솔잎 (Pinus Densiflora)부탄올 획분이 간장의 활성산소 및 제거효소에 미치는 영향

  • This study was designed to investigate the effects of a butanol (BuOH) fraction from an extract of pine (Pinus densiflora Sieb et Zucc.) needles, on oxygen radicals and their scavenger enzymes in the liver membranes of rats. Twenty-eight male Sprague-Dawley (SD) rats were divided into four groups over a 45 days study period: the control group on a basic diet, and three experimental groups on three different dietary levels of the butanol fraction, specifically 25 mg (BuOH-25), 50 mg (BuOH-50), and 100 mg (BuOH-100) butanol fraction/kg body weight/day, thereby 0.025%, 0.05%, 0.1% of butanol extract of pine needles was added to basil diet respectively. At the end of the experimental period, body weights and food intakes were not different among the four groups. The results showed that cholesterol accumulation in the mitochondria and microsomes of liver cells was significantly inhibited in the BuOH-50 and BuOH-100 groups: by 11.6% and 20.1% in the mitochondria of the BuOH-50 and BuOH-100 groups, respectively; and by 10.5%, and 13.5% in the microsomes of the BuOH-50 and BuOH-100 groups, respectively, compared with the control group. The levels of hydroxyl radicals (.OH) were significantly) lower in the liver mitochondria of the BuOH-50 and BuOH-100 groups (by 13.3% and 18.5%, respectively), while OH radicals were significantly lower in the microsomes or all three experimental groups (by 15.7% in the BuOH-25 group, 20.0% in the BuOH-50 group, and 20.6% in the BuOH-100group), compared with the control group. Superoxide radical (O$_2$) formation was also significantly inhibited in the liver cytosol of both BuOH-50 and BuOH-100 groups; the levels of these radicals were 8.0% lower for the BuOH-50 group and 11.1% lower for the BuOH-100 group, compared to the control group. Copper/Zinc - superoxide dismutase (Cu/Zn-SOD) activities were significantly increased (by 10.3% and 15.9%, respectively) in the liver cytosols of the BuOH-50 and BuOH-100 groups, but Mn-SOD activities were almost identical in the three RuOH groups, compared with the control group. Glutathione peroxidase (GPx) activities were significantly increased in the three experimental groups (by 9.0% in the BuOH-25 group, 19.4% in the BuOH-50 group, and by 25.6% in the BuOH-100 group), compared with the control group. These results suggest that the butanol extract of pine needles may play an effective role in attenuating oxygen radicals and activating scavenger enzymes; consequently, aging may be very effectively modulated and/or inhibited.

Effects of Pine Needle Butanol Fraction on Membrane Fluidity and Oxidative Stress in Liver Membranes of Rats (간장 세포막의 유동성과 산화적 스트레스에 미치는 솔잎(Pine Needle) 부탄올획분의 영향)

  • This study was designed to investigate the effects of butanol (BuOH) fraction of pine (Pinus densiflora Sieb et Zucc) needle extract on membrane fluidity (MF), basal and induced oxygen radicals (BOR and IOR), lipid peroxide (LPO) and oxidized protein (OP) as an oxidative stress, and lipofuscin (LF) in liver membranes of Sprague-Dawley male rats. The rats were fed basic diets (control group) and experimental diets (BuOH-25, BuOH-50 and BuOH-100) prepared with 25, 50 and 100 mg added to basic diet for 45 days. MFs were significantly increased (about 16∼22%) in mitochondria of BuOH-50 and BuOH-100 groups compared with control group (p<0.01∼0.001) BOR and IOR formations in mitochondria were significantly decreased (11∼17% and 11∼28%, respectively) in these three BuOH groups (p<0.05∼0.001), while BOR and IOR formations in microsomes were significantly decreased (11∼24%) in BuOH-50 and BuOH-100 groups, and (15∼24%) in these three BuOH groups compared with control group (p<0.05∼0.001; p<0.01-0.001). LPO levels were significantly decreased (9% and 9∼13%, respectively) in mitochondria of BuOH-100 and microsomes of BuOH-50 and BuOH-100 groups (p<0.05∼0.01), whereas OP levels were significantly decreased (10∼12%) in mitochondria of BuOH-50 and BuOH-100 groups compared with control group (p<0.05). LF formations were significantly decreased (8∼9%) in BuOH-50 and BuOH-100 groups (p<0.05). These results suggest that butanol fraction of pine needle extract may playa effective role in an attenuating an oxidative stress and increasing a membrane fluidity.

Effects of Pine Needle Butanol Fraction on Acetylcholine (ACh) and Its Related Enzymes in Brain of Rats (뇌 조직의 아세틸콜린 및 그 관련효소에 미치는 솔잎(Pine Needle) 부탄올획분의 영향)

  • This study was designed to investigate the effects of buthanol (BuOH) fraction of pine (Pinus densiflora Sieb et Zucc) needle on cholesterol and lipofuscin (LF) accumulations, acetylcholine (ACh) and its related enzyme activities such as choline acetyltransferase (CAhT) and acetylcholinesterase (AChE), and monoamone oxidase-B (MAO-B) activity, which destroyed the catecholamine-related neurotransmitters in brain membranes of Sprague-Dawley (SD) rats. Male SD rats were fed basic diets (control group) or experimental diets (BuOH-25, BuOH-50 and BuOH-100) for 45 days. Cholesterol accumulations in mitochondria and microsomes were significantly inhibited (about 14 - 17% and 23 - 34%, respectvely) in BuOH-50 and BuOH-100 groups, whereas LF levels were significantly inhibited (about 10 - 14%) in BuOH-50 and BuOH-100 groups compared with control group. ACh levels and ChAT activities were significantly increased (about 11 - 17% and 11 - 23%, respectively) in membranes of BuOH-50 and BuOH-100 groups compared with control group. AChE activities were significantly increased (about 14 - 17%) in membranes of BuOH-50 and BuOH-100 groups. There was no significant difference in MAO-B activities between control and experimental diet groups. The results suggest that butanol fraction of pine needle may play an effective role in an antiaging effect and improving a learning and memory impairments.

Effects of Butanol Fraction of Pine Needle (Pinus Densiflora) on Serum Lipid Metabolism and Oxidative Stress in Rats (솔잎의 부탄올획분이 SD계 Rats의 지질대사와 산화적 스트레스에 미치는 영향)

  • This study was designed to investigate the effects of a butanol extract of pine needles (Pinus densiflora Sieb et Zucc) on lipid metabolism and oxidative stress in rats. Twenty-eight male Sprague-Dawley (SD) rats were divided into four groups over a 45 days study period: the control group on a basic diet, and three experimental groups on three different dietary levels of the butanol fraction, specifically 25 mg (BuOH-25), 50 mg (BuOH-50), and 100 mg (BuOH-100) butanol fraction/kg body weight/day, thereby 0.025%, 0.05%, 0.1% of butanol extract of pine needles was added to basic diet respectively. At the end of the experimental period, body weights and food intakes were food intakes were not different among the four groups. Total and LDL-cholesterol levels were markedly decreased in the BuOH-25, BuOH-50, and BuOH-100 groups, respectively, as follows: 12.8%, 19.1% and 21.6% reductions in total cholesterol; and 10.2%, 15.6% and 23.7% reductions in LDL-cholesterol. However, HDL-cholesterol levels were significantly increased (by approximately 20%) in the serum of the BuOH-100 group only, compared with the control and other experimental groups. Atherogenic indices were also markedly decreased in the three experimental groups, by 24.8%, 30.4% and 36.2%, for each of the BuOH-25, BuOH-50, and BuOH-100 groups, respectively, compared with the control group. The levels of the hydroxyl radical (·OH) and of lipid peroxide (LPO) in the serum of the three experimental groups were significantly reduced, by 9.8%, 19.7% and 21.2%; and by 13.3%, 13.3% and 16.7%, for the BuOH-25, BuOH-50, and BuOH-100 groups, respectively Significant increases in serum superoxide dismutase (SOD) were observed in the BuOH-50 and BuOH-100 groups; specifically, 12.1% in the BuOH-50 group and 23.3% in the BuOH-100 group, compared with the control group. Significant increases in catalase (CAT) avtivities, of 24.7% in the BuOH-50 group and 29.2% in the BuOH-100 group, were also observed, compared to the control group. These results suggest that a butanol extract of pine needles could inhibit chronic degenerative disease through improving lipid metabolism, and could also effectively modulate the aging process attenuating oxidative stress.

  • 1. Purpose Through analysing simple herbs of "DongEuiBoGam JangBu", we compare JangBu of traditional oriental medicine with JangBu of Sasang Constitutional Medicine. So we can comprehend the JangBu's difference of traditional-oriental with sasang-constitional medicine. 2. Method We divide herbs into 4 sectors of sasang constitution by "DongMuYooGo", "DongEuiSuSeBoWon SinJunBang" and define constitutional herbs for each type of sasang constitution. On that base, we analyze herbs in "DongEuiBoGam JangBu" into sasang constitution, and compare with JangBu of sasang-constitution 3. Result (1) The simple herb of sasang-constition is over 50% at that of "DongEuiBoGam JangBu" (2) Only small intestine is consist of one-constitional herb, others is 2~4-constitional herb (3) At the simple herbs of "DongEuiBoGam JangBu", Soeumin's herbs many used at Spleen, Stomach, Urinary bladder, SamCho, Taeumin's herbs many used at Lung. At the Soeumin and Taemin, the Sasang JangBu of filling up BoMyungGiJu(保命之主) is connected with traditional JangBu. Soyangin's herbs many used at Heart, Gall bladder, Small intestine and there is no JangBu which Taeyangin's herbs many used. At the Soyangin and Taeyangin, the Sasang JangBu of filling up BoMyungGiJu(保命之主) is not connected with traditional JangBu.

A Study on the Physical Reactions with Administration of BU Mixed Hwangryunhaedoktang(황련해독탕) Herbal Acupucture (황련해독탕 약침액으로 희석한 BU의 신체반응에 관한 연구)

  • Objective : In the pain control, BU herbal acupuncture is highly effective but causes many phys ical reactions. This study was done to compare BU with BU mixed Hwangryunhaedoktang(HHT, 黃連解毒湯) herbal acupucture in decreasing physical reactions of BU. Methods : We injected BU Group(N=12) and BU mixed HHT Group(N=12) at four points of Fenmen(B12), Feishu(B13), Fufen(B41) and Pohu(B42). Then we observe the local thermal changes and the physical reactions at beginning, 1hour and 24 hours after administering herbal acupuncture through the D.I.T.I. and survey. Results : The following results were obtained; 1. The significant dermatothermal difference had been checked 1hour and 24hours in BU group, but 1hour in Bu mixed HHT. 2. In BU group, the difference of temperature had been continued highly until 24hours. But in BU mixed group, the difference of temperature had been continued highly until 1 hour but decreased nearly equal to beginning stage until 24hours. 3. The symptoms which appear following the herbal acupuncture administration were less appeared in BU mixed HHT than BU group.

  • Clinical observation on the diseases of Jang(臟) and Bu(腑) comes to the conclusions as follows: Variation in Jang(臟) and Bu(腑) exists diversely with every man because man is a microcosmos resembling the great universe, 'Mother Nature'. If the patient has a distinctive features of Jang(臟) and Bu(腑) in configuration, the diseas must be caused by the problems of Jang(臟) and Bu(腑). Distinctive features of Jang(臟) and Bu(腑) can be recognized by the shape and color of face, ear, eye, mouth and nose. Distinctive features of Jang(臟) and Bu(腑) should be examined preferentially when it comes to old people, children and persons who have past record of surgical operation on Jang(臟) and Bu(腑) organs.

Effect of Buffalo Follicular Fluid Alone and in Combination with PMSG and M199 on in vitro Buffalo Oocyte Maturation

  • The effect of replacement of in vitro maturation medium completely with the buffalo follicular fluid (buFF) on in vitro oocyte maturation of buffalo oocytes was studied. 5 to 8 buffalo cumulus oocyte complexes were cultured in a single drop with each of the eight media studied i.e., M199+steer serum (10% v/v), M199+steer serum (10% v/v)+PMSG, M199+buFF (10% v/v), M199+buFF (10% v/v)+PMSG, M199+buFF (50% v/v), M199+buFF (50% v/v)+ PMSG, buFF (100%) and buFF+PMSG at $39^{\circ}C$ and 5% $CO_2$ in air for 24 h. Supplementation of M199 with Steer serum alone resulted in IVM rate of 35% only. When the above medium was supplemented with PMSG, the maturation rate rallied to 82%. Significant increase in the maturation rates were observed when M199 was supplemented with increasing levels of buFF. A further increase in the maturation rate was also obtained when PMSG was incorporated into the medium of M199 supplemented with buFF. The rate of maturation was to the tune of 91% when oocytes were matured in buFF alone which was increased non significantly on the addition of PMSG. Highest maturation rate (97%) obtained with M199+buFF (50%v/v)+PMSG did not differ significantly from that obtained by either M199+buFF (10%v/v)+PMSG or buFF+PMSG. It is suggested that buFF alone without any supplementation can form the effective in vitro maturation medium for buffalo oocytes.
