Preparation of Porous Cobalt Thin Films by Using an Electrochemical Method (전기화학적 방법을 통한 다공성 코발트 박막 합성)

  • Morphology of porous cobalt electro-deposits was systematically investigated as functions of cobalt precursors in the plating bath and applied cathodic current density with a special focus on cobalt nano-rod formation. It was proved that the concentration of cobalt precursor plays little effect on the morphology of cobalt electro-deposits at relatively low plating current density while it significantly affects the morphology with increasing plating current density. Such a dependence was discussed in terms of the kinetics of two competitive reactions of cobalt reduction and hydrogen evolution. Cobalt nano-rod structure was created at specific ranges of cobalt precursor content and applied cathodic current density, and its diameter and length varied with plating time without notable formation of side branches which is usually found during dendrite formation. Specifically, the nano-rod length was preferentially increased in relative short plating time (<15 s), resulting in higher aspect ratio of nano-rod with plating time. Whereas, both the nano-rod length and diameter were increased nearly at the same level in a prolonged plating time, making the aspect ratio unchanged. From the analysis of crystal structure, it was confirmed that the cobalt nano-rod preferentially grew in the form of single crystal on a dense poly-crystalline cobalt thin film initially formed on the substrate.

A Simple Method for the Preparation of Highly Pure Conjugated Linoleic Acid(CLA) Synthesized from Safflower Seed Oil

  • A simple and rapid method was developed to prepare a large quantity of highly pure conjugated linoleic acid (CLA) chemically-synthesized from safflower seed oil (SSO). CLA-SSO(74.9% in purity) was synthesized from fresh SSO(79.9% of linoleic acid) by alkaline isomerization at 18$0^{\circ}C$. Urea(50g) and CLA-SSO (25g) were completely dissolved in ethanol (750ml) using a water bath(5$0^{\circ}C$) and followed by refluxing for 60 min. The resultant was cooled to room temperature and stored in a cold room (4$^{\circ}C$) for 24hrs. After removing the urea adduct by filtration, the filtrate was rotoevaporated under 4$0^{\circ}C$ and the residue was dissolved in hexane (200ml). The hexane extract was washed with distilled water (100ml$\times$3) and dried over sodium sulfate anhydrous. This urea treatment procedure was repeated three times. The purity of CLA recovered from the hexane extract was 95.0%. This method can be applied to prepare a large quantity of highly pure chemically-synthesized CLA (>0.5kg/a batch) from any plant oils containing high percentages (>70%) of linoleic acid.

Modulatory Effect of the Tyrosine Kinase and Tyrosine Phosphatase on the ACh-activated $K^{+}$ Channel in Adult Rat Atrial Cells

  • Acetylcholine (ACh) activates the inwardly rectifying muscarinic $K^{+}$ channel in rat atrial cells via pertussis toxin (PTX)-sensitive G-protein ($G_k$) coupled with the muscarinic receptor (mAChR). Although this $K^{+}\;(K_{ACh})$ channel function has reported to be modulated by the phosphorylation process, a kinase and phosphatase involved in these processes are still unclear. Since either PKA or PKC was not effective on this ATP-modulation, the present study examined the possible involvement of the protein tyrosine kinase (PTK) and protein tyrosine phosphatase (PTP) in the function of the $K_{ACh}$ Channel. In the inside-out (I/O) patch preparation excised from the adult rat atrial cell, when activated by 10 ${\mu}M$ ACh in the pipette and 100 ${\mu}M$ GTP in the bath, the mean open time (${\tau}_{o}$) and the channel activity ($K_{ACh}$) was 1.13 ms (n=5) and 0.19 (n=6), respectively. Following the application of 1 mM ATP into the bath, ${\tau}_{o}$ increased by 34% (1.54 ms, n=5) and $K_{ACh}$ by 66% (0.28, n=6). Channel function elevated by ATP was lasted after washout of ATP. However, this ATP-induced increase in the $K_{ACh}$ channel function did not occur in pretreated cells with genistein ($50{\sim}100 {\mu}M$), a selective PTK inhibitor, but occurred in pretreated cells with equimolar daidzein, a negative control of the genistein. On the contrary, PTP which acts on tyrosine residue conversely reversed both ATP-induced increased ${\tau}_{o}$ by 32% (1.20 ms, n=3) and $K_{ACh}$ by 41% (0.15, n=3), respectively. Taken together, these results suggest that $K_{ACh}$ channel may, at least partly, be regulated by the tyrosyl phosphorylation, although it is unclear where this process exerts on the muscarinic signal transduction pathway comprising the mAChR-$G_{k}$-the $K_{ACh}$ channel.

  • Encapsulation of lysozyme using lecithin vesicles and its stimilated release properties were studied. Lecithin vesicles were prepared by the dehydration-rehydration (DR)method. The highest encapsulation efficiency (EE) value of 80.1% was obtained by sonicating the multilamellar vesicles (MLVs) at 100 KHz for 120 min in bath sonicator. The value of entrapment progressively increased with the concentration of lysozyme, while the EE value decreased with the increase of enzyme concentration up to 50mg per 100mg per 100mg of lecithin, and then became nearly constant. At the pH of 5.9, only a small amount of lysozyme was released from DR vesicles during incubation at $37^{\circ}C$ As the pH decreased to 3.0, lysozyme was released more rapidly. Lysozyme release was accelerated for 24h and reached a plateau after 72h incubation with 1% Tween 80, $Ca^{2+}$ gave a pulse-like-release in the first hour, which was followed by a slow release.

  • Effect of the oil mixture on physical properties in soft capsule preparation with ginseng extract contained $36{\pm}1%$moisture content was investigated. The optimum condition was reached when it was emulsified for 5 minutes in $40^{\circ}C$ water bath at 10.000 r.p.m. by mixing with both palm shortening oil and corn oil in equal amount under agitation. Addible amount of ginseng extract was 50% for the total.

  • In this study, lignin/chlorinated poly(vinyl chloride)(CPVC) blended fibers have been produced for the development of low-cost carbon fiber. Carbon fiber manufacturing was accomplished through stabilization and carbonization process. The lignin/CPVC blended fibers were prepared by wet spinning method. Dimethylacetamid e(DMAc) and cychlohexanone in a ratio of 5:1(wt%) was employed as co-solvent. The ratio of lignin/CPVC was prepared at 0/10, 1/9, 2/8, 3/7, 4/6, and 5/5(wt%). The spinning solution was extruded at a rate of 0.1 to 0.4ml/min according to the blending ratio. The speed of the rollers was the same for all ratios(draw ratio=1). Analysis of fiber cross-section by scanning eletron microscopy(SEM) showed that as the lignin ratio increased in the same coagulation bath and distilled water, the pore size of the spinning fiber decreased. Therefore, the highest tensile strength of the blending fibers was 6.3±1.2MPa at the 5/5 ratio. The carbon fiber also showed the best tensile strength of 120.78±2.43MPa at 5/5 ratio.

Improvement Effect of the HyungGae-Water on Quality of Life in Children and Adolescents with Pruritus (한방입욕제(형개수) 사용을 통한 소양증을 호소 소아청소년의 삶의 질 개선효과)

  • Pruritus is the most prominent symptom of atopic dermatitis and other skin diseases. It is not easily improved and is a major factor that threatens the quality of life of patients and their families. The purpose of this study was to evaluate the effect of HyungGae-Water, which was bath preparation made from Korean medicine herbs, for children and adolescents suffering pruritus. The participants were 50 boys and girls aged from 2 to 15 years old. The HyungGae-Water was used once a day for 6 weeks, and we were followed up three times every two weeks. Of 50 participants enrolled, 47 completed the study. The outcome variables were dermatology quality of life scales in children, pruritus VAS, and investigator global assessment (IGA). The results of the study show that the dermatology quality of life is significantly improved and the VAS of pruritus has meaningful changes. In the IGA, moderate or severe improvement was observed in 48.9% and slight improvement in 17%. There were seven suspected or less relevant adverse events using HyungGae-Water. The present results suggest that the HyungGae-Water has alleviating effect for the pruritus in children and adolescents.

Preparation of an Inorganic Scintillator Loaded Film for the Measurement of Surface Contamination and its Performance Test (표면오염 측정용 무기섬광 함침 필름의 제조 및 성능 평가)

  • The smear media possible to sampling and radiation detection was prepared and evaluated for the surface contamination using indirect method. The films were made by impregnating Cerium Activated Yttrium Silicate (CAYS) in a polysulfone membrane. The membranes used solution as a dimethylformamide (DMF) and methylene chloride (MC), polysulfone as a polymer matrix and CAYS as a inorganic scintillator. The proximity membranes were prepared with single- and double-layered structure. The solidified methods were immersion to the nonsolvent bath such at water and ethanol and solvent evaporation. The measurement of the photon produced by interaction with radiation and inorganic scintillator used a photomultiflier tube (PMT), amplifier, and counter. In the comparison with the low background alpha/beta counter, the counter rate using inorganic scintillator proximity membrane for the $\^$14/C surface contamination was about 50%. Also. the $^3$H counting results revealed that the prepared membranes were efficient to monitor the surface contaminated with the low energy be-ray emitter nuclides.

  • The microcapsules containing rosmarinic acid were prepared in this research. Rosmarinic acid is known that it is effective to care the winkles. Chitosan was used as a wall material, and glutaraldehyde was used as a crosslinking agent, and the microcapsules were prepared by the water-in-oil (W/O) emulsion method. In this method Span80 was used as an emulsifier, and mineral oil was used as a medium material. Perfectly spherical microcapsules were obtained in the size range of $0.5{\sim}0.9{\mu}m$. The effects of emulsifier concentration and stirring speed on the average particle size and distribution, and encapsulation efficiency were investigated. The release behavior of the microcapsules with different amount of the crosslinking agent and different emulsifier concentrations were also investigated.

Comparison of Short-Term Toxicity Tests Based on Feeding Behavior and Temperature Control by Ceriodaphnia dubia (Ceriodaphnia dubia의 먹이섭생 기작과 온도조절에 근거한 급성독성조사법의 비교)

  • Two methods, a Ceriodaphnia algal uptake suppression test (CAUST) and a new toxicity test based on temperature control (TTBTC) which are based on feeding behaviour and temperature control, respectively, were developed and compared for the adoption as the better methodology for short-term toxicity screening. As previously published by Lee et aI., (1997), the CAUST method is based on the feeding behaviour of C. dubia and requires as little as 1 hour of contact time between C. dubia neonates and toxicant. However, even though CAUST requires only 1 hour of contact time, this method still take many hours for the preparation and measurement. Before the test starts, neonate digestive tracts were cleared by feeding yeast to the daphnids, Neonates were then exposed to toxicant, followed by addition of Scenedesmus subspiatus into the bioassay vessels. Daphnids were examined under the bright-field microscope with the presence of algae (indicated by a green colored digestive tract) or the absence of algae. Uptake indicated no toxic effect, whereas, absence of uptake indicated toxic inhibition. Unlike CAUST, the newly developed method (TTBTC) is based on just temperature control for the toxicity test of C. dubia. Initially, neonates are exposed to toxicants while the temperature of water bath containing media increased to $35.5^{\circ}C$. After 1.25 hour of contact time, the number of the daphnids, either live (no toxic effect) or dead (toxic effect), is counted without the aid of any instrument. In both methods, median effective concentrations ($EC_{50}$ values) were computed based on the results over a range of dosed toxicant concentrations. It showed that TTBTC was as sensitive as the standard 48-hour acute bioassay and CAUST. TTBTC and CAUST were much more sensitive than the I-hour I.Q. test and 30-minute Microtox. This study indicates that TTBTC is an easier and more rapid toxicity test than the standard 48-hour acute bioassay and even CAUST.
