Disinfection of Nuclear Polyhedrosis Virus and Yellow Muscardine pathogen (Beauveria bassiani) of Silkworm, Bombyx mori L., by Formalin Fumigation (누에고름병 바이러스와 흔굳음병균에 대한 포르말린 훈연소독)

  • The disinfectivity of formalin fumigation was tested against silkworm nuclear polyhedrosis virus and yellow muscardine pathogen, Beauveria bassiana. The inactivation of the virus was acquired when it was fumigated by adding 30g of potassium permanganate to 75$m\ell$ of formalin per 3.3$m^2$ of rearing room area and viability of the yellow muscardine pathogen was also lost with the same treatment of fumigation. It was also proved that the fumigation didn't give and damage to silkworm larvae when it was applied 2 or 3 times to grown larvae.

  • Beauveria bassiana infects a number of pest species and is known to produce insecticidal substances, such as the nonribosomal peptides (NRPs) beauvericin and bassianolide. However, most NRPs and their biological roles in B. bassiana remain undiscovered. To identify NRPs that potentially contribute to pathogenesis, the 21 predicted NRP synthetases (NRPSs) or NRPS-like proteins of B. bassiana ARSEF 2860 were primarily ranked into three functional groups: basic metabolism (7 NRPSs), pathogenicity (12 NRPSs), and unknown function (2 NRPSs). Based on the transcript levels during in vivo growth on diamondback moth (Plutella xylostella (Linnaeus)), half of the Group II NRPSs were likely to be involved in infection. Given that the metabolites biosynthesized by these NRPSs remain to be determined, our result underlines the importance of the NRPSome in fungal pathogenesis, and will serve as a guide for future genomic mining projects to discover functionally essential and structurally diverse NRPs in fungal genomes.

Study of the Rheological Properties of a Fermentation Broth of the Fungus Beauveria bassiana in a Bioreactor Under Different Hydrodynamic Conditions

  • Fermentation with filamentous fungi in a bioreactor is a complex dynamic process that is affected by flow conditions and the evolution of the rheological properties of the medium. These properties are mainly affected by the biomass concentration and the morphology of the fungus. In this work, the rheological properties of a fermentation with the fungus Beauveria bassiana under different hydrodynamic conditions were studied and the rheological behavior of this broth was simulated through a mixture of carboxymethyl cellulose sodium and cellulose fibers (CMCNa-SF). The bioreactor was a 10 L CSTR tank operated at different stir velocities. Rheological results were similar at 100 and 300 rpm for both systems. However, there was a significant increase in the viscosity accompanied by a change in the consistence index, calculated according to the power law model, for both systems at 800 rpm. The systems exhibited shear-thinning behavior at all stir velocities, which was determined with the power law model. The mixing time was observed to increase as the cellulose content in the system increased and, consequently, the efficiency of mixing diminished. These results are thought to be due to the rheological and morphological similarities of the two fungal systems. These results will help in the optimization of scale-up production of these fungi.

Occurrence and Characteristics of other Fungi in the Artificial Cultivation Farms of Paecilomyces tenuipes (동충하초 재배상 내 기형균 및 오염균의 발생현황 및 특성)

  • Occurrence and characteristics of infectious fungi and physiologically abnormal Paecilomyces tenuipes were examined. These unfavorable fungi are recently known to cause yield loss, decreased quality of products in artificial cultivation farms of P. tenuipes. Total 529 fungal isolates were collected from 22 farms of 7 provinces. These isolates were composed of physiologically abnormal P. tenuipes strains and other infectious fungal strains, which constituted 23.2% and 50.9%, respectively. Physiologically abnormal P. tenuipes strains showed irregular synemata, and absence of local color. They were divided into 4 types or C-l, C-2, C-3 and C-4. Other infectious fungal strains were identified to 5 species, Beauveria bassiana, Fusarium sp., P fumosoroseus, Tricothecium roseum, Aspergillus parasiticus, which were 22.1, 13.8, 7.6, 4.0 and 3.4%, respectively. All of them were hyphomycetous fungi, did not produce synemata, and revealed faster growth rate than that of P. tenuipes.

  • This study was carried out to investigate active constituents of Bombysis corpus on the neurite outgrowth from PC 12 cells led to isolate three new and a known sphingolipids from the n-hexane soluble portion and five amines from the butanol soluble portion of its methanol extract. On the basis of spectroscopic data, their structures have been elucidated as (4E,6E,2S,3R)-2-N-eicosanoyl-4,6-tetradecasphingadienine, (4E,2S,3R)-2-N-eicosanoyl-4-tetradecasphingenine,(4E,6E,2S,3R)-2-N-docosanoyl-4,6-tetradecasphingadienine,(4E,6E,2S,3R)-2-N-docosanoyl-4,6-tetradecasphingadienine,(4E,2S,3R)-2-N-octadecanoyl-4-tetradecasphingenine, 1,7-dimethyl-xanthine, uracil, urea, betaine and tyrosine, respectively. The neurite outgrowth activities of these compounds were examined in PC12 cells by measuring the length of neurites. These compounds promoted neurite outgrowth in PC12 cells significantly.

  • Biological control of lepidopterous insect pests, Agrotis segetum, Artogeia rapae, Mamestra brassicae, Plutella xylostella, Spodoptera exigua, and S. litura with entomopathogenic fungi, Beauveria bassiana and Metarhizium anisopliae isolated from Gyeongbuk province were investigated. Mean lethal concentration ($LC_50$) values of B. bassiana and M. anisopliae against $2\cdot3rd$ instar of A. segetum larvae were $1.2\times10^7\;conidia/m\ell$ and $5.2\times10^6\;conidia/m\ell$, respectively. $LC_50$ values of B. bassiana and M. anisopliae against $2\cdot3rd$ instar of A, rapae larvae were $1.2\times10^7\;conidia/m\ell$ and $5.2\times10^6\;conidia/m\ell$, respectively. $LC_50$ values of B. bassiana and M. anisopliae against $2\cdot3rd$ instar of M. brassicae and P. xylostella, larvae were $1.5\times10^6\;conidia/m\ell$, $9.7\times10^5\;conidia/m\ell$, $3.0\times10^6\;conidia/m\ell$ and $1.4\times10^6\;conidia/m\ell$, respectively. $LC_50$ values of B. bassiana and M. anisopliae against $2\cdot3rd$ instar of S. exigua, and S. litura larvae were $6.3\times10^6 \;conidia/m\ell$, $2.6\times10^6\;conidia/m\ell$, $1.6\times10^7\;conidia/m\ell$ and $3.4\times10^6\;conidia/m\ell$ respectively.

Notes on Cordyceps species Collected from the Central Region of Nepal

  • The present study was carried out to explore the Cordyceps species and other entomopathogenic fungal flora around Kathmandu Valley and a few high altitude locations of Nepal. In this paper, we report eight Cordyceps species as new to Nepal: C. gracilis, C. ishikariensis, C. liangshanensis, C. martialis, C. militaris, C. pruinosa, C. sphecocephala and C. tricentri. We also mention a few allied genera such as Beauveria, Hirsutella and Paecilomyces from Nepal. Further collections from different ecological regions of Nepal will show the richness of entomopathogenic fungal floral diversity of Nepal.

Entomopathogenicity of Simplicillium lanosoniveum Isolated in Korea

  • Fruiting bodies similar to those of the ascomycete fungi Podostroma cornu-damae and Cordyceps militaris were collected from Mt. Seunghak in Busan, Korea on August 21, 2012. The fruiting bodies were cylindrical, with tapered ends and golden red in color. The fruiting bodies contained abundant conidiophores bearing single-celled conidia, but no perithecia or asci. Pure culture of the fungal isolates was obtained through single-spore isolation. Analyses of morphological characteristics, including conidia shape, and phylogenetic traits, using internal transcribed spacer sequences, showed that these isolates belonged to the species Simplicillium lanosoniveum. Although this fungal species is known to be mycoparasitic, the isolates obtained in this study were unable to infect fungi. However, silkworms (Bombyx mori) inoculated with the fungal isolates died during the larval or pupal stages, as has been shown for the strongly entomopathogenic fungus Beauveria bassiana. This study is the first report of the entomopathogenicity of S. lanosoniveum and indicates its potential for use in biological control of insects.

  • Single ascospore isolates of Cordyceps bassiana were observed for their colony pigmentation on Sabouraud Dextrose agar plus Yeast Extract (SDAY) plates and were inoculated in a brown rice medium for production of fruiting bodies. Colony pigmentation did not show any relationship with perithecial stromata formation. The isolates were also grown on opposite sides of SDAY agar plates and were observed for vegetative compatibility. Neither vegetative compatibility nor perithecial stromata could be found to be related to each other. It was concluded that fertile fruiting body production was independent of colony pigmentation and vegetative compatibility. Synnemata formation was found to be more common than perithecial stromata formation. This might be due to its highly conidiogenous anamorphic stage, i.e., Beauveria bassiana.

A Study on the Investigation and Application of Microbial Pathogens of Major Insect Pests of Forest in Korea (중요산림해충의 병원미생물 개발에 관한 연구)

  • The study has been carried to investigate a possibility to control several major insect pest of forest by microbial pathogens existing in nature as one of the biological control measure. Microorganisms including polyhedral virus isolated from diseased fall webworm were total of 4 kinds pathogenic microbes among these 4 kinds were polyhedral virus and Bacillus .species. Control effect of these two pathogens appeared to be $70.6\%$ and $49.5\%$, respectively, when they were compared with those of control plot that was $27.8\%$. Each one of bacterium species and fungus species were isolated from diseased Japanese alder leaf beetle. Pathogenisity to the healthy beetle was recognized by the fungus species, while the bacterium showed none of pathogenisity. The fungus was identified as Beauveria sp. and its effect on the beetle control was $96.2\%$ while untreated plot showed $49.2\%$ of dead beetles in the same period. Fifteen species of microbes were isolated from diseased larvae of pine gall midge. Six species out of 15 showed certain level of insecticidal effect to the larvae of the insects. The highest efficiency was showed by a fungus species, Fusarium sp. and was followed by Bacillus SP. I, Spicaria sp. pathogens isolated from larvae of pine gall midge did not affected to both of Japanese alder leaf beetles and fall webworms in any means.
