Synthesis of New Benzylpiperidinyl Ether Derivatives as Amyloid-beta Aggregation Inhibitors (베타아밀로이드응집 억제제 도출을 위한 새로운 벤질피페리디닐에터 유도체의 합성)

  • We designed and synthesized new benzylpiperidinyl ether derivatives as beta-amyloid aggregation inhibitors for the development of novel anti-Alzheimer's disease agents. As starting material, 4-hydroxypiperidine was used. For protection of the amine group in piperidine (2), di-tert-butyl dicarbonate was reacted with 4-hydroxypiperidine in the presence of triethylamine. For introduction of benzyl group, benzylbromide was treated with compound 2 in dioxane. After deprotection of Boc group on amine in compound 3, ester (5) was synthesized by addition of ethyl-4-chlorobutyrate. The alcohol 6 was synthesized by hydride reduction of 5 using $LiAlH_4$. To obtain final products (7-14), the alcohol 6 was condensed with each of substituted benzoic acids. To screen beta-amyloid aggregation inhibition of the products, thioflavinT assay was performed using $A{\beta}1-42\;at\;37^{\circ}C$ for 26 h incubation, in vitro. From the result of screening, compound 8, 9, 11 and 12 showed effective activity about $65{\sim}85\;{\mu}M\;as\;IC_{50}$ value. Among the prepared compounds, 4-[4-(benzyloxy)piperidino]butyl-4-chlorobenzoate (8) was the most effective inhibitor having $IC_{50}\;of\;65.4{\mu}M$.

Changes of Flavor Component in Tomato Fruits during Subatmospheric Pressure Storage (감압저장 중 토마토과실의 향기성분의 변화)

  • The kinds of volatiles in tomato fruits during storage were increased with the period of climacteric rise. The increasing and decreasing rate of their contents were slower under subatmospheric pressure-low temperature (SAP-L) than under normal atmospheric pressure-normal temperature (NAP-N). High contents of 2-hexanone, n-pentanol, and ethanol were decreased at the early period of storage and then increased. The content of n-octanol and furfural were increased, while that of n-butanol, cyclohexanone, and phenylacetaldehyde was decreased during storage. On the other hand, the content of trans, trans-2,4-decadienol, benzyl alcohol, and 2-heptanone was not changed during storage.

The Solubilization Site of Some Phenyl Alkanols in Aqueous Sodium Dodecylsulfate Micelle (몇가지 페닐 알카놀의 Sodium Dodeylsulfate 수용액 미셀내에서의 가용화 위치)

  • The solubilization sites of some phenyl alkanols such as phenol, benzyl alcohol, phenethyl alcohol, 3-phenyl-1-propanol solubilized in 0.2 M aqueous sodium dodecylsulfate micelle solution was studied by two dimensional heteronuclear correlation spectroscopy (2D C-H COSY). The results show more quantitative and clear solubilization sites in the SDS micelle than previous results using $^1H$-NMR spectrum integration. We found that most of the phenyl alkanols penetrate into the core of SDS micelle, and the insertion depth was 6.5∼7.0 methylene units from ${\alpha}$-methylene.

Effects of Ketoprofen on Wound Healing in Rats (Ketoprofen이 랫트의 창상치유에 미치는 영향)

  • Ketoprofen has been used as nonsteroidal anti-inflammatory (NSAI) agent for analgesia in surgical patients and increasing numbers of surgical patients are chronically take some forms of NSAI drugs. The purpose of this study was to investigate the therapeutic effects of ketoprofen on the healing of a closed linear surgical wound and on the contraction of an opened surgical wound in rats. The experimental groups were divided into two groups and ten rats were allocated in each group. In ketoprofen-treated group, the rats were given 2.5 mg/mg/day of ketoprofen by s.c. for ten days. In control group, the rats were given 1ml of benzyl alcohol and distilled water by s.c. for ten days. After time period, all rats were sacrificed, and the breaking strength and the collagen concentrations, at the wound site, were measured. In ketoprofen-treated group, the mean breaking strength and the mean collagen concentration were significantly decreased when compared with those of controls. The ketoprofen-treated group had shown a mean rate of wound contraction less than that of the control, although not statistically significant. These results suggested that ketoprofen impaired wound healing.

  • The main purpose of this study is to analyze the characteristics of toluene removal by plasma, photocatalyst, and plasma/photocatalyst system with the major parameters such as flow rate, inlet toluene concentration and applied voltage, etc., experimentally. In the combined plasma/photocatalyst process, rates of toluene conversion are represented as 99% at flow rate 250, 500 mL/min while, below 97% at flow rate 1000 mL/min due to the low residence time(reaction time) at the same applied voltage 4173 voltage and toluene inlet concentration 50 ppm. The intermediate products are detected by GC/MS analysis showing the small amounts of benzoic acid, benzyl alcohol and residual ozone concentration $0.04{\sim}0.05$ ppm generated by plasma process in the present system.

Explorative and Mechanistic Studies of the Photooxygenation of Sulfides

  • The results of recent work on the dye-sensitized photooxygenation of sulfides is discussed. In the case of dialkyl sulfides, the weakly bonded adduct initially formed with singlet oxygen (the persulfoxide) decays unproductively unless protonation by an acid (an alcohol or a carboxylic acid) facilitates its conversion to the sulfoxide. The effect is proportional to the strength of the acid (eg., less than 0.1 % chloroacetic acid in benzene is sufficient for maximal efficiency) and corresponds to general acid catalysis, suggesting that protonation of the persulfoxide occurs. On the other hand, with sulfides possessing an activated hydrogen in ${\alpha}$ position (eg., benzyl and allyl sulfides), hydrogen transfer becomes an efficient process in aprotic media and yields a S-hydroperoxysulfoniumm ylide, possibly arising from a conformation of the persulfoxide that is different from the one protonated in the presence of acids. Calculations on some substituted sulfides support this hypothesis. This process, which leads to C-S bond fragmentation with formation of an aldehyde, may be viewed as a general method for the preparation of aryl and heteroaryl aldehydes. In this effort, mechanistic studies offered new hints on the structure of the intermediate persulfoxide.

Graft Polymerization of Methacrylic acid onto PET Film (PET필름에 Methacrylic 산의 그라프트 중합)

  • Methacrylic acid was graft polymerized with benzoyl peroxide in itiator onto the commercial PET film. Graft polymerization was carried out in emulsion, which consisted of BPO (1), chlorobenzene(8) as swelling agent, tween 80(1) and sodium lauryl sulfate(1) as emulsifier and water(1,000). Original PET film has poor reactivities and, so the film preswdled with benzyl alcohol($150^{\circ}C$, 1hr) was also examined. The graft yield of PET film was increased with reaction temperature and monomer concentration. The graft yield of preswelled PET film was higher than that of no-treated PET film. Moisture regain was linearly increased with graft yield. DSC thermodiagram showed the Tm of grafted PET film was same as that of original PET film. Grafted PET film was dyed with methylene blue solution. The photograph of the cross section showed that grafting was occured not at the center but near surfaces.

Study on the Chemical Properties of Korean Local Tobacco (Nicotiana tabacum L.) (한국 재래종 연초(Nicotiana tabacum L.)의 화학적 특성)

  • Field experiment was conducted to find out the chemical characteristics of 12 Korean local varities, Local varities were compared with oriental, flue-cured and burley tobacco. Among tested varities, Hyangcho and Matskawa were desirable in respect of flavor, leaf color and texture. Hyangcho and Matskawa were higher in the content of nicotine and petroleum ether extract. Gajacho, Byeolcho and Kukjuo were higher in the content of reducing sugar, but lower nicotine, total nitrogen and petroleum ether extract. The nicotine and total nitrogen content of Kwangcho, Daruma and Catterton were lower than that of Br. 21. In the content of volatile organic acid which were major characteristic component of oriental tobacco, the Korean local varities were lower in the content of 3-methyl butanoic and 3-methyl pentanoic acid, but hirities, Hyangcho, Matskawa and Daruma were higher in the content of volatile organic acid. Korean local varities, especially small leaf group, were higher in the content of benzyl alcohol and furfural than any other type tobacco.

  • Column chromatographic separation of the MeOH extract from the aerial parts of Paris verticillata led to the isolation of three phenolics, two terpene glucosides and two pyrrolidine alkaloids. Their structures were characterized to be methyl caffeate (1), 5-hydroxy pyrrolidin-2-one (2), vanillic acid (3), benzyl alcohol glucopyranoside (4), (6S, 9R)-roseoside (5), staphylionoside H (6) and 5-methoxy pyrrolidin-2-one (7) by spectroscopic means. The isolated compounds (1-7) were for the first time reported from this source. The isolated compounds were tested for their cytotoxicity against four human tumor cell lines by SRB method in vitro.

Comparison of Volatile Aroma Components and Non-volatile Organic Acids in Tobacco Lamina and Stems. (잎담배 엽육과 주맥의 휘발성 정유성분 및 비휘발성 유기산의 비교)

  • Volatile aroma components, non-volatile organic acids in lamina and stems of flue-cured(NC 2326) and burley ( Burley 21) were analyzed by gas chromatography and mass spectrometry, respectively. Then compositional differences of these components between lamina and stems were discussed. The contents of volatile components were higher in flue-cured than in burley tobacco, and it was also higher in lamina then in stem. The major aroma components in lamina were neophytadiene , nicotine, solanone and benzyl alcohol but those in stems were palmitic acid, neophytadiene, nicotine, solanone and phenyl ethyl acetate. On the other hand, the contents of non-volatile organic acids were higher in burley than in flue-cured tobacco, and these values of burley tobacco were higher in lamina than in stem but flue-cured tobacco were higher in stem than in lamina. The major acids in all the above four tabacco samples were malic, citric, oxalic and linolenic acid.
