Apoptosis during Rat Tooth Development

  • Teeth develop via a reciprocal induction between the ectomesenchyme originating from the neural crest and the ectodermal epithelium. During complete formation of the tooth morphology and structure, many cells proliferate, differentiate, and can be replaced with other structures. Apoptosis is a type of genetically-controlled cell death and a biological process arising at the cellular level during development. To determine if apoptosis is an effective mechanism for eliminating cells during tooth development, this process was examined in the rat mandible including the developing molar teeth using the transferase-mediated dUTP-biotin nick labeling (TUNEL) method. The tooth germ of the mandibular first molar in the postnatal rat showed a variety of morphological appearances from the bell stage to the crown stage. Strong TUNEL-positive reactivity was observed in the ameloblasts and cells of the stellate reticulum. Odontoblasts near the prospective cusp area also showed a TUNEL positive reaction and several cells in the dental papilla, which are the forming pulp, were also stained intensively in this assay. Our results thus show that apoptosis may take place not only in epithelial-derived dental organs but also in the mesenchyme-derived dental papilla. Hence, apoptosis may be an essential biological process in tooth development.

  • - I aimed to isolate trans-acting factors involved in the basal expression level of eukaryotic genes. One of the yeast histidine biosynthetic gene, HIS5 was taken as a model for this study. HIS5 gene has a substantial basal level in amino acid rich medium and is derepressed if starved for any single amino acid. The derepression is mediated by cis-acting DNA sequences 5'-TGACTC-3' found in 5' non-transcribed region of the gene and trans-acting factors including GCN4 as positive factor and its negative factor GCDI 7, and GCNZ as a negative factor of GCD17. I first investigated the role of these trans-acting factors in HIS5 basal expression level by using HIS5-pH05 fusion in which expression of pH05 gene encoding inorganic phosphate-repressible acid phosphatase (APase) is regulated by HIS5 promoter. Strain with gcn2 or gcn4 mutation showed 3 to 4 fold lower APase activity than wild type. The level of APase activity was similar in gcn2 and gcn4 mutants. Trans-acting factors involved in basal level were identified by isolating 14 mutants showing increased expression of HISSPH05 fusion from gcn4 background. All the mutants carry a single nuclear recessive mutation and fall into four complementation groups, designated as bell (basal expression level), be12, be23 and be14.

A Survey of the Management of Elementary School Foodservice - II. Satisfaction of Food Service and Food Preferences of the Elementary School Students in the Kwangju & Chonnam Area - (학교 급식 관리에 대한 실태 조사 - II. 광주ㆍ전남지역 초등학생의 급식만족도와 음식기호도 조사 -)

  • Elementary schools students'degree of satisfaction on school food service and food preference were surveyed from 352 sixth-grade students in Kwangju and Chonnam area from June 22 to July 3 to enhance the quality of foodservice. The degree of satisfaction was high with cooked rice and soup in terms of temperature and serving state. There were significant differences in the degree of satisfaction depending upon the types of foodservice (p<0.001). There was significant differences in degree of satisfaction for side dishes depending upon the temperature (p<0.05), kinds (p<0.01) and amount (p<0.001). The degree of satisfaction with Kimchi was generally high in the serving state (p<0.05) and the amounts (p<0.01). The degree of satisfaction with desserts were high in the amount (p<0.001). The amount of leftover was high with soup, side dishes, kimchi, cooked rice, and desserts in order. The improvements made on children's behavior after the implementation of the school foodsevice were the adaptation to balanced diet and spending less money on sweets. The factors of influencing the school foodservice and the children's food intake were the education in school (teachers and dietitians) and the health status of their bodies (p<0.001). Children's favorite foods were sweet and sour pork, pork cutlets, oranges, watermelons, and strawberries in order. The least preferred foods were garlic seasoned with soy sauce, raw vegetables of broad bell-flowers, crown daisy greens, and steamed peppers. Children's favorite food types were fried food, desserts, one course meal, and bread. The least preferred types were pickled food, green and seasoned vegetables, cooked rice, and hard-boiled food in order. Generally, the children in the rural area favored most foods compared with those from urban area.

Analysis of Genes with Alternatively Spliced Transcripts in the Leaf, Root, Panicle and Seed of Rice Using a Long Oligomer Microarray and RNA-Seq

  • Pre-mRNA splicing further increases protein diversity acquired through evolution. The underlying driving forces for this phenomenon are unknown, especially in terms of gene expression. A rice alternatively spliced transcript detection microarray (ASDM) and RNA sequencing (RNA-Seq) were applied to differentiate the transcriptome of 4 representative organs of Oryza sativa L. cv. Ilmi: leaves, roots, 1-cm-stage panicles and young seeds at 21 days after pollination. Comparison of data obtained by microarray and RNA-Seq showed a bell-shaped distribution and a co-lineation for highly expressed genes. Transcripts were classified according to the degree of organ enrichment using a coefficient value (CV, the ratio of the standard deviation to the mean values): highly variable (CVI), variable (CVII), and constitutive (CVIII) groups. A higher index of the portion of loci with alternatively spliced transcripts in a group (IAST) value was observed for the constitutive group. Genes of the highly variable group showed the characteristics of the examined organs, and alternatively spliced transcripts tended to exhibit the same organ specificity or less organ preferences, with avoidance of 'organ distinctness'. In addition, within a locus, a tendency of higher expression was found for transcripts with a longer coding sequence (CDS), and a spliced intron was the most commonly found type of alternative splicing for an extended CDS. Thus, pre-mRNA splicing might have evolved to retain maximum functionality in terms of organ preference and multiplicity.

  • Compression work on a pyrite powder has been carried out using energy dispersive X-ray diffraction (EDXRD) with Mao-Bell type diamond anvil cell (DAC) and synchrotron radiation(SR) at room temperature. It has been reported the bulk moduli of pyrite show the large variations depending on the experimental conditions as well as the apparatus used. Thus, two kinds of sample in different pressure transmitting media of both NaCl and MgO powder emerged in alcoholic fluids were subjected to measure their compressibilities. Bulk moduli thus obtained are 138.9 GPa and 198.2 GPa, respectively, and this result contradicts to the anticipated values according to the hydrostaticity conditions of the sample chamber. This might be due to the alcoholic fluids phase transition mainly with the side effects from the difference of both solid state detector (SSD) used and E*d value applied. All experiments were performed at the Beam Line 1B2 of Pohang Light Source (PLS).

A Farm on Broiler Chickens' Welfare in Korea (동물복지 관련 육계농가 실태조사)

  • We conducted investigations of the information and welfare of broiler chickens in Korea. The livestock housing of all broiler chicken farms were windowless type, bell feeder and nipple waterer were used, and stocking density was relatively higher (ranging from 18.2 $birds/m^2$ to 24.2 $birds/m^2$) than RSPCA's welfare standards for chicken hens (less than 19 $birds/m^2$). The ratios of sandwich panel, urethane form, slate, and steel plate in roofing materials were 61%, 21%, 13%, and 5%, respectively. The ratios of sandwich panel, urethane form, brick, and steel plate in wall materials were 61%, 21%, 13%, and 5%, respectively. The ratios of soil and concrete in flooring materials were 10% and 90%. The mist spray, fan, and cooling pad in cooling facilities were 42%, 32%, and 26%, respectively. Thus we believe that present data contribute to develop the animal welfare certification for broiler chickens and to improve animal welfare in Korea.

High Pressure Phase Transition Study of ${\alpha}$-cristobalite $GaPO_4$ (${\alpha}$-크리스토발라이트 구조의 $GaPO_4$에 대한 고압 상변이 연구)

  • High pressure x-ray diffraction patterns of ${\alpha}$-cristobalite gallium phosphate ($GaPO_4$) were acquired up to 8.9 GPa at room temperature using Mao-Bell type diamond anvil cell with high flux synchrotron radiation. Starting orthorhombic phase (phase-I) shows the splitting of peak which is possibly resulted from the pressure induced orientation disorder of the framework structure of tetrahedra. This is designated as phase-I'. This phase transforms to the orthorhombic high pressure phase-III between 2 and 3 GPa. Present phase transition sequence is not in accord with the recent high pressure X-ray diffraction results performed on the same starting sample (Ming et al., 2007). X-ray pattern of the unloaded sample to ambient pressure shows that the structure retains that of the high pressure phase prior to decompression.

  • Design parameters required for Methane/oxygen bipropellant small-rocket-engine were derived through a theoretical performance analysis. The theoretical performance of the rocket engine was analyzed by using CEA and optimal propellant mixture ratio, characteristic length, and optimal expansion ratio were calculated by assuming chemical equilibrium. A coaxial-type swirl injector was chosen because of its outstanding atomization performance and high combustion efficiency compared to other types of injector and also a bell nozzle with 80% of its full length was designed. The rocket engine configuration with 1.72 MPa of chamber pressure, 0.18 kg/s in total propellant mass flow, and O/F ratio of 2.7 was proposed as a ground-firing test model.

A Study on the Corelation between the Variation of Land Cover and Groundwater Recharge Using the Analysis of Landsat-8 OLI Data (Landsat-8 위성을 통한 토지피복 변화와 지하수 함양량 상관성 고찰)

  • Based on monthly average groundwater recharge over a nearly 10 year period, results of fully integrated hydrologic modeling of SWAT-MODFLOW, land cover, land use, soil type and hydrologic response unit (HRU) was used to assess the dominant influencing factors of groundwater recharge spatial patterns in Jangseong district. As dominant factors, land cover was FRSE (forest-evergreen) and soil type was Samgag. Landsat-8 OLI imaging spectrometer data were acquired in the period 2003 to 2004 and seasonal bare soil lines (BSL) were estimated through NIR-RED plot. Extent of slope of BSL was from 1.092 to 1.343 and the intercept was from -0.004 to -0.015. To know correlation between spatial groundwater recharge and soil-vegetation indices (PVI, NDVI, NDTI, NDRI), this study employed frequency and regression analysis. On May, RED band increased up 3 to 4 times compared to other seasons and only one turning point appeared as recharge-index with upward parabola bell shape as results of existing research. Considering precipitation, if the various studies for relationship between groundwater recharge and soil-vegetation index just like NDVI are performed, it is possible to estimate groundwater recharge through analyzing remote sensing data.

Analysis of Nursing Studies of Coping Conducted in Korea from 1978 to 1995 (대처 개념의 국내 간호연구 분석)

  • This study was done to analyze the trends of research on coping in Korea, to suggest future direction, for research on coping, and ultimately to contribute to an increase in explanation of adaptation. This article reviewed 79 nursing research papers on coping done since 1978 by examining them according to the period of publication or presentation, research design, type of subjects, measurement instruments, research for a degree or not, range of reliability, and association of coping and related variables. The results are as follows : The number of studies on coping increased rapidly from the mid-1980's and decreased slowly from the mid-1990's. The maority of the studies were surveys, comparative studies, or correlational studies. The subects of the 46 studies were healthy people, while those in the remaining studies were patients with a variety of illnesses. Thirth-eight studies on coping were done for master's thesis, three for dissertion, and 38 were not degrees. The Bell and Jalowiec coping scales have not been used since the early 1990's. In contrast, Lazarus and Folkman's W.C.C.L. has been used increasingly since that time. The reliabilities of the coping scale were reported in 37 cases and the Cronbach's alpha coefficients were .71 to .86. All subjects reported using more problem-oriented coping than emotion-oriented coping in short-term or emotion-oriented coping and healthy groups did more long-term coping. It was difficult to describe consistently the relationship between stress and coping according to the type of coping scale or research subjects, but generally moderate relationships were found. This was due to instrumental problems and no consideration of situational context. The subject group who used more short-term coping and less long-term coping reported poorer mental status, and higher scores in burnout and state anxiey than others. That is, the relationship between stress and adaptation increased the power of explanation with intervening the mediating effect of coping. The association of locus of control, mastery, social support, and self-concept with coping showed positive relationships : those of uncertainty and severity in illness with coping showed negativerelationships ; those of state anxiety and depression with short-term coping were positive, and those of self-esteem with long-term coping or problem-oriented coping were negative. There were significant differences in the scores of types of coping according to religion, level of education, and socio-economic status. That is, Presbyterians and Catholics, those with higher education levels and higher socio-economic status used more long-term or problem-oriented coping. On the basis of the above findings the following recommendations are made : 1. There is a need to test the mediating effect of coping variable in order to clarify the concept. 2. Longitudinal studies are needed to determine the patterns of change in coping strategies when stressful events are encountered. 3. It's necessary to develop a reliable and variable measurement tool for coping. 4. There is a need to identify subscales of coping to increase explanation of variance 5. It's necessary to consider personal, situational, and antecedent variables : the characteristics of subject populations, the natures of illness and treatment situations. 6. The power of explanation of studies designed to identify the stress-adaptation process should be increased using the combination model of process-oriented coping and cognitive-structural model.
