생물전기화학시스템을 이용한 염화에틸렌의 생물학적 탈염소화 (Biological Dechlorination of Chlorinated Ethylenes by Using Bioelectrochemical System)

  • 산업용제로 널리 이용되고 있는 PCE (Perchloroethylene)나 TCE (Trichloroethylene)와 같은 염화에틸렌화합물은 안정된 세정력을 가지고 있어 널리 이용되고 있지만 무분별한 사용과 부주의한 취급으로 인해 최근 토양 및 지하수 오염지역이 늘어나고 있다. 본 연구에서는 퇴적토, 슬러지, 토양, 지하수 등 다양한 지역에서 총 10개의 시료를 식종원으로 이용하여 생물학적 PCE 탈염소화 가능성을 평가하고, 가장 우수한 탈염소화 능력을 보인 낙동강 퇴적토 시료를 대상으로 PCE를 에틸렌까지 안정적으로 탈염소화 가능한 혼합미생물을 농화배양하였다. 농화배양된 탈염소화 미생물을 생물전기화학시스템(Bioelectrochemical System, BES)의 환원부에 식종하여 전극을 전자공급원으로 이용한 탈염소화 가능성을 평가한 결과, PCE가 TCE, cis-dichloroethylene, vinyl chloride를 거쳐 최종산물인 에틸렌으로 탈염소화됨을 확인할 수 있었다. Polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE)를 이용한 미생물군집 분석결과, 농화배양액에서 구축된 탈염소 미생물 군집과 BES 환원전극부내 미생물 군집 구조는 다르게 나타났으며, 전기화학적 활성을 지닌 다양한 미생물이 존재함을 확인할 수 있었다. BES 환원전극부에서 부유성장하는 미생물과 전극에 생물막을 형성하는 미생물 군집구조에도 큰 차이가 있었으며, 이는 탈염소화 메커니즘의 차이에 기인하는 것으로 판단된다. 추가적인 연구를 통해서 자세한 생물전기화학적 탈염소화 메커니즘을 밝혀낸다면 생물전기화학적 탈염소화 기술은 염화에틸렌 오염 토양/지하수의 획기적인 생물정화기술로 자리잡게 될 것이다.

病院廢水의 特性과 處理에 關한 硏究 -消毒劑가 활성스러지법에 미치는 영향- (Studies on the Wastewater of General Hospital and It에s Treatment -Effects of Disinfectants on Activated Sludge Process-)

  • The hospital wastewaters have to be so disposed as to prevent disease and to protect water resources from hazardous substances disinfectants, medicines, and chemicals. Polyvinylpyrrolidone-iodine complex (povidone-iodine) is widely used in the hospital as one of disinfectants. This study was carried out to manifest the effect of disinfectants in growth of activated sludge in treatment of the hospital wastewater by the activated sludge process. The results are as follow. 1. An average water quality of the hospital wastewater showed 7.2 in pH, 3.2 ppm in DO, 293.3 ppm in SS, 96.0 ppm in BOD, 151.1 ppm in COD, 0.4 ppm in povidone-iodine, 0.5 ppm in phenols, 5.4 ppm in surfactants, 1.6 ppm in o-phosphate, 4.6 ppm in $NH_3-N, 249\times 10^4$ counts/100 ml in coliform group organisms, and $1,369\times 10^2$ counts/ml in general counts of bacteria. And wastewater amounts discharged per bed was calculated 70 l/d/bed. 2. In batch culture activated sludge process, each of cresol and povidone-iodine was not effected in less than 0.1 ppm concentration, but the more concentration, the more inhibit the growth rate of activated sludge. In the mixture of two disinfectants, the growth was more inhibited the effect of single disinfectants. So that this reaction is considered as addition effect of two disinfectants. 3. The removal rates of the disinfectants-by continuous culture activated sludge process were 77.6% in 0.4 ppm povidone-iodine, and in BOD was 85.6%. 4. It is desirable that the hospital wastewater is planed in order to be discharged to two system separately, sewer from life system and wastewater from medical system. From those results, it has been concluded that the hospital wastewater has to be treated safely by the activated sludge process.

Carbon Storage Regulator A (csrA) Gene Regulates Motility and Growth of Bacillus licheniformis in the Presence of Hydrocarbons

  • The global carbon storage regulator (Csr) system is conserved in bacteria and functions as a regulator in the exponential and stationary phases of growth in batch culture. The Csr system plays a role in the central carbon metabolism, virulence, motility, resistance to oxidative stress, and biofilm formation. Although the Csr was extensively studied in Gram negative bacteria, it has been reported only in the control of motility in Bacillus subtilis among Gram positive bacteria. The goal of this study was to explore the role of the csrA gene of Bacillus licheniformis M2-7 on motility and the bacterial ability to use hydrocarbons as carbon source. We deleted the csrA gene of B. licheniformis M2-7 using the plasmid pCsr-L, harboring the spectinomycin cassette obtained from the plasmid pHP45-omega2. Mutants were grown on culture medium supplemented with 2% glucose or 0.1% gasoline and motility was assessed by electron microscopy. We observed that CsrA negatively regulates motility by controlling the expression of the hag gene and the synthesis of flagellin. Notably, we showed the ability of B. licheniformis to use gasoline as a unique carbon source. Our results demonstrated that CsrA is an indispensable regulator for the growth of B. licheniformis M2-7 on gasoline.

Genome-wide Analysis and Control of Microbial Hosts for a High-level Production of Therapeutic Proteins

  • The formation of insoluble aggregation of the recombinant kringle fragment of human apolipoprotein(a), rhLK8, in endoplasmic reticulum was identified as the rate-limiting step in the rhLK8 secretion in Saccharomyces cerevisiae. To analyze the protein secretion pathway, some of yeast genes closely related to protein secretion was rationally selected and their oligomer DNA were arrayed on the chip. The expression profiling of these genes during the induction of rhLK8 in fermentor fed-batch cultures revealed that several foldases including pdi1 gene were up-regulated in the early induction phase, whereas protein transport-related genes were up-regulated in the late induction phase. The coexpression of pdi1 gene increased rhLK8-folding capacity. Hence, the secretion efficiency of rhLK8 in the strain overexpressing pdi1 gene increased by 2-fold comparing in its parental strain. The oligomer DNA chip arrayed with minimum number of the genes selected in this study could be generally applicable to the monitoring system for the heterologous protein secretion and expression in Saccharomyces cerevisiae. With the optimization of fed-batch culture conditions and the alteration of genetic background of host, we obtained extracellular rhLK8 at higher yields than with Pichia pastoris systems, which was a 25-fold increased secretion level of rhLK8 compared to the secretion level at the initiation of this study.

Use of FT-IR to Identify Enhanced Biomass Production and Biochemical Pool Shifts in the Marine Microalgae, Chlorella ovalis, Cultured in Media Composed of Different Ratios of Deep Seawater and Fermented Animal Wastewater

  • Growth rates, photosystem II photosynthesis, and the levels of chlorophyll $\alpha$ and secondary metabolites of Chlorella ovalis were estimated to determine if they were enhanced by the addition of swine urine (BM) or cow compost water (EP) that had been fermented by soil bacteria to deep seawater (DSW) in an attempt to develop media that enabled batch mass culture at lower costs. Growth of C. ovalis in f/2, f/2-EDTA+BM60%, DSW+BM30%, and DSW+EP60% was enhanced and maintained in the log phase of growth for 16 days. The cell densities of C. ovalis in DSW+EP60% ($4.1{\times}10^6$ Cells/ml) were higher than those of f/2 ($2.9{\times}10^6$ Cells/ml), f/2-E+BM60% ($3.7{\times}10^6$ Cells/ml), and DSW+BM30% ($2.7{\times}10^6$ Cells/ml). The growth rate was also more favorable for C. ovalis cultured in DSW+EP60% ($0.15\;day^{-1}$) than that of C. ovalis cultured in the control medium (f/2) ($0.12\;day^{-1}$). Furthermore, the chlorophyll a concentration of C. ovalis cultured in DSW+EP60% (4.56 mg/l) was more than 2-fold greater than that of C. ovalis cultured in f/2 (2.35 mg/l). Moreover, the maximal quantum yields of photo system II at 470 nm (Fv/Fm) were significantly higher in organisms cultured at f/2-E+BM60% (0.53) and DSW+EP60% (0.52) than in the other treatment groups. Finally, Fourier transformation infrared (FT-IR) spectroscopy revealed that C. ovalis grown in DSW+EP60% had more typical peaks and various biochemical pool shifts than those grown in other types of media. Taken together, the results of this study indicate that the use of DSW+EP60% to culture C. ovalis can reduce maintenance expenses and promote higher yields.

Air-lift 및 Jar Fermenter에 의한 Hericium erinaceum 심부배양의 배양특성 및 Scale-up (Cultural Characteristics and Scale-up for Submerged Cultivation of Hericium erinaceum Through Air-lift and Jar Fermenter System)

  • H. erinaceum의 기능성 소재화 연구의 일환으로, 지금까지 체계적으로 연구된 바 없는 H. erinaceum의 액체배양을 시도하고. 이 버섯의 서로 다른 생물반응기하의 액체배양 특성을 규명하고 대량생산을 검토하였다. 5 L 용량의 jar fermenter와 air-lift fermenter에서의 액체배양의 비교실험을 통해 air-lift fermenter의 사용이 종균 배양에 적합한 것으로 나타났으며, 형태 분석을 통해 적정 크기($2{\sim}4mm$)의 pellet 형태의 유지가 대량 생산의 중요인자인 것을 규명하였다. Air-lift fermenter 하의 배양을 종균으로 하고, 적정 크기의 pellet을 유지하는 500 L까지의 jar fermenter를 사용한 회분 배양의 scale-up 실험결과, 30 및 500 L에서의 교반 및 통기속도는 각각 200 rpm, 1 vvm 및 150 rpm, 1 vvm이었다. 이 조건하에서 배양의 전 기간 중 적정 pellet을 유지하였으며, 최대 균사체량은 약 $13{\sim}14g/L$이었다.

Biological Fixation of $CO_2$ by Chlorella sp. HA-1 in a Semi-Continuous and Series Reactor System

  • Characteristics of biological $CO_2$ fixation by Chlorella sp. HA-1 were investigated in a semi-continuous and series reactor system using an internally illuminated photobioreactor to overcome shortcomings of physicochemical technologies such as adsorption and membrane separation. High $CO_2$ fixation rate was achieved in the semi-continuous reactor system, in which the dilution ratios of the culture medium were controlled. The average $CO_2$ fixation rate was maintained almost constantly when the dilution ratio increased by 0.1 increment from the initial value of 0.5. The total removal efficiency of $CO_2$ was enhanced by employing a series reactor system. The average $CO_2$ fixation rate increased until 4.013 g $CO_2\;day^{-1}$ in a series operation of four reactors, compared to 0.986 g $CO_2\;day^{-1}$ in a batch operation mode. The total $CO_2$ fixation rate was proportional to the number of reactors used in the series reactor system. In the series reactor system of semi-continuous operation, a large amount of $CO_2$ was removed continuously for 30 days. These results showed that the present reactor systems are efficient and economically feasible for a biological $CO_2$ fixation.

Aspergillus awamori와 Zymomonas mobilis로 구성된 혼합고정화 배양계의 에탄올 생산에 미치는 Neupectin-L의 영향 (Effect of Neupectin-L on Ethanol Production from Raw Starch Using a Co-Immobilized Aspergillus awamori and Zymomonas mobilis)

  • Aspergillus awamori(A)와 Zymomonas mobilis(Z)로 구성된 혼합고정화 배양계(A-Z 36계)의 특성을 이용하여 생전분으로 부터 직접 에탄올생산을 행할 때 투입되는 에너지를 절약할 목적으로 비가열살균 배양법을 검토하였다. A-Z 36계에 의한 비가열살균 배양은 배지에 함유된 세균 때문에 어려웠다. Sorbic acid, benzoic acid, dehydroacetic acid, p-hydroxybenzoic acid, Vantocil IB, Neupectin-L 등의 여러 가지 살균제 중에서 Vantocil IB와 Neupectin-L은 잡균의 오염없이 대조구와 비슷한 속도로 기질을 분해하였다. 0.1% Neupectin-L은 대조구보다 약간 높은 6.9 g/l(대조구 : 6.4 g/l)의 에탄올을 생산하였다$(Y_{p/s}:0.39)$. 0.1% Neupectin-L을 첨가한 유가배양에서는 2%기질을 기본으로 5회분의 기질을 첨가하여 34 g/l 에탄올을 얻어 2.0 g/l/day의 생산성을 나타내었다.

Construction of a Genetic System for Streptomyces albulus PD-1 and Improving Poly(ε-ʟ-lysine) Production Through Expression of Vitreoscilla Hemoglobin

  • Poly(ε-ʟ-lysine) (ε-PL) is a novel bioactive polymer secreted by filamentous bacteria. Owing to lack of a genetic system for most ε-PL-producing strains, very little research on enhancing ε-PL biosynthesis by genetic manipulation has been reported. In this study, an effective genetic system was established via intergeneric conjugal transfer for Streptomyces albulus PD-1, a famous ε-PL-producing strain. Using the established genetic system, the Vitreoscilla hemoglobin (VHb) gene was integrated into the chromosome of S. albulus PD-1 to alleviate oxygen limitation and to enhance the biosynthesis of ε-PL in submerged fermentation. Ultimately, the production of ε-PL increased from 22.7 g/l to 34.2 g/l after fed-batch culture in a 5 L bioreactor. Determination of the oxygen uptake rate, transcriptional level of ε-PL synthetase gene, and ATP level unveiled that the expression of VHb in S. albulus PD-1 enhanced ε-PL biosynthesis by improving respiration and ATP supply. To the best of our knowledge, this is the first report on enhancing ε-PL production by chromosomal integration of the VHb gene in an ε-PL-producing strain, and it will open a new avenue for ε-PL production.

재조합 효모를 이용한 항혈전 단백질 히루딘 발효 생산공정의 최적화

  • 본 연구에서는 히루딘을 생산할 수 있는 재조합 S. cerevisiae 에서 ‘히루딘 유전자의 copy number 와 히루딘 발현양과의 관계를 규명하였으며 , ${\delta}$ 서열을 이중으로 사용한 히루딘 발현벡터를 제조하여 히루딘 유전자의 효모염색체로의 도입효율을 증가시켰다. 숙주세포인 효모의 GALl 유전자를 파쇄하여 균체에 의한 갈락토스 소모를 방지하여 보다 경제적으로 히루딘을 생산할 수 있는 시스템을 개발하였으며, 재조합 H. polymorpha을 이용한 발효공정에서 히루딘 생산을 위한 최적의 메탄올 농도를 결정하였다.
