잿빛 곰팡이병의 미생물학적 제어 (Biological Control of Blue Mold by Microorganism)

  • 사과 저장중에 발생하는 푸른곰팡이병원균에 대한 길항미생물을 찾기 위하여 자연계로부터 유용미생물을 분리하여 Penicillum expansum에 대한 길항력 검정과 동정하였다. 자연계로부터 얻은 3,000여종의 미생물중에서 푸른곰팡이병원균에 대하여 길항력이 우수한 미생물을 1차적으로 11종 선발하였으며, 이중에서 가장 길항력이 뛰어난 미생물을 치종적으로 KB22를 선발하였다. 길항력이 우수한 KB22의 형태적 성질, 배양적 특성 및 생리 생화학적 성질 등을 조사하여 비교 검토한 결과 Bacillus subtilis와 유사한 균으로 동정되었다. 분리한 길항균 Baillus subtilis KB22는 푸른곰팡이병에 대한 55.9%의 높은 생장억제력을 보였으며, 한천배지에서 Penicillum expansum 접종 후 길항균처리와 열처리한 길항균배양액을 처리하였을 때 각각90%와 15%이상 길항력을 보여주었다.

Bacillus subtilis KL-57로부터 생산되는 생체계면활성제 합성 유전자 클로닝 (Cloning of Biosurfactant-Producing Gene from Bacillus subtilis KL-57)

  • A bacterium KL-57 which exhibited biosurfactant activity was isolated. This bacterium was identified as Bacillus subtilis. The biosurfactant-producing gene of B. subtilis KL-57 was cloned into R subtilis MI113 by using plasmid pTB523. The plasmid DNA from the clone was found to carry a 18 kb PstI insert. The biosurfactant-producing gene was cleaved into 4 fragments by SmaI, 3 fragments by PvulI or EcoRl, 4 fragments by PvulI and EcoRI double digestion, 5 fragments by AccI, and 2 fragments by KpnI, HindIII or BamHI. By subcloning the 18 kb Pstl insert, a 2.3 kb EcoRl fragment conferred the biosurfactant producing activity on B. subtilis cells. The 2.3 kb had one HindIII cleave site. But Two fragments, which corresponds HindIII/EcoRl termini, exhibited no biosurfactant activity.

Monitoring of Horizontal Gene Transfer from Agricultural Microorganisms to Soil Bacteria and Analysis of Microbial Community in Soils

  • To investigate the possibility of horizontal gene transfer between agricultural microorganisms and soil microorganisms in the environment, Bacillus subtilis KB producing iturin and the PGPR recombinant strain Pseudomonas fluorescens MX1 were used as model microorganisms. The soil samples of cucumber or tomato plants cultivated in pots and the greenhouse for a six month period were investigated by PCR, real-time PCR, Southern hybridization, and terminal restriction fragment length polymorphism (T-RFLP) fingerprinting. Our data from Southern blotting and T-RFLP patterns suggest that the model bacteria do not give significant impacts on the other bacteria in the pots and greenhouse during cultivation.
