길항미생물 Bacillus lentimorbus G-74의 배양을 위한 여뀌즙액 배지개발

  • 본 연구는 주변에서 흔히 구할 수 있는 여뀌를 이용하며 유용한 미생물을 대량생산 할 수 있는 값싼 배지를 개발하는데 있다. 길항미생물 B. lentimorbus G-74를 이 배지를 이용하여 최적화한 결과, 30% 여뀌즙액 배지. $35^{\circ}C$. pH 5.0의 조건에서 가장 높은 균체성장을 보였다.

Physicochemical and Sensory Properties of Appenzeller Cheese Supplemented with Shrimp Powder

  • The effects of adding shrimp (Periclimenes imperator) powder to Appenzeller cheese on quality and characteristics during ripening were investigated. Cheese samples were prepared containing 1.0%, 2.0%, and 3.0% shrimp powder. Changes in the lactic acid bacterial populations, pH, water-soluble nitrogen concentrations, consumer acceptability, colour and texture were monitored during ripening. The addition of shrimp powder did not affect the appearance or consumer sensory characteristics of the cheeses. Likewise, cheese cohesiveness, fracturability, and springiness were not significantly altered. It was concluded that the quality of the Appenzeller cheese was not affected by adding shrimp powder.

낙엽 세탈액에 따른 토양 미생물의 생장 (Growth of Soil Microorganisms for the Leachates from Leaf Litter)

  • The action and growth of soil microorganisms were studied in accordance with the leachates from leaf litter over the time lapse, and soil properties and species compositions in studied area were investigated. The investigations of soil microorganisms were made through dividing into two groupsbacterial and fungal groups. The abundance of soil microorganisms showed high correlation with organic matter(0.98) and total nitrogen(0.97) of soil. The amount of soil microorganisms was found the highest in litter layer and the next were humus and A layer in order. The bacterial growth rate in leachates from leaf letter was increased continuously by two weeks. The fungal growth rate was increased only for one week and after then it was decreased abruptly.

도시 하수내 질소와 인의 제거균주 분리 및 동정에 관한 연구 (The Isolation and Identification of Valuable Bacteria for Removal of Nitrogen and Phosphate in Municipal Waste Water)

  • Bacterial strains which have excellent removal capacity of phosphate or reduction capacity of were isolated from waste water. Among isolated strains. WR8 and WR1 strains were showed goood efficiency in removal of phosphate and reduction of respectively. When each strain was cultivated in waste water, WR8 strain removed about 85% of phosphate and WR1 strain reduced about 85% of -N. By the result of investigation of morphological and physiological characteristics, WR8 was identified as Aeromonas hydrophila and WR1 as Klebsiella pneumoniae.

Treatment of the fuel oxygenate, MTBE, contaminated ground water using Sequence Batch Bioreactor

  • A mixed bacterial culture capable of mineralizing methyl tort-butyl ether (MTBE), other fuel oxygenates ethers, tertiary carbon alcohols, benzene and toluene was used to inoculate batch reactor and sequence batch reactor (SBR) to treat gasoline contaminated ground water containing about 60 mg/L MTBE, 5 mg/L benzene, 5 mg/L toluene, and low concentrations of several other aromatic and aliphatic hydrocarbons. Respirometery studies showed that MTBE degrading mixed culture could treat MTBE contaminated ground water with addition of nitrogen and phosphate. SBR was operated to demonstrate the feasibility of using suspended growth activated system for the treatment of ground water and to confirm that the respirometry derived kinetics and stoichiometric coefficients were useful for predicting reactor performance. Theoretical performance of the reactor was predicted using mathematical models calibrated with biokinetic parameters derived from respirometry studies.

Photografting of PET Fabrics with Vinyl Pyrrolidone and Acryloyl Morpholine Monomers

  • PET fabrics were photografted under continuous UV irradiation with vinyl pyrrolidone (VP) and acryloylmorpholine(ACMO) as monomers and benzophenone as a hydrogen-abstractable photoinitiator. ACMO can be grafted onto the PET fabrics more efficiently than VP. The grafted PET surfaces were characterized by ATR, ESCA, SEM and zeta potential measurement. ATR and ESCA analysis indicated significant alterations on chemical structure and atomic composition on the surface of the grafted fabrics, where nitrogen content increased with increasing grafting yield. SEM images showed that the fabric surface was covered with the grafted polymers. The zeta potentials and the water wettability of the grafted PET increased with grafting. Also the photografted fabrics showed an increased dyeablity to reactive dyes and increased affinity to various iodine species which imparted anti-bacterial properties.

  • A bacterial strain A-6 producing the high level of an extracellular inulin fructotransfe rase(depolymerizing)(EC 2.4.1.93) which converts inulin into di-D-fructofuranose dianhydride III (DFAIII) was isolated from soil. The isolated strain could be classified as a species belonging to the genus Arthrobacter based on its morphological and physiological characteristics identified in this work. Production of the enzyme was induced by inulin, and the highest activity was detected in the slightly acidic medium supplemented with 2.5% inulin and 0.1% trypton as a sole carbon and a nitrogen source, respectively. Under the optimal conditions, the enzyme activity in the culture supernatant reached approximately 60 uints/ml after 96 hours of cultivation. The optimum pH and temperature for the crude enzyme preparation from Arthrobacter sp. A-6 were pH 5.0 and 60$\circ$C , respectively. The DFA produced by the action of the inulin fructotransferase was confirmed to be DFAIII by paper chromatography, HPLC and $^{13}$C-NMR spectroscopy.

Characteristics of Gouda cheese supplemented with fruit liquors

  • This study was conducted in order to evaluate the quality characteristics of Gouda cheeses supplemented with fruit liquor (Prunusmume or Cornus officinalis). Fruit liquor was supplemented to Gouda cheeses during preparation. Changes in chemical composition, lactic acid bacterial population, pH, water-soluble nitrogen, sensory characteristics, and proteolysis were monitored in the prepared ripened cheese. The electrophoresis patterns of cheese proteins, fruit liquor functional component concentrations, and the flavonoid content of the cheeses were also determined. The addition of fruit liquor did not affect (p> 0.05) the appearance or sensory characteristics of the cheeses. Higher amounts of crude ash, mineral, and flavonoids (p< 0.05) were observed in the liquor supplemented cheese than in the control cheese. Findings from this study suggest that wine supplemented Gouda could provide additional nutrients while maintaining flavor and quality.

Isolation and Characterization of Chitosanase-Producing Microorganism, Aureobacterium sp. YL, from Crab Shells

  • A bacterial strain producing extracellular chitosanase was isolated from crab shells and identified as a member of the genus Aureobacterium The production of chitosanase was proportionally related to the microbial growth, induced by the presence of chitosan, and repressed by glucose at 0.5% (w/v) concentration or higher. The optimal culture conditions for the production of chitosase were 3$0^{\circ}C$ and pH 7.0. Among the nitrogen sources tested, incubation with 0.25% (w/v) concentrations of tryptone and casitione showed the best production of chitosanase. The chitosanase of Aureobacterium sp. YL produced chitobiose as a major product and glucosamine, chitotriose, chitotetraose, and chitopentaose as minor products from chitosan.

  • We isolated cellulose-producing bacteria from persimmon vinegar(Korea traditional fermentation food). Some of these strains were selected for cellulose production in agitation culture. On the other hand, it was also found that strains suitable for static culture production were not necessarily suitable for agitation culture. Therefore we estimated the cellulose production of these isolates in static culture. To determine nutritional requirement for the production of bacterial cellulose, several nutrients as carbon source, nitrogen and mineral salt were tested.
