한우 Fatty Acid Synthase (FASN) 유전자 반수체형의 후대검정우 육량 및 육질에 미치는 영향 (Effect of the Fatty Acid Synthase Gene for Beef Quantity Traits in Hanwoo Breeding Stock)

  • 본 연구는 지방산합성의 완성에 관여하는 소 FASN (Fatty acid synthase) 유전자내의 g.11280G>A (Silent), g.13125T>C (Exon24-His1390Tyr)와 g.17924G>A (Exon39-Thr2158Ala) 단일염기 변이들이 한우집단 925 두를 대상으로 도체형질과의 연관성 분석, 연관불균형의 정도 및 반수체형의 분석을 위하여 수행하였다. g.11280G>A 변이의 연관성분석에 있어서는 도체중, 등지방두께 및 육량지수에 유의성이 있는 것으로 관찰되었으며 g.17924G>A변이 역시 연관성분석에서 도체중에 유의성이 있는 것으로 나타났다(P<0.05). 하지만 g.13125T>C 변이는 한우집단에서 형질과의 유의적인 연관성을 관찰 할 수 없었다. 또한 g.11280G>A, g.13125T>C와 g.17924G>A 변이들에 대한 연관불균형(Linkage disequilibrium) 분석을 한 결과 g.11280G>A와 g.13125T>C 두변이에 대한 r-square=0.177로 나타났으며 D'=0.568 로 관측되었다. 또한 g.13125T>C와 g.17924G>A 두 변이들은 r-square = 0.207로 나타났으며 D'=0.607로 나타났으며 g.11280G>A와 g.17924G>A 변이들은 r-square = 0.101으로 나타났으며 D'=0.920으로 관찰되었다. 연관불균형 유의수준(r-square>0.1, D'>0.500)에 따라 계산된 반수체형 총 7개중 그 빈도가 0.05 이상인 중요한 반수체형은 4개(-GCG- [0.378], -ATG- [0.301], -GTA- [0.191], -ACG- [0.063])를 선별하여 연관성분석을 실시하였다. FASN 반수체형 1번(-GCG-), 반수체형 2번(-ATG-)와 반수체형 4번 (-ACG-)는 한우집단에서 형질과의 유의적인 연관성을 관찰 할 수 없었으나 FASN 반수체형 3번(-GTA-)은 도체중과 연관성을 보였다(P=0.0327). 염색체상 반수체형 -GTA-.구조를 가지지 않은 동형접합체(0-copies)인 개체의 도체중은 354.70+3.58로 나타났으며, 반수체형 -GTA- 구조를 하나만 가지는 이형접합체(1-copies) 347.63+4.84으로 관찰되었으며 0-copies인 동형접합체보다 현저히 낮은 도체중을 나타내었다. 또한 반수체형 -GTA- 구조를 가지는 동형접합체(2-copies)인 개체는 22두가 나타났으며 도체중은 351.01+4.18로 0-copies 인 동형접합체 보다 낮은 도체중을 나타내었다. 이상의 결과는 소 FASN 유전자내의 g.11280G>A, g. 13125T>C와 g.17924G>A 변이들은 육질 뿐만 아니라 육량에도 관여하는 것으로 사료되며, FASN 유전자내의 g.11280G>A와 g.17924G>A 두 개의 단일염기변이는 한우개량사업소의 씨숫소의 유전자 마커를 통한 도움 선발(marker assisted selection, MAS)에 이용 될 수 있을 것이며 일반농가에서도 송아지 생산을 위한 암소 선발에 활용될 수 있을 것으로 사료된다.

한우 거세우에 있어서 생체단층조사기법에 의한 육질판정과 도축 육질 측정치와의 관계 (Relationship Between Ultrasonic and Carcass Measures for Meat Qualities in Hanwoo Steers)

  • 한우의 고급육과 관련된 비육생리를 규명하여 최적의 출하시기를 결정하고 한우 개량을 위한 종축검정에 참고자료를 제공하기 위하여1999-2000년도에 출생하고 한우 후대검정에 공시된 거세 검정우들 중 127두를 무작위 추출하여 생후 11개월령부터 24개월령 출하시까지 1개월 단위로 생체단층촬영기를 이용하여 육질을 조사하였다. 조사된 자료를 이용하여 월령별 또는 체중 증가에 따른 배장근단면적, 등지방두께 및 근내지방도의 변화를 살펴보았고 이들 측정치가 도축하여 조사된 실측치와 상관관계를 알아 보았다. 분석결과 배장근단면적의 증가는 체중의 증가와 상당히 밀접하게 관계되었으며(P<0.01) 등지방두께의 증가는 비육후기로 갈수록 월령의 이차항과 정의 관계를 보였다(P<0.01). 근내지방도의 증가는 개체의 나이와 밀접한 관계를 보였으며(P<0.01) 개체간의 변이는 생후 11개월령부터 시작되고 있었으나 17개월령 이후로 갈수록 개체간 변이 폭이 커지고 있음을 알 수 있었다. 월령별 육질 측정치가 실측정치와의 상관관계에 있어서는 나이 및 체중에 대한 보정을 실시하지 않았을 경우, 생후 17개월령 이후에 조사된 전형질에 있어서 0.6이상의 상관관계를 보이고 있었으며 22개월령 이후의 측정치와 실측치 간에는 90% 전후의 높은 상관관계를 보이고 있었다. 출하 2주 이내의 초음파 측정치와 도축 실측치와의 상관은 나이 및 체중에 대한 보정을 실시하고 추정하였을 때, 배장근단면적에 대하여는 0.91~9.92, 등지방두께에서 0.94 및 근내지방도에서 0.88으로 비교적 높게 추정되었다. 하지만 분석에 고려된 육질 관련 월령별 초음파 측정치들간에 상관은 비교적 낮게 추정되었는데 이는 적은 조사자료 및 초음파 측정의 오차 등에 기인한 것으로 사료되며 보다 많은 연구가 필요할 것으로 사료되었다. 또한 한우에 있어서 생축으로부터 육질을 판정하기 위해서는 15-17개월령 이후가 적당할 것으로 사료되었으며 22개월령 전후의 측정치는 출하시기를 결정하는데 좋은 참고자료로 사용될 수 있을 것으로 사료되었다. 한편 종축선발을 위한 육질 조기판정은 외국의 12개월령 측정치와는 달리 15개월령 측정치가 보다 타당하다고 사료되었으나 이들 측정치에 대한 유전적 변이에 대한 연구가 필요할 것으로 사료되었다.

한우에 있어서 유전체 육종가 추정 (Prediction of genomic breeding values of carcass traits using whole genome SNP data in Hanwoo (Korean cattle))

  • Genomic breeding value (GEBV) has recently become available in the beef cattle industry. Genomic selection methods are exceptionally valuable for selecting traits, such as marbling, that are difficult to measure until later in life. One method to utilize information from sparse marker panels is the Bayesian model selection method with RJMCMC. The accuracy of prediction varies between a multiple SNP model with RJMCMC (0.47 to 0.73) and a least squares method (0.11 to 0.41) when using SNP information, while the accuracy of prediction increases in the multiple SNP (0.56 to 0.90) and least square methods (0.21 to 0.63) when including a polygenic effect. In the multiple SNP model with RJMCMC model selection method, the accuracy ($r^2$) of GEBV for marbling predicted based only on SNP effects was 0.47, while the $r^2$ of GEBV predicted by SNP plus polygenic effect was 0.56. The accuracies of GEBV predicted using only SNP information were 0.62, 0.68 and 0.73 for CWT, EMA and BF, respectively. However, when polygenic effects were included, the accuracies of GEBV were increased to 0.89, 0.90 and 0.89 for CWT, EMA and BF, respectively. Our data demonstrate that SNP information alone is missing genetic variation information that contributes to phenotypes for carcass traits, and that polygenic effects compensate genetic variation that whole genome SNP data do not explain. Overall, the multiple SNP model with the RJMCMC model selection method provides a better prediction of GEBV than does the least squares method (single marker regression).

Artificial Insemination and Embryo Transfer Project to Foster Mongolia Dairy Industry

  • Mongolia has 80% livestock of total agriculture industry, 170,000 farms are engaged, 2,500,000 of cows that were beef and dairy cows are raised. Despite of Mongolian has great application with milk, there are not clear differences between cow and dairy cattle, and the production of milk is also low. But the milk suppliers are varied (horse, sheep, goat, etc), so that the total milk production is 500 thousand ton per year. It's really considerable to improve the breed of owing to many problems with big differences among milk qualities. For carrying out for first year project, artificial insemination project was operated with 3rd grade Holstein semen that were imported from S. Korea, and initiation and field training were also carried out through appropriate AI technique we developed for Mongolia environment. Local information research and MOU conclusion were done with professor D. Altangerel in May $10^{th}{\sim}13^{th}$, 2009, and development for AI technique and AI equipments were supplied for Mongolia breeding and natural environment in July $10^{th}{\sim}17^{th}$ in 2009. All cows were treated by synchronization for AI. To do this, $PGF_{2\alpha}$ injection were treated for luteal phase cow, if it wouldn't work, try again after 11 days. After confirmation of estrus, AI and AI training were carried out with sperm injection in the uterus or cervix by rectum-vagina method which is common worldwide, the most effective artificial insemination technique. If cows were return to next estrus cycle, second AI was carried out about approximately 21 days after artificial insemination. After 2 months, all cows not showing return estrus should be taken pregnancy test. Every pregnant cow will be cared thoroughly. Total 48 cows administrated by $PGF_{2\alpha}$ for synchronization and after 48 hours 45 cows (93.8%) showing estrus were detected and then artificial inseminate them within who 8 cows (27.8%) showed return estrus. Therefore, Using $PGF_2{\alpha}$ for synchronization is effective to use for Mongolia breeding conditions. There are possibility of base for food production after all, including increase of livestock production in Mongolia by improvement of breeding cow with AI and embryo transfer project.

National Genetic Evaluation (System) of Hanwoo (Korean Native Cattle)

  • Hanwoo (Also known as Korean native cattle; Bos taurus coreanae) have been used for transportation and farming for a long time in South Korea. It has been about 30 yrs since Hanwoo improvement began in earnest as beef cattle for meat yield. The purpose of this study was to determine the trend of improvement as well as to estimate genetic parameters of the traits being used for seedstock selection based on the data collected from the past. Hanwoo proven bulls in South Korea are currently selected through performance and progeny tests. National Hanwoo genetic evaluations are implemented with yearling weight (YW), carcass weight (CW), eye muscle area (EMA), backfat thickness (BF) and marbling score (MS). Yearling weights and MS are used for selecting young bulls, and EMA, BF, and MS are used for selecting proven bulls. One individual per testing room was used for performance tests, and five individuals per room for progeny tests. Individuals tested were not allowed to graze pasture, but there was enough space for them to move around in the testing room. Feeds including roughages and minerals were fed ad libitum, and concentrates were provided at the rate of about 1.8% of individual weight. Overall means of the traits were $352.8{\pm}38.56$ kg, $335.09{\pm}44.61$ kg, $77.85{\pm}8.838\;cm^2$, $8.6{\pm}3.7$ mm and $3.293{\pm}1.648$ for YW, CW, EMA, BF and MS. Heritabilities estimated in this study were 0.30, 0.30, 0.42, 0.50 and 0.63 in YW, CW, EMA, BF and MS, respectively, which are similar to results from previous research. Yearling weight was 315.54 kg in 1998, and had increased to 355.06 kg in 2011, resulting in about 40 kg of improvement over 13 yrs. YW and CW have improved remarkably over the past 15 yrs. Breeding values between 1996 and 2000 decreased or did not change much, but have moved in a desirable direction since 2001. These improvements correspond with the substantial increase in use of animal models since the late 1990s in Korea. Hanwoo testing programs have practically contributed to the improvement in aspects of quality and quantity. In sum, the current selection system is good enough to accommodate circumstances where fewer sires are used on many more cows. Although progeny tests take longer and cost more, they seem to be appropriate under the circumstances of the domestic market with its higher requirement for better meat quality. Consequently, accumulative data collection, genetic evaluation model development, revision of selection indices, as well as cooperation among farms, associations, National Agricultural Cooperative Federation, universities, research institutes, and government agencies must be applied to the Hanwoo selection program. All these efforts will assist the domestic market to secure a competitive position against imported beef under Free Trade Agreement trade system and will provide farmers with higher profits as well as the public with a higher quality of beef.

Association between Polymorphisms of MSTN and MYF5 Genes and Growth Traits in Three Chinese Cattle Breeds

  • The objective of this study was to assess the association of polymorphisms in MSTN and MYF5 genes with growth traits in three Chinese cattle breeds. Only one homozygous animal with BB genotype at MSTN locus was observed in Jiaxian population which was at Hardy-Weinberg disequilibrium (p<0.05). The frequencies of allele A at MSTN locus and allele B at MYF5 locus in the three Chinese breeds were 0.9550/0.9730/0.9720 and 0.8275/0.7581/0.7523, respectively. Allele A at MSTN locus and allele B at MYF5 locus were dominant in these three populations. No statistically significant differences in growth traits were observed between the genotypes of the Jiaxian breed at MSTN and MYF5 loci and the Nanyang breed at MYF5 locus. However, there were statistically significant differences between the genotypes at MSTN locus of the Nanyang breed for WH, HG, HGI and HGBLR (p<0.05), and of the Qinchuan breed for BLI (p<0.05). The SNP in MYF5 had significant effects on WH and HHC of Qinchuan animals (p<0.05). These results suggest that MSTN and MYF5 are strong candidate genes that influence growth traits in cattle. Other SNPs of MSTN and MYF5 or other linked genes should also be studied, which could lead to the development of selection plans to improve the performance of Chinese cattle and also promote the breeding of genuine beef cattle in China.

Dairy Cows of High Genetic Merit for Yields of Milk, Fat and Protein - Review -

  • Extensive emphasis on milk and milk fat yields with no diversion for beef performance increased the yield efficiency of North American dairy cattle. Heavy demand for North American genetics followed national strain comparison trials in Poland, and US and Canadian dairy cattle and germplasm still are an important source of genetics for many countries. Genetic improvement has accelerated in many countries because of the implementation of sampling programs for young bulls and improved evaluation procedures. Rapid access to information and more frequent calculation of genetic information also are having a positive impact on genetic improvement. Traits other than yield should be considered in a breeding program, but those traits mist have a reasonable opportunity for improvement and sufficient economic worth. Because of ever increasing efficiency, the world's milk supply comes from fewer cows each year. However, no decline in the rate of genetic improvement is apparent under current genetic practices; estimates of heritability are increasing, and a decline in yield efficiency is unlikely in the near future. As management improves, especially for subtropical conditions, many of the selection principles used in temperate climates will be adopted for more adverse environmental conditions.

Study on the Growth Pattern of Gayals (Bos Frontalis) and their Crossbred Calves

  • A study was conducted with 14 gayals to study their growth pattern from birth to 24 months of age, and gayal was compared with native calves and crossbred calves of gayals and exotic animals. A significant difference in liveweight gains up to 3 months (p<0.001) or up to 6 months (p<0.05) was found between gayal and native calves, but the difference reduced with increase of age and found non-significant (p>0.05) at 9 months and 12 months. The daily liveweight gain up to 3, 6, 9 or 12 months of age of gayals were 404.60, 306.80, 315.20 and 312.20 g/d, respectively and of native calves were 217.3, 245.3, 262.0 and 269.0 g/d, respectively. Gayal female calves had lower weights at birth (20.2 kg) and weaning (81.8 kg) and daily liveweight gains (298.5 g/d) than males (24.3 kg, 84.0 kg and 317.8 g/d, respectively) but the differences were not significant (p>0.05). Cross breeding gayal cows with Friesian bulls increased daily liveweight gain of the crossbred calves. The rate of increase of daily liveweight gain may further be increased if gayals are crossed with beef type animals, and optimum feeding may also be required which needs further research.

Reproductive Potentials of Gayal (Bos frontalis) under Semi-intensive Management

  • The assessment of reproductive potentials of female gayal under semi-intensive management was conducted with 18 animals in Bandarban hilly area of Bangladesh. Age at first estrous (days), age at first conception (days), length of estrous cycle (days), number of service per conception (natural), gestation period (days), age at first calving (days), and calving interval (natural service) (days) of gayal were $598.2{\pm}168.44$, $723{\pm}169.94$, $21.86{\pm}2.93$, 1.41, $296.05{\pm}3.87$, $1014.42{\pm}260.32$ and $465{\pm}80.48$ respectively. Daily milk yield and lactation length of gayal were $304.98{\pm}30.46ml/day$ and $116.67{\pm}8.08days$. Gayal male calves had higher birth weight (19.67 kg) than female calves (15.58 kg) in first lactation. Birth weight increases in second and third lactation than first lactation in both male and female calves. Highest conception rate observed in winter season and 70.60% successful conception occur when service was given within 21-30 h of estrous. Maximum number of female gayal use to come in estrous in winter and calved in monsoon and autumn. Cervicitis and irregular heat were the most reproductive problems followed by metritis, case of abortion, anestrous and repeat breeding. From this study it was observed that the reproductive performance of gayal is very close with the domestic cattle. It may use as beef cattle for the hilly regions of Bangladesh.

  • Revolutionary advancements, such as the reduction in DNA sequencing costs and genome editing, have transformed biotechnology, fostering progress in manipulating biomolecules, engineering cells, and computational biology. Agriculture and food production have significantly benefited from tools like high-throughput microarrays, accelerating the selection of desired traits. Genetic engineering, especially utilizing genome editing, facilitates precise alterations in plants and animals, harnessing microbiomes and fostering lab-grown meat production to alleviate environmental pressures. The emergence of new biotechnologies, notably genome editing, underscores the necessity for regulatory frameworks governing LM (living modified) organisms. Global regulations overseeing genetically engineered or genome-edited (GE) organisms, encompassing animals, exhibit considerable diversity. Nonetheless, prevailing international regulatory trends typically exclude genomeedited plants and animals, employing novel biotechnological techniques, from GMO/ LMO classification if they lack foreign genes and originate through natural mutations or traditional breeding programs. This comprehensive review scrutinizes ongoing risk and safety assessment cases, such as genome-edited beef cattle and fish in the USA and Japan. Furthermore, it investigates the limitations of existing regulations related to genome editing in Korea and evaluates newly proposed legislation, offering insights into the future trajectory of regulatory frameworks.
