Detection and Characterization of a Lytic Pediococcus Bacteriophage from the Fermenting Cucumber Brine

  • Of the twelve lytic bacteriophages recovered from five different fermenting cucumber tanks that were inoculated with Pediococcus sp. LA0281, a lytic phage, ${\phi}ps05$, was characterized in the present study. The plaques were mostly clear and round-shaped on the lawn of starter strain, indicating lytic phage. Overall appearance indicated that it belongs to the Siphoviridae family or Bradley's group B1, with a small isometric head and a flexible noncontractile tail with swollen base plate. The average size was found to be 51.2 nm in head diameter and 11.6 nm wide ${\times}$ 129.6 nm long for the tail. The single-step growth kinetics curve showed that the eclipse and the latent period were 29 min and 34 min, respectively, and an average burst size was calculated to be 12 particles per infective center. The optimum proliferating temperature ($35^{\circ}C$) was slightly lower than that of cell growth ($35\;to\;40^{\circ}C$). The structural proteins revealed by SDS-PAGE consisted of one main protein of 33 kDa and three minor proteins of 85, 58, and 52 kDa. The phage genome was a linear double-stranded DNA without cohesive ends. Based on the single and double digestion patterns obtained by EcoRI, HindIII, and SalI, the physical map was constructed. The overall size of the phage genome was estimated to be 24.1 kb. The present report describes the presence of a lytic phage active against a commercial starter culture Pediococcus sp. LA0281 in cucumber fermentation, and a preliminary study characterizes the phage on bacterial successions in the process of starter-added cucumber fermentation.

  • For the bridges managed by the municipalities, there is a need for a maintenance management system for the municipal governments for the scientific and systematic maintenance standard and the scientificization of the budget acquisition and execution through the quantified data base. The purpose of this study is to develop a system for efficient management of small bridgies maintained by the municipality and use ICT convergence technology such as BIM / GIS technology. The details of the system development include the development of the small bridgies maintenance program for mobile devices, the development of the small bridgies maintenance database, the development of the small bridgies integration maintenance management, and the reliability of the program system developed by checking the construction and operation contents of the small bridgies test-bed. The developed program system is linked to the development of smart name plate and reader module of non - power system.

Analysis of Primary Breakup Characteristics Depending on the Boss and Deflector Dimension of Fire Sprinkler Head using LES-VoF (LES-VoF를 이용한 소방용 스프링클러 헤드의 보스 및 디플렉터 치수에 따른 1차 분열 특성 분석)

  • Fire sprinkler initial spray was analyzed by Large eddy simulation (LES) and Volume of Fluid (VoF) integrated method. The IsoAdvector geometric VoF was used to identify the liquid-gas interface clearly even with the large Courant-Friedrichs-Lewy number. To reduce the computational costs, sector meshes and Adaptive Mesh Refinement up to level 3 were used. Base mesh size was 1 mm, which is roughly equivalent to the initial sprinkler droplet. Top surface radius of boss and deflector size were modified to investigate the effects of sprinkler head design on primary breakup process. When top surface radius of boss was increased, vertical liquid sheet was formed. This phenomenon reduced the sheet breakup radius, sheet thickness and velocity. Due to reduced liquid sheet thickness, a large amount of ligaments was created from the liquid sheet. As a result, there was a dramatic decrease in volume per surface area, indicating an increase in breakup process. Spray pattern viewed in radial direction also changed when top surface radius of boss increased. When top surface radius of boss was increased, a T-shaped pattern was observed while a V-shaped pattern was observed in all other cases. When the deflector size increases, the spray pattern remains V-shaped, even if the top surface radius of boss increased. Further studies on promoting atomization of the water supplied to the lower part of the sprinkler head in the T-shape pattern should be conducted.

Evaluation of Thermal Conductivity of Porous TiO2-SiO2-Base Thermal Insulation (다공성 TiO2-SiO2 복합 단열재의 열전도율 평가)

  • We developed nano-porous $TiO_2-SiO_2$ composites (commercial name : PTI, porous titania insulator) with low thermal conductivity as thermal insulating material as well as function of photocatalyst. The objectives of this paper are, firstly, to evaluate of the thermal conductivity of the PTI powder in the temperature range from -160 to $250^{\circ}C$, secondly to evaluate of thermal conductivities of insulation materials that is applied PTI powder. The structure of the PTI powder that has the pores size of 20-30 nm and the particle diameter of 2-10 nm. The PTI had a high surface area of $400m^2/g$ and a mean pore size of $45{\AA}$, which was fairly uniform. The thermal conductivity was measured by GHP(guarded hot plate) method and HFM(heat flux method). The PTI structure is a three-dimensional network nano-structures composed by a pearl-necklace that involved a precious stone in the center of the necklace. The thermal conductivities of PTI-PX powder by the GHP and HFM were 0.0366 W/m.K, 0.0314 W/m.K at $20^{\circ}C$, respectively. This is similar to values that are proportional to the square of the absolute temperature of the thermal conductivity of static air. The thermal conductivities of insulating sheets coated with PTI powder were similar results with that of the PTI powder.

Fabrication and Characterization of Cu-based Amorphous Coatings by Cold Spray Process (저온 분사를 이용한 Cu계 비정질 코팅층의 제조 및 특성 연구)

  • Cu based amorphous ($Cu_{54}Zr_{22}Ti_{18}Ni_6$) coating was produced by cold spraying as a new fabrication process. The microstructure and macroscopic properties of amorphous coating layer was investigated and compared with those of cold sprayed pure Cu coating. Amorphous powders were prepared by gas atomization and Al 6061 was used as the substrate plate. X-ray diffraction results showed that Cu based amorphous powder could be successfully deposited by cold spraying without any crystallization. The Cu based amorphous coating layer ($300{\sim}400{\mu}m$ thickness) contained 4.87% porosity. The hardness of Cu based amorphous coating represented $412.8H_v$, which was correspond to 68% of the hardness of injection casted bulk amorphous material. The wear resistance of Cu based amorphous coating was found to be three times higher than that of pure Cu coating. The 3-point bending test results showed that the adhesion strength of Cu based amorphous coating layer was higher than that pure Cu coating. It was also observed that hard Cu base amorphous particle could easily deform soft substrate by particle collisions and thus generated strong adhesion between coating and substrate. However, the amorphous coating layer unexpectedly represented lower corrosion resistance than pure Cu coating, which might be resulted from the higher content of porosity in the cold sprayed amorphous coating.

Force Assessment of Thermoformed and Direct-printed Aligners in a Lingual Bodily Movement of a Central Incisor Over Time: A 14-day In Vitro Study

  • Purpose: This study aims to investigate the force delivery profile of thermoformed aligners (TFA) compared with direct-printed aligners (DPA) and to explore the effect of different activation amounts on forces and moments of respective groups. A secondary objective is to observe the amount of stress relaxation that occurs over the 7~14 days when aligners are maintained in a simulated intraoral environment. Materials and Methods: An in vitro setup was created to quantify forces and moments. It consisted of a three dimensional-printed base plate and segmented maxillary teeth, placed in a semi-enclosed chamber to maintain a temperature of 37℃. Ninety clear aligners were divided into nine groups of ten aligners each based on material types (Zendura, ATMOS, TC-85) and activation amounts. Aligners were created with 0.00, 0.25- and 0.50-mm activations for lingual bodily movement of the upper left central incisor and kept on models in the "stressed" position in a 37℃ water bath. Three force components acting on the upper left lateral incisor, upper left central incisor, and upper right central incisor were measured for each time point, beginning from the initial baseline measurement, 8 hours, 16 hours, 24 hours, 2 days, 3 days, 4 days, 5 days, 6 days, 7 days, and lastly, 14 days. Result: TC-85 aligners in every activation group showed less force on teeth than Zendura and ATMOS. Significant force levels from 0.0 mm activation were present and stayed consistent over the course of 14 days. Comparisons made for baseline measurements to 7-days and 14-days showed statistically significant change from the baseline force level. Conclusion: TC-85 aligners demonstrated lower, more consistent forces with fewer side effects. Aligners can generate forces even when no activation is programmed. No major decreases in force levels over time were observed; the intra-oral clinical simulated environment and length of observation time could contribute to this.

DEVELOPMENT OF LIGHTWEIGHT OPTICAL TELESCOPE KIT USING ALUMINUM PROFILE AND ISOGRID STRUCTURE

  • We introduce the Transformable Reflective Telescope (TRT) kit that applies an aluminum profile as a base plate for precise, stable, and lightweight optical system. It has been utilized for optical surface measurements, developing alignment and baffle systems, observing celestial objects, and various educational purposes through Research & Education projects. We upgraded the TRT kit using the aluminum profile and truss and isogrid structures for a high-end optical test device that can be used for prototyping of precision telescopes or satellite optical systems. Thanks to the substantial aluminum profile and lightweight design, mechanical deformation by self-weight is reduced to maximum 67.5 ㎛, which is an acceptable misalignment error compared to its tolerance limits. From the analysis results of non-linear vibration simulations, we have verified that the kit survives in harsh vibration environments. The primary mirror and secondary mirror modules are precisely aligned within 50 ㎛ positioning error using the high accuracy surface finished aluminum profile and optomechanical parts. The cross laser module helps to align the secondary mirror to fine-tune the optical system. The TRT kit with the precision aluminum mirror guarantees high quality optical performance of 5.53 ㎛ Full Width at Half Maximum (FWHM) at the field center.

  • Development of the paved track is required as a low-maintenance of conventional line. The paved tracks are one of the types of the ballast reinforced tracks those are manufactured by adopting the prepacked concrete technique. The main elements of this tracks are large sleeper, low elastic pad, fastener, cement mortar, geotextile and recycled ballast. Low elastic pad is the most effective element of such tracks on the basis of stress-displacement characteristics, dynamic response and fatigue characteristics. The stiffness of the pad determine the stiffness of the track. Consequently, it is more important in case of concrete track structure such as paved track because application of low elastic pad seriously effect the durability and stability of the track. The main objective of this study is to confirm the reduction of train load, which transfer to roadbed through various pad effects. To achieve this task static, numerical analysis and real scale repeated loading test was performed while load reduction effect of low elastic pad was analyzed by using displacement, stress and strain ratio characteristics of the paved track.

EFFECT OF INSTRUMENT COMPLIANCE ON THE POLYMERIZATION SHRINKAGE STRESS MEASUREMENTS OF DENTAL RESIN COMPOSITES (측정장치의 compliance 유무가 복합레진의 중합수축음력의 측정에 미치는 영향)

  • The purpose of this study was to evaluate the effect of instrument compliance on the polymerization shrinkage stress measurements of dental composites. The contraction strain and stress of composites during light curing were measured by a custom made stress-strain analyzer, which consisted of a displacement sensor, a cantilever load cell and a negative feedback mechanism. The instrument can measure the polymerization stress by two modes: with compliance mode in which the instrument compliance is allowed, or without compliance mode in which the instrument compliance is not allowed. A flowable (Filtek Flow: FF) and two universal hybrid (Z100: Z1 and Z250: Z2) composites were studied. A silane treated metal rod with a diameter of 3.0 mm was fixed at free end of the load cell, and other metal rod was fixed on the base plate. Composite of 1.0 mm thickness was placed between the two rods and light cured. The axial shrinkage strain and stress of the composite were recorded for 10 minutes during polymerization. and the tensile modulus of the materials was also determined with the instrument. The statistical analysis was conducted by ANOVA. paired t-test and Tukey's test (${\alpha}<0.05$). There were significant differences between the two measurement modes and among materials. With compliance mode, the contraction stress of FF was the highest: 3.11 (0.13). followed by Z1: 2.91 (0.10) and Z2: 1.94 (0.09) MPa. When the instrument compliance is not allowed, the contraction stress of Z1 was the highest: 17.08 (0.89), followed by FF: 10.11 (0.29) and Z2: 9.46 (1.63) MPa. The tensile modulus for Z1, Z2 and FF was 2.31 (0.18), 2.05 (0.20), 1.41 (0.11) GPa, respectively. With compliance mode. the measured stress correlated with the axial shrinkage strain of composite: while without compliance the elastic modulus of materials played a significant role in the stress measurement.
