Effects of Supplementary Composts on Microbial Communities and Rice Productivity in Cold Water Paddy Fields

  • Cold water paddy field soils are relatively unproductive, but can be ameliorated by supplementing with inorganic fertilizer from animal waste-based composts. The yield of two rice cultivars was significantly raised by providing either chicken manure or cow dung-based compost. The application of these composts raised the soil pH as well as both the total nitrogen and ammonium nitrogen content, which improved the soil's fertility and raised its nitrification potential. The composts had a measurable effect on the abundance of nitrogencycling-related soil microbes, as measured by estimating the copy number of various bacterial and archaeal genes using quantitative real-time PCR. The abundance of ammonia oxidizing archaea and bacteria was markedly encouraged by the application of chicken manure-based compost. Supplementation with the composts helped promote the availability of soil nitrogen in the cold water paddy field, thereby improving the soil's productivity and increasing the yield of the rice crop.

Serratia sp. KH-95가 생산하는 Prodigiosin계 적색 색소의 분리 및 특성 (Isolation and Characteristics of Prodigiosin-like Red Pigment Produced by Serratia sp. KH-95.)

  • 토양으로부터 다량의 적색 색소를 생산하는 KH-95 균주를 분리하였다. 형태학적 특성 및 생리학적 특성을 조사한 결과 KH-95는 Serratia sp.로 동정되었다. 본 균주가 생산하는 적색색소를 용매 추출 및 실리카겔 칼럼 크로마토그라피를 이용하여 분리, 정제 후 기기분석을 수행한 결과 prodigiosin의 일종으로 밝혀졌다. Serratia sp. KH-95 균주가 prodigiosin계 적색색소를 생산하기 위한 최적 조건은 배양온도 28$^{\circ}C$, 초기 pH는 7.0였으며 배지 조성을 검토 결과 카제인이 색소의 생산에 가장 적합하였다. 대두유, 미강유와 같은 oil을 제외한 대부분의 탄소원과 이용하기 쉬운 질소원인 효모즙, 육즙, 펩톤 등은 균체의 생장은 촉진 하였으나 색소 생산을 현저하게 저해하였다. 최적화된 배지 및 배양조건에서 대두유를 탄소원으로 첨가할 경우 3,380 mg/l의 많은 적색색소가 생산되었다.

효소분해법에 의한 페이스트형 속성 멸치젓의 제조 및 품질에 관한 연구 (A study on the Rapid Processing of Hydrolyzed Anchovy Paste and Its Quality Stability)

  • A study on the processing method of anchovy hydrolysate paste (AHP) was carried out to improve the sensory quality of salted and fermented fish. Homogenized whole anchovy was hydrolyzed using commercial pretenses, Complex enzyme-2000 (CE, Pacific Chem. Co.) and Alcalase (AL, Novo), in a cylindrical vessel with 4 baffle plates and 6-bladed turbine impeller. Optimal pH, temperature, and enzyme concentration for the hydrolysis with CE and AL were $7.0,\;52^{\circ}C,\;7\%$, and $8.0,\;60^{\circ}C,\;6\%$, respectively. The rational amount of water for homogenization, agitation speed, and hydrolyzing time were $100\%\;(w/w)$, 100 rpm, and 210 min, respectively. To make the hydrolysate to paste type, it was effective to mix the additives, such as starch, soybean protein, agar, and carrageenan gum to the hydrolysate 5 min before the end of boiling at $100^{\circ}C$ for 30 min. Minimal NaCl concentration for long-term preservation was $15\%$, and this could be reduced to $12\%$ by adding $5\%$ of KCl. yield of the AHP based on the total nitrogen content was $94.6\~97.0\%,\;and\;86.0\~89.2\%$, of the nitrogen was amino nitrogen. Salinity, pH and histamine content of the AHP prepared with $12\%$ NaCl and $5\%$ KCl were $9.3\~9.9\%,\;6.1\~6.2$, and below 13 mg/100 g, respectively. The AHP was stable at $26{\pm}3^{\circ}C$ for 60 days on bacterial growth, and addition of $0.05\%$ of rosemary (Herbalox) extract was effective to inhibit the lipid oxidation of the AHP during storage.

Characterization of an Extracytoplasmic Chaperone Spy in Protecting Salmonella against Reactive Oxygen/Nitrogen Species

  • Antimicrobial actions of reactive oxygen/nitrogen species (ROS/RNS) derived from products of NADPH oxidase and inducible nitric oxide (NO) synthase in host phagocytes inactivate various bacterial macromolecules. To cope with these cytotoxic radicals, pathogenic bacteria have evolved to conserve systems necessary for detoxifying ROS/RNS and repairing damages caused by their actions. In response to these stresses, bacteria also induce expression of molecular chaperones to aid in ameliorating protein misfolding. In this study, we explored the function of a newly identified chaperone Spy, that is localized exclusively in the periplasm when bacteria exposed to conditions causing spheroplast formation, in the resistance of Salmonella Typhimurium to ROS/RNS. A spy deletion mutant was constructed in S. Typhimurium by a PCR-mediated method of one-step gene inactivation with ${\lambda}$ Red recombinase, and subjected to ROS/RNS stresses. The spy mutant Salmonella showed a modest decrease in growth rate in NO-producing cultures, and no detectable difference of growth rate in $H_2O_2$ containing cultures, compared with that of wild type Salmonella. Quantitative RT-PCR analysis showed that spy mRNA levels were similar regardless of both stresses, but were increased considerably in Salmonella mutants lacking the flavohemoglobin Hmp, which are incapable of NO detoxification, and lacking an alternative sigma factor RpoS, conferring hypersusceptibility to $H_2O_2$. Results demonstrate that Spy expression can be induced under extreme conditions of both stresses, and suggest that the protein may have supportive roles in maintaining proteostasis in the periplasm where various chaperones may act in concert with Spy, thereby protecting bacteria against toxicities of ROS/RNS.

  • 질소는 하수처리과정에서 제거되어야 하는 주요 오염물질 중의 하나이며, 세균 군집을 이용한 고도처리 시스템에서 생물학적 질소제거는 중요한 기술이다. 질산화반응은 생물학적 질소제거 시스템의 첫 단계로 미생물에 의해 진행된다. 암모니아는 암모니아산화세균에 의해 아질산염으로 산화되며, 그 후에 아질산염은 아질산염 산화세균에 의해 질산염으로 산화된다. 실험실 규모의 생물학적 질소제거 시스템인 변형된 eBAF 시스템, Nutrient removal laboratory 시스템과 반추기법을 적용한 rSBR 시스템의 질산화반응조 시료에서 16S rRNA 유전자를 이용한 terminal restriction fragment length polymorphism (T-RFLP) 방법으로 아질산염 산화세균군집을 분석하였다. 제한효소로 형성된 단편의 클러스터분석에서 Nitrobacter 군집은 각각의 폐수처리 시스템에 따라 군집의 차이가 있음이 나타났다. 그러나 Nitrospira 군집의 클러스터분석에서는 액체와 담체의 서식지 환경 차이에 의해 군집이 구분되었다.

어육 수리미를 첨가한 가우다 치즈의 품질특성 (Quality Properties of Gouda Cheese Added with Fish Surimi)

  • 본 연구에서는 어육 수리미를 자연 치즈에 접목하여 새로운 치즈 개발에 목표를 두고 반 경질 숙성 치즈 중에서 비교적 온화한 향미를 가진 가우다 치즈(Gouda Cheese)에 예비실험을 통하여 얻어진 최적 첨가량인 1.0% 어육 수리미를 첨가, 제조한 가우다 치즈의 전반적인 품질변화와 소비자 기호 도를 검토하였다. 본 연구의 치즈 단백질 분해도는 숙성기간이 경과함에 따라 어육 수리미 첨가구가 대조구에 비해 유의적으로 다소 높게 나타났다. 일반성분과 소비자 기호도 및 관능평가 부분에서는 대조구와 첨가구에서 유의적인 차이가 나타나지 않았다. 이상의 결과를 종합하면 어육 수리미의 첨가가 치즈 제조상의 특성이나 품질변화에는 큰 영향을 미치지 않고 치즈의 숙성과정에서 단백질 분해를 촉진하면서 치즈의 맛을 개선하였으므로 어육 수리미 이용, 신규 기능성 치즈 개발이 가능함을 확인하였다.

수리미 아펜젤러 치즈의 품질 특성 (Quality Properties of Appenzeller Cheese Added with Fish Surimi)

  • 본 연구는 한국인이 선호하는 신규 자연 치즈 개발에 목적을 두고 수리미를 이용하여 한국인에게는 비교적 온화한 치즈인 아펜젤러 치즈(Appenzeller cheese)를 제조하되 선행실험을 통해 얻은 적정 첨가량인 1.0%의 수리미 첨가가 이 치즈의 전반적인 품질 변화와 소비자 기호도, 조직도, 색도의 변화에 미치는 영향을 조사하였다. 본 연구의 치즈단백질 분해도는 숙성기간이 경과함에 따라 대조구와 첨가구에서 모두 증가함을 확인할 수 있었고 수리미 첨가 치즈에서 유의적으로 다소 높게 나타났다. 일반성분은 수리미 첨가구가 조지방 함량과 수분 함량이 높게 나타났으나, 유의적 차이는 크지 않았다. 이상의 결과를 종합하면 수리미 1.0%의 첨가가 치즈 제조상의 특성이나 품질변화에는 큰 영향을 미치지 않고 제품상의 유리한 요소들이 많음이 검토되었다. 다만 수리미 첨가로 치즈의 숙성이 촉진되었고 이로 인한 제품에서의 다소의 쓴맛이 나타났으나, 숙성기간을 1-2개월로 단축함으로써 신세대와 웰빙 추구 소비자를 위한 새로운 개념의 어육 수리미 치즈의 제품 개발이 가능할 것으로 기대되었다.

인삼의 환경 및 기주조건과 발병과의 관계 (Diseases of Ginseng: Environmental and host effect on disease outbreak and growth of pathogens.)

  • Effect of environmental factors and host on the growth and outbreak of various ginseng diseases was reviewed Environmental lectors included hydrogen ion concentration, moisture content, temperature, nutrition, and microbiol populations. Age of the ginseng plants in relation to several ginseng disease occurrence was also included in order to formulate the effective control measure for ginseng diseases. Damping-off caused by Rhizoctonia, Pythium, and Phytophthora, greymold by Botrytis, sclerotinia by Scleretinia, and phytophthora blight caused by Phytophthora were usually prevalent during the early growing season of ginseng when temperature is below 20$^{\circ}C$, while anthrac se caused by Colletotrichum, alternaria blight by Alternaria, and bacterial soft rot by Erwinia were so during the latter growing season when temperature is above 25$^{\circ}C$. However, the root rot incited by Fnarium and Cylindrocarpon caused severe damages throughout the growing season. Growth range of the temperature for a pathogen was highly related to the corresponding disease outbreak. Hydrogen ion concentration was highly related to the outbreak of sclerotinia, root rot, and red rot. Most severe outbreak of those diseases where the soil acidity was pH 4.7, pH 6.5- 7.5, and pH6.0-6.5, respectively. Nitrogen content in the soil was also related to outbreak of root rot and red rot. More red rot occurred where NH,-nitrogen is above 30 ppm and more root rot obtained when excessive nitrogen fertilizer applied. Yellow necrosis apparently was related to magnesium especially its ratio with potassium or calcium content in a soil. Fusarium Population showed significant .relations to missing rate of ginseng Plants in a Implanting ginseng field, while that of total bacteria showed similar relations in all ginseng field, However, in six year old ginseng fields, the more the Streptomyces population was, the less the Fusarium obtained. Consequently, less missing rate observed in a field where Streptomyces population was high. Damping-off, root rot, Rhytophthor a blight were mose severe on the nursery and on 2-3 years old ginseng plants, whereas sclerotinia, and grey cod, alteraria blight, anthracnose were severe on 4-6 years old ginseng plants. Root rot caused by Fusarium and Erwinia, however, was also severe regardless of the age of the plants when the roots were injured. Therefore, for the effective control of ginseng root rot most careful control of the disease during the early year should be rendered.

질소 화합물이 담배연기성분 및 안전성에 미치는 영향 (Effect of nitrogen compounds on the chemical composition and biological activity of mainstream smoke)

  • The objective of this study was to investigate the effect of nitrogen compounds such as protein on the chemical composition and toxicity of cigarette mainstream smoke. BSA protein was treated into the tobacco leaf of original 2R4F cigarette at 1~4 % level. The studies were performed which included a bacterial mutagenicity assay and a mammalian cell cytotoxicity assay for total particulate matter(TPM), and glutathione(GSH) consumption assay for gas/vapor phase(GVP) and determination of smoke chemical constitute. Cigarettes treated with protein were observed dose-dependent increase in yield of volatiles, semi-volatiles and aromatic amines compared with control cigarette. However, carbonyl compounds such as acrolein was lower than that of control cigarette when calculated on an equal TPM basis. The cytotoxicity of TPM obtained from the protein-added cigarettes was not different from that of control cigarette. However, the mutagenicity of the TPM from protein-treated cigarettes(1~4 %) was up to 10-27 % higher than that of control. On the other hand, toxicity of GVP from protein-treated cigarette(4 %) was significantly decreased compared with control cigarette. An overall assessment of our data suggests that nitrogen compounds such as protein should be important for the chemical composition and biological activity of cigarette mainstream smoke.

Piedmont토양(土壤)에서의 질소(窒素) 무기화(無機化) 및 질산화작용(窒酸化作用) 특성(特性) (Nitrogen Mineralization and Nitrification of Selected Piedmont Soils in North Carolina)

  • 토양에서 질소변화과정에 관한 보다나은 이해력 증진은 질소비료의 효용성을 증대시키기 위한 필수적 요인이다. 실험실 실험은 대표적인 Piedmont토양에 있어 질소무기화 및 질산화작용을 구명하기 위해 수행하였다. 질소무기화 및 질산화 량은 Enon, Mecklenburg, Chewcla토양의 표토층에서 60 일까지 증가되었으며, 그후로부터 90일까지는 감소하는 경향을 보였다. 그러나 Wehadkee토양에서 질소의 무기화 및 질산화 량은 숙성시간에 따라 차이가 없었다. 위에서 열거한 모든 토양의 토심이 15-30cm의 토층에서 질소무기화 및 질산화 량은 90 일까지 시간이 경과함에 따라 증가하였다. 토양에 있어 질소무기화 및 질산화 량은 토심별로 토양종류에 따라 다르게 나타났으며, 이러한 차이점은 토양통풍 및 질산화 bacteria의 수에 영향을 받은 것으로 사료된다. 전반적으로, 질소의 무기화 및 질산화 량은 subsurface lagers보다는 표토층에서 많은 것으로 나타났다.
