Carbondioxide($CO_2$) laser is one of the most widely employed lasers in oral soft tissue surgery because of its excellent affinity for water based soft tissues. It has some inherent advantages such as hemostasis, less postoperative swelling, reduction of bacterial population at surgical site, less need for suturing, less scarring, and less postoperative pain compared to conventional surgical therapies including the use of scalpel, diathermy, cryotherapy and electrosurgery. A 30-years-old male was presented with gingival swelling. Clinical examination revealed a well?defined pedunculated fibrotic mass on the buccal gingiva near right maxillary 1st molar. In radiographic examination, no remarkable abnormality was seen. Excisional biopsy was performed with $CO_2$ laser (continuous wave mode, 4.0W). Histological diagnosis was "Irritation fibroma". $CO_2$ laser has advantages those are suitable for surgical treatment of intraoral lesion. If appropriate training and experience are provided, the dentist would be able to manage intraoral lesions more efficiently and successfully with the use of the $CO_2$ laser.
Molecular methods were employed to investigate microorganisms in a thermophilic continuous stirred tank reactor(CSTR) used for continuous $H_2$ production. The reactor was inoculated with heat-treated anaerobic sludge and fed with a glucose-based medium. Denaturing gradient gel electrophoresis showed dynamic changes of bacterial populations in the reactor during 43 days of operation. Gas composition was constant from approximately 14 days but population shift still occurred. Populations affiliated with Fervidobactrium gondwanens and Thermoanaerobacterium thermosaccharolyticum were dominant on 21 and 41 days, respectively. Keeping pH of the medium at 5.0 could suppress methanogenic activity that was detected during initial operation period. $CH_4$ and mcrA detected in the samples obtained from the reactor or inoculum suggested the heat treatment condition employed in this study is not enough to remove methanogens in the inoculum. PCR using primer sets specific to 4 main orders of methanogens suggested that major $H_2$-consuming methanogens in the CSTR belong to the order Methanobacteriales.
The bloom of the genus Stephanodiscus was gradually extinguished after 18 April. Counts of bacterial population were increased as the diatom bloom was disappeared. Numbers of the heterotrophic nanoflagellates and ciliates were also increased during the disappearance of the bloom. The densities of the mesozooplankton, the major predator of the diatoms, started to increase in April. However, their growth was suppressed during the bloom period of the diatoms (from January to March). During the bloom period of the diatoms, the monthly average value of the basic productivity amounted up to 11,765.7 mgC $m^{-2}day^{-1}$, which is relatively high value considering the low temperature and light during that period. The growth rate of phytoplankton in March, when the bloom was beginning to be supressed was 0.007. The growth rate of phytoplankton was negative value in April when the decreasing of the algal density was started.
Full-length cDNAs are essential for the correct annotation of genomic sequences and for the functional analysis of genes and their products. To elucidate the functions of a large population of Chinese cabbage (Brassica rapa) genes and to search efficiently for agriculturally useful genes, we have been taking advantage of the full-length cDNA Over-eXpresser (FOX) gene hunting system. With oligo dT column it purify the each mRNA from the flower organs, leaf and stem tissue. And about 120,000 cDNAs from the library were transformed into $\lambda$-pFLCIII-F vector. Of which 115,000 cDNAs from the library were transformed into T-DNA binary vector, pBigs for transformation study. We used normalized full-length cDNA and introduced each cDNA into Arabidopsis by in planta transformation. Full-length Chinese cabbage cDNAs were expressed independently under the CaMV 35S promoter in Arabidopsis. Selfed seeds were harvested from transgenic Arabidopsis. We had selected 2,500 transgenic plants by hygromycin antibiotic tolerant test, and obtained a number of transgenic mutants. Each transgenic Arabidopsis was investigated in morphological changes, fertility and leaf colour. As a result, 285 possible morphological mutants were identified. Introduced cDNA was isolated by PCR amplification of the genomic DNA from the transgenic mutants. Sequencing result and BLAST analysis showed that most of the introduced cDNA were complete cDNAs and functional genes. Also, we examined the effect of Bromelain on enhancing resistance to soft rot in transgenic Chinese cabbage 'Osome'. The bromelain gene identified from FOX hunting system was transformed into Chinese cabbage using Agrobacterium methods. Transformants were screened by PCR, then RT-PCR and real time PCR were performed to analyze gene expression of cysteine protease in the T1 and T2 generations. The anti-bacterial activity of bromelain was tested in Chinese cabbages infected with soft rot bacteria. The results showed that the over-expressed bromelain gene from pineapple conferred enhanced resistance to soft rot in Chinese cabbage.
Postharvest decay of onion bulbs was examined by inspecting the commercial packages in the market or in storage. Bulb rot incidence was unexpectedly high, and onion bulbs with 1st quality grade were rotten most severely by 51%, followed by 32% for 2nd and 21% for 3rd grades. This indicates that larger bulbs had higher incidences of bulb rots. Major pathogens associated with basal and neck rots were Fusarium oxysporum and Aspergillus sp. or Botrytis allii, respectively, of which basal rot was most prevalent and damaging during storage. Among the epiphytic microorgani는 from onion plants, several Bacillus and Paenibacillus spp. and previously selected Pseudomonas putida and Trichoderma harzianum had inhibitory efficacy against bulb rot pathogens. Among these B. amyloliquefaciens BL-3, Paenibacillus polymyxa BL-4, and P. putida Cha 94 were highly inhibitory to conidial germination of F. oxysporum and B. allii. P. putida Cha 94, B. amyloliquefaciens BL-3, P. polymyxa BL-4, and T. harzianum TM were applied in the rhizoplane of onion at transplanting. Initially antagonist populations decreased rapidly during the first one month. However, among these antagonists, rhizoplane population densities of BL-3, Cha 94, and TM were consistently high thereafter, maintaining about 10$^4$-10$^{5}$ cells or spores per gram of onion root up to harvest time. The other bacterial antagonist BL-4 survived only for two months. TM was the most effective biocontrol agent against basal rot, with the number of rotten bulbs recorded at 4%, while that of the control was 16%. Cha 94 was effective for the first 20 days, but basal rot increased thereafter and had about the same control efficacy as that of BL-3 and BL-4. When the antagonists were applied to the topping areas of onion bulbs at harvest, TM was the most effective in protecting the stored onion bulbs from neck rotting. The second effective antagonist was BL-3. TM and BL-3 completely suppressed the neck rot in another test, suggesting that biocontrol of postharvest decay of onion using these microorganisms either at the time of transplanting or at harvesting may be promising.

This paper reports the results of the research on the effects of probiotic yogurt on the microbiological quality, pH, and sensorial characteristics of fresh chicken meat when packed with probiotic yogurt. The chicken meat pieces were packed with yogurt and were stored at $10^{\circ}C$ for 7 days. Samples were taken after 0, 2, 4, and 7 days of storage, and were analyzed for total bacterial count, E. coli, and coliform, and for the chemical parameters, including the pH. In the control group (packed without yogurt), the Pseudomonas species predominated when the spoilage was obvious after 4-day storage at $10^{\circ}C$. The yogurt-mixed chicken meat package was found to have a significantly lower total viable count and significantly fewer coliform bacteria during storage. Furthermore, the yogurt package showed a growth-inhibiting effect on the Salmonella typhimurium, which were inoculated into the chicken meat pieces for the study. The study findings indicate that probiotic yogurt can be used in packing fresh chicken meat to decrease the population of spoilage bacteria therein and to extend its shelf life.
Soil actinomycetes of 703 strains were isolated from 25 sampling points in Cheju Island using 4 different media. Arginine glycerol salts agar containing soil extract was found to be the best medium for the isolation of soil actinomycetes. Soil samples from pasture land showed higher population and diversity of the actinomycetes than those from citrus field, forest, island, hill or valley. When the antibacterial activity of the 526 isolates was tested against three bacterial strains, Escherichia coli, Staphylococcus aureus and Pseudomonas solanacearum the frequency of the isolates with antibacterial activity varied much depending upon the media used for isolation and cultivation. BL106Ba, one of the 10 isolates that showed antibacterial activity against all the above 3 test strains, was chosen based upon the pH and heat stability of its antibacterial metabolites, and was identified as Streptomyces sp. based upon its cultural, morphological and physiological characteristics. The partially purified white crystalline substance obtained from the culture supematant of BL1063a through cation exchange chromatography(AG MP-50) and three times consecutive gel filtration(Sephadex G-10) showed high antimicrobial activity against gram positive and negative bacteria, but low activity against yeasts. The partially purified substance was found to contain at least four different compounds with antibacterial activity by both thin layer chromatography and high performance liquid chromatography.

The effect of pH on the survival of R. solanacearum and its transmission via roots of tomato in hydroponic culture were studied in laboratory and greenhouse. In laboratory experiment, R. solanacearum could not survive for 24h in nutrient solution with pH of $4{\cdot}0;or\;4{\cdot}5$, while 1, 14, 51 and $62\%$ of inoculum survived at pH $5{\cdot}0,\;5{\cdot}6\;and\;6{\cdot}5$, respectively. When tomato plants were inoculated with R. solanacearum through wounds on the stems, the bacteria moved downward from the inoculation site to the roots and infectious bacteria were released from the roots into the nutrient solution. Of two pH regimes tested in greenhouse nutrient-film technique(NFT) culture, the R. solanacearum population was significantly lower in pH 5.0 than in pH 6.5 in most sampling data. In treatments in which R. solanacearum was introduced by transplanting two root-inoculated plants, significantly move plants developed wilt at pH $6{\cdot}5$(34 out of 48 plants) than at pH 5.0(11 out of 48 plants). In addition, when the bacterium was introduced by transplanting two stem-inoculated plants at pH $6{\cdot}5$, seven out of 24 plants developed wilt.

Ecological characteristics of microbial populations inhabiting heavy metal polluted soil were investigated. The samples were collected from 293 sites around an factory and industry at Gyeoungsangbuk-do. We measured the contents of seven heavy metal elements (Cd, Cu, As, Hg, Pb, $Cr^{6+}$, CN), seven sites have been seriously contaminated by mercury and chrome. A quantitative evaluation of microbial populations in mercury and chrome contaminated soil was examined by using plate count method. Bacterial numbers in polluted soil samples ranged from $7.4X10^5\;to\;9.3X10^7\;cfu\;g^{-1}$, about $10\sim100$ fold less than the count for the unpolluted soil. Moulds were not detected in chrome polluted soil. The log values of actinomycetes of each contaminated soil samples were log ranged from 6.18 to 7.52. The ratio of actinomycetes was similar to unpolluted soil. The investigation showed actinomycetes to be the major microbial population inhabiting the mercury and chrome polluted soil. Thirty-one isolates among the total isolates were examined for antibacterial activity. These isolates were identified based on a phylogenetic analysis using 16S rRNA gene nucleotide sequences, they were categorized in three major phylogenetic groups, belong to the Streptomyces (6 strains), Saccharopolyspora (3 strains), Nocardiodes (1 strain). On the phylogenetic tree, the clade consisting of five isolates were distantly related to all of the established Streptomycetes genera, indicating the possibility as members of new species.

This study aimed to understand the changes in microbial community after algicide dosing to control the fish-killing dinoflagellate Cochlodinium polykrikoides in 10L microcosm. Based on our microcosm experiments, the algicidal activity for C. polykrikoides of yellow clay at the concentrations of 4g and 10g per 10 L was < 20%. At $0.8{\mu}M$ concentration of thiazolidinedione(TD49), the population of C. polykrikoides was controlled to be > 85%. In microbial community, a significant increase in heterotrophic bacterial (HB) abundance was observed at day 1 in the TD49 and yellow clay treatments including control. The HB remained high for 2 days and then gradually decreased. In contrast, the abundance of heterotrophic nanoflagellates (HNFs) increased significantly on days 3 and 5 in the TD49 treatments, indicating that the decline in HB was probably a result of predation by the high density of HNFs. In addition, fluctuations in the aloricate ciliate Uronema sp., which feed on bacteria, was clearly correlated with fluctuations in HB abundance, with a lag period of 1-3 days. Therefore, the short-term responses of the HNF and Uronema sp. may have been a result of the rapidly increasing of HB abundance, which is related to degradation of the dense C. polykrikoides bloom, particularly in the TD49 treatment. In addition, large aloricate ciliate Euplotes sp. was significantly increased after reproduction of HNFs and Uronema sp. Consequently, the algicide TD49 had positive effect on the microbial communities, which indicates that the microbial loop was temporarily enhanced in the microcosm by energy flow from HB through HNFs to ciliate.
