동결건조커피 제조에 적합한 유산균 균주 선발 (Selection of Lactic Acid Bacteria suitable for Manufacture of Freeze-dried Coffee)

  • 정장작용, 면역체계 조절, 알레르기 저감, 콜레스테롤 경감 등 여러 기능적 효능이 알려진 프로바이오틱스 건강기능 식품이 증가하고 있다. 본 연구에서는 기능성 커피 상품개발을 위하여 커피 배합에 최적인 probiotic 균주를 선발하고자 하였다. 국내외 유통 중인 동결건조커피에서 분리한 균주 1종, 발효유스타터로 사용되는 균주 8종, Bifidobacterium 1종, Bacillus coagulans 1종으로 동결건조 후 생존율, 커피 음용 환경 생존율, 내산성 및 내담즙성을 측정하였다. Bacillus coagulans는 초기균수 $2.4{\times}10^7cfu/g$에서 동결건조 후에도 $2.0{\times}10^7cfu/g$으로 높은 생존율을 나타냈고, 커피의 음용 환경, 내산성, 내담즙성 테스트에서도 대조균인 Lactobacillus casei 균에 비하여 우수한 생존율을 나타냈다. 특히, 내산성 테스트에서는 pH 1, 120분 조건에서 Bacillus coagulans이 대조군에 비하여 3.8배 높은 생존율을 나타냈다. 이에 포자형성을 특징으로 하는 유산생성 Bacillus coagulans 균제제를 기능성 커피 배합 및 상품화에 적합한 probiotic 균주로 판단하였다.

  • Biosorption represents a technological innovation as well as a cost effective excellent remediation technology for cleaning up radionuclides from aqueous environment. In the present study, a bacteria strain FB12 with high adsorption rate of uranium ion was isolated from the vicinity of the nuclear power plant. It was tentatively identified as Bacillus sp.FB12 according to the 16S rDNA sequencing. Efforts were made to further improve the adsorption rate and genetic stability by UV irradiation and UV-LiCl cooperative mutagenesis. The improved strain named Bacillus sp.UV32 obtains excellent genetic stability and a high adsorption rate of 95.9%. The adsorption of uranium U (VI) by Bacillus sp.UV32 from aqueous solution was examined as a function of metal ion concentration, cell concentration, adsorption time, pH, temperature, and the presence of some foreign ions. The adsorption process of U (VI) was found to follow the pseudo-second-order kinetic equation. The adsorption isotherm study indicated that it preferably followed the Langmuir adsorption isotherm. The thermodynamic parameters values calculated clearly indicated that the adsorption process was feasible, spontaneous and endothermic in nature. These properties show that Bacillus sp.UV32 has potential application in the removal of uranium (VI) from the radioactive wastewater.

Molecular Cloning and the Nucleotide Sequence of a Bacillus sp. KK-l $\beta$-Xylosidase Gene

  • A gene coding for ${\beta}$-xylosidase from thermophilic xylanolytic Bacillus sp. KK-1 was cloned into Escherichia coli using plasmid pBR322. Recombinant plasmid DNAs were isloated from E. coli clones which were capable of hydrolyzing 4-methylumbelliferyl-${\beta}$-D xylopyranoside. Restriction analysis showed the DNAs to share a common insert DNA. Xylo-oligosaccharides, including xylotriose, xylotetraose, xylopentaose, and xylobiose were hydrolyzed to form xylose as an end product by cell-free extracts of the E. coli clones, confirming that the cloned gene from strain KK-1 is ${\beta}$-xylosidase gene. The ${\beta}$-xylosidase gene of strain KK-1 designated as xylB was completely sequenced. The xylB gene consisted of an open reading frame of 1,602 nucleotides encoding a polypeptide of 533 amino acid residues, and a TGA stop codon. The 3' flanking region contained one stem-loop structure which may be involved in transcriptional termination. The deduced amino acid sequence of the KK-1 ${\beta}$-xylosidase was highly homologous to the ${\beta}$-xylosidases of Bacillus subtilis and Bacillus pumilus, but it showed no similarity to a thermostable ${\beta}$-xylosidase from Bacillus stearothermophilus.

  • 위치지정 변이기법을 이용하여 Bacillus thuringiensis var. kurstaki NRD 6의 내독소 유전자의 N말단 독성부위를 변이시켜 Bacillus thuringiensis var. kurstaki NRD 6-Stu 1을 육종하였다. 내독소에 대한 염기서열 조사결과, 변이주의 내독소 유전자부위에 Stu 1 인식부위가 생성되었고 특히, 이부위의 177번 염기 A가 C로 치환된 silent mutation이 일어났음을 확인하였다. 변이주의 내독소 유전자의 항원성질은 친주와 차이가 없었고 Choristoneura femiferana-1에 대한 독성도는 0.015~0.030ng으로 친주의 0.01~0.024ng과 유사하였으나 다른 친주계열인 B.t.kurstaki NRD 5의 500~1000ng과 B.t.kurstaki NRD 4(무독성)보다 강하였다.

Characterization of Bacillus luciferensis Strain KJ2C12 from Pepper Root, a Biocontrol Agent of Phytophthora Blight of Pepper

  • In this study, we characterized the bacterial strain KJ2C12 in relation with its biocontrol activity against Phytophthora capsici on pepper, and identified this strain using morphological, physiological, biochemical, fatty acid methyl ester, and 16S rRNA gene sequence analyses. Strain KJ2C12 significantly (P=0.05) reduced both final disease severity and areas under the disease progress curves of 5-week-old pepper plants inoculated with P. capsici compared to buffer-treated controls. As for the production of antibiotics, biofilms, biosurfactant, extracellular enzyme, HCN, and swarming activity, strain KJ2C12 produced an extracellular enzyme with protease activity, but no other productions or swarming activity. However, Escherichia coli produced weak biofilm only. Strain KJ2C12 could colonize pepper roots more effectively in a gnotobiotic system using sterile quartz sand compared to E. coli over 4 weeks after treatments. However, no bacterial populations were detected in 10 mM $MgSO_4$ buffer-treated controls. Strain KJ2C12 produced significantly higher microbial activity than the $MgSO_4$-treated control or E. coli over 4 weeks after treatments. Bacterial strain KJ2C12 was identified as Bacillus luciferensis based on morphological, physiological, and biochemical characteristics as well as FAME and 16S rRNA gene sequence analyses. In addition, these results suggested that B. luciferensis strain KJ2C12 could reduce Phytophthora blight of pepper by protecting infection courts through enhanced effective root colonization with protease production and an increase of soil microbial activity.

Construction of a Recombinant Bacillus velezensis Strain as an Integrated Control Agent Against Plant Diseases and Insect Pests

  • To construct a new recombinant strain of Bacillus velezensis that has antifungal and insecticidal activity via the expression of the insecticidal Bacillus thuringiensis crystal protein, a B. thuringiensis expression vector (pHT1K-1Ac) was generated that contained the B. thuringiensis cry1Ac gene under the control of its endogenous promoter in a minimal E. coli-B. thuringiensis shuttle vector (pHT1K). This vector was introduced into a B. velezensis isolate that showed high antifungal activities against several plant diseases, including rice blast (Magnaporthe grisea), rice sheath blight (Rhizotonia solani), tomato gray mold (Botrytis cinerea), tomato late blight (Phytophthora infestans), and wheat leaf rust (Puccinia recondita), by electroporation. The recombinant B. velezensis strain was confirmed by PCR using cry1Ac-specific primers. Additionally, the recombinant strain produced a protein approximately 130 kDa in size and parasporal inclusion bodies similar to B. thuringiensis. The in vivo antifungal activity assay demonstrated that the activity of the recombinant B. velezensis strain was maintained at the same level as that of wild-type B. velezensis. Furthermore, it exhibited high insecticidal activity against a lepidopteran pest, Plutella xylostella, although its activity was lower than that of a recombinant B. thuringiensis strain, whereas wild-type B. velezensis strain did not show any insecticidal activity. These results suggest that this recombinant B. velezensis strain can be used to control harmful insect pests and fungal diseases simultaneously in one crop.

  • This report was investigate the biological characteristic of $\delta$-endotoxin of product by TH109 strain, one strain TH109 which has toxicity on Cockroach is isolate and identification. Generally the $\delta$-endotoxin of product by 3. thuringiensis strain was easily soluble in acid, alkaline and organic solvents but $\delta$-endotoxin of product by TH109 strain are insoluble in HCI, NaOH Thiol- reagent(25mM Dithiotheritol, 25mM Dithioeryritol, 25mM Nathioglycolate, 0.2M ESCN, 2% v/v $\beta$-mecaptethanol), organic solvents( acetone, $CCI_{4}$, ether, dioxin MeOH chloroforrh xylene ), Protease. Through this study of $\delta$-endotoxin produced by TH109 strain is insoluble in acid, alkaline, organic solvents and pretense etc. In the point of view, it is greater possibility that $\delta$-endotoxin will be transform into toxin by the reducible materials instead of the reaction of protease in the intestine.

Cephalosporin C Acylase 생산균주의 분리 및 특성 (Isolation and Charaterization of Microorganism Producing Cephalosporin C Acylase)

  • Twenty microbial strains producing the acylase were isolated from soil by using Micrococcus luteus ATCC 9341 as an indicator strain, using either D-($\alpha $)-phenylglycine methylester and 7-aminocephalosporanic acid (7-ACA) or glutaric acid dimethylester and 7-ACA as substrates. Among the isolates, only one strain was turned out to be the 7-ACA producer from either cephalosporin C or glutaryl 7-ACA as the substrates by using the overlay of 7-ACA sensitive strain (SS5). 7-ACA produced from cephalosporin C by an isolate (APS20) was detected by high performance liquid chromatography. The isolated strain (APS20) was identified to Bacillus macerans on the basis of cellular fatty acid profile by gas chromatography. Bacillus macerans APS20 had no $\beta $-lacta-mase activity on cephalosporin C, and that is very important for the enzymatic production process of 7-ACA. However, this strain was resistant up to 100 $\mu $g/ml of cephalosporin C.

  • For a biological control on a plant pathogen, Pryicularia Oryzae, and a mosquito, Aedes aegypti, Bacillus thuringiensis strain AF6 which produces parasporal inclusion, delta-endotoxin, was isolated. The B. thuringiensis strain AF6 was produced not only an antifungal substance(AFS) against P. oryzae, but also a mosquitocidal delta-endotoxin. The AFS of the strain AF6 in more stable at pH 4.0 than pH 10.0. At the mode of action, the AFS of the strain AF6 was inhibited hypha growth on potato agar plate(pH 5.0), and degraded cell walls of P. oryzae.

다약제내성균에 대한 항균 활성을 가지는 Bacillus subtilis MP56 균주의 분리 및 특성분석 (Isolation and Characterization of Bacillus subtilis MP56 with Antimicrobial Activity against MDR (Multi Drug Resistant) Strains)

  • 최근 국내외로 MRSA, VRE 등의 다약제 내성 균주의 분리빈도가 매년 급증하고 있고, 시판하고 있는 항생제 중 가장 효능이 강력한 제품들도 다약제 내성균 출현율이 증가함에 따라 이를 극복할 새로운 항생제의 개발이 요구되고 있다. 다약제 내성균에 유효한 항생제의 예로 vancomycin, teicoplanin, linezolid가 다약제 내성균에 유효한 의약품으로 쓰이고 있으며 방선균이나 Bacillus 등의 배양액에서 다약제 내성균에 유효한 물질의 발굴에 대한 연구가 활발히 진행되고 있다. 특히 넓은 항균범위를 가지고, 체내 축척이 적은 항생물질에 개발에 대한 연구가 활발히 진행되고 있다. 본 연구진은 국내 서해안의 갯벌에서 Bacillus 속 미생물을 수종 분리하였으며, 다약제 내성균에 항균 활성을 지니는 MP56 균주를 선발하였고, 균주 동정 결과 Bacillus subtilis 와 99.93%의 16S rRNA 상동성을 나타냄을 본 연구에서 확인하였다. 국내외 연구진에 의하여 다수의 Bacillus 속 미생물이 분리된 바가 있으며, 항균 물질에 대한 연구도 보고된 바 있다. 그러나 국내 갯벌에서 분리된 Bacillus 속 균주 배양액의 임상균주에 대한 항균 효능 평가에 관한 연구는 보고된 바가 없다. 본 연구에서 Bacillus 속 균주 MP56 배양액이 국내에서 분리한 임상 균주인 MRSA, IMP 균주에 대해 우수한 항균 활성을 나타내어 최적 발효조건과 항균범위를 본 논문을 통해 보고하였으며, 추후 MP56 균주 배양액에서 항균물질의 분리, 정제 및 구조 규명을 위한 후속 연구를 수행 할 예정이다.
