkshire)

  • The effects of slaughter weight on blood profile and pork qualities of japan berkshires were investigated A total 72 pigs were divided into 3 groups$(125\~130,\;105\~110\;or\;95\~104\;kg)$. At each slaughter weight pigs were conventionally slaughtered and then chilled overnight The carcass characteristics (carcass weight backfat thickness and grades) were determined on those carcass, the muscle longissimus dorsi was removed from each left side at 5th to 13th rib and meat qualities were evaluated. Blood profile including cortisol, creatine phos-phokinase (CPK), lactate dehydrogenase (LDH), glucose and phosphorus was not significantly (p>0.05) different among all slaughter weight, However, the calcium contents of pigs at $95\~104\;kg$ were significantly (p<0.05) higher than the other weights. The carcass weight and backfat thickness of pigs slaughtered at $125\~130\;kg$ were higher than those of $105\~110\;or\;95\~104\;kg$. The carcass grade of pigs slaughtered at $125\~130\;kg$ were significantly (p<0.05) lower than the other weight. The moisture contents of pigs slaughtered at $125\~130\;kg$ were significantly lower than the other weights, but crude protein contents were significantly (p<0.05) higher, Cooking loss and shear lone values of pigs slaughtered at $95\~104\;kg$ were significantly (p<0.05) lower than the other weight. CIE $a^*\;and\;b^*$ values of pigs slaughtered at $105\~110\;kg$ were significantly higher than the other weights. These results imply that the carcass characteristics (carcass weight and backfat thickness) could be affected by slaughter weight the cooking loss and shear force values of pigs slaughtered at $125\~130\;kg$ resulted in higher than those of $105\~110kg\;or\;95\~104\;kg$.

돈육의 이화학적 품질 특성에 미치는 산삼 배양액 급여 영향 (Effects of Artificial Culture Medium of Wild Ginseng on the Physico-chemical Characteristics of Pork)

  • 본시험은 돼지에 대한 산삼 배양액 급여 효과를 구명하기 위하여 $60{\pm}3kg(126{\pm}3$일령)의 Berkshire 종 암퇘지와 거세돼지 각각 60두씩 총 120두를 선발하여 2(성; 암 vs. 거세)${\times}2$(대조구 vs. 산삼 배양액 첨가구) 'factorial' 실험설계 하에 돈방당 30두씩 4 돈방에 임의로 배치한 다음 약 110 kg($193{\pm}4$일령)까지 70일간 비육 시험을 수행하였다. 이때 대조구는 산삼 배양액을 첨가하지 않은 구로 하였고, 시험구는 산삼 배양액을 1일 1L씩 첨가 음수시켰고, 그 결과는 다음과 같다. 처리 간에 암퇘지의 경우 수분 함량, 경도 및 씹힘성은 대조구보다 처리구가 높게 나타났으나, 보수력과 지방의 $L^*$값은 반대로 낮게 나타났다(p<0.05). 처리 간에 거세돼지의 경우 전단가와 육색 $a^*$값이 대조구보다 처리구가 높게 나타나(p<0.05) 산삼 배양액 급여에 따라 조직이 단단해지고 육의 $a^*$값은 향상되나 보수력과 지방의 명도가 낮아지는 단점을 나타내었다. 성별 간에 처리구의 경우 조지방 함량, 지방의 $a^*$$b^*$값, 경도, 응집성 및 씹힘성은 암퇘지보다 거세돼지가 낮게 나타났으나, pH는 반대로 높게 나타나(p<0.05) 암퇘지에 비하여 거세돼지가 조지방 함량은 적고, 조직이 무르나 지방의 $b^*$값은 낮고 pH가 높은 장점을 나타내었다.

Comparison of prediction accuracy for genomic estimated breeding value using the reference pig population of single-breed and admixed-breed

  • This study was performed to increase the accuracy of genomic estimated breeding value (GEBV) predictions for domestic pigs using single-breed and admixed reference populations (single-breed of Berkshire pigs [BS] with cross breed of Korean native pigs and Landrace pigs [CB]). The principal component analysis (PCA), linkage disequilibrium (LD), and genome-wide association study (GWAS) were performed to analyze the population structure prior to genomic prediction. Reference and test population data sets were randomly sampled 10 times each and precision accuracy was analyzed according to the size of the reference population (100, 200, 300, or 400 animals). For the BS population, prediction accuracy was higher for all economically important traits with larger reference population size. Prediction accuracy was ranged from -0.05 to 0.003, for all traits except carcass weight (CWT), when CB was used as the reference population and BS as the test. The accuracy of CB for backfat thickness (BF) and shear force (SF) using admixed population as reference increased with reference population size, while the results for CWT and muscle pH at 24 hours after slaughter (pH) were equivocal with respect to the relationship between accuracy and reference population size, although overall accuracy was similar to that using the BS as the reference.

The Outcomes of Selection in a Closed Herd on a Farm in Operation

  • A herd of Berkshire pigs was established in 2003 and subjected to selection without introduction of any genetic resources until 2007. The complete pedigree, including 410 boars and 916 sows, as well as the records from 5,845 pigs and 822 litters were used to investigate the results obtained from the selections. The index of selection for breeding values included days to 90 kg (D90kg), backfat thickness (BF) and number of piglets born alive (NBA). The average inbreeding coefficients of pigs were found to be 0.023, 0.008, 0.013, 0.025, 0.026, and 0.005 from 2003 to 2007, respectively. The genetic gains per year were 12.1 g, -0.04 mm, -3.13 days, and 0.181 head for average daily gain (ADG), BF, D90kg, and NBA, respectively. Breeding values of ADG, BF and D90kg were not significantly correlated with inbreeding coefficients of individuals, except for NBA (-0.21). The response per additional 1% of inbreeding was 0.0278 head reduction in NBA. The annual increase of inbreeding was 0.23% and the annual decrease in NBA due to inbreeding was 0.0064 head. This magnitude could be disregarded when compared with the annual gain in NBA (0.181 head). These results suggest that inbreeding and inbreeding depression on ordinary farms can be controlled with a proper breeding scheme and that breeding programs are economical and safe relative to the risks associated with importation of pigs.

Intestinal microbial composition changes induced by Lactobacillus plantarum GBL 16, 17 fermented feed and intestinal immune homeostasis regulation in pigs

  • In this study, Rubus coreanus (R. coreanus) byproducts with high polyphenol content were fermented with R. coreanus-derived lactic acid bacteria (Lactobacillus plantarum GBL 16 and 17). Then the effect of R. coreanus-derived lactic acid bacteria fermented feed (RC-LAB fermented feed) with probiotics (Bacillus subtills, Aspergillus oryzae, Yeast) as a feed additive for pigs on the composition of intestinal microbes and the regulation of intestinal immune homeostasis was investigated. Seventy-two finishing Berkshire pigs were randomly allotted to four different treatment groups and 18 replicates. RC-LAB fermented feed with probiotics increased the genera Lactobacillus, Streptococcus, Mitsuokella, Prevotella, Bacteroides spp., Roseburia spp., and Faecalibacterium prausnitzii, which are beneficial bacteria of the digestive tract of pigs. Also, RC-LAB fermented feed with probiotics decreased the genera Clostridium, Terrisporobacter, Romboutsia, Kandleria, Megasphaera and Escherichia, which are harmful bacteria. In particular, the relative abundance of the genera Lactobacillus and Streptococcus increased by an average of 8.51% and 4.68% in the treatment groups and the classes Clostridia and genera Escherichia decreased by an average of 27.05% and 2.85% in the treatment groups. In mesenteric lymph nodes (MLN) and spleens, the mRNA expression of transcription factors and cytokines in Th1 and Treg cells increased and the mRNA expression of Th2 and Th17 transcription factors and cytokines decreased, indicating a regulatory effect on intestinal immune homeostasis. RC-LAB fermented feed regulates gut immune homeostasis by influencing the composition of beneficial and detrimental microorganisms in the gut and regulating the balance of Th1/Th2 and Th17/Treg cells.

Quality Comparison of Pork Loin and Belly from Three-way Crossbred Pigs during Postmortem Storage

  • This study was performed to determine the meat quality characteristics of pork loin and belly from 3 different three-way crossbred pigs: Yorkshire ${\times}$ Landrace ${\times}$ Duroc (YLD), Yorkshire ${\times}$ Chester White ${\times}$ Yorkshire (YCY), and Yorkshire ${\times}$ Berkshire ${\times}$ Duroc (YBD). Each of the twenty crossed pigs were randomly selected with their live weights at the range of 110-120 kg. After being slaughtered and cooled at $0^{\circ}C$ for 24 h in a chilling room, the parts of loin and belly on the left side of the cooled carcasses were cut and prepared for analysis. The intramuscular fat contents of the loins from YLD were higher than those of the other crossbreds (p<0.05), and the YCY bellies had the highest moisture contents (p<0.05). Water holding capacity (WHC) of the loins from YCY were higher than the other crossbreds (p<0.05). Shear force values of the YBD loins were higher than the others (p<0.05). The TBARS values of YCY loins were significantly lower than the others at 0 d, but the difference disappeared after 14 d of storage. Sensory scores of YLD were ranked higher than the YCY or YBD in both the loins and bellies (p<0.05). The relatively high sensory values of YLD crossbred pork could be explained by the better WHC, the low shear forces, and the higher fat contents. The results indicated that the meat qualities could be altered by three-way crossbreeding.
