유산견(流産犬) 태아(胎兒)로부터 Brucella suis의 분리(分離) (Isolation of Brucella Suis from Aborted Fetus of a Dog)

  • 우리나라 가축중(家畜中) 우(牛) 및 돈(豚)의 brucellosis는 알려져 있으나 견(犬)에서의 brucella 분리(分離)에 관(關)한 보고(報告)는 볼 수 없다. 저자(著者)등은 1971년(年) 8월(月) German shepherd의 유산(流産)한 55일(日) 견태(犬胎)에서 brucella라고 생각(生覺)되는 균(菌)을 분리(分離)하여 그 성상(性狀)을 추구(追究)하여 Brucella suis로 동정(同定)하였다. 이로서 우리나라에도 brucella에 의(依)한 견(犬)의 유산(流産)이 있음을 알수 있다. 유산견(流産犬)의 혈청항체가(血淸抗體價)는 Br. suis에 대(對)하여 320배(倍)였다. 대구시(大邱市) 도축장(屠畜場)에서 얻은 86례(例)의 견(犬)의 혈청(血淸)의 brucella에 대(對)한 항체가(抗體價)를 검사(檢査)하였던바 Br. abortus에는 1례(例)만이 80배양성(倍陽性)이었고, Br. suis에는 3례(例)가 160배(倍), 8례(例)가 80배양성(倍陽性)였다. 분리균(分離菌)에 대(對)한 항체가(抗體價)는 대체(大體)로 Br. suis에 대(對)한 항체가(抗體價)와 평행(平行)하였다.

  • Swine respiratory diseases have induced severe economic lasses in swine industry worldwide. Therefore, several methods have been made and applied to prevent and control the diseases. However, these methods still have a problem and also induce side effects. Recently, the use of egg yolk antibody was introduced to control and prevent the diseases as one of new trials. As a study of using egg yolk antibody, antibody titers against several different antigens of major pathogens in swine respiratory diseases were compared in egg yolk and serum of hens immunized with those antigens. The titers were measured by ELISA using the antigens as coating antigens. The relationship in antibody titers between egg yolk and serum were identified by analysis of variance for linear regression. Almost of antigens used in this study showed the high relationship in antibody titers between egg yolk and serum (r = 0.87 ~ 0.93) even though the relationship in antibody titers against P. multocida A:3 IROMP was slightly low (r = 0.74)(P<0.01). These results indicated that antibody titer in egg yolk could be useful to predict the titer in serum of chicken.

  • Swine respiratory diseases have induced severe economic losses in swine industry worldwide. Several methods have been developed and applied to prevent and control the disease. However, those are still problematic in swine industry. Recently, the use of egg yolk antibodies with several advantages was introduced and applied to control diseases in animal as well as human. As the first step of the use of egg yolk antibodies in the control of the swine respiratory diseases, we investigated the immunogens of the causative agensts of the diseases and immune response in egg yolk of hens immunized with them. Bacterial antigens prepared from Bordetella bronchiseptica, Pasteurella multocida 3A and 4D, and Actinobacillus pleuropneumaniae serotype 2 and 5 were analyzed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis, Western blot and toxicity test in mice. The antigens were injected into laying hens in order to produce antibodies against them in egg yolk. After chickens were immunized three times in 2 weeks interval, the profile of antibody production was examined by ELISA. The production of antibody in egg yolk was started in 2 weeks after the first injection, reached peak in 6-8 weeks and maintained until 12 weeks. Of two adjuvants used in this study, ISA70 was more effective than aluminum hydroxide gel in enhancing immunogenecity, laying rates and safety in hens. These results suggested that egg yolk antibodies could be a good source for production of antibodies specific to pathogenic bacteria inducing respiratory diseases of swine.

Prevalence state of canine brucellosis in South Korea during 2015 and 2016

  • We investigated the prevalence of canine brucellosis in companion and stray dogs between March 2015 and December 2016 and determined the disease characteristics based on the geographic distribution, sex and age of the dogs in South Korea. We conducted a large-scale survey using serological and bacteriological tests. Samples were collected from 2,394 dogs (1,825 companions and 569 strays). Thirty (1.3%) samples were positive for Brucella canis antibodies including 16 (0.9%) from companion dogs and 14 (2.5%) from stray dogs. Two (1.0%) of the 196 samples cultured from the stray dogs were positive. When compared with male dogs, the female companion and stray dogs had a significantly higher prevalence of brucellosis. Moreover, the prevalence of canine brucellosis was significantly higher in stray dogs older than 6 years and the prevalence of the disease in companion dogs was highest in Incheon (2.1%) and Jeolla (2.1%) provinces. Stray dogs from the Daejeon metropolitan area had the highest prevalence of brucellosis (7.9%). National control measures for canine brucellosis have not previously been implemented. Our findings suggest that appropriate screening tests and control measures are necessary to improve the health of dogs and to protect public health in Korea, particularly with the rapid growth of the companion animal industry.

  • Biopulping is defined as the treatment of wood chips with lignin-degrading fungi prior to pulping. Fungal pretreatment prior to mechanical pulping reduces electrical energy requirements during refining or increases mill throughput, improves paper strength, reduces the pitch content, and reduces the environmental impact of pulping. Our recent work involved scaling up the biopulping process towards the industrial level, investigating both the engineering and economic feasibility. We envision the process to be done in either a chip-pile or silo-based system for which several factors need to be considered: the degree of decontamination, a hospitable environment for the fungus, and the overall process economics. Currently, treatment of the chips with low-pressure steam is sufficient for decontamination and a simple, forced ventilation system maintains the proper temperature, humidity, and moisture conditions, thus promoting uniform growth of the fungus. The pilot-scale trial resulted in the successful treatment of 4 tons of wood chips (dry weight basis) with results comparable to those on a laboratory. Larger, 40-ton trials were also successful, with energy savings and paper properties comparable with the laboratory scale. The overall economics of the process also look very favorable and can result in significant annual savings to the mill. Although the current research has focused on biopulping for mechanical pulping, it is also beneficial for sulfite chemical pulping and some applications to recycled fiber have been investigated.

Purification and Characterization of Cell Wall Hydrolase from Alkalophilic Bacillus mutanolyticus YU5215

  • Streptococcus mutans has the capacity of inducing dental caries. Thus, to develop a novel way of preventing dental caries, a cell wall hydrolase-producing strain was isolated and its characteristics were investigated. Among 200 alkalophilic strains isolated from soil, 8 strains exhibited lytic activities against Streptococcus mutans. However, strain YU5215 with the highest cell wall hydrolase activity was selected for further study. Strain YU5215 was identified as a novel strain of Bacillus based on analyzing its 16S rDNA sequence and Bergey's Manual of Systematic Bacteriology, and thus designated as Bacillus mutanolyticus YU5215. The optimal conditions for the production of the cell wall hydrolase from Bacillus mutanolyticus YU5215 consisted of glucose ($0.8\%$), yeast extract ($1.2\%$), polypeptone ($0.5\%$), $K_{2}HPO_{4}\;(0.1\%$), $MgSO_{4}{\cdot}7H_{2}O$ ($0.02\%$), and $Na_{2}CO_{3}\;(1.0\%$) at pH 10.0. Bacillus mutanolyticus YU5215 was cultured at 30^{circ}C for 72 h to produce the cell wall hydrolase, which was then purified by acetone precipitation and CM-agarose column chromatography. The molecular weight of the lytic enzyme was determined as 22,700 Da by SDS-PAGE. When the cell wall peptidoglycan of Streptococcus mutans was digested with the lytic enzyme, no increase in the reducing sugars was observed, while the free amino acids increased, indicating that the lytic enzyme had an endopeptidase-like property. The amino terminus of the cell wall peptidoglycan digested by the lytic enzyme was determined as a glutamic acid, while the lytic site of the lytic enzyme in the Streptococcus mutans peptidoglycan was identified as the peptide linkage of L-Ala and D-Glu.

Effect of Citrus Pectin Oligosaccharide Prepared by Irradiation on High Cholesterol Diet B6.KOR-ApoE Mice

  • Effect of citrus pectin oligosaccharides produced by irradiation was studied on the ability to improve lipid metabolism and hypercholesterolemia in mice fed high cholesterol diets. A total of 35 mice were divided into 5 groups and fed the following diets for 6 weeks: normal diet (C), 0.5% cholesterol (CH), 0.5% cholesterol+5% non-irradiated pectin (P), 0.5% cholesterol+5% irradiated pectin at 20 kGy (PIR), and 0.5% cholesterol+5% irradiated at 20 kGy and dialyzed (PIR-F). CH group had significantly higher serum triglycerides, total cholesterol, and low density lipoprotein (LDL)-cholesterol contents than pectin oligosaccharide-treated groups (p<0.05). Triglycerides and total cholesterol contents was the lowest in C and PIR-F and followed by PIR and P group, and CH group had significantly higher LDL-cholesterol. Serum high density lipoprotein (HDL)-cholesterol content in C group was not different from that in CH and P groups, but lower than that of PIR and PIR-F groups. These results suggest that pectin oligosaccharides produced by irradiation can reduce the levels of serum triglyceride, total cholesterol, and LDL-cholesterol in the blood of mice fed high-cholesterol diets and therefore, irradiation can be used as a tool to produce functional oligosaccharides from citrus pectin.

김치로부터 분리한 젖산균의 다양성과 항균성 (Diversity and Antibacterial Activity of Lactic Acid Bacteria Isolated from Kimchi)

  • 본 연구는 김치로부터 분리한 젖산균의 다양성과 항균성을 조사하기 위하여 수행되었다. 희석한 김치 국물을 CaCO$_3$가 첨가된 MRS 고체 배지에 도말 한 후, $25^{\circ}C$에서 2일 동안 배양하여 여러 가지 김치로부터 27가지의 젖산균을 분리하였다. 이렇게 분리한 젖산균들을 Bergey 분류법과 BPB 배지를 통한 생화학적인 방법과 16S rDNA PCR을 이용한 분자 생물학적인 방법으로써 분석하였다. 그러나 이들 방법으로 분석한 결과가 일치하지 않은 경우가 많았다. 이들 분리 젖산균은 16S rDNA 부분 염기서열을 결정한 결과 Leuconostoc mesenteriodes(11균주), Leu. carnosum(3균주), Lactobacillus curvatus(8균주), Lac. pentosus(2균주), Weisselia kimchi(1균주), W. cibaria(1균주), Pediococcus pentosaceus(1균주) 등의 것과 각각 매우 유사한 것으로 분석되었다. 이들중 Leu. carnosum은 이전에 김치에서 분리되었다고 보고된 바가 없다. 그리고 이들 김치에서 분리 동정한 균들의 항균작용 활성을 검사 해 본 결과 균주에 따라서 활성의 정도는 다르게 나타났으나 Bacillus subtilis, Escherichia coli, Salmonella enteritidis, S. paratyphica, S. typhi, Staphylococcus aureus, Shigella boydii, S. sonnei 등에 대해 뚜렷한 항균효과를 나타내었다.

호알칼리성 섬유소분해세균 Pseudomonas sp.의 분리 및 특성 (Isolation and Characterization of an Alkalophilic Cellulolytic Bacterium Pseudomonas sp.)

  • 알칼리성 cellulase를 분비하는 세균을 토양으로 부터 분리하고 그 중 알칼리 환경하에서 생육이 양호하며 CMCase 생성능이 우수한 균주 AC-711를 선발하였다. AC-711 균주의 형태적, 배양적 및 생리학적 특성을 조사한 결과 호알칼리성 Pseudomonas속의 균주로 동정되었으나 염색체 DNA의 G+C함량이 54.43 mol%로서 Pseudomonas속보다 다소 낮은 값이었으며, 세포벽 지방산의 주성분은 15:0 anteiso 및 17:0 anteiso이었다. AC-711 균주의 생육은 $25{\sim}37^{\circ}C$, pH 9.5~10.5에서 좋았으며 효소생산을 위한 최적 조건은 $30^{\circ}C$, pH 10.3 이었다. 또한 효소생산을 위한 배지조성은 $KH_2PO_4$ 0.1%, $CoCl_2$ 0.02%, Tween 80 0.02%, $Na_2CO_3$ 0.5%에 탄소원으로서 CMC 1%, 질소원으로서 yeast extract 0.8%를 사용하였을 때가 가장 좋았다. CMCase의 생산은 3일 배양후 가장 높았으며, 생성된 효소는 filter paer와 avicel과 같은 결정성 섬유소보다는 수용성 섬유소인 CMC에 대한 분해 활성이 현저히 높아서 세제 첨가제로서의 이용 가능성이 주목되었다.

Bacillus megaterium SFO41에 의한 Cholesterol Oxidase의 생산 및 최적 배양 조건 (Study on the Production and the Culture Condition of Cholesterol Oxidase from Bacillus megterium SFO41)

  • 우리나라 전통 발효식품(침채류 및 젓갈류)으로부터 cholesterol oxidase 생산성이 있는 균주를 분리하고 이들 분리된 균주들로부터 여러 단계의 균주 선멸시험을 통하여 cholesterol oxidase 생산성이 우수한 미생물을 선별하여 그 특성을 조사하였다. Cholesterol oxidase의 생산성이 가장 우수한 SF041의 형태학적, 배양학적 및 생리학적 특성을 조사하여 Bergey's Mannual of Systematic Bacteriology의 분류기준에 따라 동정한 결과 Bacillus megaterium 또는 그 유연균으로 동정되어 분리균을 Bacillus megaterium SFO41로 명명하였다. Cholesterol oxidase의 생산 조건을 검토한 결과 최적 배지 조성은 2.0% glucose, 0.5% yeast extact, 0.03% $MgSO_4\;7H_2O,\;0.02%\;K_2HPO_4,\;0.2%\;NH_4NO_3$, 0.2% cholesterol로 판명되었으며, 배영 조건은 $30^{\circ}C$, 초기 pH 7.0, 진탕 속도는 150 rpm에서 24시간 배양 시 효소 생성이 가장 우수하였다(2.37 U).
