  • A pixel-level readout circuit is studied for 2-dimensional microbolometer focal plane arrays (FPAs). A current mirroring injection (CMI) input circuit with 2-step current-mode bias suppression is proposed for a pixel-level architecture with high responsivity and long integration time. The proposed circuit has been designed using a $0.35-{\mu}m$ 2-poly 4-metal CMOS process for a $320{\times}240$ microbolometer array with a pixel size of $50{\mu}m{\times}50{\mu}m$. The proposed 2-step bias-current suppression has sufficiently low calibration error with wide calibration range, and the calibration range and error can be easily optimized by controlling some design parameters. Due to high responsivity and a long integration time of more than 1 ms, the noise equivalent temperature difference (NETD) of the proposed circuit can be improved to 26 mK, which is much better than that of the conventional circuits, 67 mK.

The Benefit of Supplemental Cough Suppression Therapy for Chronic Refractory Cough (만성 난치성 기침에 대한 기침 억제 요법의 효과)

  • Background and Objectives Cough suppression therapy (CST) is a physiotherapy that can be used for patients with chronic refractory cough (CRC). We aimed to investigate the efficacy of CST for CRC. Materials and Method A prospective randomized controlled trial was conducted in 27 patients with CRC. Participants were randomized to receive either standard mucolytic medications for CRC combined with supplemental CST (CST group) or standard medications alone (control group). CST consists of laryngeal hygiene management, humidification, cough suppression technique, breathing method, and counseling. We assessed the symptoms change at baseline and week 4 with the Leicester Cough Questionnaire (LCQ). Secondary efficacy outcomes included the degree of cough Visual Analog Scale (VAS) score (0 to 100 scale). Results From 2019 to 2021, 14 CST group patients and 13 control group patients were included. The improvement was significantly greater in the CST group than in the control group for cough VAS score (36.67 to 13.33 vs. 74.29 to 16.43, p<0.001). Patients in the CST group had a significant improvement in total (70.14 to 107.71, p=0.005), physical (31.42 to 43.86, p=0.015), psychological (23.57 to 40.14, p=0.003), and social (15.14 to 23.71, p=0.005) LCQ scores. However, there was not a significant pre- to post-treatment LCQ score improvement in control group. Conclusion CST might be an effective supplemental intervention for CRC.

Measurement and Monte Carlo Simulation evaluation of a Compton Continuum Suppression with low level soil Sample (저준위 토양시료를 이용한 콤프턴 연속체 억제의 측정 및 몬테카롤로 시뮬레이션 평가)

  • This study compared PENELOPE with measured values from low energy peak to high energy peak to reduce peak to compton ratio and continuum background spectrum using $^{60}Co$, $^{137}Cs$ and mixed volume source. In addition, the change in backscattering and compton edge efficiency was compared with that of PENELOPE through changes in the vicinity of low energy. The results from the mixed volume source are applied to the soil samples to determine how much the minimum detection limits of the soil samples are reduced in the suppression and unsuppressed mode. The compton suppression of the low energy region of $^{60}CO$ (1,173 keV) was considerable, and the Compton edge RF for the $^{137}Cs$ (661 keV) peak was 2.8. In particular, the $^{60}Co$ source emits coincidence gamma rays of 1,173.2 keV and 1,332.5 keV, so compton inhibition was reduced by approximately 21%. RF of compton edges of 1,173 keV and 1,332 keV emitted from a $^{60}Co$ source was 3.2 and 3.4, and the peak to compton edge ratio was improved to 8: 1. And Compared with Penelope, the uncertainty was well within 2%. In compton unsuppressed mode, MDA values of 661 keV, 1,173 keV and 1,332 keV were 0.535, 0.173 and 0.136 Bq/kg, respectively, but decreased in compton suppressed mode to 0.121, 0.00826 and 0.00728 Bq/kg. Thus, Compton suppressed could reduce the background radioactivity and the radioactivity contained in the detector itself.

EEG can Predict Neurologic Outcome in Children Resuscitated from Cardiac Arrest (심정지 후 회복된 소아 환자에서 뇌파를 통한 신경학적 예후 예측)

  • Purpose: Early prediction of prognosis of children resuscitated from cardiac arrest is a major challenge. We investigated the utility of electroencephalography (EEG) and laboratory studies for predicting of neurologic outcome in children resuscitated from cardiac arrest. Methods: We retrospectively analyzed medical records of patients who were resuscitated from cardiac arrest from 2006 to 2015 at the Gil Medical Center. Patients aged one month to 18 years were included. EEG analysis included background scoring, reactivity and seizure burden. EEG background was classified score 0 (normal/organized), score 1 (slow and disorganized), score 2 (discontinuous or burst suppression), and score 3 (suppressed and featureless). Neurologic outcome was evaluated by Pediatric Cerebral Performance Category (PCPC) at least 6 months after cardiac arrest. Results: Total 26 patients were evaluated. Nine patients showed good neurologic outcome (PCPC 1, 2, 3) and 17 patients showed poor neurologic outcome (PCPC 4, 5, 6). Patients of poor neurologic outcome group showed EEG background score 3 in 88.2%, whereas 44.4% in patients of good neurologic outcome group (P=0.028). Electrographic ictal discharges except non-convulsive status epilepticus were presented in 44.4% of good neurologic outcome group and 5.9% of poor neurologic outcome group (P=0.034). Ammonia and lactate levels were higher and pH levels were lower in poor outcome group than good neurologic outcome group. Conclusion: Suppressed and featureless EEG background is associated with poor neurologic outcome and electrographic seizures are associated with good neurologic outcome.

Detection of Underwater Transient Signals Using Noise Suppression Module of EVRC Speech Codec (EVRC 음성부호화기의 잡음억제단을 이용한 수중 천이신호 검출)

  • In this paper, we propose a simple algorithm for detecting underwater transient signals on the fact that the frequency range of underwater transient signals is similar to audio frequency. For this, we use a preprocessing module of EVRC speech codec that is the standard speech codec of the mobile communications. If a signal is entered into EVRC noise suppression module, we can get some parameters such as the update flag, the energy of each channel, the noise suppressed signal, the energy of input signal, the energy of background noise, and the energy of enhanced signal. Therefore the energy of the enhanced signal that is normalized with the energy of the background noise is compared with the pre-defined detection threshold, and then we can detect the transient signal. And the detection threshold is updated using the previous value in the noisy period. The experimental result shows that the proposed algorithm has $0{\sim}4% error rate in the AWGN or the colored noise environment.

  • Background and Objectives : The aim of this study was to investigate the anti-oxidant, anti-allergic and anti-inflammatory ability of the Taglisodog-eum(SHE) extract on the RAW 264.7 and EL4 cells Materials and Methods : Three types of experiments were implemented for this study: first, the experiment to study the anti-oxidant effect of SHE using Riboflavin; second, in vitro experiment to investigate the inhibition of Th 2 cell differentiation by SHE using EL4 cells (IL-4 mRNA expression); third, the suppression of $NF-{\kappa}B$ activation using RAW 264.7 cells (iNOS and COX-2 mRNA expression). Results : The anti-oxidant ability of SHE were dose-dependantly increased. From in vitro, the LPS-induced iNOS and COX-2 mRNA expression were dose-dependantly decreased in the RAW264.7 cells treated with SHE and the PMA-induced IL-4 mRNA expression were also dose-dependantly decreased in EL4 cells. $NF-{\kappa}B$ activation was suppressed, and iNOS & COX-2 production were inhibited by SHE Conclusion : The results suggest that SHE has dose-dependant anti-oxidant ability, and has anti-allergic and anti-inflammatory effects through the suppression of $NF-{\kappa}B$ activation and the inhibition of Th 2 cell differentiation.

Extracts of Grifola frondosa inhibit the MAPK signaling pathways involved in keratinocyte inflammation and ameliorate atopic dermatitis

  • BACKGROUND/OBJECTIVES: Grifola frondosa, commonly referred to as the maitake mushroom, has been studied extensively to explore its potential health benefits. However, its anti-inflammatory effects in skin disorders have not been sufficiently elucidated. This study aimed to elucidate the anti-inflammatory role of the ethanol extract of G. frondosa in atopic dermatitis (AD) using in vivo and in vitro models. MATERIALS/METHODS: We investigated its impact on skin and spleen inflammatory responses in Dermatophagoides farinae extract (DFE)/1-chloro-2,4 dinitrochlorobenzene (DNCB)-induced AD-like skin lesions in a mouse model. Additionally, we determined the immunosuppressive response and mechanism of G. frondosa by inducing atopic-like immune reactions in keratinocytes through tumor necrosis factor (TNF)-α/interferon (IFN)-γ stimulation. RESULTS: Our study revealed that G. frondosa ameliorates clinical symptoms in an AD-like mouse model. These effects contributed to the suppression of Th1, Th2, Th17, and Th22 immune responses in the skin and spleen, leading to protection against cutaneous inflammation. Furthermore, G. frondosa inhibited the production of antibodies immunoglobulin (Ig)E and IgG2a in the serum of AD mice. Importantly, the inhibitory effect of G. frondosa on inflammatory cytokines in TNF-α/IFN-γ-stimulated AD-like keratinocytes was associated with the suppression of MAPK (Mitogen Activated Protein Kinase) pathway activation. CONCLUSIONS: Collectively, these findings highlight the potential of G. frondosa as a novel therapeutic agent for AD treatment and prevention.

CYP1B1 Activates Wnt/β-Catenin Signaling through Suppression of Herc5-Mediated ISGylation for Protein Degradation on β-Catenin in HeLa Cells

  • Cytochrome P450 1B1 (CYP1B1) acts as a hydroxylase for estrogen and activates potential carcinogens. Moreover, its expression in tumor tissues is much higher than that in normal tissues. Despite this association between CYP1B1 and cancer, the detailed molecular mechanism of CYP1B1 on cancer progression in HeLa cells remains unknown. Previous reports indicated that the mRNA expression level of Herc5, an E3 ligase for ISGylation, is promoted by CYP1B1 suppression using specific small interfering RNA, and that ISGylation may be involved in ubiquitination related to ${\beta}-catenin$ degradation. With this background, we investigated the relationships among CYP1B1, Herc5, and ${\beta}-catenin$. RT-PCR and western blot analyses showed that CYP1B1 overexpression induced and CYP1B1 inhibition reduced, respectively, the expression of $Wnt/{\beta}-catenin$ signaling target genes including ${\beta}-catenin$ and cyclin D1. Moreover, HeLa cells were treated with the CYP1B1 inducer $7,12-dimethylbenz[{\alpha}]anthracene$ (DMBA) or the CYP1B1 specific inhibitor, tetramethoxystilbene (TMS) and consequently DMBA increased and TMS decreased ${\beta}-catenin$ and cyclin D1 expression, respectively. To determine the correlation between CYP1B1 expression and ISGylation, the expression of ISG15, a ubiquitin-like protein, was detected following CYP1B1 regulation, which revealed that CYP1B1 may inhibit ISGylation through suppression of ISG15 expression. In addition, the mRNA and protein expression levels of Herc5 were strongly suppressed by CYP1B1. Finally, an immunoprecipitation assay revealed a direct physical interaction between Herc5 and ${\beta}-catenin$ in HeLa cells. In conclusion, these data suggest that CYP1B1 may activate $Wnt/{\beta}-catenin$ signaling through stabilization of ${\beta}-catenin$ protein from Herc5-mediated ISGylation for proteosomal degradation.

Immunotherapeutic Effects of CTLA4Ig Fusion Protein on Murine EAE and GVHD (마우스 EAE, GVHD 질환에서 CTLA4Ig 융합단백의 면역치료 효과)

  • Background: CTLA4 (CD152), which is expressed on the surface of T cells following activation, has a much higher affinity for B7 molecules comparing to CD28, and is a negative regulator of T cell activation. In contrast to stimulating and agonistic capabilities of monoclonal antibodies specific to CTLA-4, CTLA4Ig fusion protein appears to act as CD28 antagonist and inhibits in vitro and in vivo T cell priming in variety of immunological conditions. We've set out to confirm whether inhibition of the CD28-B7 costimulatory response using a soluble form of human CTLA4Ig fusion protein would lead to persistent inhibition of alloreactive T cell activation. Methods: We have used CHO-$dhfr^-$ cell-line to produce CTLA4Ig fusion protein. After serum free culture of transfected cell line we purified this recombinant molecule by using protein A column. To confirm characterization of fusion protein, we carried out a series of Western blot, SDS-PAGE and silver staining analyses. We have also investigated the efficacy of CTLA4Ig in vitro such as mixed lymphocyte reaction (MLR) & cytotoxic T lymphocyte (CTL) response and in vivo such as experimental autoimmune encephalomyelitis (EAE), graft versus host disease (GVHD) and skin-graft whether this fusion protein could inhibit alloreactive T cell activation and lead to immunosuppression of activated T cell. Results: In vitro assay, CTLA4Ig fusion protein inhibited immune response in T cell-specific manner: 1) Human CTLA4Ig inhibited allogeneic stimulation in murine MLR; 2) CTLA4Ig prevented the specific killing activity of CTL. In vivo assay, human CTLA4Ig revealed the capacities to induce alloantigen-specific hyporesponsiveness in mouse model: 1) GVHD was efficiently blocked by dose-dependent manner; 2) Clinical score of EAE was significantly decreased compared to nomal control; 3) The time of skin-graft rejection was not different between CTLA4Ig treated and control group. Conclusion: Human CTLA4Ig suppress the T cell-mediated immune response and efficiently inhibit the EAE, GVHD in mouse model. The mechanism of T cell suppression by human CTLA4Ig fusion protein may be originated from the suppression of activity of cytotoxic T cell. Human CTLA4Ig could not suppress the rejection in mouse skin-graft, this finding suggests that other mechanism except the suppression of cytotoxic T cell may exist on the suppression of graft rejection.

The Literary Study on The Written Date of and The Background of Sa-Ahm's 5 Element Acupuncture Method (사암침법(舍巖鍼法)의 저작시기(著作時期) 및 형성배경(形成背景)에 관(關)한 연구(硏究))

  • The Sa-Ahm monk, who was believed to have existed in the middle of Cho-Seon Dynasty in Korea, have created an original Sa-Ahm's acupuncture method and written down, "Book of Sa-Ahm's 5 Element Acupuncture Method" using not only laws of creation and suppression in 5 element theory but also its main and other connected meridians according to the daignosis of visceras and bowels. Also the written time and background of its theory have not been known, and after obtaining Ji-San's book on Sa-Ahm's acupuncture method, I studied on them. The results are as follows : 1) The treatment chapter of Book of Sa-Ahm's 5 Element Acupuncture Method was written down by Sa-Ahm himself, and the chapter for Medical Experience was found to be written down by Ji-San. 2) It seemed that Sa-Ahm adopted the main ideas of "Eui Hak Jeong Jon(醫學正傳)" for the section of physiology, pathology and classification of disease symptoms the contents of which covered 40 of 43 parts, and the ideas of "Dong Eui Bo Gam(東醫寶鑑)" for YunGi treatise(運氣篇), and SangHan part(傷寒門) in classification of diease symptoms was intensified by the ideas of "Eui Hak Ip Moon(醫學入門)", and the contents of apoplexy, diseases characterized by acute diarrhea and vomiting, beriberi, diseases of oral cavity were intensified by the ideas of "Chim Ku Kyeong Hearn Bang(鍼灸經驗方)". 3) The written date of the Book of Sa-Ahm's 5 Element Acupuncture Method has been established as 1644 AD(22nd year of In Jo king of Cho-Seon Dynasty) after studying preface, contents and acknowledgement of Heo Im's "Chim Ku Kyeong Hearn Bang(鍼灸經驗方)". 4) The philosophic background of "Sa-Am Acupuncture Method(舍巖鍼法)" seems to be derived from the theories of "I Ching(周易)" and Yin Yang 5 Element Theory(陰陽五行學說), and to be influenced by such medical scholars as Tan Ke(丹溪), Woo Bak(虞搏), and Heo Im(許任).
