BACTERIOLOGIC FEATURES AND ANTIBIOTIC SUSCEPTIBILITY IN ISOLATES FROM ORAL AND MAXILLOFACIAL INFECTIONS (구강악안면 감염 질환에서 배양된 세균의 양상과 항생제 감수성)

  • Oral and maxillofacial infections are most commonly odontogenic in origin. The present study was implemented for patients with oral and maxillofacial infections in order to determine what differences were present in cultured bacteria, depending upon the different types of infection. For the present study, the epidemiological characteristics, the state of infection, and the results of the pus culture and antibiotic susceptibility tests were analyzed for the 159 cases where pus culture tests were performed. The patients were treated at the Oral and Maxillofacial Surgical Department of Chonnam National University Hospital during an 18-months period from March 2003 to August 2004. Among the total 159 pus culture specimens, bacteria were cultured in 111 cases (69.8%). In the 111 pus culture specimens, Streptococcus species, Neisseria species, and Staphylococcus species were cultured from 69 cases (51.1%), 21 cases (15.6%), and 15 cases (11.1%), respectively and were determined to be bacterial strains the predominant bacteria responsible for oral and maxillofacial infectious diseases. Twenty four cases (15.1%) among the 159 specimens showed mixed infections. The mostly isolated bacteria from each of the space abscess, dentoalveolar abscess, inflammatory cyst, and pericoronitis cases were the Viridans streptococci. There was little relevance between the type of infection and the type of cultured bacteria. Antibiotic susceptibility tests showed a high level of susceptibility to teicoplanin(100%), vancomycin(100%), chloramphenicol(96.4%), ofloxacin(88.3%), imipenem(83.3%), erythromycin(82.5%) and a low susceptibility to cefazolin(40.0%), oxacillin(44.7%), ampicillin(49.4%), penicillin(51.1%). These results indicate that there was no significant difference among the cultured bacteria depending on the type of infections and their susceptibility to cephalosporin and penicillin G was low.

Isolation and Characterization of a Crude oil-Degrading Strain, Nocardia sp. H 17-1 (원유 분해균주 Nocardis sp. Hl7-1의 분리 및 특성)

  • Bacterial strains which degrade crude oil were isolated by liquid culture from oil-spilled soil, and four isolates were selected among them. The strain Hl7-1 was finally selected after testing emulsifying activity and oil conversion rate. The strain Hl7-1 was identified as a Nocardia sp. based on the test for morphological, biochemical and physiological characteristics. It appears to be highly specialized for growth on crude oil in minimal salts medium since it showed preference for oil or degradation products as substrates for growth. It was found that it could grow on at least fifteen different hydrocarbons. The optimum cultural and environmental conditions were seeked. Cell growth and emulsification activity as a function of time were also determined. Crude oil degradation and the reduction of product peak was identified by the analysis of remnant oil by gas chromatography after 3 days of cultivation. Approximately 83% of oil were converted into a form no longer extractable by organic solvents.

Free Living Amoeba-Bacteria Interactions: Analysis of Escherichia coli Interactions with Nonpathogenic or Pathogenic Free Living Amoeba

  • Free-living amoebae ingest several kinds of bacteria. In other words, the bacteria can survive within free-living amoeba. To determine how Escherichia coli K1 isolate causing neonatal encephalitis and non-pathogenic K12 interact with free-living amoebae, e.g., Acanthamoeba castellanii (T1), A. astronyxis (T7), Naegleria fowleri, association, invasion and survival assays were performed. To understand pathogenicity of free-living amoebae, in vitro cytotoxicity assay were performed using murine macrophages. T1 destroyed macrophages about 64% but T7 did very few target cells. On the other hand, N. fowleri which needed other growth conditions rather than Acanthamoeba destroyed more than T1 as shown by lactate dehydrogenase (LDH) release assay. In association assays for E. coli binding to amoebae, the T7 exhibited significantly higher association with E. coli, compared with the T1 isolates (P<0.01). Interestingly, N. fowleri exhibited similar percentages of association as T1. Once E. coli bacteria attach or associate with free-living amoeba, they can penetrate into the amoebae. In invasion assays, the K1 (0.67%) within T1 was observed compared with K12 (0%). E. coli K1 and K12 exhibited high association with N. fowleri and bacterial CFU. To determine the fate of E. coli in long-term survival within free-living amoebae, intracellular survival assays were performed by incubating E. coli with free-living amoebae in PBS for 24 h. Intracellular E. coli K1 within T1 (2.5%) and T7 (1.8%) were recovered and grown, while K12 were not found. N. fowleri was not invaded and here it was not recovered.

ANTIMICROBIAL SUSCEPTIBILITY TEST ON STREPTOCOCCUS VIRIDANS IN CHILDREN'S ORAL CAVITY (소아의 구강내에서 검출된 Streptococcus viridans에 대한 항균제 감수성 연구)

  • A large number of streptococci that do not fit readily into any of the established classification schemes have been relegated to a large heterogeneous group called the Streptococcus viridans, which are members of the normal flora of the mucous membranes of the body, including the oral cavity, the nasopharynx, and genitourinary tract. This group includes S. mitis, S. oralis, S. sanguis, S. salivarius, S. milleri, etc. Surveying on the literature, it has been reported that infective endocarditis, meningitis, rhabdomyolysis, cholangitis, appendicitis caused by Streptococcus viridans, which were the most important pathogen in children with malignant hematologic disease. Various antibiotics has been chosen for treatment or prophylaxis for these infections, but were generally lower antimicrobial susceptibilities because of an abuse of antibiotics and advent of resistant group. Therefore, surveillant culture must be performed to evaluate personal antimicrobial susceptibilities of intraoral microbes for proper antimicrobial choice for dental procedures. This study examined sampling from subgingival plaque of 60 chidren's microbes. The cultured bacterial isolates, Streptococcus viridans were examined 10 antimicrobial drugs with the Kirby-Bauer agar disk diffusion method. The used drugs were Penicillin, Ampicillin, Oxacillin, Cephalothin, Imipenem, Gentamicin, Erythromycin, Vancomycin, Ciprofloxacin, Clindamycin. The results were as follows : 1. Sampling Streptococcus viridans were S. mitis(65%), S. oralis(22%), S. sanguis(5%), S. intermedius(3%), S. salivarius(2%), S acidominimus(2%), Unidentified streptococcus(2%). 2. The antimicrobial susceptibility of total Streptococcus viridans : Oxacillin< Erythromycin< Pencillin=Ciprofloxacin< Cephalothin< Ampicillin< Clindamycin< Gentamicin< Imipenem=Vancomycin. 3. The antimicrobial susceptibility of S. mitis : Oxacillin=Erythromycin< Ciprofloxacin< Cephalothin< Penicillin=Ampicillin< Gentamicin< Clidamycin< Imipenem=Vancomycin. 4. The antimicrobial susceptibility of S. oralis : Oxacillin< Erythromycin< Penicillin=Ciprofloxacin=Clindamycin< Cephalothin=Gentamicin< Ampicillin< Imipenem=Vancomycin. 5. There was no significant difference in the antimicrobial susceptibility among each Streptococcus viridans group.

A Case of Subcutaneous Salmonella Abscess Developed on Gunshot Wound Area In Lung Cancer Patient (폐암환자의 총상부위에 발생한 피하조직 살모넬라 농양 1예)

  • Salmonellosis is one of communicable disease and still occur in sporadic in Korea frequently. They are four main clinical manifestations in salmonellosis. They are gastrocnteritis, typhoidal or septisemia syndrome, focal manifestation and carrier state. Among them, focal manifestation is rarely seen. Saphra, et al. reported that localized salmonella infection is about 5%. Localized salmonella infection frequently occur during salmonella bacteremia, but may also occur with enteric fever or gastroenteritis. Gray, et al reported 3390 cases of minor gunshot wound infection that bacterial isolates from infected wounds consisted of Staphylococcus aureus(90%), Streptococcus sp.(6%), and mixed organisms(4%). Incresed host susceptibility to infection secondary to lowered resistance due to debilitating disease is an important determinant of Salmonella infection. Since salmonella is seldom suspected as a cause of soft tissue infections, there is usually a dangerous delay in the institution of appropriate antimicrobial therapy and isolation procedure. We experienced one case of subcutaneous salmonella abscess developed on gunshot wound area in lung cancer patient, which was confirmed by pus culture.

  • Two isolates of C-7 and S-34, which were identified as Bacillus licheniformis and Enterobacter cloacae, were shown the highest phytase productivities among the 23 and 44 strains isolated from the fermenting corn and soybean meals, respectively. The phytase productivity with B. lichenifrmis was maximized at pH 6.0, 3$0^{\circ}C$ after 5 days of incubation and E. cloacae was maximized at pH 1.0, 35$^{\circ}C$ after 5 days of incubation. The bacterial phytase productivity with each bacterium was significantly increased or decreased by the addition of various concentrations of 6 carbon and 7 nitrogen sources including glucose, sucrose, KNO$_3$, and NH$_4$Cl.

Application of Bacteria Isolated from Dok-do for Improving Compressive Strength and Crack Remediation of Cement-sand Mortar (독도산 탄산칼슘형성세균에 의한 모르타르 균열보수와 압축강도 증진)

  • This study shows an application of microbiologically induced carbonate precipitate for strength improvement and crack remediation of cement-sand mortar. Seven calcium carbonate-forming bacteria (CFB) were isolated from Dok-do and partially identified by DNA sequence analysis of the 16s rRNA gene. Crystal aggregates were apparent around the bacterial colonies grown on an agar medium. These strains showed strain specific $CaCO_3$ precipitation on urea-$CaCl_2$ medium. Among 7 isolates, Arthrobacter nicotinovorans KNUC601, Microbacterium resistens KNUC602, Agrobacterium tumefaciens KNUC603, Exiguobacterium acetylicum KNUC604, and Bacillus thuringiensis KNUC606 showed a repairing of artificial forced cracks in cement-sand mortar. Compressive strength of cement-sand mortar consolidated with Stenotrophomonas maltophilia KNUC605 was increased around 14.07% compared with that of negative control.

  • 2,4,5-trichlorophenoxyacetic acid (2,4,5-T)-degrading bacterial strains were isolated from rice field and field in suburbs of Taejon. Of the total 100 isolates, 19 strains were selected by fast growth on solid minimal media containing 2,4,5-T as a sole of carbon and energy, and they were identified to genus level. 11 strains were identified as Pseudomonas, 4 strains as Acinetobacter, 1 strains were as Alcaliagenes and 3 strains were not identified. Strains MU19 and MU92 which were identified as Pseudomonas were capable of degradation for 4 kinds of chlorinated aromatic hydrocarbons, 2,4-D, 2,4,5-T, MCPA and 3CB. Acinetobacter sp. MU38 showed the highest degradability in liquid minimal media at 48 hours after inoculation, and Pseudomonas spp. MU19. MU57, MU73, and MU92 were able to degrade carbon source at higher rates. As the results Acinetobacter sp. MU38 and Pseudomonas spp. MU19 and MU92 were capable of biodegradation for broad range of halogenated aromatic hydrocarbons, and had higher rates of degradation for 2,4,5-T.

Pathological, immunohistochemical, and bacteriological findings in dogs infected with Brucella canis

  • This study describes pathological, immunohistochemical, and bacteriological findings in adult dogs and fetuses naturally infected with Brucella (B.) canis. A total of 42 dogs including 40 dogs and 2 aborted fetuses were examined. The most common gross lesions in infected dogs were swelling of lymph nodes and spleen. The testes showed marked swelling with multifocal to diffuse reddish discoloration. The most significant histopathological lesions were observed in the placenta. Placental trophoblasts were markedly hypertrophied due to the accumulation of intra-cellular gram-negative bacteria. Lymphocytic inflammation of varying severity was observed in the reproductive organs such as male testis, epididymis, and prostate gland and female uterus. Strong immunolabelling was observed in the cytoplasm of most trophoblasts in the placental tissues using immunohistochemistry. However, immunohistochemical staining did not demonstrate any organisms in other organs of dogs and fetuses. B. canis isolates were most frequently obtained from the whole blood (82.5%) and superficial inguinal lymph node (77.5%) in both sexes. In addition, the isolation rate was higher in male genital organs than in those of females. Hence, management of male dogs is most important because infected dogs can play a role as carriers.

Control of Lactic Acid Bacterial Growth in Kimchi by Aromatic Amino Acids (방향족 아미노산에 의한 김치 유산균 생장의 제어)

  • Effects of amino acids on the lactic acid bacteria in kimchi were studied. 73 different lactic acid bacteria have been isolated during the kimchi fermentation at $15^{\circ}C$. Among these bacteria, dextran formers were occupied by 69.9%, of which Leuconostoc and Lactobacillus were 4.1% and 65.8%, respectively. All isolates didn't grow in a medium added with 500 ppm of tyrosine, whereas such an inhibition was not exhibited in kimchi with the same concentration of tyrosine. In kimchi added with tyrosine the lactic acid bacteria were less diverse than in the natural kimchi but the ratio of dextran formers were similar. As contrasted with natural kimchi, Leuconostoc was rather increased up to 41.4% and Lactobacillus was decreased down to 29.3%. Dominant species in each genus were Leu. mesenteroides and Lac. minor. Thus it is believed that tyrosine had inhibition effect for the growth of most Lactobacillus in kimchi.
