Characterization and Some Cultural Conditions of a Pullulanase Producing Aeromonas caviae No. S-76 (Pullulanase를 생산하는 Aeromonas caviae No.S-76의 특성과 배양조건)

  • - A bacterial strain No. S-76 which produced pullulanase powerfully was isolated frorn soil. The isolated bacterium was 0.4~$0.6\times 0.8$~1.4 $\mu\textrm{m}$ in size, gram negative, rods, motile and was identified as Aerornonas caviae by Bergey's manual of determinative bacteriology with various characteristics investigated. The highest yield of pullulanase of the strain was obtained by using the following medium: 1% pullulan, soluble starch or corn starch as a carbon sources and 0.5% yeast extract, peptone as nitrogen sources with an initial pH of 9.0. The optimal cutture conditions for production of pullulanase were at $32^{\circ}C$ for 2 days.

lsolation of Bacillus Strains Secreting Fibrinolytic Enzymes from Doen-Jang (된장으로부터 혈전용해능을 가진 Bacillus sp.의 분리 및 동정)

  • Various bacterial strains that secret extracellular fibrinolytic enzyme were screened from Doen-Jang, a traditional soybean fermented food in Korea. Five microbes of them were identified to be Bacillus sp. strains according to Bergey's manual of systematic bacteriology. The culture filtrates of B. amyloliquefaciens (2.46 plasmin unit/ml) and B. pantothenticus (3.82 plasmin unit/ml) showed a level of fibrinolytic activity that was about three times higher than that of plasmin 1.0 unit and Bacillus subtilis showed the highest fibrinolytic activity (4.94 plasmin unit/ml). All of the extracellular proteases showing the fibrinolytic activity are confirmed by SDS-PAGE followed by reverse fibrin zymogram activity assay and we proposed that some of the fibrinolytic enzymes from this work are novel enzymes.

  • For the purpose of producing cellulosic single-cell protein from the agricultural wastes, 172 strains of cellulose-assimilating bacteria were isolated from 102 samples of rotten woods, compost soils, soils and so on by the enrichment culture technique. The isolates were examined for their ability to utilize cellulose as carbon source, and then six strains were screened by their strong cellulose assimilating ability and identified as follows: 1. Among six strains of bacteria screened, five strains were identified as species belonged to the genus Cellulomonas and the remainder to the genus of Sporocytophaga. 2. The isolated Sporocytophaga species was identified as S. ellipsosporn because it has a ellipsoidal microcyst. 3. The isolated Cellulomonas species were identical to a strain of C. fimi, C. aurcgena, C. gelida, respectively and two strains to C. flavigena. 4. The isolated C. aurogena was proved to be a new variety becauuse it has different characteristics of assimilating pentoses such as arabinose and xylose from the strain discribed in Bergey's Manual.

Reevaluation of Isolation and Identification of Gram-positive Bacteria in Kimchi (김치에 서식하는 Gram 양성세균의 분리 및 동정의 재평가)

  • Attempts were made to isolate and identify Gram-positive or lactic acid bacteria in Kimchi fermentation. Species diversity depended on isolation media and temperatures, and diversity tended to be reduced with decrease of temperature. MRS and KM (natural medium prepared from Kimchi materials) were suitable respectively for isolation and present number of species. Identification of isolates was performed by dichotomous identification schemes arranged on the basis of Bergey's manual of Systematic Bacteriology (1986). Gram-positive bacteria isolated at different temperatures (5, 15, $25^{\circ}C$) were 5 species of Leuconostoc, 4 species of Streptococcus, 3 species of Pediococcus, 2 species of Bacillus and 18 species of Lactobacillus. Species with high frequency of appearance were Lactobacillus plantarum, Streptococcus raffinolactis, Leuconostoc mesenteroides subsp. mesenteroides at $25^{\circ}C$, L. plantarum, Lactobacillus fructosus, L. mesenteroides subsp. mesenteroides at $15^{\circ}C$ and L. mesenteroides subsp. mesenteroides, Leuconosotoc paramesenteroides, Lactobacillus maltaromicus at $15^{\circ}C$. In general, Kimchi fermentation was achieved by Lactobacillus spp. (59.7% frequency) at $25^{\circ}C$ and Leuconostoc spp. (65.2% frequency) at $5^{\circ}C$. Pediococcus cerevisiae and Streptococcus faecalis which have been so far known as bacteria of Kimchi fermentation were not isolated.

  • Microorganisms having beta-latamase inhibitory activity were selected from soil samples collected from 63 spots throughout the country. Screening procedures consist of two steps. Those are growth inhibition test of penicillinase-producing Staphylococcus aureus by double-layered agar plate containing penicillin G as a substrate, and that of penicillin sensitive Staphylococcus aureus ATCC 6538 in the similiar condition including penicillinase. Finally, a strain was selected from a soil sample of Pa-ju, Kyeong-gi Do. This strain was classified as a Streptomyces sp. by ISP(International Streptomycete Project) and Bergey's manual.

  • The purpose of this paper is to study on the morphological and physiological nature generally observed for the identifications, and the four strains of thermal resistanting aerobic bacteria isolated from swelled cans and the different soils collected from the wide area in Korea. The results obtained in the light of the Bergey's Manual for the identification of the bacteria, have been shown that the four strains of bacteria are pertained to Bacillus subtilis. The optimum temperature, pH and the thermal resistance (2.5 min. at $121^{\circ}C$) of the bacteria have been measured.

  • This experiment was conducted to determine the safety and feasibility of using compost-promoting-bacteria. Compost-promoting-bacteria was isolated from livestock compost containing sawdust. The isolated bacteria was identified as Bacillus subtilis LYH201 by the method of the composition of the fatty acid with MIDI system and Bergey's manual. This Bacillus subtilis LYH201 had the following characteristics : Gram-positive, straight rod ($0.5{\sim}0.7{\mu}m$ width, $2.5{\sim}3.0{\mu}m$ length), facultatively aerobic and product of xylanase, CMCase, catalase, oxidase, protease and $0.5{\sim}0.7{\mu}m$-amylase. Growth of Bacillus subtilis LYH201 at saccharose as carbon source(0.5%) was faster than other carbon source. Activity of cellulase. $0.5{\sim}0.7{\mu}m$-amylase and protease from Bacillus subtilis LYH201 after 24 hours at $50^{\circ}C$ by agar diffusion method was higher than that of low temperature. Optimum growth condition of Bacillus subtilis LYH201 was $50^{\circ}C$ and pH 6.

  • 1. According to the Bergey's manual of determinative bacteriology, the high amylase producing strain A-162 isolated from corn was similar to Bacillus subtilis in the characteristics. 2. The addition of corn powder 30%, milk casein 5% and $CaCO_3$. 5% to wheat bran was excellent as amylase producing media. 3. According to vessel content and quantities of the media, the optimum steaming condition of media was different. Excessive steaming (pressure and time) suppressed the growth of Bacillus subtilis var. A-162. 4. The optimum temperature of amylase produced was about $50^{\circ}C$ and its optimum pH 6.0.

  • Recently, we investigated characteristics of a bacterium which has never been reported as an inulin hydrolyzing microorganism. Several properties of the isolated microorganism were aerobic, rod typed, spore forming and Gram positive. According to the Bergey's manual, this bacterium tentatively appeared to be Becillus subtilis. This bacterium produced inulase constitutively in the media containing giucose, sucrose or cellulose without inulin as a sole carbon source. Also, inulase activities of the bacterium per unit culture volume showed 1.1 unit/ml comparable to 0.9 unit/ml of Kluyveromyces fragilis with relatively short culture time.

Diversity and Antibacterial Activity of Lactic Acid Bacteria Isolated from Kimchi (김치로부터 분리한 젖산균의 다양성과 항균성)

  • This study was carried out to investigate the isolation, identification, and antibacterial activity of lactic acid bacteria related to kimchi fermentation. Diluted kimchi soup was plated on the MRS agar media with CaCO$_3$ and incubated at $25^{\circ}C$ for 2 days. A total of 27 strains of lactic acid bacteria from various indigenous, spontaneously fermented vegetables (kimchi) were isolated. Combined methods of Bergey's manual of systematic bacteriology, BPB media analysis and 16S rDNA sequence analysis were applied for identification, however, their results did not coincide in several cases. Isolated lactic acid bacteria could be classified by the 16S rDNA sequence analysis as Leuconostoc mesenteriodes, Leu. carnosum, Lactobacillus curvatus, Lac. pentosus, Weisselia kimchi, W. cibaria, and Pediococcus pentosaceus. Leu. carnosum has not been reported in kimchi lactic acid bacteria. In addition, antibacterial activities of the isolates were tested with Bacillus subtilis, Escherichia coli, Salmonella enteritidis, S. paratyphica, S. typhi, Staphylococcus aureus, Shigella boydii, and S. sonnei. Some of isolates showed significant antibacterial activities to those pathogens.
